    Development of a Novel Enzyme-Linked Immunosorbent Assay and Lateral Flow Test System for Improved Serodiagnosis of Visceral Leishmaniasis in Different Areas of Endemicity

    Visceral leishmaniasis (VL) is caused by protozoan parasites of the Leishmania donovani complex and is one of the most prominent vector-borne infectious diseases with epidemic and mortality potential if not correctly diagnosed and treated. East African countries suffer from a very high incidence of VL, and although several diagnostic tests are available for VL, diagnosis continues to represent a big challenge in these countries due to the lack of sensitivity and specificity of current serological tools. Based on bioinformatic analysis, a new recombinant kinesin antigen from Leishmania infantum (rKLi8.3) was developed. The diagnostic performance of rKLi8.3 was evaluated by enzyme-linked immunosorbent assay (ELISA) and lateral flow test (LFT) on a panel of sera from Sudanese, Indian, and South American patients diagnosed with VL or other diseases, including tuberculosis, malaria, and trypanosomiasis. The diagnostic accuracy of rKLi8.3 was compared with rK39 and rKLO8 antigens. The VL-specific sensitivity of rK39, rKLO8, and rKLi8.3 ranged from 91.2% over 92.4% to 97.1% and specificity ranged from 93.6% over 97.6% to 99.2%, respectively. In India, all tests showed a comparable specificity of 90.9%, while the sensitivity ranged from 94.7% to 100% (rKLi8.3). In contrast to commercial serodiagnostic tests, rKLi8.3-based ELISA and LFT showed improved sensitivity and no cross-reactivity with other parasitic diseases. Thus, rKLi8.3-based ELISA and LFT offer improved VL serodiagnostic efficiency in East Africa and other areas of endemicity.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Identification of prognostic cell populations in patients with chronic lymphocytic leukemia by applying explainable artificial intelligence to flow cytometric data

    Einleitung: Die Chronische lymphatische Leukämie (CLL) ist die häufigste leukämische Erkrankung in den westlichen Ländern. Ein etabliertes Prognosemodell für CLL-Patienten ist der International Prognostic Index (CLL-IPI), der auf fünf Prognosefaktoren basiert (TP53-Status, IGHV-Mutationsstatus, beta2-Mikroglobulin im Serum, klinisches Stadium und das Alter). In dieser Arbeit wird untersucht, ob eine CLL-Prognose möglich ist, indem ausschließlich Multiparameter-Durchflusszytometriedaten (MPFC) von CLL-Patienten mit Hilfe von einer erklärbaren künstlichen Intelligenz (XAI) analysiert werden. Dabei sollte die XAI die Ergebnisse basierend auf der Darstellung von unterschiedlichen, für die Prognose relevanten Zellpopulationen in MPFC-Punktdiagrammen erklären können. Material und Methoden: Insgesamt erfolgte die Analyse von MPFC-Daten aus dem peripheren Blut von 157 Patienten mit CLL und der Abgleich mit klinischen Daten (Parameter des CLL-IPI, Geschlecht, ECOG-Stadium, Richter-Transformation ja/nein, Behandlung ja/nein, Todesdatum, Behandlungsversagen und Datum der letzten Nachuntersuchung). Die MPFC-Daten wurden erhoben für die routinemäßige diagnostische Analyse am Universitätsklinikum Marburg von 2014 bis 2020. Die Patienten wurden in eine Gruppe mit schlechterem und eine Gruppe mit besserem klinischen Verlauf eingeteilt. Patienten, die während der Nachbeobachtung verstarben sowie diejenigen bei denen die systemische Erstlinientherapie nicht angesprochen hat, wurden als TTF 1 (Time-to-First-Line-Treatment-Failure bzw. Zeit bis zum Versagen der Erstlinientherapie) kategorisiert, d. h. der Gruppe mit schlechterem klinischen Verlauf zugewiesen. Alle anderen Patienten wurden mit TTF 0 einem besseren Ergebnis zugeordnet. Anschließend wurde der XAI-Algorithmus ALPODS verwendet, um Zellpopulationen zu identifizieren, die bei Patienten mit schlechterem (TTF 1) oder besserem klinischen Verlauf (TTF 0) über- oder unterrepräsentiert waren und damit eine Vorhersage zur Prognose treffen können. Dabei wurde die prädiktive Fähigkeit jeder einzelnen XAI-Population mit Hilfe von Receiver-Operating-Characteristic-Kurven (ROC-Kurven) ausgewertet. Zur Überprüfung wurde dieser prädiktive Wert der XAI-Populationen mit der Häufigkeit von CD38-positiven CLL-Zellen und CLL-IPI auf den ROC-Kurven verglichen. Ergebnisse: ALPODS definierte 17 XAI-Populationen, die zusammengenommen eine genauere Prognose als der CLL-IPI-Score treffen können (AUC der XAI-Populationen: 0,95 [95%KI 0,91-0,98; p <0,0001] vs. AUC von CLL-IPI: 0,78 [95%KI 0,70-0,86; p<0,0001]). Der beste Einzelklassifikator war eine XAI-Population bestehend aus CD4+ T-Zellen (AUC 0,78; 95%KI 0,70–0,86; p<0,0001). Patienten mit weniger CD4+ T-Zellen zeigten einen schlechteren klinischen Verlauf. Bei Hinzufügen der CD4+ T-Zellpopulation steigerte sich die Vorhersagefähigkeit des CLL-IPI-Scores (AUC 0,83; [95%KI 0,77–0,90; p<0,0001]). Alleine mit einer Kombination der CD4+ mit der CD8+ T-Zellpopulation ließ sich in 79% der Fälle der betrachteten Patientenkohorte korrekt unterscheiden zwischen TTF 0 und TTF 1 (AUC 0,79; [95%KI 0,71-0,87; p<0,0001]). Dies ist insbesondere vor dem Hintergrund wichtig, dass sich diese beiden Zellpopulationen manuell leicht in durchflusszytometrischen Punktdiagrammen erfassen ließen, gerade im Vergleich zu anderen von ALPODS definierten Zellpopulationen, deren korrektes manuelles Gating zum Teil sehr anspruchsvoll ist (insbesondere das der CLL-Untergruppen). Schlussfolgerung: Der ALPODS-XAI-Algorithmus war in der Lage prädiktive Immunzellpopulationen zu identifizieren, die den klinischen Verlauf einer CLL signifikant vorhersagen können. CD4+ T-Zellen wurden als bester Einzelklassifikator identifiziert, wobei sie in Kombination mit dem CLL-IPI-Score die Vorhersagefähigkeit nochmal verbessern konnten. Darüber hinaus konnte mit dieser Arbeit gezeigt werden, dass mit dem in der Klinik relativ einfach durchführbarem Gating von CD4+ T-Zellen in Kombination mit den CD8+ T-Zellen ebenfalls eine klinische Prognose getroffen werden kann. Die Ergebnisse müssen an einer wesentlich größeren Patientenkohorte validiert werden, sind aber sehr vielversprechend was die Verfeinerung konventioneller Prognosemodelle wie den CLL-IPI betrifft.Introduction: Chronic lymphocytic leukemia (CLL) is the most common leukemic disease in Western countries. An established prognostic model for CLL patients is the International Prognostic Index (CLL-IPI), which is based on five prognostic factors (TP53 status, IGHV mutation status, serum beta2-microglobulin, clinical stage, and age). In this work, we investigate whether CLL prognosis is possible by exclusively analyzing multiparameter flow cytometry (MPFC) data from CLL patients using an explainable artificial intelligence (XAI). Here, the XAI should be able to explain the results based on the representation of different cell populations relevant for prognosis in MPFC dot plots. Material and Methods: Analysis of MPFC data from peripheral blood of 157 patients with CLL was performed in alignment with matching clinical data (parameters of CLL IPI, sex, ECOG stage, Richter transformation yes/no, treatment yes/no, date of death, treatment failure, and date of last follow-up). MPFC data were collected for routine diagnostic analysis at Marburg University Hospital from 2014 to 2020, and patients were divided into a worse outcome group and a better outcome group. Patients who died during follow-up and those who failed first-line systemic therapy were categorized as TTF 1 (time-to-first-line treatment failure), i.e., assigned to the worse outcome group. All other patients were assigned to a better outcome with TTF 0. Subsequently, the XAI algorithm ALPODS was used to identify cell populations that were over- or underrepresented in patients with worse outcome (TTF 1) or better outcome (TTF 0) and thus predictive of prognosis. Here, the predictive ability of each XAI population was evaluated using receiver operating characteristic (ROC) curves. For verification, this predictive value of the XAI populations was compared with the frequency of CD38-positive CLL cells and CLL-IPI on the ROC curves. Results: ALPODS defined 17 XAI populations that collectively can predict more accurately than the CLL-IPI score (AUC of XAI populations: 0.95 [95%CI 0.91-0.98; p<0.0001] vs. AUC of CLL-IPI: 0.78 [95%CI 0.70-0.86; p<0.0001]). The best single classifier was an XAI population consisting of CD4+ T cells (AUC 0.78; 95% CI 0.70-0.86; p<0.0001). Patients with fewer CD4+ T cells showed a worse clinical outcome. When the CD4+ T cell population was added, the predictive ability of the CLL-IPI score increased (AUC 0.83; [95%CI 0.77-0.90; p<0.0001]). Combining the CD4+ with the CD8+ T cell population alone correctly distinguished TTF 0 from TTF 1 in 79% of the cases in the patient cohort considered (AUC 0.79; [95%CI 0.71-0.87; p<0.0001]). This is particularly important in light of the fact that these two cell populations could be easily gated manually in flow cytometric dot plots, especially compared to other cell populations defined by ALPODS, some of which are very challenging to correctly gate manually (especially that of CLL subgroups). Conclusion: The ALPODS-XAI algorithm was able to identify predictive immune cell populations that significantly predicted the clinical course of CLL. CD4+ T cells were identified as the best single classifier, further improving predictive ability when combined with the CLL-IPI score. In addition, this work demonstrated that gating CD4+ T cells in combination with the CD8+ T cells, which is relatively easy to perform in the clinic, can also be used to make a clinical prediction. The results need to be validated in a much larger cohort of patients, but are very promising in terms of refining conventional prognostic models such as the CLL-IPI

    Prudential Policies in the Eurozone: A Propensity Score Matching Approach

    This paper studies the effectiveness of micro- and macroprudential policy tools in the euro area. The established empirical literature on macroprudential policy generally considers panel estimations that suffer from two estimation biases, i.e., a selection bias and a time bias. We control for the former by a propensity score matching approach. Based on a logit model, we estimate the probability of a policy tightening for every country at each point in time. Matching procedures then find one or more matching partners for every tightening event with a similar likelihood of a tightening but no shift in the prudential policy stance. An iterative approach ensures that we offset the time bias, which exists if the estimation does not control for effects of pre- ceding and subsequent prudential policy changes. We find that the announcement of a prudential policy tightening reduces credit growth significantly by about 1% on average. We further differentiate between effects along three dimensions. First, we observe that lending is more affected when policymakers have not communicated the implementation of measures before. Second, the effects are more substantial when EU/EA institutions are behind changes in the prudential policy stance. Third, microprudential policy measures have a bigger impact than macroprudential policies

    Clinical utility of the PFA-100 as preoperative screening test for detecting disorders of primary haemostasis in patients undergoing complex, elective intracranial procedures for space-occupying masses

    Das menschliche Gehirn ist durch den umgebenden Liquor und die Schädelkalotte geschützt. Eine schnelle intrakranielle Drucksteigerung führt aufgrund der engen Verhältnisse allerdings frühzeitig zu teils irreversiblen Parenchymschäden. Blutungen stellen neben dem Hirnödem eine Haupt¬ursache für Hirndruckerhöhungen dar. Eine präoperative Detektion einer erhöhten Blutungsneigung wäre daher insbesondere vor intrakraniellen Eingriffen wünschenswert. Störungen der primären Hämostase, wie beispiels¬weise die von-Willebrand-Erkrankung mit einer Prävalenz von 1 - 2 % in der Normalbevölkerung, sind häufig unerkannte Ursachen für vermehrte Blutungsneigung. Zur Beurteilung der primären Hämostase wurde von Kundu et. al. 1995 der auf einem Versuchsaufbau von Kratzer und Born basierende PFA-100® entwickelt. Dieses einfach zu bedienende Gerät der Firma Dade Behring® simuliert die Thrombozytenadhäsion und -aggregation bei hohen Scherraten und findet mittlerweile Anwendung in vielen Gerinnungslabors. In der Klinik für Neurochirurgie der Universität Marburg wurde der PFA-100® im April 2005 als Screeningtest vor jeder großen elektiven, intrakraniellen Operationen eingeführt. In dieser Studie wurde retrospektiv untersucht, ob daraufhin eine Verbesserung des Outcomes und eine Verminderung der Blutungskomplikationen stattgefunden hat. Dazu wurden insgesamt 102 Patienten, die in den neun Monaten vor der Einführung des PFA operiert wurden, mit 93 Patienten, die innerhalb der ersten neun Monate nach der Einführung ein PFA-Screening erhalten haben und operiert wurden, verglichen. Dabei fanden sich keine wesentlichen Abweichungen in der Gruppenzusammensetzung. Hinsichtlich der demografischen Werte Alter, Größe, Gewicht, BMI und Geschlecht konnte kein signifikanter Unterschied festgestellt werden. Auch bezüglich des durchschnittlichen präoperativen Gesundheits- und Bewusstseinszustandes der Patienten (beurteilt anhand des ASA- Wertes und des präoperativen GCS) unterschieden sich die Gruppen nicht signifikant. In beiden Gruppen waren die verschiedenen Tumorentitäten (nach WHO) weitgehend ähnlich häufig vertreten. Eine Thrombozytopenie als Ursache für eine gestörte Hämostase konnte bei einem Patienten identifiziert werden, die durchschnittliche Thrombozytenzahl war in beiden Gruppen vergleichbar. Auch die übrigen erfassten Parameter zur groben Beurteilung der Gerinnung (INR, aPTT, Fibrinogen) und von Organfunktionen (Kreatinin) und der Hb-Wert wiesen keine signifikanten Unterschiede auf. Schließlich gab es auch keine wesentlichen Differenzen bei der Verteilung des Erfahrungsgrades des Operateurs. Die Ausgangssituation in den Gruppen war somit vergleichbar. Als Resultat konnten keine Hinweise für eine vermehrte intraoperative Blutungsneigung in einer Gruppe festgestellt werden. Die dazu erhobenen Parameter postoperativer Hb-Wert, OP- und Anästhesiedauer und Transfusionsrate von EKs, FFPs, Thrombozytenkonzentraten, und die Anwendung von Gerinnungsfaktorkonzentraten wie Fibrinogen und PPSB unterschieden sich nicht signifikant. Die Operateure waren angehalten, eine subjektiv empfundene vermehrte Blutungsneigung des Patienten im OP-Bericht zu erwähnen, wobei auch hier bei geringer Anzahl der Vermerke (insgesamt 10) kein signifikanter Unterschied in den Gruppen festgestellt werden konnte. Der einzige signifikante Unterschied (p = 0.045) findet sich in der Applikation von DDAVP (Minirin®, Desmopressin), das in der PFA-Gruppe deutlich häufiger verabreicht wurde. Die potenziellen Nebenwirkungen des Medikamentes sollten bei fraglicher Verbesserung des Outcomes berücksichtigt werden. Das wichtigste untersuchte Kriterium dieser Studie ist die relevante (lebensbedrohliche, ggf. revisionsbedürftige) postoperative Einblutung. Es kam nur bei 4 Patienten zu solchen relevanten Einblutungen, je zwei in jeder Gruppe und somit nicht signifikant. Bei drei dieser Patienten wurde eine Revisionsoperation zur Hämatomentlastung durchgeführt. Das postoperative Outcome der Patienten wurde anhand des postoperativen GCS, der GCS-Reduktion präoperativ-postoperativ, dem GOS und der Aufenthalts¬dauer der Patienten auf der Intensivstation beurteilt, wobei beiden Gruppen durchgängig vergleichbar sind. Es ergibt sich somit keine Bestätigung des PFA-100®-Screenings als suffizientes Verfahren zur Reduktion von Blutungskomplikationen bei elektiven intrakraniellen Eingriffen. Allerdings kann aufgrund der geringen Fallzahl ein Fehler 2. Art nicht ausgeschlossen werden. Eine randomisierte, prospektive Studie wäre für die Entscheidung über ein dauerhaftes Fortsetzen oder Einstellen des PFA-Screenings sinnvoll.Background: Based on the clinical experience perioperative bleeding in complex neurosurgery can be related to an unidentified impairment of primary haemostasis. The Platelet Function Analyzer (PFA-100) has been evaluated as a screening tool for disorders of primary haemostasis. This study investigates whether the inclusion of the PFA-100 in the preoperative screening of neurosurgery patients might help reducing perioperative bleeding complications. Methods: Beginning in April 2006, patients with intracranial space occupying lesions who were scheduled for complex neurosurgical interventions underwent routine preoperative PFA-100 testing. Patients suffering from vascular mal-formations or intracranial aneurysms were excluded. In case of abnormal PFA, patients were further evaluated and/or received prophylactic treatment with desmopressin. Post-operative outcomes were compared to patients operated before introduction of PFA testing. The patients were enrolled consecutively over an 18-month period. Preoperative PFA-testing was introduced after 9 months. Inclusion criteria remained unchanged during the study period. Results: 102 patients were enrolled in the 9-month period before PFA introduction (non-PFA group), and 93 patients were enrolled thereafter (PFA group). The groups were highly comparable in terms of basic patient characteristics (age, gender, height, weight, BMI, ASA, Hb) and the spectrum of intracranial pathologies. Duration of anaesthesia or surgery, as well as experience of the operating surgeons were not significantly different between the two groups. Four patients (3.9 %) in the non-PFA group and six patients (6.5 %) in the PFA group experienced an increased intraoperative bleeding tendency according to a subjective assessment of the respective surgeon (p = 0.42). Two patients (2.0 %) in the non-PFA group and two patients (2.2 %) in the PFA group experienced a clinically relevant intracranial bleeding complication confirmed by postoperative computed tomography (odds ratio (OR) 1.05, 95% confidence intervall (95%-CI) 0.39-2.82; p = 1.0). Red blood cell, plasma or platelet transfusions were not significantly different between the two groups. Administration of coagulation factors (only fibrinogen concentrate was administered) or antifibrinolytic agents was also not significantly different between the groups. Five patients (4.9 %) in the non-PFA group and thirteen patients (14.0 %) in the PFA-group received desmopressin infusions perioperatively (OR 3.2, 95%-CI 1.1-9.2; p = 0.045). Conclusion: Prophylactic haemostatic treatment based on preoperative screening with the PFA-100 did not reduce the occurrence of perioperative bleeding complications related to complex intracranial surgical interventions, but resulted in a significant increase in the administration of desmopressin. A larger prospective randomized trial is needed to definitely determine the real clinical benefit of this approach

    Mouse models of pemphigus: valuable tools to investigate pathomechanisms and novel therapeutic interventions

    Autoimmune blistering diseases (AIBD) are paradigms of autoantibody-mediated organ-specific autoimmune disorders that involve skin and/or mucous membranes. Compared to other autoimmune diseases, the pathogenicity of autoantibodies in AIBD is relatively well described. Pemphigus is a potentially lethal autoantibody driven autoimmune disorder with a strong HLA class II association. It is mainly characterized by IgG against the desmosomal adhesion molecules desmoglein 3 (Dsg3) and Dsg1. Several murine pemphigus models were developed subsequently, each allowing the analysis of a characteristic feature, such as pathogenic IgG or Dsg3-specific T or B cells. Thus, the models can be employed to preclinically evaluate potentially novel therapies. We here thoroughly summarize past and recent efforts in developing and utilizing pemphigus mouse models for pathomechanistic investigation and therapeutic interventions.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Municipality amalgamation in Japan: A survival analysis of the timings of the amalgamation process

    This paper uses survival analysis to examine the time taken to carry out municipality amalgamation in Japan in terms of both forming the amalgamation committee and completing amalgamation. The results show that municipalities that depend on localm allocation tax grants as a revenue source, those that have an incentive to become a city that has special administrative discretions, and those that jointly manage local services form a committee and complete amalgamation more quickly. Further, municipalities that have high local public debt tend not to form committees. These findings show that the central government’s “carrot-and-stick” policy has strongly influenced municipality amalgamation

    End-of-Life Care Preferences of Patients with Advanced Urological Malignancies: An Explorative Survey Study at a Tertiary Referral Center

    Background: Many people want to die at home, but it is often not possible because they do not share their wishes with family members. This study was conducted to find out the extent to which patients with advanced urological malignancies had wishes regarding their final stage of life, made arrangements accordingly, and communicated their wishes to relatives and health care professionals. Methods: We conducted a survey among advanced urological tumor patients during their clinic visit at a German university hospital using a 31-item questionnaire. Inclusion criteria were metastatic or irresectable prostate cancer, urothelial carcinoma, or renal cell carcinoma. Results: In total, 88 patients (76 male, 12 female) completed the questionnaire, and 62 of those respondents (70%) had received their tumor diagnosis within the past 5 years. Symptoms were reported by 80%, and 18% described five or more symptoms. The majority (88%) stated that they had thought about their preferred place of death but 58% had not informed anyone about it. The preference for a hospice as the place of death correlated statistically significantly with the absence of a domestic partnership (p = 0.001) or marriage (p < 0.001) and with a high number of symptoms (≥5; p = 0.009). However, 73% had not talked with their urological oncologist about care options in case their health deteriorated though 36% of those were interested in having a conversation about it. Conclusions: Our results showed that 9 out of 10 patients reflected on their preferred place of death but only a few discussed it with anyone. Based on this finding, physicians and healthcare staff should initiate discussions about early care planning so that patients in incurable situations can express their wishes regarding their preferred place of death.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Moving Closer or Drifting Apart: Distributional Effects of Monetary Policy

    Our paper picks up the current controversial debate about increasing (income) inequality due to recent monetary policy measures in major advanced economies. We use a VAR framework identified with sign restrictions to figure out how income inequality related measures react to monetary policy in six different advanced economies. These countries differ by their absolute income inequality as well as their redistribution. We choose the U.S., Canada and South Korea as countries with very little redistribution and Sweden, the Czech Republic and Hungary as countries with relatively high redistribution. While all economies experience an increase in Gini coeffcients of gross income in the presence of an expansionary monetary policy shock, only the U.S., Canada and South Korea also show a significant response in Gini coeffcient of net income. To figure out how the transmission of monetary policy to income inequality works we pick up the two major channels dominant in the literature: The employment channel and the income composition channel. The latter is analyzed by data from national accounts concerning two different kinds of income households receive: Labor related income and capital payments, both net. While we find that capital owners profit disproportionately in the less redistributing countries, we observe a more even reaction in both income types. This indicates that the harmful effects of expansionary monetary policy on the market income distribution are mitigated if the degree of redistribution is high

    Untersuchung der Evolution von Intermediärfilament-bildenden Proteinen in Eukaryoten

    The remarkable increased complexity of eukaryotes' cytoskeleton represents one of the major transitions in life. Inherently intertwined with eukaryogenesis itself, molecular innovations that emerged during cytoskeleton evolution are pivotal for understanding Eukaryotes` evolutionary history. Despite distant homologs for two major cytoskeletal components, actin, and tubulin, identified in prokaryotes, no clear homolog for intermediate filaments has been found. Intermediate filaments represent crucial components in the eukaryotic cytoskeleton, being regarded as one of the major molecular innovations that evolved in this kingdom. Long evolutionary distances, poor sequence conservation, and many other hurdles render tracing the evolution of such proteins a daunting task. One possible route to circumvent such limitations is to investigate single-cell eukaryotes, namely protists, whose cell biology is expected to entail many overlooked fiber-forming, intermediate filament-like proteins. The filament-forming mitochondrial citrate synthase described in the series of works that prompted this study was an ideal candidate for studying the emergence of intermediate filament proteins over a known evolutionary interval. The study of such multifunctional proteins is important for several reasons. As part of a well-conserved protein family, the mitochondrial citrate synthase (mCS) makes it tractable to study transitions over known and relatively short evolutionary intervals in a phylogenetic tree. Furthermore, mCS evolution of fiber formation in protists could be the product of convergent evolution, giving rise to novel IF structures and potentially elucidating mechanisms for filament formation in non-metazoan lineages. Another possible role of fiber formation is linked to metabolism regulation. Therefore, studying the evolutionary interval along which such behavior emerged would allow us to pinpoint the evolutionary forces acting on enzyme fiber formation and their possible recruitment for structural functions. Here, we characterized the properties of in vitro fiber formation of the recombinant mitochondrial citrate synthase of Tetrahyemna thermophila. We provide orthogonal evidence for this behavior to be a buffer-driven artifact caused by a specific chemical moiety that is most likely absent in the native biological context. We also show that buffer-induced aggregation is a feature common to eukaryotic mitochondrial citrate synthase but absent in bacterial homologs. Using an in-vivo approach, we investigated the protein's localization using Strep and GFP-fibre deficient tagged mutants. We show that the protein colocalizes with mitochondria and does not form clear foci during starvation, which indicates the absence of metabolic regulation via filamentation. Furthermore, we monitored the protein's localization during conjugation, reporting the absence of any extramitochondrial structure. Finally, we show how MES-induced ordered fibers emerged in the last common ancestor of Hymenostomatida while other species and ancestral proteins form amorphous aggregates. We thereby conclude that ordered fibers likely are a product of neutral processes.Die Zunahme der Komplexität des Zytoskeletts von Eukaryonten stellt einen der wichtigsten Übergänge im Leben dar. Die molekularen Innovationen, die während der Entwicklung des Zytoskeletts entstanden sind, sind von zentraler Bedeutung für das Verständnis der Evolutionsgeschichte der Eukaryonten und sind eng mit der Eukaryogenese verbunden. Obwohl in Prokaryonten weit entfernte Homologe für zwei wichtige Komponenten des Zytoskeletts, Aktin und Tubulin, identifiziert wurden, wurde kein eindeutiges Homolog für Zwischenfilamente gefunden. Intermediärfilamente sind entscheidende Komponenten des eukaryotischen Zytoskeletts und gelten als eine der wichtigsten molekularen Innovationen, die sich in Eukaryoten entwickelt haben. Große evolutionäre Entfernungen, schlechte Qualität der Sequenz und viele andere Hürden machen die Rückverfolgung der Evolution solcher Proteine zu einer schwierigen Aufgabe. Ein möglicher Weg, solche Limitationen zu umgehen, ist die Untersuchung einzelliger Eukaryonten, nämlich der Protisten, von deren Zellbiologie erwartet wird, dass sie viele übersehene faserbildende, intermediäre filamentartige Proteine aufweisen. Die filamentbildende mitochondriale Citrat-Synthase, die in der Reihe von Arbeiten beschrieben wurde und, war ein idealer Kandidat für die Untersuchung der Entstehung von Intermediärfilament Proteinen über eine bekannte evolutionäre Zeitspanne. Die Untersuchung solcher multifunktionalen Proteine ist aus mehreren Gründen wichtig. Als Teil einer gut konservierten Proteinfamilie ist die mitochondriale Citrat-Synthase (mCS) gut geeignet, um Übergänge über bekannte und relativ kurze evolutionäre Zeitspannen in einem phylogenetischen Baum zu untersuchen. Darüber hinaus könnte die mCS-Evolution der Faserbildung bei Protisten das Produkt einer konvergenten Evolution sein, die zu neuartigen IF-Strukturen geführt haben und möglicherweise die Mechanismen der Filamentbildung in nicht-metazoischen Abstammungslinien aufklärt. Eine weitere mögliche Rolle der Faserbildung ist mit der Regulierung des Stoffwechsels verbunden. Die zeitliche Untersuchung, in dem ein solches Verhalten auftrat, würde es uns daher ermöglichen, die evolutionären Kräfte, die auf die Bildung von Enzymfasern einwirken, und ihre mögliche Rekrutierung für strukturelle Funktionen genau zu bestimmen. Hier haben wir die Eigenschaften der in vitro-Faserbildung der rekombinanten mitochondrialen Citrat Synthase von Tetrahyemna thermophila charakterisiert. Wir liefern orthogonale Beweise dafür, dass es sich bei diesem Verhalten um ein pufferbedingtes Artefakt handelt, das durch eine spezifische chemische Einheit verursacht wird, die im nativen biologischen Kontext höchstwahrscheinlich nicht vorhanden ist. Außerdem zeigen wir, dass die pufferinduzierte Aggregation ein gemeinsames Merkmal der eukaryotischen mitochondrialen Citrat-Synthase ist, dass bei bakteriellen Homologen fehlt. Mit einem in-vivo-Ansatz untersuchten wir die Lokalisierung des Proteins mit Hilfe von Strep- und GFP faserdefizienten, markierten Mutanten. Wir zeigen, dass das Protein mit Mitochondrien co-lokalisiert und während des Hungerns keine eindeutigen Foci bildet, was auf das Fehlen einer Stoffwechselregulierung durch Filamentierung hinweist. Darüber hinaus haben wir die Lokalisierung des Proteins während der Konjugation beobachtet und das Fehlen jeglicher extramitochondrialen Struktur festgestellt. Schließlich zeigen wir, wie MES-induzierte geordnete Fasern im letzten gemeinsamen Vorfahren der Hymenostomatida entstanden sind, während andere Arten und angestammte Proteine amorphe Aggregate bilden. Daraus schließen wir, dass geordnete Fasern wahrscheinlich ein Produkt neutraler Prozesse sind
