    Novel Function of Lipin and Serotonin Transporter in Development of Drosophila melanogaster

    京都工芸繊維大学博士(学術)Lipin and Serotonin transporter (SerT) are two highly conservative proteins in both human and Drosophila, which are well-known for their regular function. Lipin takes part in the mobilization of stored lipids to generate energy when the body requires. SerT, on the other hand, plays an important role in regulating Serotonin levels ? a vital neurotransmitter, responsible for multiple psychological activities in the central nervous system (CNS). Since lipin and SerT can be found in various kind of tissue in a living organism, not only in fat body or CNS for their respective roles, I suspect there are several functions of lipin and SerT remain unknown. In this thesis, I discuss novel roles of lipin and SerT in development using Drosophila model. First, I can visualize the expression of lipin in the wing disc and knockdown of this gene disrupt normal wing pattern formation. Knockdown of lipin in the 3rd instar larvae wing disc induces DNA damage. DNA damage, in turn, initializes the G2/M DNA damage checkpoint by down-regulating cyclin B expression, which then prevents the cell cycle transition from S-phase to M-phase. Additionally, the dysfunction of lipin induces cells to undergo apoptosis, leading to the formation of wing notching with a significantly smaller size. Next, SerT highly expresses in Drosophila eye discs and knockdown of SerT gene leads to a combination of rough and bald eye phenotype. SerT knockdown down-regulate the activation of the PI3K/Akt pathway, which induces excessive cell death in this tissue and in turn activates the compensatory proliferation mechanism to accumulating massive cluster of proliferating cells in the posterior region of the eye disc. This chain process induces the elevated recruitment of DNA in cone cells nuclei, impairs cone cells normal formation, and leads to rough phenotype. Taken together, these results suggest novel links of an energy-related protein (lipin) and psychological-related protein (SerT) in development

    A Study on Emotion Recognition System through Physiological Signals and Development of Smart Wearable Textile Devices for Elderly People

    京都工芸繊維大学博士(学術)The global population is ageing: virtually every country in the world is experiencing growth in the number and proportion of elderly people in their population. More and more elderly people not only pay attention to material requirements, but also to physical and mental health. While emotion plays a very important role in human’s daily life, which can be used to assess the wellbeing of people and their quality of life. With the rapid development of affective computing, and the update of computer technology and artificial intelligence, emotion recognition using physiological signals attracts a variety of attention, which reflects objective and actual internal emotional state of people. Therefore, having a solution that continuously monitors the physiological signals of the person and assesses his or her emotional well-being could be a very valuable tool to support the daily life and healthcare of the elderly people. The approaches for emotion recognition using physiological signals require the acquisition of physiological data accurately and comfortably under the state that a specific emotion is expressed. To collect physiological signals, some researchers have been doing their best to develop wearable devices, however, these systems still have some drawbacks, which are expensive, bulky, complicated and uncomfortable, with less acceptance to the elderly users. We should develop smart wearable devices for physiological signals measurement, which are comfortable, natural appearance, simple operation. Combing the deep learning methods allow the evaluation of emotion recognition algorithms on signals that can be conveniently captured in everyday scenarios, providing the means to integrate affect computing methods into a wide variety of tasks for eldercare. In this thesis, the optimal signal channels fusion: EEG and ECG signals are selected based on the related works and the design of wearable devices based on fashion and ergonomics aspects are studied. The emotion recognition model is constructed by selecting the optimal fusion of signals and optimal data collection of signals, then verify the models. The application experiment was conducted on the elderly people to verify the real-time and interactivity of the emotion recognition system using the signals collected from our designed wearable textile devices. The main contents of the thesis are as follows. Part 1, we searched and got a deep understanding of the previous related research background and works, and research development progress of emotion recognition using physiological signals; have confirmed our research objectives and significances. The existing studies, including the ageing population, the effects of the emotion recognition, emotion models, emotion eliciting methods, commonly used emotion recognition methods, as well as the processes of emotion recognition, including the acquisition and filtering of physiological signal data, feature extraction and selection, are sorted out and summarized, laying a solid theoretical foundation for the follow-up research. Part 2, we strove to build upon prior research in the field of emotion recognition for elderly people and explored new techniques for increasing the effectiveness of deep learning models for EEG and ECG signals. An experiment was performed on the young participants and collected their EEG and ECG signals with simulating video clips. It was found that with the time-frequency domain features: the spectrograms of EEG and RRI, binary emotional changes classification could be performed well by CNN, with an accuracy of 91.7% with EEG spectrograms, 93.1% with RRI spectrograms, and 94.2% with EEG and RRI spectrograms. This allows for the convenience of automatic feature optimization and feature fusion. Part 3, it’s to develop the smart wearable textile devices for EEG and ECG signals capture comfortably during long-term periods. Firstly, we developed a comfortable and user-friendly 4-channel EEG cap integrated with textiles for elderly people. The signals from the cap were strongly paralleled with the commercial wet electrodes. The flexible design improved the usability of the device, enhanced operational conveniences, reduced pressure, and made the device lighter, softer, more comfortable, and more natural in appearance. According to the factor analysis on two wearable EEG devices; the two factors: the materials properties factor and the design pattern factor were extracted when design wearable devices. This highlights the need to focus on the two factors when designing new wearable headsets in the future. According to the results of the development of the textile EEG cap, an ECG chest band combing smart textiles was developed using the same ideas and methods. The ECG signals collected from ECG chest band was highly correlated with the wet electrodes system, which is reliable and stable. Besides, the ECG chest band gained positive feedback on the assessment of the user experience. Part 4, we conducted the application experiment on elderly people to verify the emotion recognition with our designed wearable textile devices. The EEG and ECG signals of the elderly subjects were collected through our designed textile devices under varying emotional states. We applied raw signals (EEG; ECG; EEG+ECG) to end-to-end deep learning neural networks - adopting LSTM to learn features, to classify positive/negative emotions. The results show that combining EEG and ECG data to LSTM greatly improved the classification result, which is a very promising for emotion detection in future. The achievement of highly accurate on arousal and valence classification in this research suggests that emotion recognition in dimensional emotion models, even discrete emotions, will yield good results. The research of emotion recognition based on physiological signals and the application of wearable textile devices has great development potential, and even can change the pattern healthcare for the elderly in future, so it has great research significance

    Studies on screening for microbes producing vinyl compounds and its application to development of biobased polymers

    京都工芸繊維大学博士(工学)The aim of this study is development of screening method for microbes producing vinyl compounds from environmental samples and synthesis of new biobased polymers using the vinyl compounds as monomers. Thus, the following studies were carried out.In Chapter 1, structure, function, and biosynthesis of itaconic acid (IA) derivatives with the vinylidene group were summarized and their perspectives for industrial use were discussed. In Chapter 2, a selective labeling method based on Mizoroki-Heck reaction for vinyl compounds and a simple TLC?densitometric method for the quantification of the vinyl compounds were developed.In Chapter 3, the selective labeling reaction developed in Chapter 2 was confirmed by an iodine test and the screening of IA-producing fungi from soils was performed using the developed method. Using this method, 37 strains were selected as IA-producing fungi from 240 strains isolated from soils. In Chapter 4, a direct screening method based on Thiol-ene reaction for microbes producing vinyl compounds from soil samples was developed. Soil samples were spread on an agar medium supplemented with TG and VA-044. Screening of soil samples using the screening method led to isolation of IA-producing fungi from a soil. This method was named DISCOVER (direct screening method based on coupling reactions for vinyl compound producers).In Chapter 5, 37 soil microbes isolated in Chapter 3 were further characterized. One out of the 37 strains was identified as A. niger S17-5. Identification of the IA derivatives produced by S17-5 revealed that they were identical to 9-HHIA and 10-HHIA. The antibacterial, anti-inflammatory, and cytotoxic properties of two IA derivatives were evaluated.In Chapter 6, IA and 10-HHIA were copolymerized at different ratios through aqueous free radical polymerization with KPS as an initiator, resulting in poly(IA-co-10-HHIA)s with different monomer compositions. This is the first time that biobased polyvinyls were synthesized using naturally occurring IA derivatives.In chapter 7, general review was described.In conclusion, developed screening methods permit selective isolation of microbes producing vinyl compounds from environment. And the vinyl compounds produced by microbes enable the development of bio-based polymers

    学校間連携を考慮した分散型生徒情報リポジトリに対するアクセス制御システムの試作

    Study on Photos Applicable to Communication between Two Generations in the South of Jiangsu Province in China

Empty nesters have become one of the social problems, and they usually face more cognitive impairment than others due to lack of social communication. Related studies have shown that using the same photos as media for conversation can help the elderly and young people quickly find topics to promote dialogue. However, how to use pictures to overcome the difficulties of communication between the two generations is rarely studied in China. The objective of this study is to use a variety of KANSEI Evaluation Methods to analyze the impacts of photos on communication between two generations in the south of Jiangsu Province in China and classify these photos to find the features of optimal photo groups. In Chapter 2, questionnaires with 145 photos were conducted in Taizhou City and Nanjing City, and the results show that the regional economic level is an essential factor affecting the impression of the elderly for the photos. In Chapter 3, the 2-generation conversation experiment with 40 photos was conducted in Suzhou City, and the results show that preset photo categories applicable for the Japanese elders are not ideal for the Chinese elders, so we should reclassify these photos. In Chapter 4, Protocol Analysis was applied to the conversation content in Chapter 3. Four photo clusters were obtained by Principal Component Analysis (PCA) and Hierarchical Clustering Analysis (HCA). We summarized two groups of these photo clusters: Group1 (Photos Suitable for the Aged and the Young), Group2 (Photos Not Suitable for the Aged and the Young). In Chapter 5, the conversation experiment with 40 photos was conducted in Nanjing City. Sentiment Analysis was applied to the conversation content. Four photo clusters were obtained by PCA and HCA, which was summarized into three groups: Group1 (Photos Suitable for the Aged and the Young), Group2 (Photos Suitable for the Young but Not for the Aged), Group3 (Photos Not Suitable for the Aged and the Young).The results of this study can be summarized into three groups of photos: Group1 (Photos Suitable for the Aged and the Young), which can stimulate more topics for the elderly, there is no generation gap, and interaction between the aged and the young is good. Group2 (Photos Suitable for the Young but Not for the Aged), which has a generation gap between the lives of elderly and young people. Young people are interested in it, which is not attractive to older people. Group3 (Photos Not Suitable for the Aged and the Young), for which older people have nothing to talk about, and young people also feel more pressure

    The study of the ferroelectricity of odd, even, odd-even, and even-even nylons

    京都工芸繊維大学博士(工学)Nylons are one of the useful engineering plastics possessing high melting point, high heat resistance, and high crystallinity with intermolecular hydrogen bond between amide groups. Polar amide groups in the molecular chain in odd-numbered and odd-odd-numbered nylons in which the number of carbon atoms are odd have parallel packing with intermolecular hydrogen bond, and thus non-zero net polarization in odd-numbered nylons and odd-odd-numbered nylons are switched by the external alternative electric field, so-called ferroelectric switching. On the other hand, anti-parallel packing of the polar amide groups in the molecular chain of even-numbered, even-even-numbered, or odd-even-numbered nylons leads the zero net polarization and thus non-ferroelectric properties. These discussions are very simple model and the practical net polarization and it’s switching in nylons are significantly related to the each crystal forms. Thus to investigate the ferroelectric nature of nylons is challenging and thus I focused on the ferroelectric nature of even-numbered and odd-numbered nylons and even-even-numbered and even-odd-numbered nylons in this thesis. In this thesis, not only polarization ? applied electric field (P ? E ) hysteresis curve, in which the spontaneous polarization Ps can be reversed by applying external electric field E to a ferroelectrics, but also I ? E hysteresis curve of the bipolar current I against the sinusoidal applied electric field E, in which the details of the polarization reversal history of ferroelectrics are presented, are used to investigate the ferroelectric properties of even-numbered and odd-numbered nylons and even-even-numbered and even-odd-numbered nylons in detail. Furthermore, a nylon thin film is fabricated using a spin coating method in which no molecular orientation is occurred, and thus ferroelectric nature of nylons can be investigated. The purpose of this study is to elucidate the mechanism of polarization reversal of spontaneous polarization in even-numbered and odd-numbered nylons and even-even-numbered and even-odd-numbered nylons and the characteristics of ferroelectricity of even-numbered and odd-numbered nylons and even-even-numbered and even-odd-numbered nylons. Furthermore, to clarify the origin of the ferroelectricity and the antiferroelectricity of even-numbered and odd-numbered nylons and even-even-numbered and even-odd-numbered nylons, the ferroelectricity of various nylons are investigated, and the possible application to the energy harvest or the energy storage materials is also discussed

    Polyethyleneimine-functional cellulose aerogels and wood-based templates for CO2 capture

    京都工芸繊維大学博士(工学)Polyethyleneimine (PEI) functionalized mesoporous sorbent have been widely studied for chemisorb CO2. However, those porous materials of aerogels show high cost and complicated process to restrict the application in industry. In this work, the cellulose aerogel with low density and high specific surface area was prepared by the non-solvent impacted thermally induced phase separation (NITIPS) method and hydrolysis process. After PEI impregnation process, the PEI-impregnated cellulose (PIC) aerogel showed large shrinkage due to the low mechanical strength and large hygroscopicity. Hence, a novel polyethyleneimine-crosslinked cellulose (PCC) aerogel sorbent was prepared through the crosslinking reaction. The obtained porous PCC aerogel showed 3D network structure with irregularly shaped microscale and nanoscale pores and large specific surface area. The CO2 adsorption capacity of PCC-1 aerogel reached 2.31 mmol/g at 25 °C under pure dry CO2 atmosphere based on the chemical and physical absorption. The PCC aerogel showed excellent CO2 adsorption-desorption recyclability, retaining 93% of the initial value after 10 adsorption-desorption cycle tests. However, the CO2 selective and mechanical properties of materials still face huge challenges to meet the practical requirement. Therefore, a high strength delignified wood was developed to be a matrix to load PEI for CO2/N2 selective adsoroption. After lignin removal, the delignified wood showed micro- and meso-porous structure with high specific surface area (44.3 m2/g) determined with physical CO2 adsorption. With the increasing the PEI loading content, the PEI-delignified wood composite with 49 wt% of PEI (P49) showed super-high CO2/N2 ideal selectivity (S = 114), high CO2 adsorption capacity (2.96 mmol/g) and excellent CO2 cyclic performance at 120 °C. Pseudo-second order model accurately predicted the CO2 adsorption behaviors of P49 at different temperature based on chemical adsorption. More importantly, the Young`s modulus of P49 reaches to 756 MPa, over 10 times higher than that of cellulose porous materials reported in literature. The developed PEI-delignified wood would play an important role in the combustion industry as a potential CO2 adsorption sorbent

    コマンド操作履歴付き端末動画を用いたグループ演習支援システムの試作

    プレゼンテーション発表者の身体動作分析に基づく練習映像振り返りツールの作成
