Regional Market-Based Climate Policy in North America: Efficient, Effective, Fair?
Despite President Obama’s current interest in climate policy, market-based climate policy on the US federal level still appears to be deadlocked. The same is true for Canada, which has aligned its climate policy to the US. However, regional activities are more promising as British Columbia and California have started using market-based approaches recently. Against this background, the paper asks: Can state level market-based climate policy be a sustainable alternative or supplement to federal action? Which of the two programs does better in terms of fulfilling ambitious sustainability criteria? How can the programs be improved? In order to answer these questions, in a comparative fashion, the paper analyzes the design and the results of the British Columbia Carbon Tax and the California Cap-and- Trade Program and evaluates them based on sustainability criteria. By doing so, the paper provides a comparative evaluation of two North American sub-national level market-based climate policy programs, shows the significance and the challenges of such regional initiatives, identifies best-practice design elements, and provides recommendations for regional market-based climate policy design
Single‑cell analysis reveals the spatial‑temporal expression of genes associated with esophageal malformations
Understanding the molecular mechanisms of congenital diseases is challenging due to their occurrence within specific developmental stages. Esophageal malformations are examples of such conditions, characterized by abnormalities in the development of esophagus during embryogenesis. These developmental malformations encompass a range of anomalies, including esophageal atresia, and tracheoesophageal fistula. Here, we investigated the preferential expression of 29 genes that are implicated in such malformations and their immediate interactome (a total of 67 genes). We conducted our analyses across several single-cell atlases of embryonic development, encompassing approximately 150,000 cells from the mouse foregut, 180,000 cells from human embryos, and 500,000 cells from 24 human organs. Our study, spanning diverse mesodermal and endodermal cell populations and early developmental stages, shows that the genes associated with esophageal malformations show their highest cell-type specific expression in lateral plate mesoderm cells and at the developmental stage of E8.75–E9.0 days. In human embryos, these genes show a significant cell-type specific expression among subpopulations of epithelial cells, fibroblasts and progenitor cells including basal cells. Notably, members of the forkhead-box family of transcription factors, namely FOXF1, FOXC1, and FOXD1, as well as the SRY-box transcription factor, SOX2, demonstrate the most significant preferential expression in both mouse and human embryos. Lipid A in outer membrane vesicles shields bacteria from polymyxins
The continuous emergence of multidrug-resistant bacterial pathogens poses a major global healthcare challenge, with Klebsiella pneumoniae being a prominent threat. We conducted a comprehensive study on K. pneumoniae's antibiotic resistance mechanisms, focusing on outer membrane vesicles (OMVs) and polymyxin, a last-resort antibiotic. Our research demonstrates that OMVs protect bacteria from polymyxins. OMVs derived from Polymyxin B (PB)-stressed K. pneumoniae exhibited heightened protective efficacy due to increased vesiculation, compared to OMVs from unstressed Klebsiella. OMVs also shield bacteria from different bacterial families. This was validated ex vivo and in vivo using precision cut lung slices (PCLS) and Galleria mellonella. In all models, OMVs protected K. pneumoniae from PB and reduced the associated stress response on protein level. We observed significant changes in the lipid composition of OMVs upon PB treatment, affecting their binding capacity to PB. The altered binding capacity of single OMVs from PB stressed K. pneumoniae could be linked to a reduction in the lipid A amount of their released vesicles. Although the amount of lipid A per vesicle is reduced, the overall increase in the number of vesicles results in an increased protection because the sum of lipid A and therefore PB binding sites have increased. This unravels the mechanism of the altered PB protective efficacy of OMVs from PB stressed K. pneumoniae compared to control OMVs. The lipid A-dependent protective effect against PB was confirmed in vitro using artificial vesicles. Moreover, artificial vesicles successfully protected Klebsiella from PB ex vivo and in vivo. The findings indicate that OMVs act as protective shields for bacteria by binding to polymyxins, effectively serving as decoys and preventing antibiotic interaction with the cell surface. Our findings provide valuable insights into the mechanisms underlying antibiotic cross-protection and offer potential avenues for the development of novel therapeutic interventions to address the escalating threat of multidrug-resistant bacterial infections.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg
Empirical Estimation of Intraday Yield Curves on the Italian Interbank Credit Market e-MID
This paper introduces a major novelty: the empirical estimation of spot intraday yield curves based on tick-by-tick data on the Italian electronic interbank credit market (e-MID). To analyze the consequences of the recent financial crisis, we split the data into four periods, which include events before, during, and after the recent financial crisis starting in 2007. Our first result is that, from a practical point of view, the intraday yield curve can be modeled by standard models for yield curves providing advantages for intraday trading on intraday interbank credit markets. Moreover, the estimates show that the systematic dynamics in the intraday yield curves during the turmoil were highly noticeable, resulting in a significantly better goodness-of-fit. Based on this fact, we infer that investors in the interbank credit market base their investment decisions on the effects of the intraday dynamics of intraday interest rates more intensively during a financial crisis. Therefore, the systematic impact on the e-MID appears to be stronger and econometric modeling of the intraday interest rate curve becomes even more attractive during a turmoil
Chemical analysis of amorphous, pharmaceutically relevant raw materials with special consideration of Mumijo, peat and Assa-foetida
In der Pharmazie werden amorphe Substanzen wegen ihrer guten Wasserlöslichkeit und Bioverfügbarkeit eingesetzt. Mumijo, Torf und Assa-foetida sind pharmazeutische Ausgangsverbindungen, die von Natur aus in amorpher Form vorliegen und seit Jahrtausenden in der Medizin verwendet werden.
Mumijo ist eine braun-schwarze amorphe Substanz, die in Gesteinen in den Hochgebirgen - vorwiegend in Zentralasien - eingebettet ist und in Kavernen nach der Schneeschmelze aus Ritzen und Spalten austritt. Es besteht aus bis zu 85 % Humus, daneben sind Lipide, Steroide, Kohlenwasserstoffe, Alkaloide, Aminosäuren, Proteine und weitere organische Verbindungen enthalten. Die Entstehung von Mumijo wird in der Literatur kontrovers diskutiert. Ziel dieser Dissertation ist einerseits die Klärung der konkreten Genese von Mumijo und andererseits die Identifizierung pharmakologisch wirksamer Verbindungen.
Im Rahmen der Untersuchung der vorliegenden Arbeit wurden dünnschichtchromatographische Analysen mit dem wässrigen Extrakt von vier Proben durchgeführt, welche aus der Region Ostpamir stammen. Ziel dieser Analyse war es, einen Überblick über die Polaritäten des Materials zu erhalten. Im Anschluss wurden analytische HPLC-Untersuchungen durchgeführt, mit dem Ziel, ein optimiertes Programm für HPLC-HRMS-Untersuchungen zu erhalten. Diese Untersuchungen ergaben, dass in einer Probe Urolithin A und Urolithin B enthalten waren. Diese 3,4-Dibenzo-α-pyrone sind für ihre antiproliferativen Wirkungen bekannt. In drei anderen Proben konnte mit Alternariol ein Mykotoxin nachgewiesen werden, welches ebenfalls antiproliferative Eigenschaften aufweist. Um einen umfassenderen Überblick über die Genese des Materials zu gewinnen, wurden aus den Proben ein Cylohexan-Extrakt gewonnen, der mittels GC-MS auf seine Inhaltsstoffe untersucht wurde. Neben relativ unselektiven Markern wie langkettigen n-Alkanen >C23 sowie Ölsäure und Palmitinsäure wurden auch spezifischere Verbindungen entdeckt, die Aufschluss über die Genese des Materials geben. Zu diesen Verbindungen zählt cis-Vaccensäure, die in allen Proben enthalten war und als bakterieller Marker insbesondere auf Bakterien des Genus Lactobacillus hinweist. Diese Bakterien kommen in den Mägen von Säugetieren vor und sind an der Verdauung beteiligt. Eine Hypothese zur Genese von Mumijo geht davon aus, dass Mumijo überwiegend aus den Ausscheidungsprodukten von Tieren besteht. Diese Hypothese wird damit bestärkt. Außerdem enthielt eine der Proben p-Kresol, eine Verbindung, die im Urin vorkommt, was die Hypothese weiter stützt.
Ein weiteres Ziel dieser Dissertation ist der Vergleich von Mumijo und Torf mit der Fragestellung, ob sich beide Materialien hinsichtlich ihrer chemischen Zusammensetzung so ähnlich sind, dass sie jeweils durch das andere für medizinische Anwendungen substituiert werden können. Torf ist ein organisches Zersetzungsprodukt von Pflanzen und Tieren, das überwiegend in borealen Moorgebieten entsteht, wenn die Produktivität der Biomasse deren Zersetzung übersteigt. In Torf sind n-Alkane, n-Alkohole, n-Aldehyde, gesättigte und ungesättigte Fettsäuren, Methylketone, Sterole, Hopane, Vitamin E und weitere Verbindungen enthalten.
In dieser Arbeit wurde eine Torfprobe aus dem Naturschutzgebiet Woeste, Deutschland, mit den gleichen Verfahren (DC, HPLC-HRMS, GC-MS) untersucht wie zuvor die Mumijo-Proben. Mit dem Unterschied, dass der Untersuchung des Torfs eine serielle Soxhlet-Extraktion mit Cyclohexan, Dichlormethan, Ethylacetat und Methanol vorausging und dass eine verkürzte HPLC-HRMS-Methode zum Einsatz kam. Die HPLC-HRMS-Untersuchungen der Dichlormethan-, Ethylacetat- und Methanol-Extrakte wies eine pharmakologisch aktive Triterpensäure, das Hederagenin, nach. Des Weiteren konnten verschiedene Pilzmetabolite, wie beispielsweise Pulvinsäure und Terrein, nachgewiesen werden. Darüber hinaus konnten die Vitamine K1 und Riboflavin (B2) identifiziert werden. Anhand der GC-MS-Untersuchungen konnten mit Heptacosan-2-on, Pentacosan-2-on und Nonacosan-2-on spezifische Biomarker für Torfmoose der Gattung Sphagnum nachgewiesen werden. Daneben konnten auch γ-Sitosterol und Lup20(29)-en-3-on als typische Biomarker für höhere Pflanzen identifiziert werden. Diese Zusammensetzung ist typisch für ein Übergangsmoor.
Der Vergleich ergab, dass sowohl Torf als auch Mumijo Palmitin-, Öl- und cis-Vaccensäure enthalten und dass es eine Übereinstimmung in der n-Alkan-Zusammensetzung gibt, die von einer Kettenlänge von C21-C31 reicht. Allerdings sind auch Unterschiede festzustellen. Neben den genannten Biomarkern wurden in der Torfprobe auch n-Alkohole, n-Aldehyde als Abbauprodukte von Fettsäuren und Vitamin E mit seinen Abbauprodukten sowie ein Hopan-Derivat nachgewiesen, die typische Torfbestandteile sind, aber in Mumijo nicht vorkommen.
Trotz der genannten Übereinstimmungen sind die Unterschiede in der chemischen Zusammensetzung doch so umfangreich, dass die Möglichkeit der gegenseitigen Substitution von Mumijo und Torf nicht abschließend geklärt werden kann.
Die in dieser Dissertation entwickelte GC-Methode zeigt das Potenzial, amorphe Substanzen zu untersuchen und Erkenntnisse über die Genese des betreffenden Materials zu gewinnen. Es konnten mehrere Stoffgruppen parallel mit akkurater Genauigkeit untersucht werden, womit die Erstellung von Profilen gleicher Qualität möglich werden, mit deren Hilfe die Basis einer soliden Vergleichbarkeit unterschiedlichster Proben gewährleistet werden kann. Darauf aufbauend, können in Zukunft amorphe Substanzen wie Mumijo oder Torf mit hoher Qualität untersucht werden.
Als drittes amorphes Material wurde in dieser Arbeit Assa-foetida untersucht, ein Exsudat, das aus den Wurzeln von der im Iran, Afghanistan und in Teilen der ehemaligen UDSSR vorkommenden medizinischen Pflanze Ferula assa-foetida gewonnen wird. Assa-foetida besteht aus Harz (40-64 %), einer wasserlöslichen Fraktion (25 %) und ätherischen Ölen (10-17 %). Die Harz-Fraktion enthält Ferulasäure und seine Ester, Coumarine, Sesquiterpen-Coumarine und andere Terpenoide.
In der vorliegenden Arbeit wurde die Harz-Fraktion von Assa-foetida mit dem Ziel untersucht, ein Profil der enthaltenen Sesquiterpen-Coumarine und Guaianolide zu erstellen, um daraus Rückschlüsse auf den Ort seiner Herkunft zu erlangen. Analog zur Untersuchung der Torfprobe, wurde eine serielle Soxhlet-Extraktion durchgeführt. Von den erhaltenen Extrakten wurden ausschließlich die Dichlormethan-Extrakte der vier vorliegenden Proben dünnschichtchromatographisch untersucht. Die Untersuchungen zeigten unter UV-Licht bei 366 nm die für Coumarine oder Sesquiterpen-Coumarine charakteristischen Zonen.
Wie bei der Untersuchung des Torfs schlossen sich optimierte HPLC-HRMS-Untersuchungen an. Die Ergebnisse wurden mit einer im Rahmen dieser Arbeit erstellten Stoffliste, die sich auf Sesquiterpen-Coumarine und Guaianolide fokussiert, verglichen.
Mit Farnesiferol A konnte in zwei Proben eine Verbindung nachgewiesen werden, die in Ferula assa-foetida vorkommt. Neben dieser Verbindung wurden aber auch Talasin A, Fegvolid, Malaphyllinin, Malaphyll und Diversin nachgewiesen, die in dieser Zusammensetzung nur in Ferula diversivittata, einer Pflanze aus dem usbekisch-afghanischen Grenzgebiet vorkommen. Weitere nachgewiesene Verbindungen sind Cauferidin und Cauferin, die in Ferula conocaula, einer Pflanze aus Usbekistan, vorkommen. Die erhaltenen Ergebnisse weisen darauf hin, dass diese Proben nur teilweise aus Afghanistan und zu einem anderen Teil aus Usbekistan stammt.Amorphous substances are used in pharmacy due to their good water solubility and bioavailability. mumijo, peat and assa-foetida are pharmaceutical raw materials that occur naturally in amorphous form and have been used in medicine for thousands of years.
Mumijo is a brown-black amorphous substance that is embedded in rocks in the high mountains, primarily in Central Asia. After the snow melts mumijo emerges in caverns from fissures in the rocks. It consists of up to 85 % humus, and contains lipids, steroids, hydrocarbons, alkaloids, amino acids, proteins and other organic compounds. The formation process of mumijo is controversial in the literature. The objective of the dissertation is to clarify the specific genesis of Mumijo on the one hand and to identify pharmacologically active compounds on the other.
In order to gain an overview of the polarities of the material, the aqueous extract of four samples from the Eastern Pamir region was subjected to thin-layer chromatography (TLC). Thereafter, analytical High-performance liquid chromatography (HPLC) tests were carried out with the objective of developing an optimized program for HPLC-HRMS tests. These tests showed that one sample contained urolithin A and urolithin B. These 3,4-dibenzo-α-pyrones are known for their antiproliferative effects. Alternariol, a mycotoxin that also has antiproliferative properties, was detected in three other samples. In order to gain a more comprehensive overview of the genesis of the material, a cylohexane extract was obtained from the samples, which were then analyzed for their constituent components using GC-MS. Besides to relatively unselective markers such as long-chain n-alkanes (>C23), as well as oleic acid and palmitic acid, more specific compounds were also discovered that provide information about the genesis of the material. The compounds identified in this study include cis-vaccenic acid, which was present in all samples. As a bacterial marker, it is indicative particularly of bacteria belonging to the genus Lactobacillus. These bacteria are found in the stomachs of mammals and are involved in the digestive process. One hypothesis regarding the origin of mumijo proposes that it is primarily composed of the excretions of animals. This hypothesis is thus reinforced. Furthermore, one of the samples was tested positive for p-cresol, a compound commonly found in urine, which serves to reinforce the hypothesis.
A further aim of this dissertation is a comparison between mumijo and peat with the question of whether both materials are so similar in terms of their chemical composition that they can each be substituted by the other for medical applications. Peat is an organic decomposition product of plants and animals that is primarily formed in boreal peatlands when the productivity of the biomass exceeds its decomposition. Peat contains n-alkanes, n-alcohols, n-aldehydes, saturated and unsaturated fatty acids, methyl ketones, sterols, hopanes, vitamin E and other compounds.
In this study, a peat sample from the Woeste nature reserve in Germany was subjected to analysis using the same analytical methods previously employed for Mumijo, namely TLC, HPLC-HRMS and GC-MS. The key difference was the use of a serial soxhlet extraction with cyclohexane, dichloromethane, ethyl acetate, and methanol, in addition to a shortened HPLC-HRMS method. HPLC-HRMS analysis of dichloromethane, ethyl acetate and methanol extracts revealed pharmacologically active triterpenic acid, hederagenin.. Furthermore, fungal metabolites, e.g., pulvic acid and terrein, were identified. Additionally, vitamin K1 and riboflavin (vitamin B2) were detected. Based on the GC-MS analyses, specific biomarkers for mosses of the genus Sphagnum were detected with heptacosan-2-one, pentacosan-2-one and nonacosan-2-one. In addition, γ-sitosterol and lup20(29)-en-3-one were identified as typical biomarkers for higher plants. This composition of substances is characteristic of a transition peat bog.The comparison revealed that both peat and mumijo contain palmitic, oleic and cis-vaccenic acids and that there is a match in the n-alkane composition, ranging from a chain length from C21 to C31. Nevertheless, there are also notable distinctions. In addition to the biomarkers previously mentioned, n-alcohols, n-aldehydes, which are degradation products of fatty acids, and vitamin E together with its degradation products, as well as a hopane derivative, were also detected in the peat sample. These are typical components of peat, but do not occur in Mumijo.
Despite the similarities mentioned, the differences in chemical composition are so extensive that it is not possible to conclude with certainty whether mumijo and peat can be substituted for one another.
The GC method developed in this dissertation shows the potential to detect and examine amorphous substances and gain insights into the genesis of the material in question. A number of groups of substances can be analyzed in parallel with high precision, allowing the creation of profiles of an equivalent standard, which can then be used as the basis for a robust comparison of different samples. Building on this, it is possible that amorphous substances such as mumijo or peat may be analyzed with high quality in future studies.
The third amorphous material examined in this work was Assa-foetida, an exudate obtained from the roots of the medicinal plant Ferula assa-foetida. This plant is native to Iran, Afghanistan, and parts of the former USSR. Assa-foetida consists of resin (40-64 %), a water-soluble fraction (25 %) and essential oils (10-17 %). The resin fraction contains ferulic acid and its esters, coumarins, sesquiterpene coumarins and other terpenoids.
The present study aims to create a profile of the sesquiterpene coumarins and guaianolides contained in the resin fraction of Assa-foetida. This will enable conclusions to be drawn about its place of origin. A serial Soxhlet extraction was carried out analogous to the analysis of the peat sample. Of the extracts obtained, only the dichloromethane extracts of four samples underwent thin-layer chromatography analysis. The analysis revealed that the spots characteristic of coumarins or sesquiterpene coumarins under UV light at 366 nm.
Analogous to the investigation of the peat, optimized HPLC-HRMS analysis were carried out. The results were compared with a list of substances compiled as part of this dissertation, which focuses on sesquiterpene coumarins and guaianolides.
Farnesiferol A, a compound found in Ferula assa-foetida, was detected in both samples. In addition to this compound, talasin A, fegvolide, malaphyllinin, malaphyll and diversin are also present. These compounds are only found in this composition in Ferula diversivittata, a plant from the Uzbek-Afghan border region. Other compounds are cauferidin and cauferin, which are found in Ferula conocaula, a plant native to Uzbekistan. The results obtained indicate that this sample only partly comes from Afghanistan and partly from Uzbekistan
Letting off Steam! Experimental Evidence on Inappropriate Punishment
Aggression is displaced when provocations cannot be directly retaliated against and when it is redirected towards a target innocent of any wrongdoing. While this phenomenon is widespread, it has not been widely explored in experimental economics. We fill this gap and find that a sizeable proportion of subjects (37%), when treated unfairly, punish co-players who are not at all responsible for the unfairness. When in a disadvantaged position, inequity-aversion seems to be the driving force of punishment, yet when treated fairly, some subjects (17%) exhibit status-seeking behavior. Moreover, students affiliated with an armed forces university are much more likely than regular students to engage in displaced aggression
Veränderungen von Leukoztenadhäsionsfaktoren und Toll-like Rezeptoren bei Patienten mit Epilepsie
Epilepsie ist eine Erkrankung des Gehirns mit anhaltender Tendenz wiederkehrende Anfälle zu erleiden. Epileptische Anfälle sind definiert als exzessive oder synchrone neuronale Aktivität im Gehirn die ein vorübergehendes Auftreten von Symptomen hervorruft. Es gibt verschiedene Ursachen, die teilweise ineinander übergehen. Eine Kategorie stellen hierbei inflammatorische Prozesse dar. Viele Zusammenhänge zwischen Immunsystem und Epilepsie sind bereits bekannt. Eine veränderte Integrität der Blut-Hirn-Schranke spielt dabei eine große Rolle. Die genauen Mechanismen sind allerdings noch nicht bis ins Detail geklärt.
Die vorliegende Arbeit soll weitere Erkenntnisse bezüglich der immunologischen Vorgänge bei Patienten mit Epilepsie gewinnen. Dazu wurden Veränderungen von Leukozytenadhäsionsfaktoren und Toll-like Rezeptoren im peripheren Blut von Patienten mit Epilepsie mittels Durchflusszytometrie untersucht, sowohl interiktal als auch im zeitlichen Zusammenhang mit einem Anfall. Mitberücksichtigt wurden die Einflussfaktoren Anfallsart, Hippocampussklerose und Seite des Anfallsursprungs.
Es wurden insgesamt 35 Patienten mit eindeutig diagnostizierter fokaler Epilepsie aus der Video-Elektroenzephalographie-Monitoring Einheit und der Epilepsie-Ambulanz des Universitätsklinikums Marburg in die Studie eingeschlossen. Blutentnahmen bei Patienten mit Anfällen erfolgten am Aufnahmetag (Baseline), direkt postiktal, 1 Stunde postiktal und wenn möglich, 24 Stunden postiktal. Die interiktalen Blutproben wurden mit 20 gesunden Kontrollen verglichen.
Die wesentlichen Ergebnisse zeigen einen postiktalen Anstieg der Intercellular adhesion molecule-1 tragenden zytotoxischen Zellen (CD54+/CD56+ und CD54+/CD8+) und der Toll-like Rezeptor-4 tragenden Monozyten (CD284+/CD14+) sowie einen postiktalen Abfall der Intercellular adhesion molecule-1 tragenden T-Helferzellen (CD54+/CD4+). Im Vergleich mit den gesunden Kontrollen fällt interiktal eine erhöhte Anzahl der Intercellular adhesion molecule-1 tragenden zytotoxischen T-Zellen (CD54+/CD8+) auf. Der Verlauf war dabei teilweise abhängig von der Art der Anfälle, der Ätiologie und der Anfallsursprungsseite.
Der postiktale Anstieg von Natürlichen Killerzellen und zytotoxischen T-Zellen bestätigt sich in einigen Vorarbeiten und auch in neueren Arbeiten zu diesem Thema. Auch der Abfall der T-Helferzellen zeigte sich in früheren Studien. Erhöhte Level an Toll-like Rezeptor-4 tragenden Zellen bei Patienten mit Epilepsie wurden ebenfalls in anderen Studien nachgewiesen.
In tierexperimentellen Vorstudien präsentierten sich anfallssupprimierende Wirkungen von Intercellular adhesion molecule-1 und von Toll-like Rezeptor-4 Inhibitoren. Die vorliegende Arbeit unterstützt auch diese Studien und liefert einen weiteren Beitrag zum Verständnis der immunologischen Mechanismen bei epileptischen Anfällen. Damit kann auch zur Entwicklung neuer anfallssupprimierender Medikamente beigetragen werden.
Die Zusammenschau der vorliegenden und der durch Vorstudien erlangten Ergebnisse legt nahe, dass sowohl Intercellular adhesion molecule-1 als auch Toll-like Rezeptor-4 mit ihrem Einfluss auf die Blut-Hirn-Schranke eine wichtige Rolle in der Iktogenese spielen. Um noch genauere Aussagen darüber treffen zu können und um die wechselseitige Wirkung von Anfall, Stress und Veränderungen der Blut-Hirn-Schranke besser zu verstehen, sind weitere Studien mit häufigeren Blutentnahmen und einem größeren Patientenkollektiv angebracht.Epilepsy is a disorder of the brain with a persistent tendency to suffer from seizures. Epileptic seizures are defined as excessive or synchronous neuronal activity which induces a transient occurrence of symptoms. There are many different aetiologies, which are merging partially. Inflammatory processes represent one category. Many links between the immune system and epilepsy are already known. An altered integrity of the blood-brain-barrier plays an important role. The detailed mechanisms however are not fully understood.
With this study we aimed to gain further knowledge regarding immunological activities in patients with epilepsy. For that purpose, alterations of leucocyte adhesion factors and Toll-like receptors in peripheral blood of patients with epilepsy were investigated by using fluorescence activated cell sorting, interictally as well as in context with seizures. Seizure type, hippocampal sclerosis and side of seizure onset in the brain were considered as influencing factors.
We included 35 patients from the video-electroencephalography monitoring unit and the epilepsy outpatient department of the Philipps-University Marburg who were diagnosed with focal epilepsy. Blood samples were taken at admission day (Baseline), immediately postictally, 1 hour postictally and 24 hours postictally, if possible. The interictal samples were compared to 20 healthy controls.
The main findings show postictal increase of intercellular adhesion molecule-1 positive cytotoxic cells (CD54+/CD56+ and CD54+/CD8+) and Toll-like receptor-4 positive monocytes (CD284+/CD14+), as well as postictal decrease of intercellular adhesion molecule-1 positive T helper cells (CD54+/CD4+). In comparison with healthy controls, we noticed a higher rate of intercellular adhesion molecule-1 positive cytotoxic T cells (CD54+/CD8+) interictally. The course was partially dependent of seizure type, aetiology and side of seizure onset.
The postictal increase of natural killer cells and cytotoxic T cells has been described in former and in recent studies. Also, the decrease of T helper cells was shown in previous studies. Higher rates of Toll-like receptor-4 carrying cells in patients with epilepsy were noticed also in other studies.
In animal models inhibitors of intercellular adhesion molecule-1 and Toll-like receptor-4 indicated a seizure suppressing effect. With our study we can support these findings and provide a better understanding of the immunological mechanisms in epileptic seizures. This might contribute to the development of new seizure suppressing drugs in the future.
Taken together, the findings of this study and former studies suggest that with their influence on the blood-brain-barrier both, intercellular adhesion molecule-1 and Toll-like receptor-4 play an important role in ictogenesis. To give a more detailed statement and to better understand the reciprocal effects of seizures, stress and alterations of the blood-brain-barrier, more precise studies with more frequent blood samples and a larger number of patients would be needed
A mRNA panel for differentiation between acute exacerbation or pneumonia in COPD patients
Introduction: Patients suffering from chronic obstructive pulmonary disease
(COPD) are prone to acute exacerbations (AECOPD) or community acquired
pneumonia (CAP), both posing severe risk of morbidity and mortality. There is
no available biomarker that correctly separates AECOPD from COPD. However,
because CAP and AECOPD differ in aetiology, treatment and prognosis, their
discrimination would be important.
Methods: This study analysed the ability of selected candidate transcripts from
peripheral blood mononuclear cells (PBMCs) to differentiate between patients
with AECOPD, COPD & CAP, and CAP without pre-existing COPD.
Results: In a previous study, we identified differentially regulated genes
between CAP and AECOPD in PBMCs. In the present new cohort, we tested the
potential of selected candidate PBMC transcripts to differentiate at early time
points AECOPD, CAP+COPD, and CAP without pre-existing COPD. Expression
of YWHAG, E2F1 and TDRD9 held predictive power: This gene set predicted
diseases markedly better (model accuracy up to 100%) than classical clinical
markers like CRP, lymphocyte count and neutrophil count (model accuracy up
to 82%).
Discussion: In summary, in our cohort expression levels of YWHAG, E2F1 and
TDRD9 differentiated with high accuracy between COPD patients suffering

Zum Stand der Sammlungsdigitalisierung an der UMR - Quantitative Umfrageergebnisse 2023
Die Philipps-Universität Marburg entwickelt derzeit ein Konzept zur digitalen Präsentation ihrer wissenschaftlichen Sammlungen. Hierbei verfolgt im Rahmen der Landesinitiative HeFDI ein vom Hessischen Ministerium für Wissenschaft und Forschung, Kunst und Kultur (HMWK) gefördertes Projekt den Auftrag, Datenbankmanagementsysteme zur Integration wissenschaftlicher Sammlungen anzupassen. Neben der Konzeptionierung und Implementierung einer langfristig tragfähigen Datenbank hat das Projekt die Pilotierung der Sammlungsdigitalisierung an der Philipps-Universität zur Aufgabe. Der Datensatz enthält die Darstellung aller quantitativen Angaben, die unter den Sammlungen der Philipps-Universität im Dezember 2023 erhoben wurden.1.
Prevalence of Spirometry Testing among Patients with Asthma and COPD in German General Practices
The goal of this study was to estimate the prevalence of spirometry testing among patients with asthma and chronic obstructive pulmonary disease (COPD) in general practices (GPs) in Germany. This retrospective cross-sectional study was based on data from the Disease Analyzer database (IQVIA), This retrospective cross-sectional study included all patients with at least one confirmed asthma or COPD diagnosis in one of those 50 general practices in Germany between January 2020, and January 2021, as well as at least one visit to these general practices between January 2021, and January 2022. The main outcomes of the study aimed to ascertain the proportion of spirometry testing among asthma and COPD patients between January 2021, and January 2022, overall, and separately, in men, women, six age groups (≤30, 31–40, 41–50, 51–60, 61, 70, >70), and patients who received at least one prescription of anti-asthma or anti-COPD drugs. This study included 8835 patients with asthma only, 5597 with COPD only, and 1897 with both asthma and COPD diagnoses. Of these, 27.2% of COPD patients, 7% of asthma patients, and 54.7% of asthma + COPD patients, received spirometry testing during the study period. Among COPD and asthma + COPD patients, the prevalence of spirometry testing was much higher in women than in men (COPD: 31.6% vs. 23.2%; asthma + COPD: 59.6% vs. 46.3%) and much higher in treated than in non-treated patients (COPD: 31.7% vs. 15.0%; asthma + COPD: 57.5% vs. 27.8%). The prevalence of spirometry testing was relatively low among COPD and asthma patients followed in GP practices, but usually higher in female patients, treated patients, and patients suffering from both asthma and COPD.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg
