    Fast Discrimination of Chocolate Quality Based on Average-Mass-Spectra Fingerprints of Cocoa Polyphenols

    This work aims to sort cocoa beans according to chocolate sensory quality and phenolic composition. Prior tothe study, cocoa samples were processed into chocolate in a standard manner, and then the chocolate was characterized bysensory analysis, allowing sorting of the samples into four sensory groups. Two objectives were set:first to use average massspectra as quick cocoa-polyphenol-extractfingerprints and second to use thosefingerprints and chemometrics to select themolecules that discriminate chocolate sensory groups. Sixteen cocoa polyphenol extracts were analyzed by liquidchromatography−low-resolution mass spectrometry. Averaging each mass spectrum provided polyphenolicfingerprints,which were combined into a matrix and processed with chemometrics to select the most meaningful molecules fordiscrimination of the chocolate sensory groups. Forty-four additional cocoa samples were used to validate the previous results.Thefingerprinting method proved to be quick and efficient, and the chemometrics highlighted 29m/zsignals of known andunknown molecules, mainlyflavan-3-ols, enabling sensory-group discrimination

    Introduction : The induced abortion rate is high in France, Reunion island in particular (20,7 for every 1000 in 2017). The problem isn’t accessibility to contraception, rather the daily management of it. The number one contraception method continues to be the pill which observance may be irregular. The IUD is an efficient contraceptive method, with excellent longevity. Generalist doctors are on the frontline for the contraception counseling. Our objective was to identifie the reluctances of generalist to using IUD in Reunion Island and the possibilities of dismantling methods.Method : Qualitative descriptive study, by semi-directive interviews with generalist doctors in the Reunion Island who didn’t insert IUDs.Results and discussion : This study is carried out from August to November 2018 with 13 generalist doctors. The overall opinion of them was positive. The observance was the primary benefit of this. Many stereotypes persist and were similar with others french and international studies, result from doctors, the IUD insertion, patients, and environnemental concerns.Conclusion : There is a probable link between practical training of generalist doctors to the IUD insertion, and a context low demand and good health care facilities.There are some perspectives that exist to overcome those concerns in order to expand IUD access : information and counseling for patients, an easier and quicker access to echography, trainings on other IUD insertion method, screening for sexually transmitted diseases before IUD insertion to prevent pelvic inflammatory diseases.Contexte : l’incidence des IVG reste importante en France métropolitaine, notamment à La Réunion (20,7 pour 1 000 en 2017). Le souci n’est pas l’accessibilité du contraceptif mais sa gestion. La première contraception prescrite reste la pilule dont l’observance est variable. Le dispositif intra-utérin (DIU) est un moyen de contraception efficace et de longue durée d’action. Les médecins généralistes sont en première ligne dans le discours contraceptif mais pratiquent peu la pose de DIU. Notre objectif était d'identifier les freins des médecins réunionnais à la pose de DIU ainsi que les possibilités de « déconstruction » de ces freins.Méthode : étude descriptive qualitative, par entretiens semi-dirigés auprès de médecins généralistes installés à La Réunion ne posant pas de DIU. Résultats et discussion : l’étude a été réalisée entre août et novembre 2018 auprès de 13 généralistes. L’opinion générale sur le DIU était positive. L’observance était le principal avantage. De nombreux a priori persistaient relativement similaires à ceux de métropole et dans la littérature internationale, liés aux médecins, patientes, geste de la pose et contexte environnemental. Conclusion : il existe probablement un lien entre la formation pratique à la pose de DIU, jugée insuffisante, et le contexte actuel de faible sollicitation/offre de soins suffisante. Des perspectives d’avenir se dessinent, afin d’élargir l’accès à ce contraceptif, comme augmenter l'information et counseling des patientes, un accès facilité à l’échographie, un apprentissage de techniques de pose moins douloureuses et un dépistage des IST en pré-pose pour baisser le risque infectieux

    Introduction: Chikungunya is an arboviral infection transmitted by Aedes mosquitoes (commonly called Asian tiger mosquitoes) which has been expanding largely since its re-emergence in 2004. During an unprecedented outbreak on the Reunion Island between 2005 and 2006, paediatricians described for the first time a mother-to-child vertical transmission of the virus during childbirth. An assessment of the toddlers at the age of two years revealed that consequences were mainly neurological, suggesting a global retardation in psychomotor acquisitions subsequent to the neonatal infection. Objectives: The main objective of our study was to evaluate the neurocognitive performances of children infected by the chikungunya virus during delivery, by the age of 10 years. The secondary objectives were to identify associated neuro-sensory disorders, and to assess the children’s schooling and the use of specialized care such as speech therapy, orthoptics or psychomotor care. Methods: In 2015, we examined 21 children from the CHIMERE cohort, who got infected by chikungunya in the neonatal period, at the approximate age of 10 years. Based on EDA evaluation scale (assessing children’s cognitive and learning outcomes), children performed a variety of neurocognitive tests in comparison with a reference group of 626 children in order to track neurocognitive impairments.Results: The results showed lower performances in the infected group, inferior to -1DS on both verbal and non-verbal abilities and slightly superior to -1DS on learning skills on the EDA scale (p < 0.001). 4 children out of 21 (19%) were affected by severe encephalopathy with psychomotor retardation, and educate at special school. In addition, MR imaging performed on most affected children, allowed to localise cerebral hypo-metabolism in the frontal white matter where the coordination and language functions are regulated.Conclusion: Mother-to-child infection to the chikungunya virus has a major impact on children’s neurocognitive functions despite a great brain plasticity. Neonatologists, obstetricians and paediatricians should be on the frontline for prevention and surveillance of the arbovirus epidemics.Introduction : Le chikungunya est une arbovirose transmise par les moustiques tigres qui connait actuellement une expansion pandémique depuis sa réémergence en 2004. Lors d’une épidémie sans précédent sur l’île de la Réunion entre 2005 et 2006, les pédiatres ont décrit pour la première fois une transmission verticale materno-fœtale de ce virus au moment de l’accouchement. Les principales conséquences pour le nouveau-né étaient d’ordre neurologique, avec une évaluation des enfants à l’âge de deux ans qui suggérait un retard global des acquisitions psychomotrices secondaire à cette infection néonatale. Objectifs : L’objectif principal de notre étude était d’évaluer vers l’âge de 10 ans les performances neurocognitives des enfants infectés au virus du chikungunya par transmission materno-fœtale. Les objectifs secondaires étaient d’évaluer la présence de séquelles neurosensorielles associées, d’évaluer le parcours scolaire de l’enfant et le recours ou non à des soins spécialisés tels que l’orthophonie, l’orthoptie ou la psychomotricité. Méthodes : Nous avons revu en 2015, 21 enfants de la cohorte CHIMERE alors âgés de 10 ans, infectés par le virus du chikungunya en période néonatale. À partir de l’échelle d’évaluation EDA (Évaluation des fonctions cognitives et Apprentissages de l’enfant), nous leurs avons fait passer une batterie de tests neurocognitifs en comparaison d’une population de référence de 626 enfants, afin de dépister une déficience neurocognitive.Résultats : Les résultats montraient une performance moindre des enfants infectés, inférieure à -1DS sur les fonctions verbales et non verbales et une performance légèrement supérieure à -1DS sur les fonctions d’apprentissage de l’EDA (p < 0,001). 4 enfants sur 21 (soit 19 %) avaient une encéphalopathie sévère avec un retard psychomoteur et une scolarisation en centre adapté au handicap. Les IRM réalisées chez les enfants les plus atteints, montraient un hypo-métabolisme de la substance blanche frontale où se situent les aires cérébrales qui contrôlent la coordination et le langage. Conclusion : L’infection materno-foetale au virus du chikungunya a des conséquences majeures sur les fonctions neurocognitives de l’enfant malgré une grande plasticité cérébrale. Les obstétriciens, les néonatologues et les neuro-pédiatres doivent être en première ligne pour la prévention et la surveillance de ces épidémies d’arboviroses

    OntoSIDES: Ontology-based student progress monitoring on the national evaluation system of French Medical Schools

    International audienceWe introduce OntoSIDES, the core of an ontology-based learning management system in Medicine, in which theeducational content, the traces of students’ activities and the correction of exams are linked and related to itemsof an official reference program in a unified RDF data model. OntoSIDES is an RDF knowledge base comprised ofa lightweight domain ontology that serves as a pivot high-level vocabulary of the query interface with users, andof a dataset made of factual statements relating individual entities to classes and properties of the ontology.Thanks to an automatic mapping-based data materialization and rule-based data saturation, OntoSIDES containsaround 8 millions triples to date, and provides an integrated access to useful information for student progressmonitoring, using a powerful query language (namely SPARQL) allowing users to express their specific needs ofdata exploration and analysis. Since we do not expect end-users to master the raw syntax of SPARQL and toexpress directly complex queries in SPARQL, we have designed a set of parametrized queries that users caninstantiate through a user-friendly interface

    A comparative study of some kaolinites surface properties

    International audienceThe surface properties of five kaolinites of various origins were analyzed using potentiometric titration, electrokinetic measurements and low-pressure gas adsorption associated to derivative isotherm summation (DIS) modeling. The data show that sample structure is important in determining the surface properties. The combined analytical results clearly shows the existence of a permanent negative layer charge for all the samples. The general features that could be observed for the five kaolinites are: the titration curves are shifted to lower pH with increasing ionic strength due to the layer charge; the amount of consumed proton is influenced by the permanent layer charge; although low, the permanent charge determines the electrokinetic behavior. The shape anisotropy and the charge distribution on the basal and edge surfaces are crucial parameters to understand the behavior of kaolinite particles in aqueous media. The Cameroonian sample of the set exhibits a significant permanent charge together with large specific surface area associated to fine particle size and considerable shape anisotropy

    Several formal methods of supervisory control of discrete events systems (DES), based on Petri nets (PN) have been developed. However, they are partially structural because of the need to construct the marking graph, which is an automaton, to determine the states forbidden by the specification. Some of those methods are limited to a particular class of Petri nets. Our work focused on the labeled Petri nets (LPN), which offers a better structure for defining the languages and automata. Among the based methods based on Petri nets, the invariant place method (simple and efficient) is the most used, although it does not guarantee in the general an optimal solution. In order to overcome this problem, we have developed a completely structural method, without construction of the marking graph for ordinary labeled Petri nets. It consists in determining in a way structural, admissible constraints for the invariant method place. It is based on the structural condition of controllability of closed loop SED LPN states, got by synchronous product of the process and specification LPN. In addition, we have proved that the structural condition of controllability, defined by the marking condition of entrance places of an uncontrollable synchronous transition, is equivalent to the condition of controllability defined by the languages of the LPN. This condition defines the equation of a separator hyperplan which ensures separation of the admissible states from the forbidden states in the SED. Constraints derived from hyperplane separator are admissible and can be simplified systematically. They provide a one-to-one link between the theory of supervisory control of Ramadge and Wonham and the calculation of maximum permissive controller by the invariant place method. Although for some complex structures (great scale), the maximum permissive solution cannot be guarantee directly; our approach gave the optimal controller in the real case studies considered.Plusieurs méthodes formelles de commande par supervision des systèmes à événements discrets (SED) basées sur les réseaux de Petri (RdP) ont été développées. Cependant, elles sont partiellement structurelles à cause de la nécessité de construire le graphe de marquage, qui est un automate, pour déterminer les états interdits par la spécification. Certaines de ces méthodes sont limitées à une classe particulière de RdP. Notre travail s'est focalisé en particulier sur le RdP synchronisé (RdPS), qui offre une meilleure structure pour définir les langages et les automates. Parmi les méthodes basées sur les RdP, la méthode des invariants (simple et efficace) est la plus utilisée bien qu’elle ne garantisse pas en général une solution de contrôle optimale. Pour pallier ce problème, nous avons développé une méthode complètement structurelle, sans construction du graphe de marquage, pour les RdPS ordinaires ou généralisés. Elle consiste à déterminer de manière structurelle les contraintes admissibles pour la méthode des invariants. Elle est basée sur la condition de contrôlabilité structurelle des états du RdPS du SED en boucle fermée, obtenu par produit synchrone des RdPS du procédé et de la spécification. En outre, nous avons prouvé que la condition de contrôlabilité structurelle, définie par la condition de marquage des places d’entrée d’une transition synchrone incontrôlable, est équivalente à la condition de contrôlabilité définie par les langages du RdPS. Cette condition définit l’équation d’un hyperplan séparateur qui assure la séparation des états admissibles des états interdits dans le SED. Les contraintes dérivées de d’hyperplan séparateur sont admissibles et peuvent être simplifiées systématiquement. Elles permettent de faire un lien biunivoque entre la théorie de commande par supervision de Ramadge et Wonham et la méthode des invariants pour le calcul d’un contrôleur optimal au sent maximal permissif. Même si pour certaines structures très complexes (à grande échelle), la solution maximale permissive ne peut être garantie directement, notre approche a donné le contrôleur optimal dans les études de cas réels considérés

    A Data Integration Multi-Omics Approach to Study Calorie Restriction-Induced Changes in Insulin Sensitivity

    International audienceBackground: The mechanisms responsible for calorie restriction (CR)-induced improvement in insulin sensitivity (IS) have not been fully elucidated. Greater insight can be achieved through deep biological phenotyping of subjects undergoing CR, and integration of big data.Materials and Methods: An integrative approach was applied to investigate associations between change in IS and factors from host, microbiota, and lifestyle after a 6-week CR period in 27 overweight or obese adults (ClinicalTrials.gov: NCT01314690). Partial least squares regression was used to determine associations of change (week 6 – baseline) between IS markers and lifestyle factors (diet and physical activity), subcutaneous adipose tissue (sAT) gene expression, metabolomics of serum, urine and feces, and gut microbiota composition. ScaleNet, a network learning approach based on spectral consensus strategy (SCS, developed by us) was used for reconstruction of biological networks.Results: A spectrum of variables from lifestyle factors (10 nutrients), gut microbiota (10 metagenomics species), and host multi-omics (metabolic features: 84 from serum, 73 from urine, and 131 from feces; and 257 sAT gene probes) most associated with IS were identified. Biological network reconstruction using SCS, highlighted links between changes in IS, serum branched chain amino acids, sAT genes involved in endoplasmic reticulum stress and ubiquitination, and gut metagenomic species (MGS). Linear regression analysis to model how changes of select variables over the CR period contribute to changes in IS, showed greatest contributions from gut MGS and fiber intake.Conclusion: This work has enhanced previous knowledge on links between host glucose homeostasis, lifestyle factors and the gut microbiota, and has identified potential biomarkers that may be used in future studies to predict and improve individual response to weight-loss interventions. Furthermore, this is the first study showing integration of the wide range of data presented herein, identifying 115 variables of interest with respect to IS from the initial input, consisting of 9,986 variables
