    Single-Center Experience in Microsurgical Resection of Acoustic Neurinomas and the Benefit of Microscope-Based Augmented Reality

    Background and Objectives: Microsurgical resection with intraoperative neuromonitoring is the gold standard for acoustic neurinomas (ANs) which are classified as T3 or T4 tumors according to the Hannover Classification. Microscope-based augmented reality (AR) can be beneficial in cerebellopontine angle and lateral skull base surgery, since these are small areas packed with anatomical structures and the use of this technology enables automatic 3D building of a model without the need for a surgeon to mentally perform this task of transferring 2D images seen on the microscope into imaginary 3D images, which then reduces the possibility of error and provides better orientation in the operative field. Materials and Methods: All patients who underwent surgery for resection of ANs in our department were included in this study. Clinical outcomes in terms of postoperative neurological deficits and complications were evaluated, as well as neuroradiological outcomes for tumor remnants and recurrence. Results: A total of 43 consecutive patients (25 female, median age 60.5 ± 16 years) who underwent resection of ANs via retrosigmoid osteoclastic craniotomy with the use of intraoperative neuromonitoring (22 right-sided, 14 giant tumors, 10 cystic, 7 with hydrocephalus) by a single surgeon were included in this study, with a median follow up of 41.2 ± 32.2 months. A total of 18 patients underwent subtotal resection, 1 patient partial resection and 24 patients gross total resection. A total of 27 patients underwent resection in sitting position and the rest in semi-sitting position. Out of 37 patients who had no facial nerve deficit prior to surgery, 19 patients were intact following surgery, 7 patients had House Brackmann (HB) Grade II paresis, 3 patients HB III, 7 patients HB IV and 1 patient HB V. Wound healing deficit with cerebrospinal fluid (CSF) leak occurred in 8 patients (18.6%). Operative time was 317.3 ± 99 min. One patient which had recurrence and one further patient with partial resection underwent radiotherapy following surgery. A total of 16 patients (37.2%) underwent resection using fiducial-based navigation and microscope-based AR, all in sitting position. Segmented objects of interest in AR were the sigmoid and transverse sinus, tumor outline, cranial nerves (CN) VII, VIII and V, petrous vein, cochlea and semicircular canals and brain stem. Operative time and clinical outcome did not differ between the AR and the non-AR group. However, use of AR improved orientation in the operative field for craniotomy planning and microsurgical resection by identification of important neurovascular structures. Conclusions: The single-center experience of resection of ANs showed a high rate of gross total (GTR) and subtotal resection (STR) with low recurrence. Use of AR improves intraoperative orientation and facilitates craniotomy planning and AN resection through early improved identification of important anatomical relations to structures of the inner auditory canal, venous sinuses, petrous vein, brain stem and the course of cranial nerves.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Ueberbringen schlechter Nachrichten in der Neonatologie Breaking Bad News in Neonatology

    Die Dissertation beschäftigt sich mit dem Überbringen schlechter Nachrichten im Kontext der Neonatologie. In der Medizin bezieht sich das Überbringen schlechter Nachrichten auf negative Informationen in Bezug auf Diagnose, Therapie und Prognose. Die Aufklärung darüber erfolgt in ärztlichen Gesprächen. Basierend auf einer systematischen Literatursuche wurde in der vorliegenden Arbeit die Sicht neonatologischer Ärzt*innen und betroffener Eltern kranker Früh- und Neugeborener zum Überbringen schlechter Nachrichten erhoben. Die Aufnahme eines Früh- oder kranken Neugeborenen auf einer neonatologischen Intensivstation (NICU) stellt für die Eltern eine Ausnahmesituation dar, mit Gefühlen von Angst, Hilflosigkeit und Kontrollverlust (Al Maghaireh et al., 2016; Gateau et al., 2021; Schecter et al., 2020). Weltweit wird etwa jedes 16. Neugeborene temporär auf einer NICU aufgenommen (Haidari et al., 2021). In Deutschland verbrachten 2021 etwa 13% aller Neugeborenen einige Zeit auf einer NICU (Bundesqualitätsbericht 2022 Perinatalmedizin, Institut für Qualität und Transparenz im Gesundheitswesen, 2022; Statistisches Bundesamt, 2023). Studien zeigen generell, dass die Kommunikation zwischen Ärzt*innen und Patient*innen/Angehörigen deren psychisches Wohlbefinden beeinflusst und Stress und negative Emotionen (experimentelle Kontrollstudien (Schmid Mast et al., 2005; Zwingmann et al., 2017), Metaanalyse (Schnur et al., 2008)) auslösen kann. Auch in der Neonatologie beeinflusst die Kommunikation elterliches Coping (Review; Labrie et al., 2021), Stresserleben (Orzalesi & Aite, 2011) oder Selbstmanagement (Review; Wreesmann et al., 2021). Das Überbringen schlechter Nachrichten führt dabei sowohl bei Überbringer*innen als auch Empfänger*innen zu Stress, weshalb Gesprächsempfehlungen hilfreich sind. Eine der am häufigsten gelehrten Empfehlungen ist das SPIKES-Protokoll von Baile (Setting, Perception, Invitation, Knowledge, Emotion, Strategy & Summary; Baile et al., 2000). Jedoch stammt die Literatur zum Überbringen schlechter Nachrichten überwiegend aus der Onkologie, mit vorrangig expertengenerierten Empfehlungen zum patientenzentrierten Überbringen schlechter Nachrichten (Baile et al., 2000; Witt & Jankowska, 2018). In der Neonatologie gibt es nur wenige Studien zu Grundlagen, Empfehlungen und Auswirkungen des Überbringens schlechter Nachrichten (z.B. Bernstein & Lemmon, 2021; Boss et al., 2017; Marçola et al., 2020; Strauss et al., 1995). Daher wurde eine qualitative Inhaltsanalyse (Mayring, 2015) von Expert*innenmeinungen (neonatologische Ober-/Chefärzt*innen) durchgeführt und wichtige Determinanten für das gelungene Überbringen schlechter Nachrichten generiert (Seifart et al., 2022): Der konzeptionelle Rahmen NEO-SPEAK (Neonatale prognostische Unsicherheit, Begegnung (Encounter), Organisation und Teamarbeit - Situativer Stress, Prozesshaftigkeit, Emotionale Belastung, Beachtung der Individualität (Attention), Wissen und Erfahrung (Knowledge)) benennt wichtige Bestandteile für gelungenes Übermitteln schlechter Nachrichten in der Neonatologie aus Expert*innensicht (ebd., 2022). Die Elternsicht wurde anhand einer prospektiv angelegten querschnittbasierten quantitativen Fragebogenstudie erhoben, mit Vergleich der elterlichen Präferenzen zum Überbringen schlechter Nachrichten mit ihren tatsächlichen Erfahrungen und Erfassung der Gesprächsqualität (Wege et al., 2023a). Zudem wurde untersucht, ob sich die psychische Gesundheit von Eltern und ihre Beziehung zu Ärzt*innen in Abhängigkeit von ihrem Bildungsstand im Kontext des Überbringens schlechter Nachrichten unterschied (Wege et al., 2023b). Der für die Elternbefragung konzipierte Fragebogen orientierte sich an den einzelnen SPIKES-Komponenten, sowie vorangegangenen Expert*innen- und Elterninterviews (Wege et al., 2023a). Die Mehrheit der elterlichen Präferenzen für das Überbringen schlechter Nachrichten unterschied sich bedeutsam von ihren berichteten Erfahrungen mit ärztlichen Aufklärungsgesprächen. Die Gesprächsqualität, in Form von Zufriedenheit, korrelierte am stärksten mit einer mitfühlenden Art des Arztes/der Ärzt*in, sowie einer verständlichen Erklärung der Erkrankung des Kindes (Wege et al., 2023a). Eltern mit niedrigerem Bildungsstand berichteten signifikant weniger Vertrauen in die Ärzt*innen sowie ein geringeres Gefühl von Sicherheit aufgrund des Gesprächs, in welchem schlechte Nachrichten überbracht wurden, als Eltern mit höherem Bildungsstand. Beide Gruppen berichteten von großer Erschöpfung sowie höherer Hilflosigkeit im Vergleich zur Normgruppe. Depressivität und Ängstlichkeit waren im Vergleich zur Allgemeinbevölkerung für beide Gruppen erhöht. Die Möglichkeit Fragen zu stellen korrelierte bei allen Eltern hoch mit der durch das Gespräch vermittelten Orientierung und Sicherheit. Nur bei weniger gebildeten Eltern korrelierte das Sicherheitsgefühl hoch mit der im Gespräch vermittelten Hoffnung (Wege et al., 2023b). Ärzt*innen sollten daher neben der Vermittlung von Informationen darauf achten, auch Hoffnung zu vermitteln.The dissertation deals with the breaking of bad news in the context of neonatology. In medicine, breaking bad news refers to negative information regarding diagnosis, treatment and prognosis. This information is given in medical consultations. Based on a systematic literature search, this study investigated the views of neonatologists and affected parents of sick premature and newborn infants on breaking bad news. The admission of a premature or sick newborn to a neonatal intensive care unit (NICU) is an exceptional situation for parents, with feelings of fear, helplessness and loss of control (Al Maghaireh et al., 2016; Gateau et al., 2021; Schecter et al., 2020). Worldwide, around one in 16 newborns is temporarily admitted to a NICU (Haidari et al., 2021). In Germany, around 13% of all newborns spent some time in a NICU in 2021 (Federal Quality Report 2022 Perinatal Medicine, Institute for Quality and Transparency in Healthcare, 2022; Federal Statistical Office, 2023). Studies generally show that communication between physicians and patients/relatives influences their psychological well-being and can trigger stress and negative emotions (experimental control studies (Schmid Mast et al., 2005; Zwingmann et al., 2017), meta-analysis (Schnur et al., 2008)). In neonatology, communication also influences parental coping (review; Labrie et al., 2021), stress experience (Orzalesi & Aite, 2011) or self-management (review; Wreesmann et al., 2021). Delivering bad news leads to stress for both the messenger and the recipient, which is why recommendations for conversation are helpful. One of the most commonly taught recommendations is Baile's SPIKES protocol (Setting, Perception, Invitation, Knowledge, Emotion, Strategy & Summary; Baile et al., 2000). However, the literature on breaking bad news comes predominantly from oncology, with primarily expert-generated recommendations on patient-centered breaking of bad news (Baile et al., 2000; Witt & Jankowska, 2018). In neonatology, there are few studies on the principles, recommendations and effects of breaking bad news (e.g. Bernstein & Lemmon, 2021; Boss et al, 2017; Marçola et al, 2020; Strauss et al, 1995). Therefore, a qualitative content analysis (Mayring, 2015) of expert opinions (senior/chief neonatologists) was conducted and important determinants for the successful delivery of bad news were generated (Seifart et al., 2022): The conceptual framework NEO-SPEAK (Neonatal Prognostic Uncertainty, Encounter, Organization and Teamwork - Situational Stress, Processuality, Emotional Stress, Attention to Individuality, Knowledge and Experience) identifies important components for successful delivery of bad news in neonatology from an expert perspective (ibid., 2022). Parents' views were assessed using a prospective cross-sectional quantitative questionnaire study, comparing parental preferences for breaking bad news with their actual experiences and recording the quality of the conversation (Wege et al., 2023a). In addition, it was investigated whether the mental health of parents and their relationship with physicians differed depending on their level of education in the context of breaking bad news (Wege et al., 2023b). The questionnaire designed for the parent survey was based on the individual SPIKES components and the previous expert and parent interviews (Wege et al., 2023a). The majority of parental preferences for the delivery of bad news differed significantly from their reported experiences with delivery of bad news. The quality of the conversation, in terms of satisfaction, correlated most strongly with a compassionate way of disclosure and an understandable explanation of the infant’s illness (Wege et al., 2023a). Parents with a lower level of education reported significantly less trust in the physicians and a lower sense of security due to the conversation in which bad news was delivered than parents with a higher level of education. Both groups reported high levels of exhaustion and helplessness compared to the norm group. Depressiveness and anxiety were higher in both groups compared to the general population. For all parents, the opportunity to ask questions correlated highly with orientation and security provided by the interview.     Identification and characterization of the cellular interactome of the Lassa virus matrix protein Z

    Das hochpathogene Lassavirus (LASV) stellt eine große Gefahr für die öffentliche Gesundheit in Westafrika dar. Protein-Protein-Interaktionen (PPI) zwischen Virus- und Wirtsproteinen spielen eine wichtige Rolle in verschiedenen Phasen des viralen Lebenszyklus, einschließlich dem Viruseintritt, der Virusreplikation und der Virusfreisetzung. Darüber hinaus können diese PPI auch eine Modulation von zellulären Signalwegen und biologischen Prozessen während einer Infektion induzieren. Das LASV- Matrixprotein Z hat wichtige strukturelle und funktionelle Aufgaben im viralen Lebenszyklus, wie z. B. die Regulierung der viralen RNA-Synthese, die Orchestrierung der viralen Assemblierung und Virusfreisetzung sowie die Hemmung der Interferon-vermittelten Immunantwort. Das Verständnis über die molekularen Mechanismen und Wirtsprotein-Interaktionen durch die das Z-Protein seine vielfältigen Funktionen ausübt, ist jedoch noch begrenzt. In dem vorliegenden Promotionsprojekt wurde die Proximity-abhängige Biotinidentifizierung 2 (BioID2) als Methode der Affinitätsaufreinigung in Verbindung mit massenspektrometrischen Analysen für den Nachweis und die Identifizierung des PPI-Netzwerks des LASV-Z-Proteins in lebenden Zellen eingesetzt. Eine stringente statistische Filterung ergab 584 potenzielle zelluläre Interaktionskandidaten des Z-Proteins. Einer der identifizierten Wirtsfaktoren war der Septinregulator Cdc42EP1, ein Effektorprotein der Rho-GTPase Cdc42. Die funktionelle Validierung mittels Ko-Immunpräzipitationsanalysen (Ko-IP) und konfokaler Mikroskopie bestätigte eine physische Interaktion und subzelluläre Ko-Lokalisation zwischen LASV Z und Cdc42EP1. Die weitere molekulare Charakterisierung ergab, dass die Interaktion zwischen dem Z-Protein und Cdc42EP1 von der CRIB (Cdc42 and Rag interactive binding)-Domäne von Cdc42EP1 abhängt, die normalerweise die Bindung an Cdc42 vermittelt. Interessanterweise reguliert die rekombinante Expression von Cdc42EP1 durch die Modulation von Septinfilamenten die Freisetzung von Z-induzierten Virus-ähnlichen Partikeln (virus-like particles, VLPs). Infektionsstudien zeigten ferner, dass LASV eine Umlagerung des zellulären Septinzytoskeletts induziert. Die pharmakologische Hemmung der Septinpolymerisation mit dem Wirkstoff Forchlorfenuron führte zu einem gestörten Plasmamembrantransport des Z-Proteins sowie einer reduzierten Freisetzung von VLPs und authentischem LASV. Dies unterstreicht die Bedeutung eines dynamischen Septinnetzwerks während einer LASV-Infektion. Septine regulieren das Mikrotubuli- als auch das Aktinzytoskelett, die ebenfalls eine wichtige Rolle im LASV-Replikationszyklus spielen. Wohingegen das Mikrotubulinetzwerk beim Plasmamembrantransport des Z-Proteins beteiligt ist, scheint ein dynamisches Aktinzytoskelett bei dem Viruseintritt und der Virusfreisetzung an der Plasmamembran notwendig zu sein. Insbesondere die Virusausbreitung von Zelle-zu-Zelle über Aktin-reiche Filopodien konnte in dieser Arbeit als weiterer Freisetzungsmechanismus von LASV aufgezeigt werden. Zusammenfassend lässt sich sagen, dass Cdc42EP1 als ein wichtiger Wirtsfaktor identifiziert werden konnte, der die Freisetzung von Z-induzierten VLPs und infektiösen LASV-Partikeln reguliert. Der zelluläre mTOR-Signalweg war einer der am stärksten angereicherten Signalwege im LASV Z-Interaktom. So wurde beispielsweise die Ragulatoruntereinheit LAMTOR1 als möglicher Interaktionspartner des Z-Proteins in den Interaktomanalysen identifiziert. Die wichtigste Funktion des Ragulatorkomplexes ist die lysosomale Gerüstfunktion für die Aminosäure-abhängige mTORC1-Rekrutierung und Aktivierung. Mittels Ko-IP-Analysen und Ko-Lokalisationsstudien konnte der Ragulatorkomplex als physischer Interaktionspartner des Z-Proteins bestätigt werden. Funktionelle Validierungen wiesen LAMTOR1 als antiviralen Faktor bei der Z-vermittelten VLP-Freisetzung aus. Der mögliche Mechanismus deutet auf die Rolle von LAMTOR1 bei der intrazellulären Positionierung von späten Endosomen und Lysosomen hin. Seine Funktion bei der mTORC1-Aktivierung scheint hingegen keine Rolle zu spielen. Das lassavirale Oberflächenglykoprotein GP, das Matrixprotein Z sowie das Nukleoprotein NP als Marker für Ribonukleoproteinkomplexe ko-lokalisierten mit späten Endosomen an der Plasmamembran in infizierten Zellen. Die Bedeutung der späten Endosomen und der Exosomenbiogenese für die Freisetzung von LASV konnte zusätzlich durch die elektronenmikroskopische Charakterisierung von LASV Z-positiven extrazellulären Vesikeln und LASV-infizierten Zellen bestätigt werden. Zusammenfassend konnten in diesem Promotionsprojekt bislang unbekannte Freisetzungsmechanismen des LASV aufgezeigt werden. Eine umfassendere Kenntnis der Virus-Wirt-Interaktionen könnte neue Möglichkeiten für die Entwicklung neuer Behandlungsmöglichkeiten gegen das hochpathogene LASV erschließen.Highly pathogenic Lassa virus (LASV) poses a major public health threat in West Africa. Protein-protein interactions (PPI) between viral and host proteins play an important role in various stages of the viral life cycle, including virus entry, virus replication, and virus release. In addition, these PPIs can also modulate cellular signaling pathways and biological processes during infection. The LASV matrix protein Z has important structural and functional roles in the viral life cycle, such as regulating viral RNA synthesis, orchestrating viral assembly and release, and inhibiting the interferon-mediated immune response. However, our understanding of the molecular mechanisms and host-protein interactions by which the Z protein exerts its diverse functions is still limited. In the present dissertation project, proximity-dependent biotin identification 2 (BioID2) was used for affinity purification in combination with mass spectrometric analysis for the detection and identification of PPI networks of the LASV Z protein in living cells. Stringent statistical filtering revealed 584 potential host interaction candidates of Z protein. One of the identified host factors was the septin regulator Cdc42EP1, an effector protein of the Rho GTPase Cdc42. Functional validation using co-immunoprecipitation (co-IP) analyses and confocal microscopy confirmed a physical interaction and subcellular co-localization between LASV Z and Cdc42EP1. Further molecular characterization demonstrated that the interaction between the Z protein and Cdc42EP1 depends on the Cdc42 and Rac interactive binding (CRIB) domain of Cdc42EP1, which normally mediates binding to Cdc42. Interestingly, recombinant expression of Cdc42EP1 regulates the release of Z-induced virus-like particles (VLPs) by modulating septin filaments. Infection studies further showed that LASV induces rearrangement of the cellular septin cytoskeleton. Pharmacological inhibition of septin polymerization with the drug forchlorfenuron resulted in impaired plasma membrane transport of Z protein and reduced release of VLPs and authentic LASV. This underscores the importance of a dynamic septin network during LASV infection. Septins regulate the microtubule as well as the actin cytoskeleton, which also play an important role in the LASV replication cycle. Whereas the microtubule network is involved in plasma membrane transport of the Z protein, a dynamic actin cytoskeleton appears to be required in virus entry and release at the plasma membrane. This work revealed cell-to-cell virus spread via actin-rich filopodia as a novel release mechanism of LASV. In summary, Cdc42EP1 was identified as an important host factor regulating the release of Z-induced VLPs and infectious LASV particles. The cellular mTOR pathway was one of the most enriched signaling pathways in the LASV Z interactome. For example, the ragulator subunit LAMTOR1 was identified as a putative interaction partner of the Z protein in the interactome analyses. The major function of the ragulator complex is lysosomal scaffolding for amino acid-dependent mTORC1 recruitment and activation. Using co-IP analyses and co-localization studies, the ragulator complex was confirmed as a physical interaction partner of the Z protein. Functional validations indicated that LAMTOR1 is an antiviral factor in Z-mediated VLP release. The possible mechanism suggests the role of LAMTOR1 in the intracellular positioning of late endosomes and lysosomes. In contrast, its function in mTORC1 activation does not appear to play a role in the antiviral activity against Z. The lassaviral surface glycoprotein GP, matrix protein Z, and nucleoprotein NP, as a marker for ribonucleoprotein complexes, co-localized with late endosomes at the plasma membrane in infected cells. The importance of late endosomes and exosome biogenesis for LASV release was also confirmed by electron microscopic characterization of LASV Z-positive extracellular vesicles and LASV-infected cells. Together, this doctoral project revealed previously unknown release mechanisms of LASV. A more comprehensive knowledge of virus-host interactions could open new possibilities for the development of treatment options against the highly pathogenic LASV

    Frühdiagnose akuter systemischer Infektionen und Prädiktion der bronchopulmonalen Dysplasie bei Frühgeborenen mittels einer elektronischen Nase (Cyranose)

    Jedes Jahr kommen in Deutschland rund 60.000 Kinder als Frühgeborene zur Welt, 1,3 % von ihnen gelten mit einem Geburtsgewicht von < 1500 g als sehr kleine Frühgeborene (IQTIG, 2021). Zu den häufigsten Gründen für Mortalität in diesem Patientenkollektiv gehören akute systemische Infektionen sowie chronische Atemwegserkrankungen wie die bronchopulmonale Dysplasie (BPD), welche europaweit mit einer Inzidenz von 10-25 % aller sehr kleinen Frühgeborenen auftritt (Gortner et al., 2011). Diese beruht auf einer durch die Frühgeburt bedingten gestörten Lungenentwicklung in Kombination mit prä- und postnatal einwirkenden Noxen, welche eine Entzündungreaktion hervorrufen. Besonders aufgrund der großen Vulnerabilität von Frühgeborenen ist die möglichst frühe Diagnose einer akuten systemischen Infektion sowie Prädiktion der BPD für den klinischen Verlauf der Erkrankungen und die Prognose des Frühgeborenen entscheidend. Die Etablierung einer geeigneten Methode zur gezielten Frühdiagnose und Prädiktion von Erkrankungen bei Frühgeborenen stellt die medizinische Forschung vor eine große Herausforderung. Durch Voruntersuchungen konnte gezeigt werden, dass die elektronische Nase (Cyranose), ein elektronisches Äquivalent zum biologischen Geruchssinn, möglicherweise als neues und nicht-invasives Diagnostiktool in der Neonatologie einsetzbar ist. Diese ist in der Lage, volatile organische Substanzen (volatile organic compounds, VOCs) über einer Probe zu detektieren und zu einem „Smellprint“, einer Art Fingerabdruck der jeweiligen Probe, zusammenzufassen. Diese Smellprints können daraufhin statistisch miteinander verglichen werden. Das übergeordnete Ziel dieser Studie war es, zu untersuchen, ob mittels Cyranose zwischen Proben von Frühgeborenen mit und ohne sich entwickelnder BPD und Frühgeborenen mit und ohne akuter systemischer Infektion differenziert werden kann. Da besonders nicht-invasiv gewonnene Proben für die potenzielle klinische Nutzung interessant sind, wurden Urin-, Stuhl-, Speichel- und Trachealaspiratproben untersucht. Zu diesem Zweck wurden 21 Frühgeborene des Universitätsklinikums Marburg über einen Zeitraum von sechs Wochen postnatal oder bis zum Zeitpunkt ihrer Entlassung aus der stationären Behandlung beobachtet. Wenn möglich, wurden wöchentlich Urin, Stuhl und Speichel sowie im Rahmen der Erstintubation, kurz vor der Extubation oder bei klinischer Indikation zum Absaugen Trachealaspirat gewonnen. Diese Proben wurden daraufhin mittels Cyranose untersucht und durch eine lineare Diskriminanzanalyse statistisch miteinander verglichen. Hierbei konnte anhand von Trachealaspirat-, Urin- und Stuhlproben zwischen Smellprints von Kindern mit und ohne einer akuten systemischen Infektion sowie mit und ohne sich entwickelnder BPD differenziert werden. Die im Rahmen dieser Dissertation durchgeführte Pilotstudie konnte die Machbarkeit bestätigen und wichtige Ergebnisse liefern, welche nun mittels größerer Studien überprüft werden müssen. Zusammenfassend ermutigen die in dieser Arbeit gefundenen Hinweise zu weiterer Forschung, um das Potenzial der Cyranose zur Frühdiagnose akuter systemischer Infektionen und Prädiktion der BPD bei Frühgeborenen anhand von Urin-, Stuhl- und Trachealaspiratproben weiter zu untersuchen.Every year approximately 60,000 children are born prematurely in Germany, 1.3 % of whom are considered very small preterm infants with a birth weight of < 1500 g (IQTIG, 2021). The most common causes of mortality in this patient population include acute systemic infections as well as chronic respiratory diseases such as bronchopulmonary dysplasia (BPD), which has an incidence of 10-25 % of all very small preterm infants across Europe (Gortner et al., 2011). This is due to a disturbed lung development caused by the premature birth in combination with pre- and postnatally acting noxae, which cause an inflammatory reaction. Especially due to the high vulnerability of preterm infants, the earliest possible diagnosis of acute systemic infection as well as prediction of BPD is crucial for the clinical course of the diseases and the prognosis of the preterm infant. The establishment of a suitable method for targeted early diagnosis and prediction of diseases in preterm infants is a major challenge for medical research. Preliminary studies have shown that the electronic nose (cyranose), an electronic equivalent to the biological sense of smell, may be applicable as a new and non-invasive diagnostic tool in neonatology. The cyranose is able to detect volatile organic compounds (VOCs) over a sample and aggregating them into a "smellprint", a kind of fingerprint of the sample. These smellprints can then be statistically compared with each other. The overall aim of this study was to investigate whether the cyranose can be used to differentiate between samples from preterm infants with and without developing BPD and preterm infants with and without acute systemic infection. Because noninvasively obtained samples are of particular interest for potential clinical use, urine, stool, saliva, and tracheal aspirate samples were examined. For this purpose, 21 premature infants of the University Hospital Marburg were observed over a period of six weeks postnatally or until the time of their discharge from inpatient treatment. When possible, urine, stool, and saliva were obtained weekly, as well as tracheal aspirate during initial intubation, shortly before extubation, or when clinically indicated for aspiration. These samples were then analyzed by the cyranose and statistically compared by linear discriminant analysis. Here, tracheal aspirate, urine, and stool samples could be used to differentiate between smellprints from children with and without an acute systemic infection and with and without developing BPD. The pilot study conducted as part of this dissertation was able to confirm the feasibility and provide important results, which now need to be verified by means of larger studies. In summary, the evidence found in this work encourages further research to investigate the potential of the cyranose to detect acute systemic infections and predict BPD in preterm infants using urine, stool, and tracheal aspirate samples

    Harmonization of Meteosat First and Second Generation Datasets for Fog and Low Stratus Studies

    Operational weather satellites, dating back to 1970s, currently provide the best basis for climatological investigations, such as an analysis of changes in the cloud cover. Because clouds are highly dynamic in time, temporally high-resolution data from the geostationary orbit are preferred in order to take variations in the diurnal cycles into account. For such studies, a consistent dataset in space and time is mandatory, but not yet available. Ground-based point measurements of various cloud parameters, such as ceiling, visibility, and cloud type are often sparsely spread and inconsistent, making it difficult to derive reliable spatio-temporal information over large areas. The Meteosat program has generally provided suitable data from over Europe since 1977, but different spatial, spectral, and radiometric resolution of the instruments of the individual satellites, including early-years calibration uncertainties, makes harmonization necessary to finally derive a time series applicable to any kind of climatological study. In this study, a machine learning-based approach has been employed to generate a long-term consistent dataset with high spatio-temporal resolution and extensive coverage over Europe by the harmonization of Meteosat First Generation (MFG) and Meteosat Second Generation (MSG) satellite datasets (1991–2020). A random forest (RF) regressor is trained on the overlap period (2004–2006), where datasets of both satellite generation (MFG and MSG) are available to predict MFG Water Vapour (WV) and Infrared (IR) channels brightness temperature (BT) values based on MSG channels. The aim of the study is to synthesize MFG MVIRI data from MSG SEVIRI to generate a consistent MFG time series. The results indicate a good match of MFG synthesized data with the original MFG data with a mean absolute error of 0.7 K for the WV model and 1.6 K for the IR model, and an out-of-bag (OOB) R² score of 0.98 for both the models. Based on the trained models, the MFG scenes are synthesized from the MSG scenes for the years from 2006 to 2020. The long-term homogeneity of the generated time series is analyzed. The harmonized dataset will be applied to generate a continuous time series on fog and low stratus (FLS) occurrence for a climatological time scale of 30 years.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Binaural acoustic stimulation in patients with Parkinson's disease

    Acoustic stimulation can improve motor symptoms in Parkinson’s disease (PD) and might therefore represent a potential non-invasive treatment option. Scalp electroencephalography studies in healthy subjects indicate that specifically binaural beat stimulation (BBS) in the gamma frequency range is associated with synchronized cortical oscillations at 40 Hertz (Hz). Several studies suggest that oscillations in the gamma-frequency range (>30 Hz) serve a prokinetic function in PD. In this double-blind, randomized study, 25 PD patients were recruited. The study was conducted with (ON) and without dopaminergic medication (OFF). Each drug condition consisted of two phases (no stimulation and acoustic stimulation). The acoustic stimulation phase was divided into two blocks including BBS and conventional acoustic stimulation (CAS) as a control condition. For BBS, a modulated frequency of 35 Hz was used (left: 320 Hz; right: 355 Hz) and for CAS 340 Hz on both sides. We assessed effects on motor performance using Movement Disorder Society-Unified Parkinson’s Disease Rating Scale (MDS-UPDRS) and two validated commercially available portable devices (Kinesia ONE™ and Kinesia 360™) measuring motor symptoms such as dyskinesia, bradykinesia, and tremor. Repeated measures ANOVA revealed that BBS improved resting tremor on the side of the more affected limb in the OFF condition, as measured by wearables (F(2,48) = 3.61, p = 0.035). However, BBS did not exert a general positive effect on motor symptoms as assessed via MDS-UPDRS (F(2,48) = 1.00, p = 0.327). For CAS, we did not observe an improvement in specific symptoms but rather an overall beneficial effect on motor performance (MDS-UPDRS total score OFF medication: F(2,48) = 4.17, p = 0.021; wearable scores: F(2,48) = 2.46, p = 0.097). In this study, we found an improvement of resting tremor when applying BBS in the gamma frequency band OFF medication. Moreover, the positive effects of CAS underline the general positive potential for improvement of motor function by acoustically supported therapeutic approaches.     Enabling Navigation and Augmented Reality in the Sitting Position in Posterior Fossa Surgery Using Intraoperative Ultrasound

    Despite its broad use in cranial and spinal surgery, navigation support and microscopebased augmented reality (AR) have not yet found their way into posterior fossa surgery in the sitting position. While this position offers surgical benefits, navigation accuracy and thereof the use of navigation itself seems limited. Intraoperative ultrasound (iUS) can be applied at any time during surgery, delivering real-time images that can be used for accuracy verification and navigation updates. Within this study, its applicability in the sitting position was assessed. Data from 15 patients with lesions within the posterior fossa who underwent magnetic resonance imaging (MRI)-based navigation-supported surgery in the sitting position were retrospectively analyzed using the standard reference array and new rigid image-based MRI-iUS co-registration. The navigation accuracy was evaluated based on the spatial overlap of the outlined lesions and the distance between the corresponding landmarks in both data sets, respectively. Image-based co-registration significantly improved (p < 0.001) the spatial overlap of the outlined lesion (0.42 ± 0.30 vs. 0.65 ± 0.23) and significantly reduced (p < 0.001) the distance between the corresponding landmarks (8.69 ± 6.23 mm vs. 3.19 ± 2.73 mm), allowing for the sufficient use of navigation and AR support.     Acceptance of Climate-Oriented Policy Measures in Times of the COVID-19 crisis

    Based on data from a representative survey among citizens in Germany during the peak of the COVID-19 crisis, this paper empirically examines the acceptance of climate-oriented economic stimulus programs and several further climate policy measures. Our descriptive analysis shows no general lower acceptance of climate policy measures compared to the time before the crisis. However, the econometric analysis reveals that individuals with higher negative emotions towards the crisis are significantly less supportive of at least some climate-oriented policy measures. Economic concerns are of particular relevance. For example, a perceived deterioration of the general economic situation due to the COVID-19 crisis has a significantly negative effect on the acceptance of climate-oriented economic stimulus programs. Concerns about the own personal economic and financial situation due to the crisis are significantly negatively correlated with the support of climate-oriented policy measures that directly lead to higher costs in daily life. Besides the relevance of this perceived self-interest, our estimation results also highlight the relevance of social aspects since individuals with a social policy identification are significantly more likely to agree with climate-oriented policy measures that are also financially beneficial for socially de-prived groups, but significantly less likely to support measures that are financially unfa-vorable for them. We discuss several climate policy implications. For example, our estima-tion results suggest that successful climate policy should, especially in times of the COVID-19 crisis, also be socially oriented and consider distribution effects, for example, through financial compensations for costly measures like taxes

Damit können teilweise auch christliche Bedeutungsdimensionen verbunden werden. • Diese Schemata erscheinen vertraut, generieren erhöhte Aufmerksamkeit und nicht selten auch Empathie. • Christliche Codes erfahren durch Medienikonen eine Aktualisierung im Medium der Fotografie. • Durch das Anknüpfen an die christliche (Leidens-)Ikonografie wird der Ikonisierungsprozess von Bildern begünstigt, teilweise sogar erst ermöglicht.The work pursues the following theses: - Schemata that have been shaped by the Christian pictorial tradition are present in the medium of photography (even outside of religious contexts).     (Thio)Carbonyl Pseudohalides – Synthesis, Spectroscopy and Reactivity

    Der Fokuspunkt der Forschung in dieser Dissertation war die Synthese und Charakterisierung (erster Teil), sowie die Reaktivität (zweiter Teil) von Carbonyldipseudohalogeniden (CDPsH). Die Kristallstrukturen von Carbonyldiisothiocyanat (CO(NCS)2) und Oxalyldiisothiocyanat ((CONCS)2) wurden ebenso wie ihre Hirshfeld Oberflächen und spektroskopischen Signaturen aufgeklärt. Diese Erkenntnisse erweitern das Wissen über Syntheseverfahren, Konformation und Packung der Moleküle im Festkörper von kleinen Molekülen. Des Weiteren wurde eine Konformationsanalyse in Kooperation mit der Arbeitsgruppe SCHNELL (DESY in Hamburg) von Carbonyldiisothiocyanat mittels chirped-pulse Fourier-Transformation Mikrowellenspektroskopie durchgeführt, in welcher ein syn-syn zu syn-anti Verhältnis von 10:1 beobachtet wurde. Das syn-syn Konformer existiert dabei wie ein Boson in den zwei Kernspinisomeren ortho und para. Darüber hinaus wurde die photochemisch induzierte Zersetzung von Oxalyldiisothiocyanat in einer festen Argon-Matrix mittels IR-Spektroskopie in Zusammenarbeit mit der Arbeitsgruppe WAGNER (Eberhard Karls Universität Tübingen) untersucht. Bei Bestrahlung mit UV Licht wird Kohlenstoffmonoxid abgespalten und Carbonyldiisothiocyanat in seinem energetisch höherliegendem syn-anti Konformer gebildet. Tempern der Matrix nach Bestrahlung bei erhöhten Temperaturen (30 K) führt zu einer Konformationsanderung zum stabileren syn-syn Konformer. Viele Syntheseversuche von CDPsH wie z,B. Carbonyldiselenocyanat oder Thiocarbonyldiisocyanat führten nicht zu isolierbaren Produkten. Carbonyldiselenocyanat (CO(SeCN)2) wurde möglicherweise in Lösung durch 13C-NMR-Spektroskopie bei einer chemischen Verschiebung von 185.3 ppm und 98.1 ppm beobachtet. Das erhaltene feste Produkt zersetzt sich jedoch oberhalb von −30 °C oder bei Solvatisierung. Aufgrund dessen wurde das alkylsubstituierte tert-Butylcarbonylisoselenocyanat (CO(NCSe)tBu) synthetisiert, um die Existenz von Carbonylselenocyanaten zu überprüfen. Die Verbindung konnte als festes Rohprodukt aufgrund Zersetzung während dem Entfernen des Lösungsmittels nicht isoliert werden. In Lösung ist die Verbindung jedoch handhabbar und kann mittels 13C-NMR-Spektroskopie mit Resonanzsignalen bei 171.4 ppm und 144.0 ppm detektiert werden. Die Synthese von Pseudohalogeniden mit Thiocarbonylgruppe wurde zuerst für Thiocarbonyldiisocyanat (CS(NCO)2) versucht. Es konnte auf den durchgeführten Wegen nicht erhalten werden, aber es wurde in der Zwischenzeit von HENNES GÜNTHER (Arbeitsgruppe TAMBORNINO) in Lösung beobachtet und ein definiertes Folgeprodukt mit Ethanol erzeugt. Die Synthese der anschließenden Verbindung Thiocarbonyldiisothiocyanat (CS(NCS)2) führte zur Isolierung von dem bekannten, wenn auch bisher schlecht charakterisierten, Thiocarbonyldithiocyanat (CS(NCS)2). Obwohl erstes das thermodynamisch stabilere Reaktionsprodukt ist, war es nicht möglich eine synthetische Herangehensweise, welche zu seiner Isolation führte, zu finden. DFT Rechnungen bestätigten, dass das kinetische Reaktionsprodukt Thiocarbonyldithiocyanat (CS(NCS)2) über einen niedrigeren Übergangszustand gebildet wird. Dessen Kristallstruktur wurde zusammen mit der des einfach substituierten Chlorthiocarbonylthiocyanats (CS(SCN)Cl) bestimmt. Dies resultiert in der sechsten bekannten Molekülstruktur von CDPsH. Zuletzt wurden Versuche zur Synthese von Thiocarbonyldiselenocyanat (CS(SeCN)2) unternommen, die zur möglichen Detektion durch 13C-NMR-Spektroskopie mit Resonanzsignalen bei 185.8 ppm und 103.2 ppm führte. Die Verschiebungen stimmen mit den Beobachtungen zum Syntheseversuch von Carbonyldiselenocyanat (CO(SeCN)2) überein und bekräftigen die oben genannten Schlussfolgerungen. Zum Abschluss des ersten Teiles wurden Oxalylchloridisothiocyanat (Cl(CO)2NCS) und Chlorcarbonylisothiocyanat (CO(NCS)Cl) synthetisiert. Im Verlauf der Synthese von Oxalylchloridisothiocyanat wurde ein Gleichgewicht von diesem und 2-Chlorothiazol-4,5-dion in Lösung festgestellt. Weitere Untersuchungen mit Einkristalldiffraktometrie und Schwingungsspektroskopie führte zu der Erkenntnis, dass nur letzteres im Festkörper vorliegt. Beide Verbindungen zersetzen sich in Lösung bei Raumtemperatur zu Chlorocarbonylisothiocyanat, welches genauer von SVEN RINGELBAND (Arbeitsgruppe TAMBORNINO) untersucht wurde. Der zweite Teil dieser Dissertation beschäftigt sich mit der Reaktivität von CDPsH. Carbonyldiisocyanat (CO(NCO)2) und Carbonyldiisothiocyanat (CO(NCS)2) reagieren mit Halogenwasserstoffen jeweils zu Carbonylbis(carbamoylhalogeniden) und halogensubstituierten 1,3,5-Thiadiazin-4-onen. Carbonyldiisocyanat addiert zwei Äquivalente des Nukleophils, wohingegen Carbonyldiisothiocyanat unter intramolekularem Ringschluss nur ein Äquivalent addiert. Die verschiedenen Reaktionspfade sind thermodynamisch gesteuert. Während Sauerstoff die Bildung von ex-Ring Doppelbindungen mit Kohlenstoff präferiert, bevorzugt Schwefel in-Ring Einfachbindungen zu bilden. Beide Verbindungsklassen zeigen im Festkörper ausgedehnte Netzwerke von Wasserstoffbrückenbindungen, welche im Detail anhand der Reaktionsprodukte zwischen Carbonyldiiso(thio)cyanat und Nukleophilen wie Alkoholen, Thiolen und Aminen erläutert wurden. N,N’-Carbonylbis(carbamate), -bis(S-thiocarbamate) und N,N’-Biscarbamoylharnstoffe werden jeweils in der Reaktion mit Carbonyldiisocyanat erzeugt. Alle Moleküle bilden eine intramolekulare und zwei intermolekulare Wasserstoffbrückenbindungen zur Bildung von Dimeren oder Ketten. Aus der Reaktion von Nukleophilen mit Carbonyldiisothiocyanat entstehen substituierte 1,3,5-Thiadiazin-4-one, die alle über intermolekulare Wasserstoffbrückenbindungen Dimere bilden. Dies steht im Gegensatz zu den Ketten bildenden halogensubstituierten 1,3,5-Thiadiazin-4-onen. Die weitere Packung der Moleküle aller Verbindungsklassen ist abhängig von der Größe der substituierten Gruppe, z.B. Methyl gegen Phenyl. Bei kleinen Alkylgruppen überwiegen polare Wechselwirkungen, während mit zunehmender Größe der Alkyl- oder Arylgruppe steigend unpolare Wechselwirkungen vorherrschen. Einige Nebenprodukte der Reaktionen von Carbonyldiisocyanat und Carbonyldiisothiocyanat mit Nukleophilen konnten isoliert und als z.B. Derivate der Isocyanursäure und des Biurets identifiziert werden. Diese konnten Hinweise auf mögliche reaktive Spezies in der Lösung während der Reaktion sein. Nukleophile Additionen von Organolithium-Verbindungen an Carbonyldiisocyanat wurden mit Methyllithium, tert-Butyllithium und Lithiummethanolat getestet, aber definierte Reaktionsprodukte wurden nicht erhalten. Es wird angenommen, dass die gebildeten Anionen zur Isolierung zu instabil sind, weshalb eine saure Aufarbeitung zur Protonierung dieser notwendig sein wird. Überraschend aufregend war das Ergebnis der Reaktion von Carbonyldiisocyanat mit Eisenbis(diisopropylphenyltrimethylsilylamid), in welcher eine Reaktion mit dem Liganden und die Kristallisation eines neuartigen Metallkomplexes erreicht wurde. Dieser neue Komplex mit einem substituiertem Biuret-Liganden und einem tetraedrisch koordiniertem Eisen ist der erste über den berichtet wird. Kristalle aus derselben Reaktion mit Cobalt oder Nickel, ebenso wie Carbonyldiisothiocyanat wurden nicht erhalten. Allerdings wurde ein äquivalenter Metallkomplex aus Nickel und Carbonyldiisothiocyanat durch Massenspektrometrie beobachtet. Dies beweist, dass die Reaktions- oder Kristallisationsbedingungen angepasst werden müssen, um mehr dieser Komplexe zu erhalten. Zukünftige Perspektiven dieser Dissertation sind zunächst in Kooperation das Rotationsspektrum von Carbonyldiisocyanat, das IR-Spektrum von Oxalylchloridisothiocyanat beziehungsweise 2-Chlorthiazol-4,5-dion in fester Argon-Matrix und deren Verhalten bei Bestrahlung mit UV Licht zu untersuchen. Des Weiteren sollte die Isolierung einer reinen Probe und die Charakterisierung der Festkörper von Carbonyldiselenocyanat und Thiocarbonyldiselenocyanat durch Filtration über eine gekühlte Schlenk-Fritte oder die Verwendung von anderen Losungsmitteln wie z.B. Dioxan möglich sein. Ein komplett neuer Ansatz wäre die Synthese ausgehend von (Thio)Harnstoff, Kohlenstoffdiselenid und Quecksilberchlorid. Darüber hinaus ist es lohnenswert einen Blick auf die fehlenden NMR spektroskopischen und Molekülstrukturdaten von Carbonyldicyanid (CO(CN)2), das in der Einleitung erwähnt wurde, zu werfen. Asymmetrische Carbonyldipseudohalogenide finden in dieser Dissertation keine Beachtung, obwohl Studien zu ihrer Synthese, Charakterisierung und Reaktivität das Wissen über Konformation und Kristallpackung von kleinen Molekülen deutlich verbessern würden. Abseits der Synthese von neuen Carbonylpseudohalogeniden ist auch die Untersuchung der Edukte Phosgen, Thiophosgen und Oxalylchlorid von Interesse. So sind bereits Experimente zur Neutronenbeugung an Phosgen und Thiophosgen am ISIS in Oxford geplant.In this dissertation the synthesis and characterization (first part), as well as the reactivity (second part) of carbonyl dipseudohalides (CDPsH) were the focal point of research. The crystal structures of carbonyl diisothiocyanate (CO(NCS)2) and oxalyl diisothiocyanate ((CONCS)2) were elucidated alongside their Hirshfeld surfaces, as well as their spectroscopic signatures. These findings extend the knowledge about synthetic procedure, conformation and molecule packing in the solid state of small molecules. Furthermore, a conformer study of carbonyl diisothiocyanate was performed by chirped-pulse Fourier transform microwave spectroscopy in cooperation with the working group SCHNELL (DESY in Hamburg) and led to the observation of a syn-syn to syn-anti ratio of 10:1. The syn-syn conformer exists as two nuclear spin isomers ortho and para like a boson. In addition, the photochemical induced decomposition of oxalyl diisothiocyanate in solid argon matrices was investigated with IR spectroscopy in collaboration with working group WAGNER (Eberhard Karls University Tubingen). Upon irradiation with UV light carbon monoxide is released and forms carbonyl diisothiocyanate in its less favored syn-anti conformer. Annealing the matrix after irradiation at higher temperature (30 K) leads to a conformer switch to the more stable syn-syn conformer. Many attempts to synthesize other carbonyl dipseudohalides (CDPsH) e.g. carbonyl diselenocyanate or thiocarbonyl diisocyanate did not end in isolable products. Carbonyl diselenocyanate (CO(SeCN)2) could possibly be observed in solution by 13C-NMR spectroscopy with chemical shifts of 185.3 ppm and 98.1 ppm, but the collected solid product decomposes above −30 °C or during dissolution. Therefore, the alkyl substituted tert-butylcarbonyl isoselenocyanate (CO(NCSe)tBu) was synthesized to confirm the existence of carbonyl selenocyanates. The isolation of this compound as crude solid was not possible due to decomposition during solvent removal, but it is manageable in solution and detectable by 13C-NMR spectroscopy with signals at 171.4 ppm and 144.0 ppm. The synthesis of thiocarbonyl group containing pseudohalides was first attempted for thiocarbonyl diisocyanate (CS(NCO)2). It was not possible to synthesize this compound through the employed synthetic routes, but in the meantime HENNES GÜNTHER (working group TAMBORNINO) could observe it in solution and isolate a defined reaction product with ethanol. The next compound is thiocarbonyl diisothiocyanate (CS(NCS)2) from which the synthesis yielded in the isolation of the known, albeit hitherto poorly characterized, thiocarbonyl dithiocyanate (CS(SCN)2). Despite the former being the thermodynamically more stable reaction product, it was not possible to find a synthetic pathway resulting in the isolation of the pure compound. DFT calculations confirmed that the kinetic reaction product thiocarbonyl dithiocyanate (CS(SCN)2) is formed via a lower transition state. Its crystal structure was determined along with the monosubstituted chlorothiocarbonyl thiocyanate (CS(SCN)Cl), which concludes to the sixth known structure of CDPsH. Lastly, an effort to synthesize thiocarbonyl diselenocyanate (CS(SeCN)2) was made and resulted in the possible detection by 13C-NMR spectroscopy with signals at 185.8 ppm and 103.2 ppm. The shifts match the observation made for the synthesis attempt of carbonyl diselenocyanate (CO(SeCN)2) and underline the above stated conclusions. At the end of part one, oxalylchloride isothiocyanate (Cl(CO)2NCS) and chlorocarbonyl isothiocyanate (CO(NCS)Cl) were synthesized. During the synthesize of oxalylchloride isothiocyanate an equilibrium with 2-chlorothiazol-4,5-dione in solution was observed. Further investigations with SC-XRD analysis and vibrational spectroscopy led to the conclusion that only the latter one exists in the solid state. Both compounds decompose in solution at room temperature to chlorocarbonyl isothiocyanate, which was extensively studied by SVEN RINGELBAND (working group TAMBORNINO). The second part of this dissertation deals with the reactivity of CDPsH. Carbonyl diisocyanate (CO(NCO)2) and -diisothiocyanate (CO(NCS)2) react with hydrogen halides to form carbonyl bis(carbamoylhalides) and halogenido substituted 1,3,5-thiadiazine-4- ones, respectively. Carbonyl diisocyanate adds two equivalents nucleophile, while carbonyl diisothiocyanate only adds one and undergoes intramolecular ring-closure. The different reaction pathways are thermodynamically driven by the preference of oxygen to form an ex-ring double bond versus the sulfur to form in-ring single bonds with carbon atoms. Both compound classes show extended hydrogen bonding networks in the solid state, which were examined in detail by the reaction products between carbonyl diiso(thio)cyanate and nucleophiles e.g. alcohols, thiols and amines. N,N’-Carbonyl bis(carbamates), -bis(S-thiocarbamates) and N,N’-biscarbamoyl ureas are formed during the reaction with carbonyl diisocyanate, respectively. All molecules form an intramolecular hydrogen bond and intermolecular hydrogen bonds to form either dimers or chains. The reaction of nucleophiles with carbonyl diisothiocyanate yields substituted 1,3,5-thidiazine-4-ones, all of which form dimers via intermolecular hydrogen bonds. This is in contrast to the chain forming halogenido substituted 1,3,5-thiadiazine-4-ones. Further packing of the molecules of all compound classes depends on the size of the substituted group, e.g. methyl versus phenyl. For small alkyl groups polar interactions predominate, while non-polar interactions dominate as the size of the alkyl or aryl groups increases. These reactions of carbonyl diisocyanate and carbonyl diisothiocyanate with nucleophiles led to the crystallisation of some side products like derivatives of isocyanuric acid and biuret, which could possibly hint at reactive species being present in solution during the reaction. Nucleophilic additions of organolithium compounds to carbonyl diisocyanate were tested with methyl lithium, n-butyl lithium and lithium methoxide, but defined reaction products were not collected. It is assumed that the formed anions are too unstable to isolate, therefore an acidious workup to protonate them is suggested. Surprisingly exciting was the result of the reaction of carbonyl diisocyanate with iron bis(diisopropylphenyltrimethylsilyamide), in which a reaction with the ligands and crystallization of a new type of metal complex was observed. This new complex, featuring a substituted biuret ligand and tetrahedral coordination of iron, is the first one to be reported. While crystals of reactions with cobalt or nickel, as well as carbonyl diisothiocyanate were not obtained, the equivalent metal complex from nickel and carbonyl diisothiocyanate was observed in mass spectrometry. This proofs that the reaction or crystallization conditions need to be adjusted in order to obtain more of these complexes. Future prospects of this dissertation are to investigate in collaboration the rotational spectrum of carbonyl diisocyanate, the IR spectrum of oxalyl chloride isothiocyanate or rather 2-chlorothiazol-4,5-dione in solid argon-matrix and its behavior upon irradiation with UV light. Further, the isolation of a pure sample and solid-state characterization of carbonyl diselenocyanate and thiocarbonyl diselenocyanate should be possible by filtration over a cooled frit or the use of other solvents e.g. dioxane. A complete new approach would be the synthesis from (thio)urea, carbon diselenide and mercury chloride. In addition, it is worth taking a look at the missing NMR spectroscopic and molecular structure data of carbonyl dicyanide (CO(CN)2) mentioned in the introduction. In this work asymmetrical carbonyl dipseudohalides were not considered, but to study their synthesis, characterization and reactivity should enhance the knowledge of conformations and crystal packing of small molecules even further. Aside from the synthesis of new carbonyl pseudohalides an investigation of the starting materials phosgene, thiophosgene and oxalyl chloride is also of interest. For instance, neutron diffraction experiments of phosgene and thiophosgene at ISIS in Oxford are planned
