    Design and synthesis of novel transcriptional modulators

    Im ersten Projekt (GANT-Projekt) sollten Strukturvorschläge aus einem phänotypischen Screening synthetisiert und auf die biologische Aktivität getestet werden. Nach Etablierung der Synthesen für die ausgewählten Testmoleküle, konnten diese, mit Ausnahme der Verbindung 1, erfolgreich synthetisiert werden. Die Ergebnisse stuften jedoch die meisten potenziellen Kandidaten als eher schlechte GLI-Inhibitoren ein. Bis auf den sehr potenten und literaturbekannten GLI-Antagonisten 39, zeigte lediglich Verbindung 6 ein geringes Hh/GLI-signaling. Aufgrund der Ergebnisse wurde letztendlich dieses Projekt zurückgestellt, um den beiden anderen Projekten der Dissertation den Vorzug zu lassen. Im zweiten Projekt (Photoaffinitätsmarkierung von PPARβ/δ-Liganden) gelang aus synthetischer Perspektive, nach zuvor gescheiterten Versuchen, die Darstellung zweier aktiver PA-Liganden 68 und 69 (Abbildung 152) für die Photoaffinitätsmarkierung von PPARβ/δ durch Modifizierung der etablierten Syntheseroute. Die lichtempfindlichen Gruppen konnten late-stage eingeführt und durch den effektiven Einsatz der Schutzgruppen Chemie die Bildung von schwer trennbaren Regioisomeren unterbunden werden, was letzten Endes zum synthetischen Erfolg geführt hat. Leider war es nicht möglich, einen von beiden Liganden an das Protein zu binden, um im anschließenden Enzymverdau die gebundenen Peptide bis hin zu einzelnen markierten Aminosäuren zu identifizieren, um eine Bindungshypothese aufstellen zu können. Im dritten Projekt (Neuartiger PRMT1-Modulator) wurde im virtuellen Screening die Substanz 80 als potenzieller PRMT4/6-Inhibitor gefunden. Im biologischen Assay war diese Verbindung inaktiv, zeigte allerdings wider Erwarten eine selektive und agonistische PRMT1-Aktivität. Im Rahmen dieses Projektes wurde die Struktur des initialen Hits 80 durch analytische Methoden wie der Massenspektrometrie sowie NMR-Spektroskopie vollständig aufgeklärt und die falsch postulierte Struktur revidiert. Zudem konnten einige sehr vielversprechende und potente Aktivatoren der PRMT1 synthetisiert werden, die sich strukturell von dem bis dato in der Literatur beschriebenen stark unterscheiden. Darunter sind drei Verbindungen (80, 115, und 128, siehe Abbildung 153), die sehr selektiv an die PRMT1 binden und zu einer Aktivierung führen. Durch die Ableitung von Struktur-Wirkungs-Beziehungen ausgehend von dem initialen Hit 80 konnte in verschiedenen SERIEN gezeigt werden, dass für die Aktivität das planare Isochinolin-1,3(2H,4H)-dion-Grundgerüst, die Enamin-Partialstruktur sowie ein am Stickstoff substituiertes/alkyliertes Benzylamin essenziell ist. Die Verbindungen 193, 195, 200, 131 x HCl, 192 x HCl, 202 sowie 184 x HCl sind hochaffine PRMT-Aktivatoren mit unterschiedlichem Selektivitätsprofil und führen zu einer gesteigerten Aktivität von 80-100% (Abbildung 154). Basierend auf der abgeleiteten SAR konnte ein vielversprechender Photoaffinitätsligand entworfen werden, der für die Aktivität notwendigen basischen Stickstoff trägt und dadurch die ersten Markierungsversuche ermöglichen würde. Eine erste Synthese verlief leider erfolglos, es konnte jedoch eine alternative Synthesesequenz erarbeitet werden. Bei erfolgreichem Experiment würden hilfreiche Informationen über den Bindungsmodus erhalten werden. Zum Zeitpunkt der Niederschrift dieser Dissertation wurden erste Versuche gestartet, um eine Röntgenkristallstruktur einiger potenter PRMT1-Aktivatoren zu erhalten. Diese Proteinkristallstrukturen würden, begleitend neben der Photoaffinitätsmarkierung, die Aufstellung einer Bindungshypothese erlauben und dadurch das Struktur-basierte Wirkstoffdesign eröffnen, um eine zielgerichtete Weiterentwicklung der entsprechenden Moleküle zu ermöglichen. Außerdem würde die Identifizierung des neuartigen Bindungsmodus die hit-to-lead-Optimierung zugänglich machen, um dadurch Ansatzpunkte für die Entwicklung klinisch nutzbarer PRMT1-Aktivatoren liefern.In the first project (GANT project), structural proposals from a phenotypic screening were to be synthesised and tested for biological activity. After establishing the synthesis for the selected test molecules, these were successfully synthesized. For compound 1, complete analytical characterization and testing of biological activity was not possible because of insolubility. However, the results categorised most of the potential candidates as rather poor GLI inhibitors. As expected, the very potent and well-known GLI antagonist 39 showed very good inhibitory effects. For all other substances that were tested, only compound 6 had a low level of hh/GLI signalling. Based on these results, this project was postponed in favour of the other two projects in this thesis. In the second project (Photoaffinity labeling of PPARβ/δ ligands), two active PA ligands 68 and 69 (Figure 152) were successfully synthesized for the photoaffinity labelling of PPARβ/δ by modifying the established synthesis route after previous unsuccessful attempts. The photosensitive groups could be introduced late-stage and the formation of regioisomers, which are difficult to separate, could be prevented by the effective use of the protecting group chemistry, which resulted in the successful synthesis. Unfortunately, it was not possible to bind one of the two ligands to the protein in order to identify the bound peptides down to individual labelled amino acids in the subsequent enzyme digestion in which a binding hypothesis could be formulated. In the third project (Novel PRMT1 modulator), substance 80 was identified as a potential PRMT4/6 inhibitor through virtual screening. Contrary to expectations, this compound was inactive in the biological assay for PRMT4/6 but showed selective and agonistic PRMT1 activity. Within this work, the structure of the initial hit 80 was fully elucidated using analytical methods such as mass spectrometry and NMR spectroscopy. In addition, some very promising and potent activators of PRMT1 were synthesized, which are structurally different from those previously described in the literature. These include three compounds (80, 115 and 128; Figure 153) that bind very selectively to PRMT1 and lead to activation. By deriving structure-activity relationships starting from the initial hit 80, it was shown in various series that the planar isoquinoline-1,3(2H,4H)-dione backbone, the enamine partial structure and a nitrogen-substituted/alkylated benzylamine are essential for the activity. The compounds 131 x HCl, 184 x HCl, 192 x HCl, 193, 195, 200 and 202 (see figure 154) are high-affinity PRMT activators with different selectivity profiles and lead to an increased activity of 80-100%. Based on the derived SAR, a promising photoaffinity ligand could be designed, which carries the basic nitrogen necessary for the activity and would thus enable the first labeling experiments. Unfortunately, an initial synthesis was unsuccessful, but an alternative synthesis sequence was developed. A successful experiment would provide helpful information about the binding mode. At the time of writing this thesis, initial attempts were being made to obtain an X-ray crystal structure of some potent PRMT1 activators. In addition to photoaffinity labeling, these protein crystal structures would enable the establishment of a binding hypothesis and thus open up a structure-based drug design for the targeted further development of the corresponding molecules. Furthermore, the identification of the novel binding mode would make hit-to-lead optimisation accessible in order to provide starting points for the development of clinically usable PRMT1 activators

    The influence of alpha angle and completeness of cam resection on outcome after hip arthroscopy

    Die Behandlung des Femoroazetabulären Impingement (FAI) mittels einer Hüftarthroskopie hat in den letzten Jahren stetig an Popularität gewonnen. Durch ein anatomisches Missverhältnis im Bereich des Oberschenkelkopf-Hals-Überganges und der Hüftpfanne kann es zur Entstehung eines solchen FAI kommen. In der vorliegenden Arbeit wird anhand von klinischen und radiologischen Parametern untersucht, welches Ausmaß an Cam-Resektion notwendig ist, um ein bestmögliches klinisches Ergebnis bei den Patienten zu erzielen. Dabei wurde vor allem die Veränderung des alpha-Winkels beurteilt. In die Studie aufgenommen wurden Patienten, bei denen im Zeitraum zwischen 2006 und 2014 ein FAI diagnostiziert und operativ mittels Hüftarthroskopie behandelt wurde. Von 529 Patienten wiesen 234 Patienten ein isoliertes Cam-Impingement auf und bei 190 Patienten konnte eine sogenannte Mischform identifiziert werden. Bei einer Mischform handelt es sich um eine Deformität der femoralen- und azetabulären Komponente des Gelenkes. Die klinischen und radiologischen Ergebnisse wurden anhand eines Fragebogenkatalogs und der Evaluation der angefertigten Röntgenbilder erhoben. In diesem Fragebogenkatalog waren der WOMAC Score, die VAS für Schmerzen, der SF-36 questionnaire, der HHS sowie der HOS enthalten. Zusätzlich wurden intra- und postoperative Komplikationen und die Zufriedenheit der Patienten festgehalten. Komplette Datensätze konnten von 484 Patienten (91,4 %) erhoben werden. Das durchschnittliche Follow-up betrug 42,5 Monate und die Patienten waren im Schnitt 47,8 Jahre alt. Bei 83 Studienteilnehmern (17,7 %) folgte eine erneute Operation. In allen erhobenen Scores konnten statistisch signifikante postoperative klinische Verbesserungen erzielt werden. Der Median des postoperativen alpha-Winkels lag bei 46,8°. Die Regressionsanalysen demonstrierten, dass besonders hohe präoperative alpha-Winkel mit einer erneuten Operation assoziiert sind. Im Gegensatz dazu hatten die postoperativen alpha-Winkel keinen Einfluss auf das Ergebnis „erneute Operation“. Auch in den Daten der Subgruppenanalyse, die lediglich Patienten mit reiner Cam-Deformitäten umfasste, konnte kein signifikanter Einfluss hinsichtlich der Vollständigkeit einer Cam-Resektion auf das Outcome der Scores verzeichnet werden. Die Untersuchung der Veränderungen der prä- zu den postoperativen alpha-Winkeln zeigte, dass die großen Resektionsausmaße (über 30°) oder kleine Veränderungen, die allerdings von hohen präoperativen alpha-Winkeln ausgehen, mit einem erhöhten Risiko einer erneuten Operation verbunden sind. Auf der einen Seite machen die Daten der vorliegenden Arbeit den hohen Stellenwert des alpha-Winkels als diagnostisches Tool zur prädiktiven Analyse der präoperativen Situation deutlich. Auf der anderen Seite könnten sie möglicherweise darauf hinweisen, dass zur Erfassung der postoperativen klinischen Situation die Festlegung eines Schwellenwertes für den alpha-Winkel ungeeignet ist. Anhand dieser Ergebnisse wird die Bedeutung einer adäquaten Patientenselektion besonders deutlich. Insbesondere bei Patienten, bei denen Veränderungen des Hüftgelenks vorliegen, die mit einer Arthrose assoziiert sind, beim Auftreten niedriger präoperativer klinischer Scores und hoher präoperativer alpha-Winkel sollte eine besonders sorgfältige Operationsindikation gestellt werden.The treatment of femoroacetabular impingement (FAI) with hip arthroscopy has steadily gained popularity in recent years. An anatomical imbalance in the area of the femoral head-neck transition and the acetabulum can lead to the development of such an FAI. In the present study, clinical and radiological parameters are used to determine the extent of cam resection necessary to achieve the best possible clinical outcome for patients. In particular, the change in the alpha angle was assessed. The study included patients who were diagnosed with FAI between 2006 and 2014 and treated surgically with hip arthroscopy. Out of 529 patients, 234 showed an isolated cam impingement and 190 patients showed a so-called mixed form. A mixed form is a deformity of the femoral and acetabular components of the joint. The clinical and radiological results were collected by means of a questionnaire catalogue and the evaluation of the radiographs taken. The questionnaire included the WOMAC score, the VAS for pain, the SF 36 questionnaire, the Harris hip score and the hip outcome score. In addition, intra- and postoperative complications and patient satisfaction were recorded. Complete data sets were collected from 484 patients (91.4 %). The average follow-up was 42.5 months and the patients were 47.8 years old on average. 83 study participants (17.7 %) underwent surgery again. Statistically significant postoperative clinical improvements were achieved in all scores. The median postoperative alpha angle was 46.8°. Regression analysis demonstrated that particularly high preoperative alpha angles were associated with a new operation. In contrast, the postoperative alpha angles had no influence on the outcome "reoperation". Also in the data of the subgroup analysis, which included only patients with pure cam deformities, no significant influence regarding the completeness of a cam resection on the outcome of the scores could be recorded. The investigation of the changes of the preoperative to the postoperative alpha angles showed that the large resection dimensions (over 30°) as well as small changes, which however assume high preoperative alpha angles, are associated with an increased risk of a renewed operation. On the one hand, the data of the present work highlight the high value of the alpha angle as a diagnostic tool for predictive analysis of the preoperative situation. On the other hand, they could possibly indicate that the definition of a threshold value for the alpha angle is inappropriate for the assessment of the postoperative clinical situation. These results show the importance of adequate patient selection. Especially in patients with changes of the hip joint that are associated with arthrosis, low preoperative clinical scores and high preoperative alpha angles, a particularly careful indication for surgery should be provided

    Tax Pass-through in the European Beer Market

    We study the pass-through of indirect taxes on beer prices in the European Union (EU). Exploiting the variation of value added tax rates, beer excise tax rates, and beer prices in a panel of monthly data from 1996 to 2016 of all current 28 EU member states, we estimate the tax pass-through of specific beer excise taxes and ad valorem value added taxes, respectively. Ad valorem taxes are under-shifted at a rate of approximately 70%. Specific excise taxes are almost fully shifted to prices in the EU, but, in contrast to the empirical findings for the US, there is no evidence of over-shifting. Nevertheless, the difference between ad valorem and specific tax pass-through rates indicates that imperfect competition plays an important role in the European beer market

    The Debt Brake in the Eyes of the German Population

    In response to the recent sovereign debt crisis, the member states of the European Union agreed to enact balanced budget rules in their national legislation. However, little is known about the public’s opinion of balanced budget rules. To fill this gap, we conducted a survey among 2,000 representatively chosen German citizens. Our findings suggest that 61% of the German population supports the debt brake, whereas only 8% oppose it. However, approval rates differ notably among various subgroups of the population. The debt brake enjoys greater support among high-income earners and among those well-informed about the future costs of deficit spending. People who do not trust politicians would like to see the government’s hands tied even more tightly. Opinions about the debt brake also differ markedly across the supporters of different political parties

    Open-Access-Publikationsfonds der Universitätsbibliothek Marburg. Ein Praxisbericht zum neuen DFG-Förderprogramm „Open-Access-Publikationskosten“

    2022 veränderten sich die Rahmenbedingungen für die Open-Access-Publikationsförderung an der Universitätsbibliothek Marburg. Das neue Förderprogramm „Open-Access-Publikationskosten“ der Deutschen Forschungsgemeinschaft erfordert ein deutlich ausgeweitetes Publikationsmonitoring. Open-Access-Verlagsmonographien wurden erstmals förderfähig. Der Praxisbericht beschreibt, wie die UB Marburg Fördermöglichkeiten und Vorgaben konkret umgesetzt hat und wie sich die Publikationsförderung im ersten Jahr 2022 entwickelt hat. Neben den erzielten Erfolgen werden die bestehenden Herausforderungen etwa in den Bereichen Affiliationsprüfung und Erfassung des Forschungsförderkontextes erörtert.In 2022, the framework to support open access with publication funding at Universitätsbibliothek Marburg changed. The Deutsche Forschungsgemeinschaft announced the funding programme "Open Access Publication Funding" that requires significantly expanded publication monitoring. Open access monographs became eligible for funding for the first time. This report describes how Universitätsbibliothek Marburg has implemented the funding requirements and how publication funding has developed in 2022. In addition to the successes achieved, the existing challenges, for example in the areas of affiliation checks and research funding acknowledgements, are discussed

    Does a good central banker make a difference?

    The personalities of central bankers moved center stage during the recent financial crisis. Some central bankers even gained \superstar" status. In this paper, we evaluate the pivotal role of superstar central bankers by assessing the difference an outstanding governor makes to economic performance. We employ school grades given to central bankers by the financial press. A superstar central banker is one receiving the top grade. In a probit estimation we first relate the grades to measures of economic performance, institutional features, and personal characteristics. We then employ a nearest neighbor matching approach to identify the central bankers which are closest to those receiving the top grade and compare the economic performance across both groups. The results suggest that a superstar governor indeed matters: a topgraded central banker faces a significantly more favorable output-in ation trade-off than his peers. This effect is driven by outstanding central bankers in emerging markets

    Have Pharmaceutical R&D Project Success Rates Decreased? A Critical Review and New Empirical Results

    In the context of the ongoing debate about an innovation crisis in the pharmaceutical industry, we study the success rates of pharmaceutical R&D projects as a measure of innovative productivity. The empirical literature suggests success rates have been decreasing during recent decades. We critically review this literature and only find few studies with a focus on the development of success rates over time. Further, the empirical analysis of success rates imposes difficulties with respect to methodological aspects like data censoring, the definition of success, and the range of firms included in the samples. These difficulties are generally not discussed by the literature. We therefore discuss these issues when critically reviewing the empirical studies and complement this discussion with own empirical results. While most other studies use samples containing a small number of firms and cover just a short time period, we use a broad sample containing firms of different sizes over an observation period of more than 20 years (1989-2010). Descriptive results suggest a declining success rate of pharmaceutical projects during recent years. Correcting for censored observations shows there has been a stabilization of success rates, but at a lower level than before. The main underlying reason for a lower success rate is the start of many more projects in more recent time periods. Results from hazard rate models even suggest there has only been a temporary drop in the success rate for projects between 1995 and 2002

    How to get photovoltaics on the roofs? Empirical evidence on the public support for a residential solar mandate in Germany

    This study evaluates whether a residential solar mandate in the case of roof renovation is a useful complement to economic incentives for further photovoltaics (PV) adoption. Analyzing determinants affecting PV ownership and installation intentions among single-family homeowners, as well as factors influencing support for a solar mandate and perceptions of its effectiveness, our empirical results, based on a survey of German utility customers, show that a residential solar mandate is a rather unpopular policy measure among homeowners. However, a solar mandate addresses two important factors which, according to our results, increase the willingness to install PV: firstly, the perception that the personal environment expects more PV, and secondly, an upcoming roof renovation. Both social desirability and a favorable time window can be institutionalized through a solar mandate. In terms of support for a solar mandate, we find that the perceived effectiveness of such a mandate has a strong influence on homeowner support. Perceived effectiveness, in turn, is closely related to perceived cost savings and perceived environmental benefits of PV. Based on these results, we conclude that an active information policy regarding the environmental and cost implications of PV expansion is essential to increase the acceptance of a solar mandate

    Metadaten für Objekte in wissenschaftlichen Sammlungen

    Die nachfolgenden Ausführungen behandeln das Thema Metadaten für Sammlungsrepositorien. Im Zentrum steht ein weiterentwickelter Vorschlag (Version 2.0) für ein gemeinsames Metadatenschema der digitalen Daten wissenschaftlicher Sammlungen an der Philipps-Universität. Dieses Schema soll bei der Umsetzung eines sammlungsübergreifenden Objektportals verwendet werden. Diese Publikation stützt sich auf die initiale Entwicklung (Version 1.0) des Metadatenschemas von Alexander Maul. Um die Weiterentwicklung zu ermöglichen, haben mehrere Fachinstitutionen und Partnereinrichtungen das Metadatenschema Version 1.0 einem Review unterzogen: So haben Anmerkungen und Hinweise von NFDI4Culture, der Koordinierungsstelle wissenschaftliche Sammlungen in Deutschland, dem Datenkompetenzzentrum Sammlungen, Objekte, Datenkompetenzen (SODa), vom Sachgebiet Digitalisierung und Metadaten der Universitätsbibliothek der Philipps-Universität Marburg sowie vom initialen Autor Alexander Maul Eingang in die Weiterentwicklung des Metadatenschemas gefunden. Allen Beteiligten danken wir sehr herzlich für den profunden fachlichen Review. Die vorliegenden beschreibenden Abschnitte zu den Metadatenfeldern wurden entsprechend gegenüber Version 1.0 wie folgt teilweise abgeändert: (1) Die Weiterentwicklung des Metadatenschemas berücksichtigt nun Herausforderungen im Bereich der Provenienz und Sensibilität. Insbesondere komplexe Provenienzen, wie sie beispielsweise in kolonialen Kontexten oder bei medizinhistorischen Humanpräparaten auftreten, werden explizit vermerkbar. Zudem adressiert das Metadatenschema nun systematisch ethische und rechtliche Aspekte. Sensible Inhalte, wie menschliche Überreste oder historisch problematisches Fachvokabular etwa in Form von antisemitischen oder rassistischen Begriffen, können durch dedizierte Disclaimer, eingeschränkte Sichtbarkeiten und Zugriffsrechte kontrolliert, dokumentiert und geschützt werden. Die Flexibilität der Konfiguration des Metadatenschemas ermöglicht eine differenzierte Anpassung an sensible Kontexte mit Regelungen zur Zugriffssteuerung und zur Vermeidung unerwünschter Veröffentlichung. Damit berücksichtigt das Metadatenschema nun die gemeinsame Verpflichtung, die koloniale Vergangenheit in Museen und Sammlungen aufzuarbeiten. Dieser Ansatz entspricht damit auch schon den Empfehlungen der gemeinsamen Erklärung vom 14. Oktober 2025, die die Staatsministerin des Bundes für Kultur und Medien, die Staatsministerin im Auswärtigen Amt, die Kulturministerinnen und Kulturministern der Länder sowie die kommunalen Spitzenverbänden ausgesprochen haben. (2) Weiterhin haben wir für die vorliegende Fassung auf eine erneute Publikation der technischen Beschreibung der kontrollierten Vokabulare und Normdaten verzichtet, da keine signifikanten Änderungen vorgenommen wurden.Diese überarbeitete Ausgabe basiert auf der Publikation „Sammlungsdigitalisierung an der Philipps-Universität Marburg 3“, ursprünglich veröffentlicht unter CC BY 3.0.     Untersuchung biotechnologisch relevanter Enzyme derNaturstoffbiosynthese in Microorganismen

    Microorganisms are omnipresent on our planet. They produce natural products whose derivatives make up a considerable proportion of the drugs in use today. Natural products are divided into primary metabolites, which are essential for survival, and secondary metabolites (SM), which are not essential but represent a selective advantage for the producer. Secondary metabolites are found in a large number of living organisms and have a wide variety of biological and pharmacological effects. The biosynthesis of SM begins mainly through the formation of a backbone structure. This structure allows various modifications by so-called modifying enzymes. The modifying enzymes in SM-associated biosynthesis pathways include prenyltransferases (PTs) and cytochrome P450 enzymes (P450s). The initial section of the thesis examines these PTs. PTs have proven to be invaluable tools in the synthesis of new chemical structures and drug candidates. These enzymes are widely distributed in nature and participate in a multitude of biosynthetic pathways. While numerous dimethylallyl tryptophan synthase-PTs from ascomycetes have been identified and characterized, surprisingly little is known about these enzymes from basidiomycetes. At the time of this study, only two members of this class had been described. The prenyltransferase ShPT from the basidiomycete Stereum hirsutum exhibited a striking degree of similarity to a previously described PT involved in the biosynthesis of vibralactone. It was therefore postulated that ShPT might also prenylate 4-hydroxybenzyl alcohol. We received a codon- optimized plasmid containing the PT from our cooperation partners at the Interfaculty Institute of Biochemistry at the University of Tübingen, for the heterologous expression in E. coli. In the initial phase of this project, I conducted a genomic analysis of ShPT and overexpressed the PT in E. coli. A biochemical analysis of this PT was conducted to ascertain its acceptability of different substrates. The results demonstrate that benzyl alcohol derivatives are not accepted as substrates for enzymatic catalysis by ShPT. Instead, hydroxynaphthalenes are identified as the preferred major prenyl acceptors. Consequently, ShPT is the first basidiomycete prenyltransferase to be characterized as accepting hydroxynaphthalenes as substrates. Furthermore, the results illustrate that ShPT exhibits a broad substrate specificity towards DMAPP and GPP as prenyl donors. Subsequently, further studies were conducted on the neighboring cytochrome P450 enzyme by heterologously expressing it in Aspergillus nidulans LO8030 and subsequently culturing the transformant in the presence of prenylated hydroxynaphthalene. While the natural substrate of ShPT remains unidentified, our findings indicate that ShPT catalysis expands the chemical arsenal of enzymes from the DMATS superfamily, enabling the production of new regioselectively prenylated naphthalenes. This provides a new approach for the future discovery and synthesis of new drug candidates. The second part of this work deals with P450 enzymes. Six orthologous two-gene BGCs for mycocyclosin and guatyromycine biosynthesis were identified by our research group. Biochemical studies have confirmed that the P450 enzymes GymBx act as intramolecular oxidases and intermolecular nucleobase transferases. These enzymes facilitate oxidative C-C coupling in cYY, or alternatively, link it to guanine or hypoxanthine. This represents a distinctive attribute of CDP-modifying P450 enzymes. In collaboration with the University of Tübingen, we undertook an investigation into the structure of this unusual enzyme family and sought to elucidate the mechanism of its chemoselective catalysis. Jasmin Freytag and Dr. Georg Zocher were able to solve the crystal structures of two members, GymB1 and GymB5, in their unbound state, as well as that of GymB5 in complex with hypoxanthine and cYY. The structural data of the ternary complex facilitated an understanding of CDP binding and the nucleobase transfer reaction. This enables the identification of crucial positions for substrate recognition and modification of the bifunctional enzymes. By side-directed and saturation mutagenesis performed by me, a GymB5 variant was created that exclusively exhibits intramolecular coupling enzyme activity. These data provide a robust basis for future protein modification of P450 enzymes to synthesize new CDP nucleobase adducts and other "unnatural" natural products. Through genome mining, we identified several interesting cytochrome P450 enzymes that may be involved in tailoring reactions in the third part of this thesis. Through heterologous expression in S. albus J1074 I elucidated the function of a CDPS backbone gene and demonstrated the involvement of a P450 enzyme in hydroxylation reactions. Precursor feeding experiments led to the generation of the same new products in the native host strain. Furthermore, an attempt was made to elucidate the function of another previously uncharacterized actinomycete BGC by heterologous expression in S. coelicolor M1146. The resulting accumulated product was isolated and the structure elucidated.Mikroorganismen sind allgegenwärtig auf unserem Planeten. Sie produzieren Naturstoffe, deren Derivate einen beträchtlichen Anteil der aktuell in Verwendung befindlichen Medikamente darstellen. Naturstoffe werden in überlebenswichtige Primärmetabolite und nicht überlebenswichtige, jedoch einen Selektionsvorteil für den Produzenten darstellende Sekundärmetabolite (SM) eingeteilt. Sekundärmetabolite kommen in einer Vielzahl von Lebewesen vor und zeigen verschiedenste biologische und pharmakologische Wirkungen. Die Biosynthese von SM beginnt in der Regel mit der Formation einer Grundgerüststruktur. Diese Struktur erlaubt diverse Modifikationen durch sogenannte Modifizierungsenzyme. Zu den Modifizierungsenzymen in SM-assoziierten Synthesewegen gehören unter anderem Prenyltransferasen (PTs) und Cytochrom P450-Enzyme (P450s). Der erste Teil der Arbeit beschäftigt sich mit eben diesen PTs. PTs haben sich als wertvolle Werkzeuge für die Generierung neuer chemischer Strukturen und Wirkstoffkandidaten erwiesen. Diese Enzyme sind in der Natur weit verbreitet und an vielen verschiedenen Biosynthesewegen beteiligt. Während viele Dimethylallyltryptophansynthase (DMATS)-PTs aus Ascomyceten bereits bekannt und charakterisiert sind, weiß man überraschend wenig über diese Enzyme aus Basidiomyceten, von denen zum Zeitpunkt dieser Arbeit nur zwei bekannt waren. Die Prenyltransferase ShPT aus dem Basidiomyceten Stereum hirsutum zeigte eine sehr große Ähnlichkeit mit einer zuvor beschriebenen PT, die an der Biosynthese von Vibralacton beteiligt ist. Daher wurde vermutet, dass ShPT auch 4-Hydroxybenzylalkohol prenylieren könnte. Von unseren Kooperationspartnern am Interfakultären Institut für Biochemie der Universität Tübingen wurde uns ein codonoptimiertes Plasmid zur heterologen Expression der PT in Escherichia coli zugeschickt. Zu Beginn der Arbeit habe ich den genomischen Locus von ShPT analysiert und die Prenyltransferase in E. coli überproduziert. Durch die Untersuchung der Akzeptanz verschiedener Substrate führten wir eine biochemische Analyse dieser Prenyltransferase durch. Unsere Ergebnisse zeigen, dass Benzylalkoholderivate nicht als Substrate für die enzymatische Katalyse durch ShPT akzeptiert werden. Stattdessen werden Hydroxynaphthaline als Hauptprenylakzeptoren bevorzugt. Damit ist ShPT die erste beschriebene Basidiomycetenprenyltransferase, die Hydroxynaphthaline als Substrate prenyliert. Darüber hinaus zeigen unsere Ergebnisse, dass ShPT eine breite Substratspezifität gegenüber DMAPP und GPP als Prenyldonoren aufweist, was zur Identifikation dreier neuer geranylierter Produkte führte. Anschließend wurden weitere Untersuchungen an dem benachbarten Cytochrom P450 Enzym durchgeführt, indem es heterolog in Aspergillus nidulans LO8030 exprimiert und dieser Stamm dann in Anwesenheit von prenyliertem Hydroxynaphthalin kultiviert wurde. Obwohl das natürliche Substrat von ShPT noch nicht Zusammenfassung identifiziert wurde, zeigen unsere Untersuchungen, dass die Charakterisierung von ShPT das chemische Arsenal der Enzyme aus der DMATS Superfamilie erweitert, um neuartige regioselektiv prenylierte Naphthaline zu produzieren. Dies bietet einen neuen Ansatz für die Entdeckung und Synthese neuer Wirkstoffkandidaten in der Zukunft. Der zweite Teil der Arbeit handelt von P450 Enzymen. Sechs orthologe Zwei-Gen-BGCs für die Mycocyclosin- und Guatyromycin-Biosynthese wurden von unserer Arbeitsgruppe identifiziert. Es wurde bewiesen, dass die P450 Enzyme GymBx als intramolekulare Oxidasen und intermolekulare Nukleobasetransferasen fungieren. Sie katalysieren eine oxidative C-C- Kopplung in cYY oder verknüpfen es mit Guanin beziehungsweise Hypoxanthin. Dies ist eine einzigartige Eigenschaft der CDP-modifizierenden P450 Enzyme. In Zusammenarbeit mit der Universität Tübingen haben wir in diesem Teil der Arbeit die Struktur dieser ungewöhnlichen Enzymfamilie untersucht und den Mechanismus ihrer chemoselektiven Katalyse aufgeklärt. Jasmin Freytag und Dr. Georg Zocher haben die Kristallstrukturen von zwei Mitgliedern, GymB1 und GymB5, in ihrem ungebundenen Zustand sowie von GymB5 im Komplex mit Hypoxanthin und cYY gelöst. Die Strukturdaten des ternären Komplexes lieferten Einblicke in die CDP- Bindung und die Nukleobasen-Transferreaktion, was zur Identifizierung von Schlüsselpositionen für die Substraterkennung und Modifikation der bifunktionellen Enzyme führte. Durch von mir durchgeführte gerichtete Mutagenese und Sättigungsmutagenese wurde eine GymB5-Variante etabliert, die die Chemoselektivität strikt auf ein intramolekulares Kopplungsenzym umschaltet. Diese Daten bilden eine solide Grundlage für die zukünftige Proteinmodifizierung von Cytochrom P450-Enzymen zur Synthese von neuartigen CDP- Nukleobase-Addukten und anderen „unnatürlichen“ Naturstoffen. Durch tiefe Genomanalyse habe ich im dritten Teil dieser Arbeit mehrere interessante Cytochrom-P450-Enzyme identifiziert, die an Modifizierungsreaktionen an CDPs oder anderen Grundstrukturen beteiligt sind. Durch heterologe Expression in S. albus J1074 konnte ich die Funktion eines CDPS-Backbone-Gens aufklären und die Beteiligung eines P450-Enzyms an Hydroxylierungsreaktionen nachweisen. Durch Vorstufenfütterungsexperimente wurden die gleichen neuen Produkte im nativen Wirtsstamm nachgewiesen. Darüber hinaus wurde versucht, die Funktion eines weiteren, bisher nicht charakterisierten BGC aus Actinomyceten durch heterologe Expression in S. coelicolor M1146 zu klären. Das resultierende akkumulierte Produkt wurde isoliert und die Struktur aufgeklärt
