When Does the Introduction of a New Currency Improve Welfare?
In recent years, cryptocurrencies such as Bitcoin have emerged, in upcoming
years, corporate currencies such as Libra (Diem) and central bank digital currencies will emerge even in low-inflation developed economies. Veränderungen der Schlafarchitektur bei Epilepsiepatienten - Eine klinische Studie
Epilepsie ist mit über 70 Millionen Betroffenen eine der häufigsten chronischneurologischen
Erkrankungen weltweit. Eine der häufigsten Komorbiditäten der
Epilepsie sind Schlafstörungen. Studien, die sowohl subjektive als auch objektive
Schlafparameter untersuchten, zeigten bei den Epilepsiepatienten Veränderungen des
Schlafes. Schlafparameter wie die Schlafeffizienz oder die verschiedenen Schlafphasen
waren in früheren Studien verändert.
Bisher gab es nur wenige, kleine Studien zu dieser Thematik. Sie verglichen
Epilepsiepatienten mit gesunden Kontrollen. Ziel der vorliegenden Studie war es, die
Schlafarchitektur von Epilepsiepatienten mit Kontrollpatienten zu vergleichen, die an
dissoziativen nicht-epileptischen Anfällen leiden. Die daraus resultierenden Hypothesen
für diese Studie lauteten: 1. Die Anteile der einzelnen Schlafstadien unterscheiden sich
bei Epilepsiepatienten und der Kontrollgruppe signifikant und 2. Epilepsiepatienten
zeigen eine schlechtere Schlafeffizienz als die Patienten der Kontrollgruppe.
Um die Fragestellungen zu beantworten, untersuchten wir in dieser Studie 54 Patienten
mit insgesamt 79 PSGs. Mittels Video-EEG-Monitoring und Polysomnographie wurde
der Schlaf der 42 Patienten mit epileptischen Anfällen und der 12 Patienten mit
dissoziativen Anfällen analysiert. Dabei wurden die fünf Hauptparameter Schlafeffizienz,
Anteil Schlafstadium 1 (N1), Anteil Schlafstadium 2 (N2), Anteil Schlafstadium 3 (N3)
und Anteil Schlafstadium REM mittels Welsch-Test untersucht. Die Analyse der
Nebenparameter Zeit im Bett (TIB), Gesamtschlafzeit (TST), Einschlaflatenz, REMLatenz,
Wachzeit nach Einschlafen, Arousal-Index und Respiratory Disturbance Index
(RDI) wurde explorativ vorgenommen.
Bei den Epilepsiepatienten zeigte sich ein signifikanter Unterschied im Anteil des
Schlafstadiums 2, 3, REM, in der TIB und im Arousal-Index. Das Schlafstadium 2 war
hierbei in der Epilepsiegruppe signifikant geringer als bei den Kontrollen (p = 0,014),
genau wie die TIB (p = 0,027) und Arousal-Index (p = 0,049). Der Anteil vom
Schlafstadium 3 war bei den Epilepsiepatienten signifikant höher als bei den
Kontrollprobanden (p = 0,022), so auch der des REM-Stadiums (p = 0,013).
Für die übrigen Haupt- und Nebenparameter ergaben sich nicht-signifikante
Unterschiede.
Die Erkenntnisse aus vorherigen Studien konnten in der vorliegenden Untersuchung
teilweise bestätigt werden. Es ist eine ausgeprägte Heterogenität der Schlafveränderungen
bei Epilepsiepatienten in den verschiedenen Studien zu beobachten. Jedoch kommen alle
Ergebnisse zu dem Schluss, dass die Schlafarchitektur bei Epilepsie verändert ist.
Ein nächster Ansatzpunkt für weitere Studien könnte ein Vergleich der Schlafarchitektur
von Epilepsiepatienten mit gesunden Kontrollen und Patienten mit dissoziativen Anfällen
sein. Im Zuge dessen könnte die Frage beantwortet werden, ob die epileptische Aktivität
oder die Anfälle an sich eine größere Auswirkung auf den Schlaf der Patienten haben.With more than 70 million patients, epilepsy is one of the most common chronic
neurological diseases worldwide. One of the most common comorbidities of epilepsy are
sleep disturbances. Studies that examined both subjective and objective sleep parameters
showed changes in sleep in epilepsy patients. Sleep parameters such as sleep efficiency
or the different sleep phases were altered in earlier studies.
So far, there have only been a few small studies on this topic. They compared epilepsy
patients with healthy controls. The aim of the present study was to compare the sleep
architecture of epilepsy patients with control patients suffering from dissociative nonepileptic
seizures. The resulting hypotheses for this study were: 1. The proportions of the
individual sleep stages differ significantly in epilepsy and control patients, and 2.
Epilepsy patients show a poorer sleep efficiency than the patients in the control group.
The study included 54 patients and a total of 79 PSGs. The sleep of 42 patients with
epileptic seizures and 12 patients with dissociative seizures was analysed using video
EEG monitoring and polysomnography. The five main parameters sleep efficiency,
proportion of sleep stage 1 (N1), proportion of sleep stage 2 (N2), proportion of sleep
stage 3 (N3) and proportion of sleep stage REM were examined using the Welch test. The
analysis of the side-parameters time in bed (TIB), total sleep time (TST), sleep onset
latency, REM latency, wake time after sleep onset, arousal index and respiratory
disturbance index (RDI) was carried out exploratively.
In the epilepsy patients, there was a significant difference in the proportion of sleep stages
2, 3, REM, in the TIB and the arousal index. The sleep stage 2 was significantly lower in
the epilepsy group than in the controls (p = 0.014), as were the TIB (p = 0.027) and arousal
index (p = 0.049). The proportion of sleep stage 3 was significantly higher in the epilepsy
patients than in the control subjects (p = 0.022), as well as the proportion of REM stage
(p = 0.013).
There were non-significant differences for the other main and side-parameters. The
findings from previous studies were partially confirmed in the present study.
A pronounced heterogeneity of the sleep changes in epilepsy patients can be observed in
the different studies. However, all studies come to the conclusion that sleep architecture
is altered in patients with epilepsy.
A next starting point for further studies could be a comparison of the sleep architecture
of epilepsy patients with healthy controls and patients with dissociative seizures. This
could answer the question whether the epileptic activity or the seizures themselves have
a greater impact on patients' sleep
Bridging the Gap - how Effective are Remedial Math Courses in Germany?
Almost all German universities complain that the math skills of
students entering the higher education system do not equal the math
skills that are demanded by the universities. To bridge the gap, almost
all universities offer some remedial math courses. However, there is
only weak empirical evidence for the effectiveness of these courses in
Germany. This paper aims to fill this gap by evaluating some math
bridging courses given at the Economics Department of the University
of Kassel. A key finding: taking a math bridging course on a regular
basis will enhance students' math skills and increase the probability of
passing the final math exam by 35 percent
Heterodisulfid-Reduktase-Megakomplexe, die in einem CO2-reduzierenden Methanogen der Klasse I unter Ni-suffizienten und Ni-limitierten Bedingungen gebildet werden
Methanogenic archaea are the major producers of the greenhouse gas methane on Earth. In the methanogenic pathway from CO2 and H2, low-potential electrons for CO2 reduction are formed by a flavin-based electron bifurcation (FBEB) reaction catalyzed by heterodisulfide reductase (Hdr) in complex with Hdr-associating [NiFe] hydrogenase (Mvh). In addition, F420-reducing [NiFe]-hydrogenase (Frh) provides electrons to the methanogenic pathway via an electron carrier F420. It is known that under moderately nickel-limited conditions (Ni2+ ~200 nM), the production of Frh in Methanothermobacter marburgensis is strictly down regulated and the catalytic function is replaced by a coupled reaction of Hmd ([Fe]-hydrogenase) and F420-dependent methylene-tetrahadromethanopterin (methylene-H4MPT) dehydrogenase (Mtd). Proteomic analysis of M. marburgensis in this study indicated that under strictly nickel-limited conditions (Ni2+ < 50 nM), close to the natural environment, Mvh is also not produced, and the production of uncharacterized proteins encoded in a gene cluster are induced [Chapter 2]. We refer to these proteins as F420-dependent electron-donating proteins (ElpABC) and hypothesize that these proteins form a complex with Hdr to donate electrons to the FBEB reaction in this methanogen under strictly nickel-limited conditions. Indeed, the purified Hdr complex contains ElpABC and catalyzed the F420 dependent Hdr reaction. Thus, under strictly nickel-limited conditions often found in nature, Hmd provides all electrons for the reducing metabolism instead of the cytosolic [NiFe]-hydrogenases (Frh and Mvh). Conservation of the elp and hmd genes in the genome of hydrogenotrophic methanogens suggests that this nickel-dependent transition in electron flow occurs in nature. Unexpectedly, the Elp-Hdr complex was purified as a megacomplex with formyl-methanofuran dehydrogenase (Fmd). Cryo-electron microscopy (cryo-EM) revealed the structure of the Elp-Hdr, confirming the electronic interaction between Elp and Hdr although the Fmd module of this megacomplex was dissociated due to the instability of the protein complex. The finding of Elp-Hdr-Fmd megacomplex prompted us to purify the Mvh-Hdr-Fmd complex from M. marburgensis cultivated under nickel-sufficient standard laboratory condition. We successfully purified the Mvh-Hdr-Fmd megacomplex, which was more stable than the Elp-Hdr-Fmd complex. The purified megacomplex catalyzed the H2-dependent electron bifurcation reduction of [13C]-CO2 to [13C]-formyl-methanofuran in the absence of free ferredoxin [Chapter 3]. This result demonstrated that the subunits in the Mvh-Hdr-Fmd megacomplex are electronically linked to transfer electrons for the reduction of CO2. This result also supports the physiological function of the Elp-Hdr-Fmd complex in CO2 reduction using electrons from the reduced form of F420. Most recently, we elucidated the cryo-EM structure of the Mvh-Hdr-Fmd complex from M. marburgensis (unpublished results). The topology of the asymmetric Mvh-Hdr-Fmd complex is distinct from the previously published symmetric Fdh-Hdr-Fmd complex from Methanospirillum hungatei. In the asymmetric Mvh-Hdr-Fmd structure, two FmdABCDFGH dimers form an hourglass-like tetramer, which associates with the Mvh-Hdr dimer. The electron output domain of HdrA and electron input domain of FmdF is connected by an extra polyferredoxin MvhB, which contains twelve [4Fe-4S]-clusters.Methanogene Archaeen sind die Hauptproduzenten des Treibhausgases Methan auf der Erde. Während des methanogenen Wegs aus CO2 und H2 werden Elektronen mit niedrigem Potenzial für die CO2-Reduktion gebildet. Dies wird durch eine flavinbasierte Elektronenverzweigungsreaktion (FBEB), die von Heterodisulfid-Reduktase (Hdr) im Komplex mit Hdr-assoziierender [NiFe]-Hydrogenase (Mvh) katalysiert wird, ermöglicht. Darüber hinaus stellt die F420-reduzierende [NiFe]-Hydrogenase (Frh) über den Elektronenträger F420 Elektronen für den methanogenen Weg zur Verfügung. Es ist bekannt, dass unter moderater Nickellimitierung (Ni2+ ~200 nM) die Produktion von Frh in Methanothermobacter marburgensis streng herunterreguliert und die katalytische Funktion durch eine gekoppelte Reaktion von Hmd ([Fe]-Hydrogenase) und F420-abhängiger Methylen-Tetrahadromethanopterin (Methylen-H4MPT)-Dehydrogenase (Mtd) ersetzt wird. Die Proteomanalyse von M. marburgensis in dieser Studie ergab, dass unter strenger Limitierung der Nickelmenge (Ni2+ < 50 nM), welcher der natürlichen Umgebung nahe kommen, auch Mvh nicht produziert wird und die Produktion unbekannter, in einem Gencluster kodierter Proteine induziert wird [Kapitel 2]. Wir bezeichnen diese Proteine als F420-abhängige elektronendonierende Proteine (ElpABC) und stellen die Hypothese auf, dass diese Proteine, unter Bildung eines Komplexes mit Hdr und strikter Limitierung der Nickelmenge, Elektronen für die FBEB-Reaktion in diesem Methanogen spenden. In der Tat enthält der gereinigte Hdr-Komplex ElpABC und zeigte die F420-abhängige Hdr-Reaktion. Unter streng nickelbegrenzten Bedingungen, wie sie in der Natur häufig vorkommen, liefert Hmd, anstelle der cytosolischen [NiFe]-Hydrogenasen (Frh und Mvh), alle Elektronen für den reduzierenden Stoffwechsel. Die Konservierung der elp- und hmd-Gene im Genom von hydrogenotrophen Methanogenen deutet darauf hin, dass dieser nickelabhängige Übergang im Elektronenfluss ebenfalls in der Natur stattfindet. Unerwarteterweise wurde der Elp-Hdr-Komplex nach der Aufreinigung als Megakomplex mit Formyl-Methanofuran-Dehydrogenase (Fmd) erhalten. Die Analyse der Struktur von Elp-Hdr, welche unter Verwendung von Cryo-Elektronenmikroskopie (Cryo-EM) erhalten wurde, bestätigte die elektronische Wechselwirkung zwischen Elp und Hdr, obwohl das Fmd-Modul dieses Megakomplexes aufgrund der Instabilität des Proteinkomplexes dissoziierte. Die Entdeckung des Elp-Hdr-Fmd-Megakomplexes veranlasste uns, den Mvh-Hdr-Fmd-Komplex aus M. marburgensis, der unter nickelhaltigen Standard-Laborbedingungen kultiviert wurde, zu reinigen. Die Aufreinigung des Mvh-Hdr-Fmd-Megakomplexes, welcher eine höhere Stabilität als der the Elp-Hdr-Fmd.Komplex aufweiste, konnte erfolgreich durchgeführt werden. Der gereinigte Megakomplex katalysierte die H2-abhängige Elektronen-Bifurkationsreduktion von [13C]-CO2 zu [13C]-Formyl-Methanofuran in Abwesenheit von freiem Ferredoxin [Kapitel 3]. Dieses Ergebnis zeigt, dass die Untereinheiten im Megakomplex Mvh-Hdr-Fmd, welche für die Übertragung von Elektronen für die Reduktion von CO2 zuständig sind, elektronisch verbunden sind und stützt auch das Wissen über die physiologische Funktion des Elp-Hdr-Fmd-Komplexes bei der CO2-Reduktion unter Verwendung von Elektronen aus der reduzierten Form von F420. Vor Kurzem gelang es uns die Cryo-EM-Struktur des Mvh-Hdr-Fmd-Komplexes aus M. marburgensis aufzuklären (unveröffentlichte Ergebnisse). Die Topologie des asymmetrischen Mvh-Hdr-Fmd-Komplexes unterscheidet sich von dem zuvor veröffentlichten symmetrischen Fdh-Hdr-Fmd-Komplex aus Methanospirillum hungatei. In der asymmetrischen Mvh-Hdr-Fmd-Struktur bilden zwei FmdABCDFGH-Dimere ein sanduhrartiges Tetramer, das mit dem Mvh-Hdr-Dimer assoziiert ist. Influence of PACAP or PAC1 deficiency on the morphology and expression of inflammation/apoptosis/atherosclerosis-related proteins/cytokines in Mm. soleus/gastrocnemius in hypercholesterolemic ApoE-/- mice under regular feed.
Das Hypophysen-Adenylat-Cyclase-aktivierende-Polypeptid (engl. „Pituitary Adenylate Cyclase-Activating Polypeptide“; PACAP) ist ein 38 AS großes Neuropeptid, das über die Bindung an verschiedene Rezeptoren vor allem dem PAC1-Rezeptor wirkt und seine Zielfunktionen über die Aktivierung einer Adenylat-cyclase (AC) ausführt.
Im Zusammenhang mit Atherosklerose konnte gezeigt werden, dass PACAP bei glatten Muskelzellen die Bildung von Lipidperoxiden und die morphologischen Veränderungen, die durch Hyperlipidämie (ApoE-/-) verursacht wurden, signifikant verminderte. Atherosklerose spielt eine wichtige pathophysiologische Rolle bei einigen relevanten Erkrankungen. Im Bereich der Skelettmuskulatur, kann sie zu erhöhtem oxidativem Stress, Muskelfaserdegeneration, Fibrose sowie einer verringerten Kapillarisierung der Muskelfasern führen. In dieser Arbeit wurde der Einfluss von PACAP- bzw. PAC1-Defizienz auf die Morphologie und Expression Inflammations- / Apoptose- / Atherosklerose-relevanter Proteine/ Zytokine in Mm. soleus/gastrocnemius bei hypercholesterinämischen ApoE-/--Mäusen unter Standardfütterung analysiert. Hierfür wurden folgende Maus-Genotypen generiert und untersucht: 1. PACAP+/+ / PAC1+/+ / ApoE-/- (kurz „ApoE-/-“; Kontrolle); 2. PACAP-/- / ApoE-/-; 3. PAC1-/- / ApoE-/-. Alle Mäuse erhielten für 30 Wochen Standardfutter. Zur Messung der Triglycerid- und Cholesterinkonzentrationen wurden Blutproben entnommen. Die weiteren o.g. Parameter wurden überwiegend mittels Immunhistomorphometrie untersucht. Das Körpergewicht der PACAP-/- / ApoE-/-- und PAC1-/- / ApoE-/-- Mäuse war ähnlich zu dem der ApoE-/--Mäuse. Die Plasma-Cholesterin- und -Triglyceridspiegel der PACAP-/- / ApoE-/-- bzw. PAC1-/- / ApoE-/--Mäuse waren ähnlich wie bei ApoE-/--Mäusen. Im M. gastrocnemius fanden wir bei PAC1-/- / ApoE-/--Mäusen eine signifikante Erhöhung der Faserdichte um 28,5% (p=0,042) bei gleichzeitiger Abnahme der Faserquerschnittsfläche um 22,9% (p=0,043) im Vergleich zu ApoE-/-. Im M. soleus der PAC1-/- / ApoE-/--Mäuse war die Faserdichte um 19% (p=0,045) höher als bei ApoE-/--Mäusen. Die Faserquerschnittsflächen aller Fasertypen waren bei PACAP-/- /ApoE-/- -bzw. PAC1-/- / ApoE-/--Mäusen im M.soleus um 15 % (p=0,016) bzw. 17,2%

(p<0,001) erniedrigt im Vergleich zu den ApoE-/--Mäusen. Die Faserquerschnittsflächen der einzelnen Fasertypen waren bis auf die der Typ I Fasern signifikant verringert. Zudem zeigte sich im M. soleus der PACAP-/- / ApoE-/--Mäuse eine Verschiebung der Faserkomposition mit 5%iger Abnahme der Typ I Fasern und 5,9%iger Zunahme der IIxa-Fasern im Vergleich zu ApoE-/--Mäusen. Die Dichte der MuRF-1+-Zellen war im M. soleus der PAC1-/- / ApoE-/--Mäuse signifikant um 30,7% (p=0,018) höher als bei ApoE-/--Mäusen. Die für die Innervation bedeutenden neuromuskulären Endplatten waren im M. soleus bei PACAP-/- / ApoE-/-- als auch bei PAC1-/- / ApoE-/--Mäusen signifikant um 33,3% reduziert im Vergleich zu ApoE-/--Mäusen.
Unsere Daten zeigen, dass bei PAC1-/- / ApoE-/--Mäusen im Vergleich zu ApoE-/--Mäusen die Dichte der COX-2+-Zellen im M.soleus um 18,3% (p=0,030) erniedrigt war. Auch die Anzahl COX-2+-Zellen pro Faser war bei den PAC1-/- / ApoE-/--Mäusen im Vergleich zu ApoE-/--Mäusen um 33,2% bzw. im M. gastrocnemius um 38% erniedrigt. Unsere Daten zeigen, dass bei PACAP-/- / ApoE-/--Mäusen die Dichte der IL-1ß+-Zellen im M. soleus und M. gastrocnemius um 24,6% (p=0,047) bzw. 19,6% (p=0,050) niedriger war im Vergleich zu ApoE-/-. Auch die Anzahl der IL-1ß+-Zellen pro Faser waren bei beiden Muskeltypen signifikant reduziert im Vergleich zu ApoE-/-. Die Kapillardichte war im M. soleus bei PACAP-/- / ApoE-/--Mäusen signifikant (p=0,045) um 30,4% höher als bei ApoE-/--Mäusen. Ferner fanden wir im M. gastrocnemius von PACAP-/- / ApoE-/-- und PAC1-/- / ApoE-/--Mäusen signifikant höhere Konzentrationen der proteinogenen Aminosäuren Methionin, Leucin, Tyrosin und Phenylalanin bezogen auf das Gesamtprotein (nmol/mg Protein). Der intrazelluläre Gluthation-Redoxstatus der PACAP-/- / ApoE-/-- und PAC1-/- / ApoE-/--Mäusen zeigte keine signifikanten Veränderungen im Vergleich zu ApoE-/--Mäusen.
Unsere Ergebnisse zeigen erstmals, dass eine PACAP-/PAC1-Defizienz zu einer Abnahme der Faserquerschnittsfläche bei gleichzeitiger Zunahme der Faserdichte führt, was auf eine mögliche Atrophie mit begleitender Hyperplasie hindeutet. Da wir bei PAC1-/- / ApoE-/--Mäusen eine signifikant erhöhte MuRF-1-Expression im M. soleus beobachteten, könnte dies eine

Ursache für atrophische Prozesse sein. Die durch PACAP bzw. PAC1-Defizienz induzierte Abnahme der Innervation könnte zudem für die Abnahme der Faserquerschnittsfläche/Zunahme der Faserdichte verantwortlich sein. Eine verringerte Durchblutung als eine weitere mögliche Ursache der Atrophie im M. soleus konnte in unseren Untersuchungen nicht nachgewiesen werden, da sich unter PACAP-/PAC1-Mangel eher eine Zunahme der Kapillardichte zeigte. Erstaunlicherweise war im Gegensatz zur Herzmuskulatur (Gohlke, 2017) in der Skelettmuskulatur kein begleitendes erhöhtes pro-inflammatorisches Environment beobachtet worden. Atrophische Prozesse erfolgten begleitet von einer reduzierten Dichte an Entzündungsparametern (COX-2; Il-1β). Die erhöhten Konzentrationen an proteinogenen Aminosäuren im M. gastrocnemius deuten auf vermehrte Proteolyseprozesse hin, welche in Verbindung mit der verringerten Faserquerschnittsfläche bei PAC1-/- / ApoE-/--Mäusen stehen könnten. Hier könnte der Untersuchungszeitpunkt im M. soleus zu früh gewählt gewesen sein, sodass sich hier noch keine Veränderungen in der Aminosäureanalyse zeigten.
Summa summarum bestätigt sich unsere Annahme zu Beginn der Arbeit, dass ein PACAP-/PAC1-Mangel in der Skelettmuskulatur pathologische Veränderungen im Bezug auf Atrophie, Kapillarisierung und Inflammation zur Folge hat.
Aus unseren Daten deuten wir, dass PACAP in der Skelettmuskulatur eher protektive Eigenschaften hinsichtlich Atrophie, Innervation und Proteolyse innerhalb der Zellen zu haben scheint; hier wäre es künftig spannend, Studien mit PACAP- und PAC1-Agonisten durchzuführen. Womöglich eignen sich diese zur Behandlung und Regeneration von nervalen Muskelatrophien. Die unter PACAP-Mangel beobachtete Hyperplasie deutet daraufhin, dass PACAP- und PAC1-Agonisten auch in der Neoplasieforschung eine nicht unwichtige Rolle spielen könnten.The Pituitary Adenylate Cyclase-activating Polypeptide (PACAP) is a 38 amino acid neuropeptide that acts by binding to various receptors, most notably the PAC1 receptor, and performs its target functions via activation of an adenylate cyclase (AC). In the context of atherosclerosis, PACAP was shown to significantly decrease lipid peroxide formation and morphological changes induced by hyperlipidemia (ApoE-/-) in smooth muscle cells. Atherosclerosis plays an important pathophysiological role in several important diseases. In skeletal muscles, it can lead to increased oxidative stress, muscle fiber degeneration, fibrosis, as well as decreased capillarization of muscle fibers. This paper analyses the influence of PACAP or PAC1 deficiency on the morphology and expression of inflammation/apoptosis/atherosclerosis-related proteins/cytokines in Mm. soleus/gastrocnemius in hypercholesterolemic ApoE-/- mice under regular feed. For this purpose, the following genotypes were generated and studied: 1. PACAP+/+ / PAC1+/+ / ApoE-/--mice (short as "ApoE-/-"; control); 2. PACAP-/- / ApoE-/--mice; 3. PAC1-/- / ApoE-/--mice. All mice received regular feed for 30 weeks. Blood samples were taken from the animals for the measurement of triglyceride and cholesterol concentrations. The other parameters mentioned above were mainly examined by immunohistomorphometry. The body-weights of the PACAP-/PAC1-deficient mice were similar to those of the ApoE-/--mice. Plasma cholesterol- and triglyceride levels of PACAP-/PAC1-deficient mice were similar to those of ApoE-/- mice. In the gastrocnemius muscle, we found a significant increase in fiber density of 28.5% (p=0.042) with a concomitant decrease in fiber cross-sectional area of 22.9% (p=0.043) in PAC1-/- / ApoE-/- mice compared to ApoE-/-. In the soleus muscle of PAC1-/- / ApoE-/--mice fiber density was 19% (p=0.045) higher than in ApoE-/- mice. The fiber cross-sectional areas of all fibertypes were significantly decreased 15% and 17,2% in PACAP-/- / ApoE-/- as well as PAC1-/- / ApoE-/-- mice compared to ApoE-/- mice. The fiber cross-sectional areas of each fiber type were significantly decreased except for those of type I fibers. In addition, the soleus muscle of PACAP-/- / ApoE-/--mice showed a shift in fiber composition with 5% decrease in type I fibers and 5.9% increase in IIxa fibers compared

to ApoE-/--mice. The density of MuRF-1+ cells was significantly higher by 30.7% (p=0.018) in soleus muscle of PAC1-/- / ApoE-/--mice than ApoE-/--mice. Neuromuscular junctions, which are important for innervation, were significantly reduced by 33.3% in soleus muscle of PACAP-/- / ApoE-/- as well as PAC1-/- / ApoE-/-- mice.
Our data show that in PAC1-/- / ApoE-/--mice, the density of COX-2+ cells in the soleus muscle was decreased by 18.3% (p=0.030) compared to ApoE-/- mice. The number of COX-2+ cells per fiber was also decreased by 33.2% in the soleus muscle respectively by 38% in the gastrocnemius muscle compared to ApoE-/- mice. IL-1β+ cells were significantly reduced in density and number/fiber in both muscle types and in PACAP- and PAC1-deficient mice compared with ApoE-/--mice. Our data show that in PACAP-/- / ApoE-/--mice, the density of IL-1ß+ cells was lower in soleus and gastrocnemius muscle by 24.6% (p=0.047) respectively by 19.6% (p=0.050) compared to ApoE-/-. The number of IL-1ß+ cells per fiber were also significantly reduced in both muscle types compared to ApoE-/- mice. We found a 30.4% (p=0.045) higher Capillary density in soleus muscles of PACAP-/- / ApoE-/--mice compared to ApoE-/--mice. Furthermore, we found significantly higher concentration of proteinogenic amino acids methionine, leucine, tyrosine and phenylalanine relative to total protein (nmol/mg protein) in the gastrocnemius muscle of PACAP-/- / ApoE-/-- and PAC1-/- / ApoE-/--mice. No significant changes in intracellular gluthation redox status were observed under PACAP or PAC1 deficiency compared to ApoE-/- mice.
Our results show for the first time that PACAP-/PAC1-deficiency leads to a decrease in fiber cross-sectional area with a concomitant increase in fiber density, suggesting possible atrophy with accompanying hyperplasia. We observed markedly increased MuRF-1 expression in the soleus muscle in PAC1-/- / ApoE-/--mice, this could be a cause of the atrophic processes. Moreover, the decrease in innervation induced by PACAP or PAC1 deficiency could be responsible for the decrease in fiber cross-sectional area/increase in fiber density. Decreased blood flow as another possible cause of atrophy in soleus muscle could not be demonstrated in our studies, because PACAP/PAC1 deficiency rather showed an increase in capillary

density. Surprisingly, no concomitant increased pro-inflammatory environment was observed in skeletal muscle compared to cardiac muscle (Gohlke, 2017). The atrophic processes were accompanied by a reduced density of inflammatory parameters (COX-2; Il-1β). The increased concentrations of proteinogenic amino acids in the gastrocnemius muscle indicate increased proteolysis processes, which could be related to the reduced fiber cross-sectional area in PAC1-/- / ApoE-/--mice. In this case, the time of investigation in soleus muscle might have been too early, so that no change was observed yet.
In summary, our assumption at the beginning of this work that PACAP-/PAC1 deficiency in skeletal muscle results in pathological changes in terms of atrophy, capillarization and inflammation is confirmed.
From our data, we suggest that in skeletal muscle PACAP seems to have rather protective properties in terms of atrophy, innervation, and proteolysis within cells; here, it would be exciting to perform further studies with PACAP and PAC1 agonists. It is possible that these may be suitable for the treatment and regeneration of nervous muscle atrophies. The hyperplasia observed under PACAP deficiency suggests that PACAP and PAC1 agonists may also play a not insignificant role in neoplasia research

We uncover an inhibitory role for IgA in mast cell degranulation. We conclude that IgA serves as a regulator of mast cell degranulation, including anaphylaxis, providing a new therapeutic potential for IgE-mediated allergic disease. Additionally, through the development of mice deficient in IgA but with an intact IgE locus, we have been able to investigate the consequences of IgA deficiency on immune equilibrium and systemic antibody responses in the context of allergic conditions. We conclude that the role of IgA in maintaining the gut microbiome plays a crucial role in regulating adaptive immune responses, particularly in the induction of IgE antibodies.Wir enthüllen eine hemmende Rolle für IgA bei der Mastzellendegranulation. New Ecological Paradigm Meets Behavioral Economics: On the Relationship between Environmental Values and Economic Preferences
This paper empirically examines whether environmental values are correlated with eco-nomic preferences from behavioral economics and considers possible consequences when independence is assumed. The data for this analysis stem from a large-scale com-puter-based survey among more than 3700 German citizens. Our indicators for environ-mental values are based on the New Ecological Paradigm (NEP), which is a standard instrument in social and behavioral sciences and increasingly common in economic stud-ies. The econometric analysis with Generalized Poisson regression models reveals strong correlations between two NEP scales and several economic preferences, which are based on established experimental measures: While social preferences (measured in an incen-tivized dictator game) and positive reciprocity are significantly positively correlated, trust and (less robust) negative reciprocity are significantly negatively correlated with the NEP scales, respectively. Only risk and time preferences (also measured in an incentiv-ized experiment) are not robustly significantly correlated with the NEP scales. These estimation results strongly recommend the additional inclusion of economic preferences in econometric analyses that use a NEP scale as explanatory factor of main interest for environmentally relevant behavior. In particular, not considering social preferences, trust, and positive and negative reciprocity can lead to strong distortions due to omitted variable biases. This conclusion is illustrated in an empirical example that reveals biased estimation results for the effect of a NEP scale on donation activities if not all relevant economic preferences are included as control variables
Choices with Delayed Consequences: Pleasing or Fighting Future Tastes?
Many choices concern consumption in future periods. If preferences are state-dependent, a fundamental question is whether people consider their preferences at the time of consumption or decision as more important. Assuming the first, previous studies apparently demonstrate that people systematically mispredict their future tastes. Most of this evidence, however, is also consistent with the idea that people understand, but do not approve of their future preferences. To disentangle both approaches, we conducted a framed field experiment with a commitment option. Commitment in our experiment was not a device against weak will. It was a judgment, which one planning self imposed on another planning self. The results suggest that people are not willing to neglect their preferences at the time of the decision. People may sometimes experience a con ict between two far-sighted selves. This has profound implications in the area of consumer sovereignty and questions the main justification of paternalism

The importin beta superfamily member RanBP17 exhibits a role in cell proliferation and is associated with improved survival of patients with HPV+ HNSCC
More than twenty years after its discovery, the role of the importin beta superfamily member Ran GTP-binding protein (RanBP) 17 is still ill defined. Previously, we observed notable RanBP17 RNA expression levels in head and neck squamous cell carcinoma (HNSCC) cell lines with disruptive TP53 mutations.
We deployed HNSCC cell lines as well as cell lines from other tumor entities such as HCT116, MDA-MB-231 and H460, which were derived from colon, breast and lung cancers respectively. RNAi was used to evaluate the effect of RanBP17 on cell proliferation. FACS analysis was used for cell sorting according to their respective cell cycle phase and for BrdU assays. Immunocytochemistry was deployed for colocalization studies of RanBP17 with Nucleolin and SC35 (nuclear speckles) domains. TCGA analysis was performed for prognostic assessment and correlation analysis of RanBP17 in HNSCC patients.
RNAi knockdown of RanBP17, significantly reduced cell proliferation in HNSCC cell lines. This effect was also seen in the HNSCC unrelated cell lines HCT116 and MDA-MB-231. Similarly, inhibiting cell proliferation with cisplatin reduced RanBP17 in keratinocytes but lead to induction in tumor cell lines. A similar observation was made in tumor cell lines after treatment with the EGFR kinase inhibitor AG1478. In addition to previous reports, showing colocalization of RanBP17 with SC35 domains, we observed colocalization of RanBP17 to nuclear bodies that are distinct from nucleoli and SC35 domains. Interestingly, for HPV positive but not HPV negative HNSCC, TCGA data base analysis revealed a strong positive correlation of RanBP17 RNA with patient survival and CDKN2A.
Our data point to a role of RanBP17 in proliferation of HNSCC and other epithelial cells. Furthermore, RanBP17 could potentially serve as a novel prognostic marker for HNSCC patients. However, we noted a major discrepancy between RanBP17 RNA and protein expression levels with the used antibodies. These observations could be explained by the presence of additional RanBP17 splice isoforms and more so of non-coding circular RanBP17 RNA species.
