    In-vitro Untersuchung zum Vergleich verschiedener intraoraler Keramikpoliersysteme an unterschiedlichen zahnfarbenen Werkstoffen

    Keramische Restaurationen kommen in der Zahnmedizin in den letzten Jahren immer häufiger zum Einsatz, da die heute verwendeten Keramiken nicht nur hohen ästhetischen Ansprüchen gerecht werden, sondern z.T. auch sehr hohe Festigkeiten aufweisen. Bei dentalen Keramiken ist eine gut polierte Oberfläche wesentlich, um zum einen eine geringe Plaqueretention zu erreichen und zum anderen die Festigkeit der Restauration zu erhöhen. Zahntechnische, keramische Restaurationen, dazu gehören Inlays, Onlays, Overalys, Teilkronen, Veneers, Verblendkronen und vollkeramische Kronen, werden nach der Herstellung vom Zahntechniker im Labor hochglanzpoliert und häufig mit einem Glanzbrand versehen. Die so bearbeitete Keramikoberfläche muss vom Zahnarzt vor dem Einsetzen nicht mehr bearbeitet werden. Jedoch müssen die Restaurationen vom Zahnarzt vor oder nach dem Einsetzen im Patientenmund angepasst werden, um Ungenauigkeiten vom Herstellungsprozess auszugleichen. Potentielle Fehlerquellen sind hierbei unpräzise Abformungen und Abformmaterialien, fehlerhafte Lagerung und Transport der Abformung sowie ungenaues Arbeiten bei der Modellherstellung und der Anfertigung der Restauration. Passungenauigkeiten können auch durch Änderung der Zahnstellung bei nicht exakt sitzenden Provisorien entstehen. Einschleifmaßnahmen, die vor dem Einsetzen der Restauration vorgenommen werden, können extraoral mit dem gleichen Poliersystem, wie es der Zahntechniker verwendet hat, bearbeitet werden. Letzte Korrekturen oder das Einschleifen von z.B. Lithium-Disilikat-Keramiken erfolgen jedoch erst nach Eingliederung im Patientenmund. Beim Einschleifen der Keramiken werden hier meist ein Diamantschleifer (Gelbring oder Rotring) verwendet. Diese in-vitro Untersuchung soll Keramikpoliersysteme für die intraorale Anwendung verschiedener Hersteller miteinander vergleichen. Dabei werden Keramikplättchen mit verschiedenen Rauigkeiten mit dem jeweiligen System poliert und die Oberflächenrauigkeit ausgemessen

    Die Rolle des mTOR-Signalweges in einem in vivo-Modell der neonatalen hypoxisch-ischämischen Hirnschädigung

    Perinatale Aspyxie stellt mit einer Inzidenz von 1-4 pro 1000 Lebendgeburten eine Hauptursache für Langzeitmorbidität und –mortalität bei Neugeborenen dar, wobei die Zahl in Entwicklungsländern deutlich höher ist. Eine der möglichen Folgen, die zu langfristiger Behinderung führen kann, ist die neonatale hypoxisch-ischämische Enzephalopathie (HIE). Der mTOR-Signalweg fungiert als zellulärer Energiesensor. Er steuert an zentraler Stelle Protein-, Lipid- und Nukleotidmetabolismus und beeinflusst die Entzündungs-reaktion sowie die Autophagieleistung der Zelle. Pathophysiologisch spielen diese Parameter bei der HIE eine Rolle und stellen deshalb mögliche Angriffspunkte zukünftiger Therapien dar. In unserem Rattenmodell der neonatalen HIE konnten wir zeigen, dass der mTOR-Signalweg im Kortex und Hippocampus nach 6 und 24 h durch den hypoxisch-ischämischen Insult deutlich beeinflusst wird. Hypoxie-Ischämie führte in den genannten Arealen ebenfalls zu einem Zytokinsturm, der durch Hypothermie-Behandlung abgeschwächt werden konnte. Diese Ergebnisse belegen eine Beteiligung des mTOR-Signalwegs und der Hyperinflammation in der Pathophysiologie der hypoxisch-ischämischen Hirnschädigung bei Neugeborenen, sodass eine Modulation dieser Parameter in Zukunft neuroprotektive Effekte erzielen und Mortalität und Morbidität bei Neugeborenen mit HIE verbessern könnte

    Investigating the Conformational Landscape of Cas13a

    Bacteria and bacteriophages are in a constant arms race to develop strategies to coexist. One adaptive defense system, which bacteria and archaea have developed against phages is the CRISPR-Cas (Clustered Regularly Interspaced Short Palindromic Repeats – CRISPR Associated proteins) system. Out of a variety of known CRISPR-Cas types, the type VI CRISPR effector protein Cas13a (Cas-associated 13a), composed of a recognition (REC) lobe and a nuclease (NUC) lobe, has unique properties. In contrast to the most prominent Cas protein representative Cas9, Cas13a binds and cleaves RNA and not DNA. In addition, it catalyzes the maturation of a precursor-CRISPR RNA (pre-crRNA) and the cleavage of the target RNA, which activates a unique sequence non-specific collateral RNA cleavage. This collateral cleavage was harnessed as nucleic acid detection tool, to detect viral RNA, causing human diseases. Further Cas13a has therapeutic applications for example in inhibiting cancer cell growth. Despite the high medical relevance and application, the underlying coordinating mechanism and its structure-dynamics-function relationship was not investigated. Structures from all functionally relevant complexes are described (apo, pre-crRNA bound, crRNA bound, and cr- and target RNA bound), but they origin from proteins of different organisms and contain different extends of truncation. Additionally, it is unknown how the structures relate to functional aspects of this system. Thus, to identify and follow conformational changes on the molecular level, information of all states from one organism are needed. In this work, mainly Pulsed Electron-Electron Double Resonance (PELDOR) spectroscopy was used to analyze conformational changes during the functional pathway of Cas13a. In the first part of this thesis, basic biochemical techniques were used to optimize the expression and purification approach for Cas13a from Leptotrichia buccalis (Lbu) and to optimize RNA cleavage assays in our laboratory. The second part of this thesis focuses on the development of active double labelled protein constructs, for PELDOR studies. A mutational analysis was used, to find replacements for three native cysteines, generating a protein construct that retained cleavage activity after spin labelling. The third part of the thesis focuses on validating the structures of LbuCas13a and following its conformational changes through the entire functional pathway. The PELDOR measurements showed a high flexibility in the apo state of the REC lobe. This flexibility was not described by any known structure or prediction. In contrast, the apo state of the NUC lobe is rigid, with the helical-2 domain being the exception. The REC and NUC lobes are flexible towards each other. The pre-crRNA bound structure is not known. We reveal this structure to match the crRNA bound state. The expected conformational changes from the experimentally known binary crRNA-bound complex to the experimentally known ternary complex were found to occur in frozen solution. Interestingly, by using a combination of PELDOR spectroscopy and AFM measurements, a previously unknown dimeric structure of the protein complex with cr- and target RNA was found. This structure is different than the structure observed in the asymmetric unit of the cr- and target RNA bound LbuCas13a. By summing up, for the first time, the entire functional cycle of the Cas13a protein from one organism is structurally and dynamically investigated and new details are added

    Postoperative Nervus femoralis Läsionen in der gynäkologischen Onkologie

    Die meisten Verletzungen des Nervus femoralis sind iatrogen bedingt und treten nach chirurgischen Eingriffen auf. Auch wenn sie meist eine gute Prognose haben, kann eine spontane Heilung Monate andauern und somit den postoperativen Heilungsprozess beeinflussen. Daher wurden in dieser Studie neben der Prävalenz für postoperative Nervus femoralis Verletzungen in der gynäkologischen Onkologie weitere Kriterien untersucht, um mögliche Risikofaktoren für das postoperative Auftreten einer Nervus femoralis Verletzung zu ermitteln. Die Prävalenz für das Auftreten postoperativer Nervus femoralis Schäden lag bei Operationen in der gynäkologischen Onkologie im Zeitraum von 06 / 2006 und 12 / 2017 bei 1,54 %. Es zeigte sich ein signifikanter Zusammengang zu der Operationsdauer, der Lagerung der Patientinnen in Maquetstiefeln gegenüber der Lagerung in GöpelBeinhaltern, dem Vorhandensein eines intraoperativen arteriellen Zugangs via Arteria femoralis, einer vorausgegangenen Chemotherapie, dem Nikotinkonsum, einer vorausgegangene Chemotherapie und einer vorbestehenden Polyneuropathie. Während Nikotinkonsum, eine vorausgegangene Chemotherapie sowie eine vorbestehende Polyneuropathie zum Operationszeitpunkt unveränderbare Faktoren darstellen, können die Operationsdauer, die Lagerung der Patientinnen sowie das Vorhandensein eines arteriellen Zugangs beeinflusst werden. In der Universitätsfrauenklinik wurden nach Ermittlung o.g. Daten folgende klinische Konsequenzen gezogen: 1. Die Retraktorblätter wurden mit kleinerer intraabdomineller Auflagefläche neu entwickelt. 2. In der Regel werden lange zytoreduktive Operationen – wie sie beim Ovarialkarzinom erforderlich sind – im Oberbauch begonnen, um die Zeit einer möglicherweise nicht bemerkten ungünstigen Beinlagerung zu verkürzen. 3. Auf arterielle Femoraliskatheter wird zu Gunsten von arteriellen Radialiskathetern verzichtet. 4. Eine standardisierte Lagerungsüberprüfung wurde in den OP-Ablauf integriert

    In Search for Authenticity : Village tourism, community resilience and cultural preservation in Indonesia

    The authenticity of nature and culture is the main attraction of rural tourism. However, this does not mean that the attractions in these destinations are untamed. Instead, tourism attractions have been managed and set up in such a way as to be attractive, memorable, and safe for tourists. The hope is that these attractions will provide a satisfying travel experience, encouraging visitors to return or recommend the destination to their social circles. This reality makes rural tourism a contest between preservation and commodification. Samiran Tourism Village is a contemporary case study from Southeast Asia. This destination is one of the most popular rural tourism spots at the foot of Mount Merapi in Central Java, Indonesia. The majority of visitors are domestic tourists from cities in Central Java. Most of them come to enjoy the mountainous landscape with the comfort of various amenities, such as cafes, restaurants, cottages, or local theme parks. Many tourists are also seeking a closer experience with rural life in the area. Those who wish to sample the unique Merapi-style local lifestyle participate in the Samiran Village Tour guided by a local tour operator named Guyub Rukun. Both segments of tourists seek authentic experiences in a familiar, comfortable, and organized manner. This study assesses authenticity as a social construct. Various tourism scholars argue that authenticity is a perceptual experience based on the interaction between tourists and tourist attractions. A genuine attraction will evoke more admiration than its replica. Tourists may accept artificial attractions as substitutes when the original is unavailable or inaccessible. On the other hand, the hosts find the momentum from tourism to preserve their traditions. Economic diversification resulting from tourism contributes to the capacity of the community to sustain its traditions. Based on this conceptual foundation, this study proposes a hypothesis that tourists and hosts mutually contribute to the construction of the authenticity image of rural tourist attractions. Tourists enjoy the beauty of mountainous landscapes, farmlands, and enduring Javanese traditions, which validate their imagination of the village's authenticity. Tourism provides hosts with a reason to revisit the relevance of old values. Traditions in religious-based festivals and local art troupes reviving folk dances are examples of how cultural preservation intersects with the practices of the Samiran Village Tourism program. Staged authenticity serves as a compromise between tourists seeking authenticity and hosts pursuing progress

    Asymmetrische Abhängigkeit, exkludierende Inklusion, skalenfreie Netzwerke : theoretische Ressourcen zur Erklärung moderner Sklaverei; sexueller Knechtschaft und des Menschenhandels in der Weltgesellschaft

    Nigerianische Sexhandelsnetzwerke sind durch komplexe Strukturen geprägt, die tief in der Weltgesellschaft verwurzelt sind und schwer zu durchdringen bleiben. Ungeachtet der zunehmenden Aufmerksamkeit für Phänomene wie moderne Sklaverei, sexuelle Ausbeutung und Menschenhandel zeigen die Zahlen der zu verzeichnenden Betroffenen, wie hartnäckig sich solche Strukturen halten. Dies wirft nicht nur praktische, sondern auch wissenschaftlich und theoretisch wichtige Implikationen für die soziologische Forschung auf. Theoretische Ansätze zu asymmetrischer Abhängigkeit, exkludierender Inklusion und skalenfreien Netzwerken bieten einen differenzierten Blickwinkel, um die Strukturen und Dynamiken dieser Netzwerke tiefergehend zu analysieren. Die Analyse „skalenfreier Netzwerke“ ermöglicht ein tieferes Verständnis davon, wie Machtverhältnisse und Abhängigkeitsstrukturen innerhalb dieser Systeme stabilisiert und reproduziert werden. Dabei zeigt sich, dass das System durch Flexibilität und Anpassungsfähigkeit eine bemerkenswerte Widerstandsfähigkeit aufweist. Unter Anwendung der Theorien von Rudolf Stichweh wird untersucht, wie Inklusions- und Exklusionsmechanismen sowie (starke) asymmetrische Abhängigkeit die Stabilität dieser Netzwerke gewährleisten. Eine besondere Bedeutung kommen dabei den Ausgleichsoperationen zu, welche eigentlich der Verringerung von asymmetrischer Abhängigkeit dienen. Paradoxerweise tragen diese jedoch häufig zur Selbstproduktion des Systems und einer Verstärkung der Machtverhältnisse bei, anstatt sie zu destabilisieren. Dieser Modus Operandi unterstreicht den totalen Charakter dieser Institution der (starken) asymmetrischen Abhängigkeit in Form einer exkludierenden Inklusion. Die Analyse verdeutlicht, wie relevant eine gezielte Auseinandersetzung mit den Mechanismen der Reproduktion dieser Netzwerke ist. Zudem macht sie deutlich, dass eine holistische Herangehensweise notwendig ist, die sowohl offensichtliche als auch subtilere Machtstrukturen in Betracht zieht, um nachhaltige Strategien zur Bekämpfung von Menschenhandel und sexueller Ausbeutung zu entwickeln

    Nonstationary Least-Squares Collocation

    Many geodesists worldwide deal with the modelling of functions to approximate or interpolate measured data. For this purpose, a functional model is usually set up and adjusted by parameters so that it fits as precisely as possible to the data. One of the most important questions is how to select the functions of the model. One method, which has the advantage of being as independent as possible from the choice of functions is provided by least-squares collocation. Here, function values are estimated from the dependencies (covariances) between the observations themselves. Even though, when predicting values between the data, an analytic model, which describes the track of the covariances, still must be specified. This is where this dissertation starts. After a general introduction and the discussion of the crucial terms (chapter 1) as well as the current state of the field (chapter 2), thus it is firstly shown how the estimate of autoregressive (AR) processes can be used to create a continuation of the sequence of variances, while the resulting function also must satisfy all requirements of a covariance function (chapter 3). The benefit of this method lies in the fact that the estimate of AR processes is data-driven. After finding the most suitable AR process of order p, a continuous continuation of the discrete covariances is to be found which is (1) clearly assigned to the AR process, (2) reproduces the discrete covariances and (3) is positive definite. As the spectrum of these covariance functions differ from the spectrum of the discrete variances, I show how they can be transformed into one another by a multiplication with a dirac comb. It must be mentioned that this thesis is limited to one dimensional time series associated with time. The results of chapter 3 shows that instead of depending on the coefficients directly, the constructed covariance function depends on the roots of the characteristic polynomial of the AR process. These roots are also used to compute the spectrum of the AR process. As they offer the advantage of the coefficient being variable in time, the approach of using a TVAR process with known root motion guarantees that the roots of the characteristic polynomial of the process stay within the unit circle which implies that the variance of the resulting process stays finite. From this, we derive three methods to estimate TVAR processes, i.e. with linear, quadratic and piecewise linear root motion. Following the transformation from the coefficients of the AR process into the corresponding roots with linear or quadratic motions, only TVAR processes of order one, two and three are estimated directly. Since there is no analytic solution for computing the roots of a polynomial of order five or higher, the TVAR processes of higher order are estimated by a successive calculation using TVAR(1) and TVAR(2) processes. As its title suggests, this study provides a method to compute the least-squares collocation under the assumption of nonstationary time series. As such, chapter 5 combines the findings of chapter 3 and chapter 4. Therefore the discrete covariances of a TVAR process are derived. In contrast to the discrete covariances of the stationary AR process these discrete covariances not only depend on the lag, but also on the epoch of the first observation as well as on the fact whether the second epoch relative to the first is in the past or in the future. As a result, the continuous continuation of the discrete covariances embodies a function of two parameters; the epoch of the first observation and the difference between the second and the first epoch including the sign of the difference. In chapter 6, the different theories are tested on discrete measurements. First, sea level anomalies are estimated either by the Gauss-Markov model and trigonometric base functions, or by the Least-squares collocation with a covariance function from AR processes. Second, TVAR processes with predetermined root motions are estimated by GNSS elevation measurements and validated over the roots of stationary AR processes estimated for a moving window. In a final example, functions are estimated by temperature anomalies, wherein the covariances for the least-squares collocation come once from an AR(1) process estimate and another time from a TVAR(1) process estimate
