    Get PDF
京都工芸繊維大学博士(工学)To reveal the sulfur cross-linking reaction of isoprene rubber, modern approaches such as synchrotron X-ray analysis and density functional theoretical (DFT) calculation were subjected. As a result, it was revealed that the dinuclear bridging bidentate zinc/stearate complexes play an important role in the sulfur cross-linking reaction of isoprene rubber

Studies on Effects of Coating Process Conditions on Structures and Properties of Pressure-Sensitive Adhesive Layers Composed of Di- and Tri-Block Copolymers

    Get PDF
京都工芸繊維大学博士(工学)I have revealed the effects of coating process conditions on structures and properties of pressure-sensitive adhesive layers composed of di- and tri-block copolymers forming spherical microdomains. Two different types of the coating process have been examined; the solution coating (by solvent evaporation from solutions) and the hot-melt coating (by solidification from melt). Although the latter solvent-free process is much preferred, the coated layer by this method has poorer mechanical properties. Since it is important to understand the structure-property relationship to improve their mechanical properties, I have conducted precise structural analyses by two-dimensional small-angle X-ray scattering measurements and by atomic force microscopy observations

Isolation and evaluation of anti-inflammatory constituents from lichen Dirinaria applanata and Lasia spinosa leaves

京都工芸繊維大学博士(学術)Natural products or traditional medicines play essential roles in therapeutic agents as well as a resource for the discovery of new drugs. Lichen Dirinaria applanata and Lasia spinosa (L.) Thwaites were used as traditional medicines to treat many diseases for centuries. However, chemical components and their effects on inflammatory response are not yet characterized. Thus, the goal of this dissertation is to explore chemical positions and investigate the anti-inflammatory activities of these extracts in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages. Chemical investigation of the lichen Dirinaria applanata methanol extract led to isolating two hopane derivatives including 1β-acetoxy-21α,3β-hydroxyhopane-29-oic acid (1), hopane-3β,6β,21α-triol (2) and a depside namely 2-O′-methylnordivaricatic acid (3). Their structures were elucidated by spectroscopic data in combination with published literature. Notably, compound 2 was isolated from this lichen species for the first time. Unfortunately, only compound 1 shows the anti-inflammatory effect by reducing nitric oxide (NO) production in LPS-induced inflammation response. Besides, ethanol extracts of L. spinosa leaves showed antioxidant activity due to the presence of high levels of polyphenolic compounds. Therefore, L. spinosa leaves may exhibit anti-inflammatory properties. In particular, treatment with the leaf extract significantly repressed the production of inflammatory mediators such as nitric oxide and reactive oxygen species and the expression of pro-inflammatory cytokines in the LPS-stimulated RAW264.7 cells. Moreover, pretreatment of L. spinosa leaf extract prevented activation of the nuclear factor-kappa B (NF-κB) pathway by inhibiting nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor, alpha (IκBα) degradation. Furthermore, the mitogen-activated kinase (MAPK) and phosphoinositide-3-kinase/protein kinase B (PI3K/Akt) pathways were suppressed upon treatment with the leaf extract. In addition to suppressing inflammatory factors, the extract also promoted the activated nuclear factor erythroid 2-related factor 2/heme-oxygenase-1 (Nrf2/HO-1) pathway. Taken together, the significant findings of these herbs will be highlighted and discussed throughout this dissertation. Result obtained highlights critical findings in assessing the potential of anti-inflammatory constituents. Thereby, we propose to further develop them as novel pharmacological strategies in order to defeat inflammation diseases

Rheology and Colloidal Gelation of Dense Suspensions of Thermo-Responsive Soft Microgels

    Get PDF
京都工芸繊維大学博士(工学)In the present thesis, the rheological properties and colloidal gelation behavior of temperature (T)-responsive soft microgel suspensions are investigated. In Chapter I, the basic introduction of the characteristics of T-responsive microgels, the viscoelasticity of their microgel suspensions, and the colloidal gelation phenomena is provided. In Chapter II, sample synthesize, preparation and experimental method used in this study are illustrated. The poly(N-isopropyl acrylamide) (denote as N) and poly(N-isopropyl methacrylamide) (denote as NM) microgels with various values of particle size and crosslink densities are employed in the experiments. In Chapter III, the elastic and flow properties of binary N/N microgel pastes with size and stiffness disparities are investigated. The effects of the size and stiffness disparities on elasticity, yield point, and steady-state flow properties are elucidated. The local fast dynamics of microgels is correlated to the characteristic time in the steady-state flow of the pastes. In Chapter IV, the yielding and flow properties of binary N/NM microgel pastes with chemically dissimilar surfaces are studied. We reveal that the contribution of the N-NM contacts to yield strain (γc) of the mixtures. We also show that large size disparity results in anomalies not only in the composition dependence of γc and modulus, but also in steady state flow behavior, due to the characteristic packing efficiency where small particles enter in the gap between large particles. In Chapter V, the formation criteria and viscoelasticity of colloidal gels are revealed for the N microgel suspensions. We examine how each of surface charge density, T and particle concentration affects the colloidal gelation, and elucidate the correlation between them. The rheological properties of the colloidal gels are revealed by large amplitude oscillatory shear, and long-term linear creep and recovery measurements. In Chapter VI, viscoelasticity and colloidal gelation of binary N/NM microgel suspensions with different transition T are investigated. We analyze the measured T-dependence of linear viscoelasticity by converting into the dependence on each particle volume fraction or the total particle volume fraction. This analysis elucidates the contribution of the packing effect to the viscoelasticity, and it also gives a clue to clarify the criterion for the onset gelation temperature. In Chapter VII, the important results of present thesis are concisely summarized
