    Impact of N-terminal domain on the sHSP Lo18 function

    International audienceExternal stress conditions can induce disturbances in the internal environment of cells, sufficient for protein misfolding or membrane disorganization. To cope with this phenomenon, cells have developed several strategies, including the production of heat stress proteins (HSPs). These include small heat shock proteins (sHSPs). sHSPs share a characteristic three-dimensional architecture, with a central α-crystalline domain (ACD) flanked by two terminal domains. This structural organization enables them to form a variety of oligomeric complexes, giving these proteins their function. As large oligomers, all sHSPs act as chaperones. Conversely, some of them adopt a lipochaperone role when present in dimeric form. While the role of the ACD domain in oligomerization and the mechanism of action of sHSPs has been studied extensively, the functions of the terminal domains, in particular the N-terminal domain, remain poorly explored. In this context, the role of the N-terminal domain of sHSP Lo18, produced by the bacteria Oenococcus oeni, was investigated. To assess its importance, a truncated protein, depleted of the first 27 amino acids, was compared with the wild-type protein. Analyses focused on the oligomeric structure, the ability to prevent protein aggregation, and the ability to maintain membrane fluidity. In parallel, the involvement of the N-terminal domain in the interaction between Lo18 and membranes was examined by RP-HPLC. The results reveal that Lo18’s N-terminal domain plays a decisive role, both in the formation and stabilization of oligomeric structures, and in interaction with protein and lipid substrates, essential parameters in chaperone and lipochaperone activity

    Wine in a can: A consumer study in a traditional wine-producing region. Effect of the level of consumer involvement

    International audienceIt is imperative that the wine industry embraces innovation, given the sector’s traditionally conservative nature. To succeed, innovators must address consumer expectations, which are pivotal in shaping preferences and perceptions of quality. The present study investigated the effect of consumer involvement on wine perception, using packaging innovation as a starting point. One hundred and twenty-five non-professional wine consumers from the traditional Rioja wine-producing region in Spain took part in a tasting of two wines (one red and one white) under two different conditions. Participants were informed that under one experimental condition, the wine was bottled, whereas in the other the wine was canned. The samples were drawn from the same bottle to ensure only the provided information was modified, without altering the intrinsic properties, thus allowing a focus on consumers’ cognitive responses. The two containers, a 200 mL can and a 125 mL bottle, were designed with the same label and packaging colour. The participants were first asked to evaluate each sample’s quality and liking on a 10-point scale. They described each sample using the Rate-All-That-Apply (RATA) methodology. The participants then completed a validated questionnaire designed to measure their level of involvement with wine, which was used to segment consumers into four groups. Finally, the participants were asked to provide demographic information. The results of the study confirm that in the case of red wine, the more innovative packaging, the aluminium can, has a negative impact on the perceived quality of the product. This remains consistent regardless of the level of involvement, wine colour, or the preference for red wine. Conversely, in the case of white wine, no effect of the packaging was observed on consumer’s liking except for highly involved consumers. This suggests that canned young white wine could be a suitable alternative for traditional consumers. Furthermore, the “canned” wines, though they were never in actual contact with the aluminium packaging, were perceived as more metallic and less fruity, regardless of the wine. The results are discussed in terms of cognitive mechanisms influencing wine perception, including the desire for unique products (DFUP), the concept of “image mold” and consumer expectations

    International audienceThe nitrogen nutrition index (NNI) is central to monitoring and managing the nitrogen statusof crops. It can be estimated using a sampling of the aboveground biomass or optical devices likeSPAD chlorophyll meter, though these are impractical for large areas. A dark green colour index(DGCI), derived from visible images was calculated and refined into a normalized nDGCI, therebyremoving the need for light calibration. Trials conducted in 2023 and 2024 on rapeseed crops revealeda linear relationship (R²=0.64) between NNI and nDGCI at the E–F1 growth stage (BBCH scale57–60).     International audienceEn réponse aux préoccupations environnementales croissantes et à la recherche de ressources locales durables, cette étude examine l’émergence de micro-secteurs biosourcés à travers l’exemple de la valorisation de la laine de mouton en France. Traditionnellement considérée comme un déchet, en particulier lorsqu’elle provient de races à viande, la laine brute reste largement sous-exploitée en dehors de l’industrie textile. Pourtant, ses propriétés physico-chimiques offrent un potentiel prometteur pour des utilisations alternatives telles que l’isolation thermique et acoustique, le paillage agricole et les composites écologiques. S’appuyant sur une revue de la littérature et une étude de cas de Vert Laine, une start-up basée en Bourgogne, cette recherche explore la structuration de ces nouvelles filières, les acteurs impliqués et les obstacles auxquels ils sont confrontés. À l’aide du cadre de modèle d’affaire circulaire BM3C2, l’étude analyse les modèles économiques alignés sur les principes de l’économie circulaire. Elle vise à identifier les dynamiques d’innovation et les partenariats territoriaux nécessaires pour développer de telles initiatives. Cette recherche contribue au débat plus large sur les modèles de production durable, la valorisation des ressources locales et la transition vers une bioéconomie circulaire
