Cartel Formation With Endogenous Capacity and Demand Uncertainty
This article analyzes the strategic decisions of firms whether to establish and adhere to a cartel when they can also shape competition by investing into production capacity while being subject to unexpected demand shocks with persistence. The model shows that a negative demand shock can facilitate cartel formation despite lowering collusive profits. This is because lower demand reduces capacity utilization and makes competition more intense especially when capacities are durable and when demand falls much within a short time. The model also shows that firms with a low discount rate strive for a dominant position in the market which results in asymmetric capacity distributions. These obstruct collusive strategies. This is interesting because a low discount rate is usually considered a facilitating factor for collusion
Immune tolerance and the prevention of autoimmune diseases essentially depend on thymic tissue homeostasis
The intricate balance of immune reactions towards invading pathogens and
immune tolerance towards self is pivotal in preventing autoimmune diseases,
with the thymus playing a central role in establishing and maintaining this
equilibrium. The induction of central immune tolerance in the thymus involves
the elimination of self-reactive T cells, a mechanism essential for averting
autoimmunity. Disruption of the thymic T cell selection mechanisms can lead
to the development of autoimmune diseases. In the dynamic microenvironment
of the thymus, T cell migration and interactions with thymic stromal cells are
critical for the selection processes that ensure self-tolerance. Thymic epithelial
cells are particularly significant in this context, presenting self-antigens and
inducing the negative selection of autoreactive T cells. Further, the synergistic
roles of thymic fibroblasts, B cells, and dendritic cells in antigen presentation,
selection and the development of regulatory T cells are pivotal in maintaining
immune responses tightly regulated. This review article collates these insights,
offering a comprehensive examination of the multifaceted role of thymic tissue
homeostasis in the establishment of immune tolerance and its implications in the
prevention of autoimmune diseases. Additionally, the developmental pathways
of the thymus are explored, highlighting how genetic aberrations can disrupt
thymic architecture and function, leading to autoimmune conditions. The impact
of infections on immune tolerance is another critical area, with pathogens
potentially triggering autoimmunity by altering thymic homeostasis. Overall,
this review underscores the integral role of thymic tissue homeostasis in the
prevention of autoimmune diseases, discussing insights into potential
therapeutic strategies and examining putative avenues for future research on
developing thymic-based therapies in treating and preventing

Inzidenz und Outcome von Zytomegalievirus- und BK-Virusinfektionen nach Nieren- und kombinierter Nieren- und Pankreastransplantation - Eine retrospektive monozentrische Datenanalyse -
In Deutschland sind etwa 95.000 Menschen dialysepflichtig, wobei eine Nierentransplantation für viele der Patient:innen die zu präferierende Therapieoption darstellt, da sie der Dialyse im Hinblick auf Mortalität, Morbidität und Lebensqualität überlegen ist. Zur Aufrechterhaltung von Lebensqualität und im Hinblick auf die limitierte Anzahl an verfügbaren Organen ist es von fundamentaler Relevanz, die Transplantatfunktion zu sichern. Durch eine potente Immunsuppression kann das Risiko einer Rejektion verringert werden. Im Gegenzug steigt das Risiko opportunistischer Infektionen u.a. mit CMV und BKV, welche potenziell mit einer gesteigerten Rate an Transplantatverlusten sowie einer erhöhten Mortalität
einhergehen können. Deshalb sind eine frühzeitige Detektion und Einleitung einer adäquaten Therapie von elementarer Bedeutung. Ziel der vorliegenden Arbeit war es,
die Häufigkeit beider Infektionen und der assoziierten Krankheitsbilder am Transplantationszentrum Marburg zu erfassen sowie Risikofaktoren und das Outcome
zu evaluieren. Dazu wurden retrospektiv die Daten von Patient:innen, welche sich im Zeitraum vom Januar 2012 bis Juli 2020 einer Nieren- oder kombinierten Nieren- und
Pankreastransplantation am Transplantationszentrum Marburg unterzogen haben, analysiert. Im Einzelnen wurden neben den demografischen Daten die Inzidenz von
CMV- und BKV-Infektionen sowie ggf. eine histologisch gesicherte renale Beteiligung erfasst und mit dem renalen Outcome anhand des Serum-Kreatinins, des Transplantat-Überlebens und der Mortalität assoziiert.
Das Alter der 152 Patient:innen betrug im Median 53 Jahre und die Nachbeobachtungsdauer 40,1 Monate (IQR = 49). Bei 38,8 % konnte keine Virusreplikation, bei 28,9 % eine Replikation von CMV, bei 17,8 % von BKV und bei 14,5 % beider Viren im Blut oder Urin nachgewiesen. Insgesamt lag die Inzidenz der CMV-Infektionen bei 43,4 % und die der BKV-Infektionen bei 32,2 %. Als Risikofaktoren für CMV-Infektionen konnte die serologische Konstellation
D+/R- und eine Anzahl von ³ 12 HWI in den ersten 24 Monaten nach Transplantation identifiziert werden. Rejektionen und die serologische Konstellation D+/R- waren mit CMV-Erkrankungen assoziiert. Außerdem zeigte sich ein Zusammenhang zwischen BKV-Infektionen und Rejektionen sowie zwischen PD und BKAN. Personen mit einem additiven Risiko wie serologischer D+/R- Konstellation, vermehrten HWI nach Transplantation und Rejektionen könnten somit von einem vermehrten Screening
profitieren, um die Virusreplikation frühestmöglich zu detektieren. Des Weiteren konnte in unserem Patientenkollektiv nachgewiesen werden, dass die
exkretorische Transplantatfunktion bei Personen mit alleiniger BKV-Infektion auf vergleichbarem Niveau mit derer von Personen ohne Virusreplikation lag. Außerdem
war eine alleinige BKV-Infektion oder BKAN erfreulicherweise nicht mit gesteigerter Mortalität oder Transplantatverlusten assoziiert. Dem gegenübergestellt
ging eine Koinfektion mit CMV und BKV mit einer geminderten Transplantatfunktion, gesteigerter Mortalität und einem höheren Risiko für Transplantatverluste einher. In dieser Kohorte kam es bei Vorliegen einer BKAN zusätzlich vermehrt zu Transplantatverlusten. Analog dazu führte eine alleinige CMV-Infektion oder -Erkrankung ebenfalls zu einem schlechteren Outcome verglichen mit der Kohorte ohne Virusreplikation.
Die Ergebnisse dieser Studie legen nahe, dass insbesondere CMV-Infektionen und CMV+BKV-Koinfektionen eine große Krankheitslast am Transplantationszentrum Marburg darstellen. Alleinige BKV-Infektionen hingegen sind bei frühzeitiger Erkennung und Einleitung einer Therapie, im Sinne einer Dosisreduktion der
immunsuppressiven Medikation und ggf. unter Hinzunahme von Leflunomid, nicht mit einem minderen Outcome assoziiert
Firm Selection and the Role of Union Heterogeneity
Empirical evidence suggests that high-productivity firms face stronger
trade unions than low-productivity firms. Then a policy that puts all unions
into a better bargaining position is no longer neutral for firm selection as
in models with a uniform bargaining strength across firms. Using a Melitztype
model, we show that firm selection becomes less severe. Since more
low-productivity firms enter the market, the negative employment effect of
unionization is mitigated. Neglecting inter-union differences in bargaining
power leads to an overestimation of the negative labor market effects. However, trade liberalization increases unemployment because firms with the least powerful labor unions have to leave the market
Internal Migration and House Prices in Australia
Australia is one of the most mobile countries in the world through internal migration, which is an overlooked part of population change. This study provides the first empirical evidence for the impact of internal migration inflow on house price changes across 237 statistical regions in Australia from 2014 to 2019. Employing a spatial correlation approach and constructing a novel instrumental variable that matches the shift-share instrument used in the immigration literature, the paper indicates that population mobility increases housing prices in migrationreceiving regions. Internal migration that amounts to 1% of the initial local area population is associated with a 0.7% to 0.8% increase in house prices in the three most populated states of Australia. The results provide further suggestive evidence that migration inflow has a significant positive effect on house price changes in metropolitan areas of New South Wales and Victoria – i.e., Capital Cities of Sydney and Melbourne – rather than non-metropolitan regions
Are Eastern European Taylor Reaction Functions Asymmetric in Inflation or Output: Empirical Evidence for four Countries
Do central banks in Eastern European countries react asymmetrically
and in a non-linear fashion to changes in inflation and output? We tackle
this question by expanding the standard Taylor reaction function for the
four inflation targeting countries Czech Republic, Hungary, Poland and Ro-
mania. We do so taking explicitly inflation rates below or above target and
output below or above potential, the so-called state of the economy, into
account. The results reveal that there are indeed substantial asymmetries in
the reaction function of the Czech, Polish and Romanian central bank, which
are only evident when the combination of inflation and output thresholds is
explicitly modelled in one estimation equation. For these three central banks
also non-linearities in the inflation and output response could be verified
Hippokampale MikroRNA-Expression bei der Entstehung epileptischer Toleranz im Rattenmodell
Epilepsie gehört mit einer Prävalenz von 0,5-1% (ca. 400.000 bis 800.000 Menschen in Deutschland) und einer Inzidenz von 30.000 Neuerkrankten pro Jahr zu den häufigsten chronischen neurologischen Erkrankungen und kann teilweise mit erheblichen Einschränkungen im Alltag der Menschen verbunden sein. Die bisher zur Verfügung stehenden Therapieoptionen beinhalten vor allem die Pharmakotherapie, welche jedoch nebenwirkungsreich und vor allem bei Temporallappenepilepsien oftmals unzureichend sein kann. Epileptische Toleranz beschreibt einen Zustand einer vorrübergehenden erhöhten Resistenz gegenüber der Entwicklung einer Epilepsie und kann durch verschiedene Präkonditionierungsschemata erlangt werden. Die zugrundeliegenden Mechanismen der epileptischen Toleranz sind bisher jedoch nur unzureichend verstanden. MiRNAs sind kurze nicht-kodierende Ribonukleinsäuren, die durch Kontrolle der Translation von mRNA maßgeblichen Einfluss auf die Proteinsynthese nehmen. Auch im Bereich der Epilepsie und epileptischen Toleranz wurde eine Regulierung ihrer Expression festgestellt. In der vorliegenden Arbeit wurden am Tractus-perforans- Modell der Epileptogenese in Ratten Veränderungen in der miRNA-Expression im Hippocampus unter vier verschiedenen Bedingungen (preconditioning, injury, tolerance, control) untersucht. Hierfür wurden die Versuchstiergruppen nach unterschiedlichen Paradigmen stimuliert. Nach Sequenzierung der miRNAs zeigten sich insgesamt 21 miRNAs signifikant hoch- und 15 miRNAs herunterreguliert, wovon ausschließlich in tolerance zehn miRNAs hoch- und sechs miRNAs herunterreguliert waren. Die mit diesen miRNAs verbundenen Prozesse betreffen vor allem die Regulierung von Apoptose, Morphologie und Bildung von Axonen, Dendriten und Synapsen. Auf dieser Grundlage bedarf es nun weiterer Studien, um die genauen Funktionsmechanismen der mit epileptischer Toleranz assoziierten miRNAs zu untersuchen und ihr therapeutisches Potential zu explorieren.With a prevalence of 0.5-1% (approx. 400,000 to 800,000 people) and an incidence of 30.000 cases per year, epilepsy is one of the most common chronic neurological diseases world wide. Patients suffering from epilepsy sometimes experience considerable restrictions regarding their daily lives. Current therapeutic options include, above all, pharmacotherapy, which can cause severe side effects and, especially in the case of temporal lobe epilepsy, often proves ineffective in controlling seizures. Furthermore, such seizure suppressive medications are not disease modifying. Epileptic tolerance is a state of temporarily increased resistance to epileptic seizures and can be achieved through preconditioning. However, the underlying mechanisms of epileptic tolerance have not yet been fully understood. MiRNAs are short, non-coding ribonucleic acids which, by controlling the translation of mRNA, have a decisive influence on protein synthesis. Changes in miRNA expression have been shown in epilepsy and epileptic tolerance-related research. In the present study, changes in miRNA expression in the hippocampus under four different conditions (preconditioning, injury, tolerance, control) were investigated using the perforant pathway stimulation model. For this purpose, test animals were stimulated following different paradigms. After sequencing the miRNAs, a total of 21 miRNAs were found to be significantly upregulated and 15 miRNAs downregulated, of which ten miRNAs were exclusively upregulated and six miRNAs were downregulated in tolerance. The processes associated with these miRNAs primarily concern the regulation of apoptosis, morphology and the formation of axons, dendrites and synapses. Based on these findings further research is needed to investigate the specific mechanisms through which those miRNAs lead to epileptic tolerance and to explore their therapeutic potential
Who Supports War for Justice and Why? Evidence from Russia and Ukraine
We study the acceptability of war as a necessary tool to obtain justice under certain conditions across individuals from Russia and Ukraine in 2011. We discuss which socio-economic, political and individual characteristics shape the support for using destructive military force to achieve justice. Overall, the acceptance of war for justice is relatively low in both countries. Using logistic regressions, we found that there are characteristics that significantly reduce the support for war for justice in both countries, such as gender and level of happiness. Support in both countries is also significantly larger among respondents who are interested in politics and are married. Additionally, there are conditions which produce different results between the countries, such as religiosity, country aims, employment, confidence in the government, concern over possible war and political orientation
Immunreaktion bei Integration bakterieller DNA im Gehirn der Maus
Die Integration artfremder Nukleinsäuren in eukaryotische Zellen findet in der Natur ebenso wie im Labor häufig statt, vor allem im Rahmen der Klonierung von Konstrukten im experimentellen Setting, aber auch bei viralen Infektionen mit Retroviren, die in der Lage sind, ihre RNA in das Genom der Wirtszelle zu integrieren. Das Einbringen bakterieller DNA in eukaryotisches Genom ist hingegen kaum beschrieben. Einzelne Untersuchungen zeigen, dass zum Beispiel die Aufnahme bakterieller DNA über den Darmtrakt mittels Ingestion und anschließender Integration für einen begrenzten Zeitraum auch ohne gezielte Transfektions- oder Transduktionsmechanismen im experimentellen Setting möglich ist.
Im Rahmen der Erschaffung der transgenen Mauslinie F2b, in welche ein GFAP-TIMP-1-Konstrukt eingebracht wurde, kam es zu einer akzidentiellen Ko-Integration bakterieller E.-coli-DNA gemeinsam mit dem transgenen Konstrukt. Da diese Mauslinie eine im Vergleich zu den ebenso TIMP-1-transgenen Mauslinen F11 und F60 eigene Pathologie mit ausgeprägter Entzündungsreaktion aufwies, sollte der Einfluss des bakteriellen Abschnitts untersucht werden. Aufgrund der Immunreaktion mit Hochregulation der Zytokine CCL3, CCL4, CXCL1, Il-1ß, Il-6, Il-7 und TNFα, die vor allem den Signalwegen der angeborenen Immunität mittels PRRs wie TLRs zuzuordnen sind, wurden PAMPs wie einzelsträngige RNA, dsRNA und freie DNA genauso wie bakterielle Proteine als mögliche antigene Strukturen und somit Auslöser der beobachteten Immunreaktion angesehen.
Zunächst sollte die eingebrachte Sequenz der bakteriellen DNA genau charakterisiert werden. Es sollte geklärt werden, welchen Abschnitt der E.-coli-DNA das eingebrachte Stück umfasst und in welchem Zusammenhang das unwissentlich integrierte Konstrukt mit dem gewünschten Transgen steht. Zudem war es Ziel der Arbeit herauszufinden, ob die bakterielle DNA in der F2b-Maus transkribiert und auch zu einem Protein translatiert wird. Zuletzt sollte in primären Astrozytenkulturen anhand exemplarisch vom E.-coli-Abschnitt ausgewählter Gene untersucht werden, ob und das dieser Gene zu einer Hochregulation der Zytokine führt, wie sie in der F2b-Maus beobachtet wurde.
Mittels iPCR und multipler Sequenzierungen konnte gezeigt werden, dass ein ca. 11.580 bp langer E.-coli-Abschnitt in die Maus-DNA integriert wurde. Dieser umfasst die Gene von yciT bis anteilig SapB und geht sowohl in 5’- als auch in 3’-Richtung in das ursprünglich für die Integration intendierte GFAP-TIMP-Konstrukt über. Durch überlappende PCRs mit aus RNA-Extrakten der F2b-Mäuse hergestellter cDNA war der bakterielle Abschnitt von SapC bis mod auf RNA-Ebene nachweisbar, der möglicherweise als zusammenhängender Strang transkribiert wird. Für zwei der vier ausgewählten Gene, fab1 und rnb, konnte in E.-coli-Kulturen eine ausreichende Proteinmenge hergestellt werden, um damit eine Immunisierung von Kaninchen zur Antikörperherstellung durchzuführen. Mit diesen aufgereinigten selbst hergestellten Antikörpern konnte die Antigene fab1 und rnb ebenso wie die nativen Proteine in E.-coli-Proteinextrakten detektiert werden. Sowohl fab1 als auch rnb konnten jedoch nicht als Proteine in den F2b-Extrakten nachgewiesen werden. Gleichermaßen zeigte sich auch keine Expression der gesuchten Proteine in den transduzierten Astrozytenkulturen. Die transduzierten primären Astrozytenkulturen wiesen bei der quantitativen Untersuchung auf Zytokine, welche in der F2b-Maus hochreguliert waren, keine signifikanten Unterschiede zu den nicht-transduzierten oder mit Leervektor transduzierten Astrozytenkulturen auf.
Die Abfolge der letztlich integrierten Konstrukte mit je einem GFAP-TIMP-1-Konstrukt an jedem Ende der E.-coli-Sequenz birgt mit dem vorhandenen Promotor die Möglichkeit, dass die bakterielle DNA sowohl in Sense- als auch in Antisense-Richtung abgelesen wird. Somit stehen bakterielle DNA, bakterielle einzelsträngige mRNA (möglicher-weise in Sense- und Antisense-Richtung) genauso wie ein langer zusammenhängender RNA-Strang oder aneinander gelagerte komplementäre RNA-Stränge (sense und anti-sense) als mögliche Antigene für das Auslösen der Immunreaktion zur Diskussion. Die Proteine konnten mittels der selbst hergestellten Antikörper gegen fab1 und rnb nicht als Antigene detektiert werden. Dies könnte daran liegen, dass diese entweder nicht translatiert werden oder nur in Mengen unterhalb der Nachweisgrenze oder in anderer Form – zum Beispiel posttranslationale Modifikationen – für den Antikörper undetektierbar vorliegen. Die Ergebnisse der qPCRs deuten darauf hin, dass die in Sense-Richtung untersuchten Gene nicht für die Immunreaktion der F2b-Maus verantwortlich sind, da keine Hochregulation der entsprechenden Zytokine beobachtet werden konnte.
Die Ergebnisse dieser Arbeit weisen auf ein möglicherweise im Laboralltag auftretendes methodisches Problem bei der Erzeugung transgener Tiere hin. Da die Ko-Integration sehr wahrscheinlich auf einer Verunreinigung und versehentlichen Exzision gleichlanger Banden in der Gelextraktion beruht, ist nicht auszuschließen, dass solche Fehlintegrationen häufiger vonstattengehen und Auswirkungen auf die Untersuchungen transgener Mauslinien haben, ohne wissentlich berücksichtigt zu werden. Weiterhin weist die Mauslinie F2b eine Autoimmunreaktion auf, welche in dieser Arbeit in einem ersten Schritt näher eingegrenzt wurde. Weitere Untersuchungen sowohl mit Sense- als auch mit Antisense-Produkten der ausgewählten Gene geben Hinweise darauf, dass ein Antisense-Produkt des Gens SapC eine Zytokinhochregulation triggern könnte. In diesem Zusammenhang geben die Ergebnisse dieser Arbeit Anstoß für neue Untersuchungen zur Entstehung autoimmuner Prozesse und Erkrankungen in Zusammenhang mit dem Kontakt und Vorhandensein bakterieller Antigene.The integration of foreign nucleic acids into eukaryotic cells occurs frequently in nature as well as in the laboratory, especially in the context of cloning constructs in experimental settings, but also in viral infections with retroviruses capable of integrating their RNA into the host cell genome. The introduction of bacterial DNA into the eukaryotic genome, on the other hand, is scarcely described. Individual studies show, for example, that the uptake of bacterial DNA through the intestinal tract via ingestion and subsequent integration is possible for a limited period even without targeted transfection or transduction mechanisms in experimental settings.
In the context of creating the transgenic mouse line F2b, into which a GFAP-TIMP-1 construct was introduced, accidental co-integration of bacterial E. coli DNA occurred together with the transgenic construct. Since this mouse line exhibited its own pathology compared to the similar TIMP-1-transgenic mouse lines F11 and F60, with a pronounced inflammatory reaction, the influence of the bacterial segment was to be investigated. Due to the immune reaction with upregulation of cytokines CCL3, CCL4, CXCL1, Il-1ß, Il-6, Il-7, and TNFα, which are primarily associated with innate immunity pathways through PRRs such as TLRs, PAMPs such as single-stranded RNA, dsRNA, and free DNA as well as bacterial proteins were possible antigenic structures triggering the observed immune reaction.
First, the exact integrated sequence of the bacterial DNA should be characterized. This included determining which portion of the E. coli DNA the integrated fragment encompassed, as well as the relationship between the unintentionally integrated construct and the intentionally integrated transgene. Additionally, the aim of this dissertation project was to determine whether the bacterial DNA in the F2b mouse is transcribed and translated into a protein. Finally, primary astrocyte cultures were used to investigate whether and which gene present in the E. coli segment leads to upregulation of cytokines as observed in the F2b mouse.
Results: Using iPCR and sequencing, it was shown that an approximately 11.580bp long E. coli segment was integrated into mouse DNA. This included the genes from yciT to partially SapB and extended into both 5’ and 3’ directions into the originally intended GFAP-TIMP construct. By overlapping PCRs on cDNA from RNA extracts of F2b mice, the bacterial segment from SapC to mod was detected at the RNA level, possibly transcribed as a contiguous strand.
For two of the four selected genes, fab1 and rnb, sufficient protein amounts were produced in E. coli cultures to immunize rabbits for antibody production. With purified self-made antibodies, the antigens fab1 and rnb, as well as the native protein in E. coli protein extracts, could be detected. However, neither fab1 nor rnb could be detected as proteins in the F2b extracts. Similarly, no expression of the desired proteins was observed in the transduced astrocyte cultures. The transduced primary astrocyte cultures showed no significant differences in the quantitative examination of cytokines, which were upregulated in the F2b mouse, compared to untransduced or empty-vector-transduced astrocyte cultures.
The sequence of the ultimately integrated constructs, with a GFAP-TIMP-1 construct containing the GFAP promoter at each end of the E. coli sequence, provides the possibility that bacterial DNA is transcribed in both sense and antisense direction. Thus, possible antigens triggering the immune reaction include bacterial DNA, bacterial single-stranded mRNA, possibly in sense and antisense directions, as well as a long contiguous RNA strand or concatenated complementary RNA strands (sense and antisense). The proteins could not be detected as antigens by the self-synthesized antibodies for fab1 and rnb, indicating that they are either not translated or are present below the detection limit or undetected by the antibodies due to post-translational modifications. The qPCR results suggest that the genes investigated in the sense direction are not responsible for the immune reaction in the F2b mouse, as no upregulation of the corresponding cytokines was observed.
The results of this work point to a possible methodological problem encountered in the generation of transgenic animals in everyday laboratory practice. Since co-integration most likely results from contamination and accidental excision of equally long bands in gel extraction, it cannot be ruled out that such misintegrations occur more frequently and have effects on the investigations of transgenic mouse lines without being consciously considered. Furthermore, the F2b mouse line describes an autoimmune reaction, which was narrowed down in this work in a first step. Further investigations on both sense and antisense products of the selected genes suggest that an antisense product of the SapC gene could trigger cytokine upregulation. In this context, the investigations of this work provide impetus for new investigations into the development of autoimmune processes and diseases in relation to the contact and presence of bacterial antigens
Identification of Dysregulated microRNAs in Glioblastoma Stem-like Cells
Glioblastoma multiforme (GBM) is the most common malignant primary brain tumor in adults. Despite multimodal therapy, median survival is poor at 12–15 months. At the molecular level, radio-/chemoresistance and resulting tumor progression are attributed to a small fraction of tumor cells, termed glioblastoma stem-like cells (GSCs). These CD133-expressing, self-renewing cells display the properties of multi-lineage differentiation, resulting in the heterogenous composition of GBM. MicroRNAs (miRNAs) as regulators of gene expression at the post-transcriptional level can alter many pathways pivotal to cancer stem cell fate. This study explored changes in the miRNA expression profiles in patient-derived GSCs altered on differentiation into glial fiber acid protein (GFAP)-expressing, astrocytic tumor cells using a polymerase chain reaction (PCR) array. Initially, 22 miRNAs showed higher expression in GSCs and 9 miRNAs in differentiated cells. The two most downregulated miRNAs in differentiated GSCs were miR-17-5p and miR-425-5p, whilst the most upregulated miRNAs were miR-223-3p and let-7-5p. Among those, miR-425-5p showed the highest consistency in an upregulation in all three GSCs. By transfection of a 425-5p miRNA mimic, we demonstrated downregulation of the GFAP protein in differentiated patient-derived GBM cells, providing potential evidence for direct regulation of miRNAs in the GSC/GBM cell transition.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg
