However, there has been an increasing interest in the variability of hydrological processes that induced nonstationarity in data. There is a consensus in the scientific community that climate change has accelerated over the past decades and will continue in the future. Therefore, water engineers have been faced with the practical problems of designing a hydrologic system against future extreme events including droughts. Therefore, the risk assessment framework based on nonstationary modeling is necessary for a changing climate to deal with unexpected damages from extreme droughts. The changing statistical characteristics are the most critical aspects of nonstationary modeling. Generalized Additive Models in Location, Scale, and Shape (GAMLSS) provides the modeling technique that the distribution parameters can be assumed to be linear, nonlinear, or nonparametric smoothing functions of the explanatory variables. GAMLSS allows not only the location parameter but all parameters can be modeled as parametric terms. The overall drought risk assessment is comprised of three phases, which are risk identification, risk analysis, and risk evaluation. Firstly, risk identification (Chapter 3) mainly consists of defining the event and what would be its consequences. The risk condition is identified that daily streamflow under the observed low flow, and hence the prolonged duration and its total volume of flow deficit are estimated as drought properties. The modeling variables for drought risk assessment are decided as water supply performance indexes i.e., reliability and vulnerability, defined from durations and severities during the specific design period, which is a 5-year moving window in the dissertation. Reliability generally ranges from 0.05 to 0.40, and the maximum values of reliability are observed generally in P2 (2041-2070) and P3 (2071-2099) rather than P1 (2011-2040) On the other hands, vulnerabilities estimated the average values range from 10 to 20 cms, and unlike reliability, vulnerability tends to decrease in P2 and P3, and the maximum values are generally observed in P1. Therefore, it is expected that drought duration would increase, while the magnitude of the deficit would decrease in the future. Secondly, the risk analysis (Chapter 4) involves the understanding of the riskhow the hazard cause the damage and how severe the damage will be. The time-dependent copula model based on GAMLSS framework is built to analyze the future drought risk. Reliability and vulnerability are used for marginal distributions for copula and assumed to be time-varying. The parameters of each marginal and joint distribution are assumed to be a constant, linear or polynomial function of time to capture nonstationary features of droughts. The hydrologic risk is evaluated using the exceedance probability of an event that has return period T within the N-year design period. The occurrence probability is changing with time, which makes ambiguous to select the design criteria. Therefore, the drought occurrence probabilities of average drought events are estimated to represent the changing risk and compared with the results of stationary models. The occurrence probabilities of each scenario generally have the ranges from 0.25 to 0.30 under the drought conditions that have average reliability and vulnerability. Lastly, the risk evaluation (Chapter 5) is the process of comparing the results of risk analysis with predefined risk criteria to determine whether the harm of risk is acceptable or not. Hence, the magnitude and occurrence probability of the possible damage should be estimated. Joint Drought Management Index (JDMI) is proposed to quantify the risk using a nonstationary copula model. JDMI is unique drought multi-index because it can quantify the range of variables that can produce the same quantile of bivariate copula using Kendall distribution. JDMI can classify drought conditions into five states, i.e., normal, mild, moderate, severe, and extreme. It is designed that the larger JDMI, the higher the drought risk. The risk quantification method is also developed using JDMI, which is the estimation of potential values of drought duration and severity. It is achieved through the backward process of JDMI estimation, and hence the minimum values that trigger each state of drought that possibly occurs in each scenario are obtained. For example, during 5 years (1825 days), the dry condition would be problematic when it is persisted from 70 to 180 days and has daily flow deficit of 0.15 to 0.40 million tons per day. Finally, the expected amount of water deficit per five years is estimated based on the potential drought properties by simply multiplying duration and severity. It is a fundamental question of how drought risk should be modeled under changing environmental condition. The risk assessment framework proposed in the dissertation would provide the decision criteria through nonstationary quantities of drought risk. Thus the results enable to support establishing drought management or mitigation plans. However, the socio-economic condition of the local community is not in consideration, and hence it should be revealed in the future study in order to reduce drought risk in practice.Docto

    Korea has serious youth unemployment problems due to low global growth and technological development. As a result, the younger generation is staying at school longer than in the past. Also, young Koreans have so many things to give up that they are called the N-Po generation, but they are also in the age group of the trend of a steady increase in overseas trip. The purpose of the study was to understand the essential meaning of the experience of people in their 20s and 30s on overseas trips, called the N-Po Generation to understand phenomenon, in-depth interviews were conducted with 12 participants in the study using Giorgi's phenomenological research methods. In this analysis, 30 key meanings and 8 themes were presented. five categories have been identified. The results how that N-po generations’s meaning of experience in overseas trip is as follows: (1)A struggle to survive in a cruel reality, (2)overseas trip as a career strategy, (3)the pursuit of psychological comfort through nature, (4)memories of the happy past, (5)opportunities to restore confidence. N-Po generations’s overseas trip was an act of trying to prepare for his future as a positive mind without succumbing to the difficult reality. Also, these actions were the result of human nature's pursuit of happiness

    Amino acid signatures of HLA Class-I and II molecules are strongly associated with SLE susceptibility and autoantibody production in Eastern Asians

    Human leukocyte antigen (HLA) is a key genetic factor conferring risk of systemic lupus erythematosus (SLE), but precise independent localization of HLA effects is extremely challenging. As a result, the contribution of specific HLA alleles and amino-acid residues to the overall risk of SLE and to risk of specific autoantibodies are far from completely understood. Here, we dissected (a) overall SLE association signals across HLA, (b) HLA-peptide interaction, and (c) residue-autoantibody association. Classical alleles, SNPs, and amino-acid residues of eight HLA genes were imputed across 4,915 SLE cases and 13,513 controls from Eastern Asia. We performed association followed by conditional analysis across HLA, assessing both overall SLE risk and risk of autoantibody production. DR15 alleles HLA-DRB1*15:01 (P = 1.4x10(-27), odds ratio (OR) = 1.57) and HLA-DQB1*06:02 (P = 7.4x10(-23), OR = 1.55) formed the most significant haplotype (OR = 2.33). Conditioned protein-residue signals were stronger than allele signals and mapped predominantly to HLA-DRB1 residue 13 (P = 2.2x10(-75)) and its proxy position 11 (P = 1.1x10(-67)), followed by HLA-DRB1-37 (P = 4.5x10(-24)). After conditioning on HLA-DRB1, novel associations at HLA-A-70 (P = 1.4x10(-8)), HLA-DPB1-35 (P = 9.0x10(-16)), HLA-DQB1-37 (P = 2.7x10(-14)), and HLA-B-9 (P = 6.5x10(-15)) emerged. Together, these seven residues increased the proportion of explained heritability due to HLA to 2.6%. Risk residues for both overall disease and hallmark autoantibodies (i.e., nRNP: DRB1-11, P = 2.0x10(-14); DRB1-13, P = 2.9x10(-13); DRB1-30, P = 3.9x10(-14)) localized to the peptide-binding groove of HLA-DRB1. Enrichment for specific amino-acid characteristics in the peptide-binding groove correlated with overall SLE risk and with autoantibody presence. Risk residues were in primarily negatively charged side-chains, in contrast with rheumatoid arthritis. We identified novel SLE signals in HLA Class I loci (HLA-A, HLA-B), and localized primary Class II signals to five residues in HLA-DRB1, HLA-DPB1, and HLA-DQB1. These findings provide insights about the mechanisms by which the risk residues interact with each other to produce autoantibodies and are involved in SLE pathophysiology. In spite of multiple SLE association signals identified in the HLA region, only amino-acid residues within HLA-DRB1 have been specifically described previously. In this study, we performed an imputation-based analysis on individuals with East Asian ancestry, and characterized SLE risk within the HLA region for all involved independent genes (HLA-DRB1, HLA-DPB1, HLA-DQB1, HLA-A, and HLA-B).
