    IsoFMiR: An unsupervised anomaly detection framework for biomarker discovery in rare cancers

    &lt;Description of the project&gt;Identifying microRNA (miRNA) biomarkers in rare cancers remains a major challenge due to limited patient cohorts and high tumor heterogeneity. To overcome these limitations, we developed IsoFMiR — an unsupervised anomaly detection framework designed to discover cancer-specific miRNA signatures without relying on large, labeled datasets. Built upon the Isolation Forest algorithm, IsoFMiR leverages the concept that samples from rare cancers such as sarcomas exhibit distinct molecular patterns when compared to more common malignancies. By training the model on miRNA expression profiles from abundant cancer datasets and subsequently applying it to sarcoma samples, IsoFMiR isolates anomalous expression signatures that may represent potential biomarkers.This approach addresses key limitations of conventional supervised learning methods, which often fail when applied to data-scarce conditions. Through this unsupervised design, IsoFMiR facilitates robust biomarker discovery, even in cases where annotated samples are minimal. The framework was implemented using Python 3.11.9 and R 4.4.2, with the Scikit-learn library employed for building the Isolation Forest model. Sequencing data were sourced from publicly available repositories such as The Cancer Genome Atlas (TCGA) and XenaBrowser, ensuring transparency and reproducibility.Overall, IsoFMiR represents a scalable and data-efficient framework that enables the identification of clinically relevant miRNA biomarkers in rare cancers.     Photochemical Functionalization of 4-Diazoisoquinoline-1,3(2H,4H)-diones and Their 1-Sulfoxide Analogues

    Isoquinoline-1,3(2H,4H)-diones have been the subject of numerous studies toward new pharmaceuticals, and therefore novel, mild functionalizations of this scaffold are highly desirable. As diazo compounds are versatile reagents in organic synthesis, 4-diazoisoquinoline-1,3(2H,4H)-diones should give access to a broad range of various derivatives. Indeed, they enable the introduction of fluorinated moieties via photochemical O–H insertion reactions in serviceable yields, typically in under 2 h. Exchanging the 1-carbonyl group with the sulfoxide moiety causes a hypsochromic shift in the absorption of the diazo compounds, and thus, violet light is required for effective O–H and S–H insertion reactions.</p

    Multi-state emission of TICT luminophores

    In this work, we address the challenge of designing versatile luminophores that maintain their emissive properties across various phases, including solutions and solid states (dual-state emission, DSE). We report the synthesis and detailed characterization of a series of Twisted Intramolecular Charge Transfer (TICT)-type luminophores with D-A-D molecular geometries composed of triphenylamine (TPA) or dimethylphenylamine fragments as donors (D) and aromatic ortho-substituted aldehydes as acceptors. In solutions, they show polarity-dependent solvatochromism, with emissions ranging from 500 to 600 nm. Importantly, their luminescence persists even in restrictive environments, such as crystalline, aggregated states, mechanically treated solids, and solid polymer matrices (PMMA). This durable condensed-phase luminescence results from their non-planar structure, particularly the ortho-substituted acceptor and propeller-shaped donor components, which effectively hinder harmful H-type stacking interactions linked to aggregation-induced quenching (AIQ), leaning more toward characteristics similar to aggregation-induced emission (AIE). Crystallographic data and theoretical calculations confirm the presence of twisted biaryl D-A fragments with D-A angles of 41-45°, a structural feature vital for preventing efficient solid-state stacking. Additionally, we find that these luminophores can undergo further twisting, showing a mechanochemically induced batochromic shift in luminescence (mechanochromism). The most sterically hindered compound consistently exhibits higher photoluminescent quantum yields under various conditions. Our results, supported by structural analysis, suggest that multi-state luminescence is a common trait among D-A-D-type twisted molecules, with significant potential for future development.</p

The birds were fed diets containing fat from Hermetia illucens larvae, soybean oil, rapeseed oil, palm oil, palm fatty acid distillate, poultry fat, pig lard, and beef tallow. On the last day of the experiment (35d), biological material, i.e., cecal content was collected for next generation sequencing (NGS).Data contains a ReadMe.txt file.</p
