Imidacloprid decreases the total energy production in western honeybees although in sublethal doses it increased the values of six of the nine compounds in the respiratory and citric cycle
Neonicotinoids, including imidacloprid (ID), harm Apis mellifera in a number of ways, among others by impairing body maintenance and immunity. Energy resources are important to prevent this, particularly, as we hypothesized, in the fat body tissue. But hormesis was also reported in the energy-dependent traits of bees exposed to sublethal doses of ID. Consequently, concentrations/activities of respiratory and citric cycle compounds (Acetyl-CoA, IMH-2, AKG, succinate, fumarate, NADH2, CoC, COX, and ATP) were compared in the hemolymph and fat bodies of bees fed with diets containing 200 ppb (IM-200), 5 ppb (IM-5; sublethal), and 0 ppb of ID. The titers of the compounds were higher in the fat body than in the hemolymph, where the variability was higher. The pattern of response to ID was the same in both tissues, but differed between IM-200 and IM-5. The concentrations of the strongly correlated NADH2, ATP and acetyl-CoA decreased in ID-200 and ID-5, whereas the levels of the remaining compounds decreased in ID-200 but increased in ID-5. Decreased ATP levels in each diet show that the pesticide impairs the fat-body energy metabolism in spite of hormesis in some respiratory and citric cycle compounds even in the low, residual doses.</p
Arbuscular mycorrhizal symbiosis under short- and long-term drought in Lotus japonicus
[ENG:]The dataset contains results from experiments investigating the impact of arbuscular mycorrhizal fungi (Rhizophagus irregularis) on the responses of Lotus japonicus under full irrigation and during short-term (1 week) and long-term (3 weeks) drought. The data include morphological measurements such as shoot and root length, leaf number, and fresh and dry biomass, as well as root architecture parameters including total root length, volume, diameter, and number of tips. Physiological indicators such as leaf relative water content (RWC) and proline concentration are also included, along with chlorophyll fluorescence parameters such as Fv/Fm, Y, ETR, and NPQ. Additionally, the dataset provides information on the level of root colonization by AMF assessed using microscopy (gridline-intersect method) and quantified by qPCR. The dataset covers six experimental treatments: plants inoculated with mycorrhizal fungi and control plants supplemented with two phosphorus concentrations, under drought and full irrigation. Experiments were conducted in a phytotron at the Poznań University of Life Sciences, in the Biocentrum building at Dojazd 11, Poland. Methods included root scanning and image analysis (WinRHIZO), chlorophyll fluorescence measurements using an OS5p fluorometer, leaf water status assessment by the Weatherley method (1950), proline determination following Bates et al. (1973), and AMF colonization evaluation by microscopy and qPCR using species-specific gene markers.For more information, please see the ReadMe file.[PL:]Zbiór danych obejmuje wyniki eksperymentów dotyczących wpływu arbuskularnej mikoryzy (Rhizophagus irregularis) na reakcje Lotus japonicus w warunkach pełnego nawodnienia oraz podczas krótkotrwałej (1 tydzień) i długotrwałej (3 tygodnie) suszy. Dane zawierają pomiary morfologiczne, takie jak długość pędów i korzeni, liczba liści oraz biomasa świeża i sucha, a także parametry architektury korzeni, w tym całkowitą długość, objętość, średnicę i liczbę włośników. Uwzględniono również wskaźniki fizjologiczne, takie jak względna zawartość wody w liściach (RWC) i zawartość proliny, oraz parametry fluorescencji chlorofilu, w tym Fv/Fm, Y, ETR i NPQ. Dodatkowo zebrano dane dotyczące poziomu kolonizacji korzeni przez grzyby AMF ocenianego metodą mikroskopową (gridline-intersect) oraz ilościowo przy użyciu qPCR. Zbiór obejmuje sześć wariantów eksperymentalnych, w tym rośliny inokulowane grzybem mikoryzowym, oraz rośliny kontrolne, suplementowane dwoma stężeniami fosforu, w warunkach suszy i pełnego nawodnienia. Badania przeprowadzono w fitotronie na terenie Uniwersytetu Przyrodniczego w Poznaniu, w budynku Biocentrum przy ulicy Dojazd 11, Polska. Metody obejmowały skanowanie korzeni i analizę obrazu (WinRHIZO), pomiary fluorescencji chlorofilu za pomocą fluorometru OS5p, ocenę zawartości wody w liściach metodą Weatherley (1950), oznaczanie proliny metodą Bates i wsp. Tear fluorescein clearance rate assessment with modified slit lamp biomicroscopy
The data were generated as a result of clinical measurements in the MINIATURA project financed by the National Science Centre. The project aimed to demonstrate whether it is possible to determine tear clearance rate (TCR) using a slit-lamp biomicroscope and whether the TCR parameter determined in this way can be used to distinguish ophthalmologically healthy subjects from patients with Dry Eye Disease.Dataset includes:Two ZIP files containing 5-minute videorecordings of the ocular surface of participants with a fluorescein-stained tear film recorded through a modified slit lamp biomicroscope. The participants were divided into a study group (subjects diagnosed with Dry Eye Disease) and a control group according to the clinical protocol recommended in the Dry Eye Workshop II report.a Python code created to track the tear fluorescence intensity over time based on the average brightness of the recorded fluorescein image and, as a result, to plot graphs showing tear clearance over time and save numerical data used to determine Tear Clearance Rate (TCR). The code was deposited together with the user manual.</ul

Improvement of the quality of goat's milk products, including their nutritional value, through the addition of plant juices
Analysis of physicochemical and rheological properties of kefir obtained by hydrolysis and transgalactosylation of lactose and the addition of plant juices (buckthorn puree, mountain ash, beet, mulberry, burdock, aloe and kalanchoe pinnata) in various proportions (0.5%; 1.5% and 3%) in the process of lactic-alcohol fermentation. Analysis of the obtained goat cheeses with the addition of plant juices. the effect of adding plant juices on the change in pH, water activity and colour of kefir and goat milk cheese was assessed. Research carried out at the Faculty of Food Science and Nutrition, Poznań University of Life Sciences.</p
Analysis of Filtration Coefficient of Selected Recycled Materials on the Example of Concrete Aggregate and Rubber Waste
This dataset contains the results of permeability tests on recycled materials, specifically fine recycled concrete aggregate mixed with rubber granulate from waste car tires (3 mixtures marked as M1_R, M3_R, and M3) and shredded tires (3 mixtures marked as RTWS1, RTWS2, and RTWS3). For permeability testing, two different equipment were used. The permeability coefficient of the three tested mixtures: M3, M1_R, and M3_R, was investigated using the Humboldt Flex Panels model “HM-4150”. The permeability coefficient of the shredded tires was investigated employing the homemade apparatus.The dataset includes the results from variable-gradient tests under various consolidation presssures.</p

Ex vivo permeation of agomelatine from DLP 3D-printed cone microneedle systems containing a 3% AGM-drug resin mixture
Raw data from ex vivo permeation test (IVPT) of agomelatine (AGM) from microneedle systems.Microneedle system specifications:3D printing method: DLPmicroneedle shape: conecoating-gel type: - (no coating) 3% AGM mix within resin (served as printing material)Drug permeation studies:The permeation of agomelatine (AGM) was evaluated using Franz Diffusion Cells (Teledyne Hanson 376 Research, USA) with full-thickness human skin as the diffusion membrane. The skin was thawed in PBS at room temperature, mounted on the diffusion cells, and conditioned for 30 min. After this time the microneedle system was placed onto the chamber opening (1 cm²) with full-thickness human skin as the diffusion membrane. The study lasted for 7 days and sodium azide 0.02% w/v was added to the acceptor fluid as an antimicrobial agent. At specific time points, 0.3 mL samples were taken and analyzed using High-Performance Liquid Chromatography (HPLC). The amount of the acceptor medium taken for the analysis was immediately replaced with the fresh portion of the fluid. Six replications were performed for each formulation. After the study, the epidermis and dermis from each skin sample were separated manually using scissors and forceps and inserted in tubes with 3 mm zirconium beads (Benchmark Scientific Inc., Sayreville, NJ, USA). Next, 1 mL of water and ethanol solution (50:50 v/v) was added to each tube, and the samples were homogenized for 9 min using a BeadBug Microtube Homogenizer (Benchmark Scientific Inc., USA). Then, the samples were centrifuged for 5 min at 10,000 rpm, and the supernatant was analyzed using HPLC to examine the amount of the drug in the tissue.Quantification of AGM:The amount of AGM permeated was analyzed using High-Performance Liquid Chromatography (HPLC) (Shimadzu Nexera-I LC-2040C, Japan) with LabSolution Lite software.Chromatographic Conditions:Column: Reversed-phase C18 (HyperClone BDS C18, 5 µm, 4.6 × 250 mm, Phenomenex, Torrance, CA, USA)Column Temperature: 30.0 ± 0.2°CMobile Phase: Acetonitrile and 0.05 M potassium dihydrogen phosphate solution (pH = 2.9, adjusted with 85% orthophosphoric acid) in a 35:65 ratioMode: IsocraticFlow Rate: 1.0 mL/minDetection Wavelength: 230 nmThe set contains data in LCD/ LCB format, created using the LabSolutions software (HPLC, Nexera LC 2040C).
