3International audienceAbstract Specific data regarding outcome of cats with high‐grade and large granular lymphocyte alimentary lymphoma (HGAL and LGL, respectively) treated with multi‐agent chemotherapy are scarce. The aims of this multi‐centric, retrospective study were to describe the outcome of cats with HGAL and LGL treated with COP‐ or CHOP‐based chemotherapy and to identify potential prognostic factors. Cats with a cytological or histological diagnosis of HGAL or LGL lymphoma treated with COP‐ or CHOP‐based protocol as first‐line chemotherapy were included. Data regarding diagnosis, staging, treatment and follow‐up were collected. Fifty‐seven cats treated with CHOP ( n = 37) or COP ( n = 20) protocols were included. Complete response (CR), partial response (PR), stable disease (SD) and progressive disease (PD) were observed in 20%, 22%, 36% and 22% of cats, respectively, for an overall response rate of 42%. Median progression‐free interval (PFI) was 148 days and overall median survival time (OST) was 131 days. Cats achieving CR, PR or SD showed significantly longer PFI ( p < .01) and OST ( p < .015) compared with cats with PD. Other positive prognostic factors in multi‐variate analysis were rescue treatment ( p < .001) and absence of lymph node involvement ( p < .03). Negative prognostic factors were diffuse infiltration of the gastrointestinal tract ( p = .035) and infiltration of a non‐haematopoietic organ ( p < .01)

International audienceAwareness of the negative externalities and vulnerability of today's food systems has led to the development of numerous initiatives on food issues by a wide range of local actors. “Territorial food projects” (PAT), conceived as institutional arrangements for strengthening and enhancing local food systems, are now recognized as levers for agricultural and food transition. In particular, they aim to federate players around a common food-related territorial project, strengthen local food chains and act as levers for integrating different issues (social, environmental, economic or health-related) and areas of public action. The aim of this paper is to analyze how PATs meet these objectives by studying the dynamics of coordination between the players involved in these schemes. The innovative analytical framework used articulates two approaches to social network analysis: that of relational chains using the quantified narratives method, and that of complete networks. The results show that the PAT mobilizes a wide range of resources and federates the actors involved. Links between agricultural sector organizations and inter-municipalities are particularly strong, with agricultural and food issues being placed on the political agenda and local authorities becoming involved in structuring localized supply chains. COVID has highlighted new challenges, fostering the emergence of the social sector. The aim of this paper is to contribute to the debate on the evaluation of PATs and their role in the transition of territorial agricultural and food systems.

No full text
International audienceThe definition of reference levels for radiological protection of the environment is dependent on the methodologies and applicability domains considered by the relevant expert groups (ICRP, IAEA, ERICA European Consortium). However, these methodologies share common limitations regarding the derived reference levels, mainly due to the use of available data, coming from laboratory experiments with the observation of biological endpoints that are not necessarily representative of the wildlife chronically exposed to various sources and levels of ionising radiation. Furthermore, the potential presence of other pollutants and stressors (abiotic parameters, etc) that interact with radionuclides makes it challenging to assess the risk to wildlife in realistic exposure scenarios. The Adverse Outcome Pathways (AOP) framework appears to be a valuable tool that helps to structure a sequence of causally linked events at different biological levels (Molecular Initiating Event, Key Events (KE), and Adverse Outcome (AO)). In the context of ecological risk assessment, AOP provide a way of linking molecular data, usually obtained in laboratory conditions, to ecological levels, i.e., community and ecosystem, for which the definition of relevant key events is still challenging. In addition, AOP networks can be proposed to help to compare effects between different types of stressors; to deconvolute biological effects in the context of exposures to multiple stressors. AOP provides an efficient summary of existing data through a clear and standardized framework accessible via the open access database AOP-KB, shared by research and regulators communities. This work proposes a review of the challenges of developing AOP for radioecology. A few guidelines are proposed to overcome some of the identified challenges. These include the assessment of possible relevant ecological endpoints regarding ionising radiation for AOP development, proposal of integrated and multidisciplinary approaches to fuel AOP development, and discussions of AOP applications to inform ecological risk assessment

Lineage-dependent partitioning of activities in chemoclines defines Woesearchaeota ecotypes in an extreme aquatic ecosystem

2/1International audienceBackground DPANN archaea, including Woesearchaeota, encompass a large fraction of the archaeal diversity, yet their genomic diversity, lifestyle, and role in natural microbiomes remain elusive. With an archaeal assemblage naturally enriched in Woesearchaeota and steep vertical geochemical gradients, Lake Dziani Dzaha (Mayotte) provides an ideal model to decipher their in-situ activity and ecology. ResultsUsing genome-resolved metagenomics and phylogenomics, we identified highly diversified Woesearchaeota populations and defined novel halophilic clades. Depth distribution of these populations in the water column showed an unusual double peak of abundance, located at two distinct chemoclines that are hotspots of microbial diversity in the water column. Genome-centric metatranscriptomics confirmed this vertical distribution and revealed a fermentative activity, with acetate and lactate as end products, and active cell-to-cell processes, supporting strong interactions with other community members at chemoclines. Our results also revealed distinct Woesearchaeota ecotypes, with different transcriptional patterns, contrasted lifestyles, and ecological strategies, depending on environmental/host conditions. ConclusionsThis work provides novel insights into Woesearchaeota in situ activity and metabolism, revealing invariant, bimodal, and adaptative lifestyles among halophilic Woesearchaeota. This challenges our precepts of an invariable host-dependent metabolism for all the members of this taxa and revises our understanding of their contributions to ecosystem functioning and microbiome assemblage.</div

Clinical practice and effect of carbon dioxide on outcomes in mechanically ventilated acute brain-injured patients: a secondary analysis of the ENIO study

No full text
International audiencePurpose: The use of arterial partial pressure of carbon dioxide (PaCO 2 ) as a target intervention to manage elevated intracranial pressure (ICP) and its effect on clinical outcomes remain unclear. We aimed to describe targets for PaCO 2 in acute brain injured (ABI) patients and assess the occurrence of abnormal PaCO 2 values during the first week in the intensive care unit (ICU). The secondary aim was to assess the association of PaCO 2 with in-hospital mortality. Methods:We carried out a secondary analysis of a multicenter prospective observational study involving adult invasively ventilated patients with traumatic brain injury (TBI), subarachnoid hemorrhage (SAH), intracranial hemorrhage (ICH), or ischemic stroke (IS). PaCO 2 was collected on day 1, 3, and 7 from ICU admission. Normocapnia was defined as PaCO 2 > 35 and to 45 mmHg; mild hypocapnia as 32-35 mmHg; severe hypocapnia as 26-31 mmHg, forced hypocapnia as < 26 mmHg, and hypercapnia as > 45 mmHg.Results: 1476 patients (65.9% male, mean age 52 ± 18 years) were included. On ICU admission, 804 (54.5%) patients were normocapnic (incidence 1.37 episodes per person/day during ICU stay), and 125 (8.5%) and 334 (22.6%) were mild or severe hypocapnic (0.52 and 0.25 episodes/day). Forced hypocapnia and hypercapnia were used in 40 (2.7%) and 173 (11.7%) patients. PaCO 2 had a U-shape relationship with in-hospital mortality with only severe hypocapnia and hypercapnia being associated with increased probability of in-hospital mortality (omnibus p value = 0.0009). Important differences were observed across different subgroups of ABI patients. Conclusions: Normocapnia and mild hypocapnia are common in ABI patients and do not affect patients' outcome. Extreme derangements of PaCO 2 values were significantly associated with increased in-hospital mortality.</div
