    Complement Factor H-Related 3 Enhanced Inflammation and Complement Activation in Human RPE Cells

However, the non-canonical local, cellular functions of FHR-3 remained poorly understood. Here, we report that FHR-3 bound to oxidative stress epitopes and competed with FH for interaction. Furthermore, FHR-3 was internalized by viable RPE cells and modulated time-dependently complement component (C3, FB) and receptor (C3aR, CR3) expression of human RPE cells. Independently of any external blood-derived proteins, complement activation products were detected. Anaphylatoxin C3a was visualized in treated cells and showed a translocation from the cytoplasm to the cell membrane after FHR-3 exposure. Subsequently, FHR-3 induced a RPE cell dependent pro-inflammatory microenvironment. Inflammasome NLRP3 activation and pro-inflammatory cytokine secretion of IL-1ß, IL-18, IL-6 and TNF-α were induced after FHR-3-RPE interaction. Our previously published monoclonal anti-FHR-3 antibody, which was chimerized to reduce immunogenicity, RETC-2-ximab, ameliorated the effect of FHR-3 on ARPE-19 cells. Our studies suggest FHR-3 as an exogenous trigger molecule for the RPE cell “complosome” and as a putative target for a therapeutic approach for associated degenerative diseases

    Central Banks’ Predictability: An Assessment by Financial Market Participants

    In this paper, we examine the relationship between market participants’ perception of central bank predictability and their assessment of central bank communication skills and success in conveying objectives as well as the importance of transparencyenhancing measures, such as voting records, transcripts or minutes of policy meetings, and conditional interest rate projections. Our analysis is based on a unique dataset of almost 500 market participants worldwide who were asked questions with respect to the performance of the Bank of England, the Bank of Japan, the European Central Bank, and the Federal Reserve. Our results indicate a positive and economically notable relationship between central banks’ ability to convey their objectives and their overall communication skills on the one hand, and market participants’ perception of the banks’ predictability on the other hand, for all four central banks. The dissemination of more specific information does not appear to contribute to better central bank predictability. This raises doubts about the widely-held notion that implementing ever more transparency-enhancing measures will improve central bank predictability

    Neural correlates of temporal recalibration to delayed auditory feedback of active and passive movements

    When we perform an action, its sensory outcomes usually follow shortly after. This characteristic temporal relationship aids in distinguishing self‐ from externally generated sensory input. To preserve this ability under dynamically changing environmental conditions, our expectation of the timing between action and outcome must be able to recalibrate, for example, when the outcome is consistently delayed. Until now, it remains unclear whether this process, known as sensorimotor temporal recalibration, can be specifically attributed to recalibration of sensorimotor (action‐outcome) predictions, or whether it may be partly due to the recalibration of expectations about the intersensory (e.g., audio‐tactile) timing. Therefore, we investigated the behavioral and neural correlates of temporal recalibration and differences in sensorimotor and intersensory contexts. During fMRI, subjects were exposed to delayed or undelayed tones elicited by actively or passively generated button presses. While recalibration of the expected intersensory timing (i.e., between the tactile sensation during the button movement and the tones) can be expected to occur during both active and passive movements, recalibration of sensorimotor predictions should be limited to active movement conditions. Effects of this procedure on auditory temporal perception and the modality‐transfer to visual perception were tested in a delay detection task. Across both contexts, we found recalibration to be associated with activations in hippocampus and cerebellum. Context‐dependent differences emerged in terms of stronger behavioral recalibration effects in sensorimotor conditions and were captured by differential activation pattern in frontal cortices, cerebellum, and sensory processing regions. These findings highlight the role of the hippocampus in encoding and retrieving newly acquired temporal stimulus associations during temporal recalibration. Furthermore, recalibration‐related activations in the cerebellum may reflect the retention of multiple representations of temporal stimulus associations across both contexts. Finally, we showed that sensorimotor predictions modulate recalibration‐related processes in frontal, cerebellar, and sensory regions, which potentially account for the perceptual advantage of sensorimotor versus intersensory temporal recalibration.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Diagnostic stability in autism spectrum disorders and possible predictors of diagnosis change at the item level of the Diagnostic Observation Scale for Autistic Disorders- a descriptive analysis

    Die Autismus-Spektrum-Störung (Autism spectrum disorders, ASD) gehört zu den neuronalen Entwicklungsstörungen und beeinträchtigt Betroffene deutlich in ihrem Alltag. Daher ist eine präzise Diagnostik unabdingbar, um die Betroffenen bestmöglich unterstützen und therapieren zu können. Allerdings weisen ASD keine vollständige Diagnosestabilität auf. Das bedeutet, dass PatientInnen eine zunächst erhaltene ASD-Diagnose zu einem späteren Zeitpunkt wieder abgeben, da sie die diagnostischen Kriterien nicht mehr erfüllen oder dass PatientInnen zunächst eine andere oder keine Diagnose erhalten und erst zu einem späteren Zeitpunkt eine ASD diagnostiziert wird. Daraus ergeben sich vier Diagnosegruppen: ASD stabil (PatientInnen, welche konstant eine ASD-Diagnose erhalten), NON-ASD stabil (PatientInnen, welche konstant keine ASD-Diagnose erhalten), ASD zu NON-ASD (PatientInnen, welche von einer ASD-Diagnose zum ersten Messzeitpunkt zu keiner ASD-Diagnose zum zweiten Messzeitpunkt wechseln) und NON-ASD zu ASD (PatientInnen, welche von keiner ASD-Diagnose zum ersten Messzeitpunkt zu einer ASD-Diagnose zum zweiten Messzeitpunkt wechseln). Verschiedene Faktoren, die die Diagnosestabilität beeinflussen oder Aussagen darüber zulassen, ob jemand eine Diagnose im Verlauf abgibt bzw. erhält, werden in dieser Arbeit beschrieben (vgl. Kapitel 3 bis 3.3.5). Die vorliegende Arbeit untersucht einen zentralen Baustein der Autismusdiagnostik – die Diagnostische Beobachtungsskala für autistische Störungen (ADOS). Im Sinne einer deskriptiven Übersichtsarbeit wird die Fragestellung bearbeitet, ob es bestimmte Items des ADOS gibt, welche sich in ihrer Ausprägung bzw. Entwicklung zwischen den Diagnosegruppen unterscheiden und so zur Charakterisierung der einzelnen Gruppen dienen können. Dies soll als Grundstein dienen, um in weiteren Untersuchungen valide Prädiktoren für die Zuordnung zu einzelnen Diagnosegruppen zu identifizieren. In diesem Sinne werden 12 modulübergreifende Items sowie die einzelnen Skalen des ADOS untersucht. Zudem prüft die Arbeit zwei der transdiagnostischen Faktoren, die Einfluss auf die Diagnosestabilität nehmen. Zum einen das Alter bei Diagnosestellung, zum anderen den Zusammenhang zwischen der Entwicklung von sprachlichen Fähigkeiten und der Kommunikation bzw. der sozialen Interaktion. Die Untersuchungen zeigen, dass es unter den modulübergreifenden Items zwei Items gibt, welche sich signifikant zwischen den Diagnosegruppen unterscheiden: die Items ENJ (gemeinsame Freude an der Interaktion) und EYE (ungewöhnliche Blickkontakte). Auf Skalenebene zeigt sich eine unterschiedliche Ausprägung in den Diagnosegruppen für die Skala „Sozialer Affekt“ (SA) sowie den Comparison Score (Vergleichswert des ADOS, um ADOS-Werte über verschiedene Module hinweg vergleichbar zu machen). Die Ausprägung der Skala „Restriktive und Repetitive Verhaltensweisen“ (RRV) unterscheidet sich nicht signifikant zwischen den Diagnosegruppen. Ein positiver Zusammenhang zwischen der Entwicklung von sprachlichen Fähigkeiten (gemessen an den Items NESL; Gesamtniveau der nicht-echolalischen Sprache und IECHO; unmittelbare Echolalie) und der Entwicklung der der Kommunikation bzw. der sozialen Interaktion (gemessen am SA-Score) kann für das Item NESL gezeigt werden. Die aufgestellte Hypothese, dass Betroffene der Diagnosegruppe NON-ASD zu ASD zum ersten Diagnosezeitpunkt jünger sind, als die in den anderen Diagnosegruppen, kann in der vorliegenden Arbeit nicht belegt werden, jedoch zeigt sich eine positive Korrelation zwischen dem Abstand der Messzeitpunkte und der Abnahme der Skala SA bzw. des Comparison Scores. Die Ergebnisse der Arbeit auf Skalenebene unterstreichen die Validität des ADOS sowie seine zentrale Rolle im diagnostischen Prozess. Allerdings wird deutlich, dass die Skala RRV sich nicht eignet, um die Diagnosegruppen voneinander zu unterscheiden. Dies liegt vorwiegend daran, dass restriktive und repetitive Verhaltensweisen über alle Diagnosegruppen hinweg nur geringfügig ausgeprägt sind. Auf Itemebene rückt durch die Ergebnisse insbesondere der Bereich der wechselseitigen sozialen Interaktion der Skala SA in den Fokus. Die Items ENJ und EYE sollten als mögliche Prädiktoren evaluiert werden, sowie weitere nicht-modulübergreifende Items aus dem Bereich des Sozialen Affektes. Obwohl unter den untersuchten Items keines der sprachbezogenen Items Unterschiede in der Ausprägung vorweist, lässt sich durch den beschriebenen Zusammenhang zwischen sprachlichen Fähigkeiten und SA-Score vermuten, dass die sprachlichen Fähigkeiten trotzdem Einfluss auf die Diagnosestabilität nehmen. Die Untersuchung weiterer nicht-modulübergreifender sprachbezogenener Items ist an dieser Stelle sinnvoll. Auch mögliche Gründe für die positive Entwicklung bei Betroffenen mit größerem Abstand zwischen den beiden Messzeitpunkten (beispielsweise Therapiedauer oder Reifung) sollten in weiteren Arbeiten untersucht werden. Ausgehend von den Ergebnissen der vorliegenden Arbeit, ergeben sich weitere Hypothesen, insbesondere in Bezug auf die Items ENJ und EYE. Die Ergebnisse bilden einen wichtigen Grundstein auf dem Weg zur Identifizierung von validen Prädiktoren für die Diagnosegruppenzugehörigkeit.Autism spectrum disorders (ASD) belong to the group of neuronal developmental disorders and significantly impair affected individuals in their daily lifes. Therefore, a precise diagnosis is essential in order to provide the best possible support and therapy for those affected. However, ASDs do not exhibit complete diagnostic stability. This means that patients who initially receive an ASD diagnosis may later relinquish it because they no longer meet the diagnostic criteria, or that patients may initially receive a different diagnosis or no diagnosis at all and only later be diagnosed with an ASD. This results in four diagnostic groups: ASD stable (patients who stably receive an ASD diagnosis), NON-ASD stable (patients who stably do not receive an ASD diagnosis), ASD to NON-ASD (patients who change from an ASD diagnosis at the first measurement point to no ASD diagnosis at the second measurement point), and NON-ASD to ASD (patients who change from an ASD diagnosis at the first measurement point to an ASD diagnosis at the second measurement point). Various factors that influence diagnostic stability or allow conclusions to be drawn about whether someone gives up or receives a diagnosis in the course of time are described in this thesis (cf. Chapters 3 to 3.3.5). The present thesis examines a central component of autism diagnostics - the ADOS. In the sense of a descriptive review, the question is addressed whether there are certain items of the ADOS which differ in their expression or development between the diagnostic groups and can thus serve to characterize the individual groups. This will serve as a cornerstone to identify valid predictors for assignment to individual diagnostic groups in further studies. With this in mind, 12 cross-module items as well as the individual scales of the ADOS are examined. In addition, the thesis examines two of the transdiagnostic factors that influence diagnostic stability. First, the age at diagnosis, and second, the relationship between the development of language skills and communication or social interaction. The research shows that among the cross-module items there are two items that differ significantly between the diagnostic groups: the items ENJ (shared enjoyment of interaction) and EYE (unusual eye contact). On the scale level, there is a different expression in the diagnostic groups for the scale "Social Affect" (SA) as well as the Comparison Score (comparison value of the ADOS to make ADOS values comparable across different modules). The expression of the scale "Restrictive and Repetitive Behaviors" (RRV) does not differ significantly between the diagnostic groups. A positive correlation between the development of language skills (measured by the items NESL ; total level of non-echolalic speech and IECHO ; immediate echolalia) and the development of communication or social interaction (measured by the SA score) can be shown for the item NESL. The raised hypothesis that affected persons in the diagnosis group NON-ASD to ASD are younger at the time point of the first diagnosis than those in the other diagnosis groups cannot be proven in the present work, but a positive correlation between the distance of the measurement time points and the decrease of the scale SA respectivly the Comparison Score is shown. The results of the work at the scale level underline the validity of the ADOS as well as its central role in the diagnostic process. However, it is clear that the RRV scale is not suitable for distinguishing the diagnostic groups from one another. This is predominantly due to the fact that restrictive and repetitive behaviors are only slightly pronounced across all diagnostic groups. On the item level, the results bring the area of reciprocal social interaction of the scale SA particulary into focus. The items ENJ and EYE should be evaluated as possible predictors, as well as other non-crossmodule items from the domain of social affect. Although none of the language-related items shows differences in expression among the examined items, the described correlation between linguistic abilities and SA score suggests that linguistic abilities nevertheless influence diagnostic stability. The investigation of further non-crossmodule language-related items is reasonable at this point. Also, possible reasons for the positive development in affected individuals with a larger gap between the two measurement time points (for example, therapy duration or maturation) should be investigated in further work. Based on the results of the present work, further hypotheses arise, especially with regard to the items ENJ and EYE. The results form an important cornerstone on the way to identifying valid predictors for diagnostic group membership

    Charakterisierung und Training frontaler Dysfunktionen bei juveniler myoklonischer Epilepsie (JME)

    Das Ziel der vorliegenden klinischen Studie war es, erstmals ein Schulungsprogramm für Patienten mit JME zu schaffen, welches die eigene Wahrnehmung- und Achtsamkeit trainiert, um geeignete Kompensationsmechanismen zu finden, Lösungsansätze zu erlernen, die psychosozialen, alltagsrelevante Schwierigkeiten auszugleichen und das Leben mit der Krankheit zu erleichtern. Maßstab der Wirksamkeit waren psychometrische und neuropsychologische Testbatterien, die vor und nach dem Interventionsprogramm angewendet wurden. Gleichwohl nicht alle Testergebnisse Signifikanzen aufweisen und die Stichprobe mit n = 13 relativ gering ist, gibt es einige Indikatoren, die auf eine Wirksamkeit dieser Intervention hinweisen. Im FERUS ist ein signifikanter Anstieg der Selbstwirksamkeit unmittelbar nach dem Interventionstraining und im Follow up FU6 zu verzeichnen. Dabei ist die Selbstwirksamkeit (Bandura & Adams, 1977) ein Maßstab für die Änderung von Denk- und Verhaltensweisen. Es geht um die Überzeugung aus eigener Kraft Herausforderungen und Krankheiten zu beeinflussen oder bewältigen zu können. Es ist eine Art Schlüsselrolle, die Menschen hilft aus ihrer Passivität herauszukommen und aktiv Verhaltensweisen bestmöglich zu verändern. Neben indirekter Messung über den FERUS, weisen auch direkte Fragen beim FB des Epilepsiemanagement im Langzeit-follow up auf ein besseres Verständnis über, und einen besseren Umgang mit der Erkrankung hin. Über das Selbstwirksamkeitstraining hinaus konnten auch signifikante Effekte in der Wahrnehmung von Gefühlen, als Teilbereich sozialer Kognitionen, festgestellt werden. Neben Signifikanzen weisen auch Trends auf eine Verbesserung der sozialen Kognition nach Intervention hin, wie die Skalen der Reflexibilität und Durchsetzungsvermögen im ISK-K sowie im extern orientierten Denkstil des TAS-26. Bei diesen verbesserten sich die Werte auch nach dem Langzeit-follow up stetig. Dies deutet auf ein verbessertes Bewusstsein kognitiver Vorurteile und Verzerrungen hin und erfüllt damit das Kernziel des MKT. Die Schritte nach dem Erkennen und Bewusstwerden der eigenen Selbstwirksamkeit und den stereotypischen Denk- und Handelsweisen ist der Umgang mit diesen zu erlernen und bei Bedarf im Alltag anzupassen. Dies ist ein Teilaspekt der Entspannungsübung, welche über alle Module hinweg zur signifikanten Reduktion von Spannungszuständen führte und die Motivation zur eigenständigen Ausführung auch sechs Monate nach der Intervention aufrechterhalten konnte. Der erhoffte Effekt dieser Entspannungsübung, eine Reduktion der Provokationsfaktoren und Verminderung der Anfallsfrequenz, konnte nicht erreicht werden. Trotz der theoretisch optimalen Kombination aus Psychoedukation (Vermittlung von Wissensinhalten in Bezug auf Epilepsie, Anfälle und deren Umgang) und anschließender Entspannungsübung als praktische Umsetzung des Gelernten, veränderte sich die Anfallsfrequenz der Untersuchungsgruppe nicht. Womöglich ist der ausschlaggebende Faktor die Homogenität der Gruppe. In dieser lagen die Anfälle über ein Jahr zurück. Zudem nahmen die Probanden konsequent ihre Medikation ein, da sie berufstätig sind und auf eigenständiges Autofahren (Fahrerlaubnis) angewiesen sind. Im Gegensatz dazu wurden Gedächtnisübungen in dieser Intervention nicht direkt trainiert. Wie erwartet veränderten sich die Ergebnisse der Gedächtnistests nur wenig. Die Trends im WMS-R, genau dem Block Span Test vorwärts und Digit Span Test rückwärts deuten auf Verbesserungen des Kurzeit- und Arbeitsgedächtnisses hin. Es ist anzunehmen, dass weitere Zykluseinheiten oder angehängte Trainingsmodule diese Effekte in Zukunft verstärken können. Sicherlich sind zusätzliche Forschungsansätze notwendig, um die Wirksamkeit zu validieren, weitreichendere Erkenntnisse (in Bezug auf Effekte der Exekutivfunktion und Anfallsreduktion) einzuholen und das Interventionsprogramm dahingehend auszuweiten. Zusammenfassend zeigt sich, dass solche Interventionsprogramme einen möglichen neuen Therapieansatz in der Behandlung von JME darstellen, welche langfristig ergänzend zur pharmakologischen Therapie – im ambulanten oder stationären Setting – angeboten werden können. Insbesondere um Therapiemöglichkeiten zu schaffen, die neben symptomorientierter antikonvulsiver Behandlung psychosoziale und kognitive Defizite abdecken und die Lebensqualität steigern.The main goal of the study was the creation of an intervention program specifically aimed at people with juvenile myoclonic epilepsy trying to improve their aware- and mindfulness in order to better cope with daily psychosocial struggles caused by this disease. To measure the effectiveness psychometric and neuropsychological tests were used at the start of, end of, and six months after the intervention. On the one hand alexithyma, mood and social skills were key aspects to the psychometric tests. On the other hand cognition, attention, memory, and executive functions were tested in the neuropsychological assessments. This intervention program is based on a metacognitive training program for schizophrenia which also includes theory of mind (ToM) elements. Therefore the training was specifically modified for JME. That meant each person in the sample participated in a five week training program which initially taught the psychoeducational basis of JME, followed by relaxation and ToM exercises based on MCT. Additionally, the participants were given homework for each week manifesting the learned methods. All that combined enhanced the social cognition and overall coping with JME. Not all tests were significant, but some results definitely indicated the effectiveness of this intervention. For example, the significance in self-efficacy increased immediately after the training and also in the six months later follow up session. The results of the epilepsy management questionnaire showed a better understanding and coping with JME. Scores for overall alexithymia and difficulties in emotion identification increased from post training to the six month follow up session. Furthermore, significant effects of alexithyma, like the perception of feelings, indicated an improvement in awareness and social cognition. Some tests weren’t significant but trends in reflexibility, externally oriented thinking or neuropsychological tests like attention and working memory confirmed the efficiency of the training in different kinds of ways. Other than the results just mentioned, the patient’s stress levels decreased significantly after each module. Due to long term seizure-free periods, experienced by all participants, a further reduction in this matter could not be properly measured. With the only exception being one patient who had a single seizure within the 12 months prior the intervention. 150 Presently there’re only few therapy options for patients with JME. The method discussed in this paper still needs further validation and elaboration to confirm it’s results. However, the most effective therapy for JME patients would most likely be a combination of this specific MCT and pharmacological treatments

    Life Satisfaction, Contract Farming and Property Rights: Evidence from Ghana

    Recently, large-scale land acquisition has increased dramatically in the developing world. The question whether land deals can benefit both the local population and the investor is therefore high on the international agenda. Contract farming is discussed as a possible solution but studies identifying the causal effects are rare. Using data from a quasi-natural experiment in contract allocation, we compare the subjective well-being of outgrowers and independent farmers in the sphere of the biggest palm oil producer in Ghana. We identify a positive causal effect of the outgrower scheme which increases subjective well-being by 1.5 points on a scale of 0 to 10. We find a substitutive relationship between having an outgrower contract and having property rights, and thus we argue that by increasing security a contract increases well-being, as secure rights to land matter substantially for the overall life satisfaction of non-contract but not of contract farmers

    The Impact of Incentive Pay on Corporate Crime

    This paper presents a moral hazard model analyzing the agent's incentive to commit corporate crime. The principal can only observe profits which the agent can increase by committing crime or exerting effort. It is shown how different incentive contracts, i.e., thresholdlinear, capped bonus and linear contracts, can be adjusted in order to promote agent's law abiding behavior. Any adjustment implies a loss in internal efficiency which decreases in individual sanctions imposed on the agent

    Cognitive functioning associated with acute and subacute effects of classic psychedelics and MDMA - a systematic review and meta-analysis

    Classic psychedelics and MDMA have a colorful history of recreational use, and both have recently been re-evaluated as tools for the treatment of psychiatric disorders. Several studies have been carried out to assess potential long-term effects of a regular use on cognition, delivering distinct results for psychedelics and MDMA. However, to date knowledge is scarce on cognitive performance during acute effects of those substances. In this systematic review and meta-analysis, we investigate how cognitive functioning is affected by psychedelics and MDMA during the acute drug effects and the sub-acute (“afterglow”) window. Our quantitative analyses suggest that acute cognitive performance is differentially affected by psychedelics when compared to MDMA: psychedelics impair attention and executive function, whereas MDMA primarily affects memory, leaving executive functions and attention unaffected. Our qualitative analyses reveal that executive functioning and creativity may be increased during a window of at least 24 h after the acute effects of psychedelics have subsided, whereas no such results have been observed for MDMA. Our findings may contribute to inform recommendations on harm reduction for recreational settings and to help fostering differential approaches for the use of psychedelics and MDMA within a therapeutic framework.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Investigation of the Structural and Optoelectronic Properties of Fluorinated Organic Semiconductors

    Die rasche Entwicklung der organischen Elektronik und die Vielzahl ihrer Anwendungen, seien es Leuchtdioden, Transistoren oder Sensoren, führen zu einem kontinuierlichen Anstieg der Anforderungen an die zugrundeliegenden molekularen Materialien. Während heutzutage vielseitige chemische Funktionalisierungen möglich sind und die molekularen Eigenschaften durch theoretische Berechnungen gut vorhergesagt werden können, ist die Kontrolle von Festkörpereigenschaften durch das Moleküldesign nicht vollumfänglich möglich. Denn diese Festkörpereigenschaften werden durch das Packungsmotiv bestimmt, welches wiederum durch intermolekulare Wechselwirkungen festgelegt wird. Die theoretische Beschreibung dieser intermolekularen Wechselwirkungen und die Vorhersage der molekularen Packung ist nur für einfache Systeme möglich, sodass die experimentelle Korrelation von Moleküldesign und realisierter Struktur notwendig ist. In dieser Arbeit werden die intermolekularen Wechselwirkungen von den gut untersuchten polyzyklischen aromatischen Kohlenwasserstoffen (PAK) durch Heteroatomsubstitution gezielt verändert, um die Auswirkungen auf das Packungsmotiv und somit die Festkörpereigenschaften zu untersuchen. Dabei ist Fluorierung die gewählte Substitution, da Fluor das Atom mit der höchsten Elektronegativität ist und dadurch die unpolaren Kohlenstoff-Wasserstoff-Bindungen durch polare Kohlenstoff-Fluor-Bindungen ersetzt werden. Dies verändert die Elektronendichte der PAK und somit auch das molekulare elektrostatische Potential. Folglich können die intermolekularen elektrostatischen Wechselwirkungen durch regioselektive Fluorierung gezielt modifiziert werden. Das Einbauen von lokalen, stark polaren Bindungen wirkt sich nicht nur auf das molekulare elektrostatische Potential aus, sondern auch auf die optoelektronischen Moleküleigenschaften. So führt der elektronenziehende Charakter der Fluoratome zu einer Reduktion der Elektronendichte im Inneren der PAK und induziert somit positiv geladene Kohlenstoffatome. Dadurch sind die Rumpfelektronen stärker gebunden, was anhand einer starken chemischen Verschiebung in den Röntgenphotoelektronenspektren messbar ist. Ebenso erfahren die π-Elektronen eine stärkere Coulomb-Wechselwirkung mit den Kohlenstoffatomen, was zu einer Absenkung der Grenzorbital-Energieniveaus führt. Mithilfe von optischer Absorptions- und der Röntgennahkantenabsorptionsspektroskopie kann gezeigt werden, dass die Rumpf- und Grenzorbital-Energieniveaus gleichermaßen abgesenkt werden. Das veränderte elektrostatische Potential der fluorierten Moleküle beeinflusst die intermolekularen Wechselwirkungen dahingehend, dass das Packungsmotiv geändert wird. So führt im Falle der Acene eine Teilfluorierung zum Aufbrechen des typischen Herringbone Packungsmotives und zum Auftreten von planar gestapelten Molekülen. Die Kristallzucht polarer Acene in einer Umgebung von polaren Lösungsmolekülen ermöglicht die Ausbildung einer Dipol-parallelen Anordnung der Acene, da die Dipolmomente der Lösungsmoleküle die Dipolmomente der polaren Acene beim Wachstum kompensieren. Dagegen führt die Kristallzucht in Abwesenheit polarer Moleküle, beispielsweise in unpolarem Toluol oder durch Vakuumsublimation, zur Kristallisation polarer Acene in der stabileren Dipol-antiparallelen Packung. Die unterschiedlichen Festkörperpackungen von Tetracen, das im Herringbone Packungsmotiv kristallisiert und fluorierten Tetracenen, welche im Festkörper planar gestapelte Moleküle aufweisen, legen die spektroskopische Charakterisierung nahe. Die Untersuchung der Festkörperphotolumineszenz zeigt, dass die fluorierten Tetracene eine gegenüber dem ursprünglichen Tetracen und den Molekülen in Lösung verbreiterte und rot-verschobene Lumineszenz aufweisen. Dies wird auf eine Exzimerbildung, also eine strukturelle Relaxation der angeregten Zustände, welche für planar gestapelte Moleküle auftreten kann, zurückgeführt. Dieses Beispiel zeigt eindrucksvoll, dass Fluorierung über die Modifikation des elektrostatischen Potentials die Festkörperpackung verändert und dadurch die optoelektronischen Festkörpereigenschaften, wie die Photolumineszenz, bestimmt. Vor dem Hintergrund, dass die Elektrostatik maßgeblich das Packungsmotiv beeinflusst, wird eine Analyse der Packungsmotive bekannter PAK, sowie deren funktionalisierten Gegenstücke vorgenommen. Dabei werden die beobachteten Kristallstrukturen in vier grundsätzliche Packungsmotive klassifiziert und diese wiederum mit der molekularen Elektrostatik korreliert. Basierend auf diesen Überlegungen werden teilfluorierte Tetracene entworfen, welche den gleichen Fluorierungsgrad aufweisen, für die aber verschiedene Packungsmotive und damit unterschiedliche optoelektronische Festkörpereigenschaften erwartet werden. Somit weist diese Arbeit einen Weg auf, Festkörpereigenschaften durch die Variation des Fluorierungsmusters gezielt zu modifizieren. Allgemein erweitern die in dieser Arbeit gewonnenen Erkenntnisse das Verständnis über die molekularen Eigenschaften, sowie die intermolekularen Wechselwirkungen organischer Halbleiter. Dieses Wissen erlaubt es, die optoelektronischen Eigenschaften im Festkörper gezielt durch das Moleküldesign zu kontrollieren. Denn die Energieniveaus, welche über den Fluorierungsgrad eingestellt werden können, beeinflussen die Injektionsbarrieren und den Ladungstransport in organischen Bauelementen. Die molekulare Orientierung, welche über das Fluorierungsmuster gegeben ist, bestimmt die Richtung des emittierten Lichtes in Leuchtdioden oder den anisotropen Ladungstransport in Transistoren. Somit ermöglicht diese Arbeit neue Strategien, die molekularen Eigenschaften und die Festkörperpackung zu kontrollieren und dadurch organische Bauelemente wie Sensoren, Transistoren oder Leuchtdioden zu verbessern.The rapid development of organic electronics and the multitude of its applications, be it light emitting diodes, transistors or sensors, leads to a continuous increase in the demands on the underlying molecular materials. While versatile chemical functionalizations are possible nowadays and the molecular properties can be predicted well by theoretical calculations, controlling solid-state properties by molecular design is not fully possible, as these solid-state properties are governed by the packing motif. The packing motif in turn is determined by intermolecular interactions. The theoretical description of these intermolecular interactions and the prediction of molecular packing is only possible for simple systems, raising the need to experimentally correlate molecular design and the realized structure. In this work, the intermolecular interactions of the well-studied polycyclic aromatic hydrocarbons (PAHs) are specifically modified by heteroatom substitution in order to investigate the effects on the packing motif and thus the solid-state properties. Fluorination is the chosen substitution, as fluorine is the atom with the highest electronegativity and thus the non-polar carbon-hydrogen bonds are replaced by polar carbon-fluorine bonds. This changes the electron density in the PAHs and thus also the molecular electrostatic potential (MEP). Consequently, the intermolecular electrostatic interactions can be specifically modified by regioselective fluorination. The incorporation of local, strongly polar bonds not only affects the molecular electrostatic potential, but also the molecular optoelectronic properties. The electron withdrawing character of the fluorine atoms leads to a reduction in the electron density inside the PAH and thus induces more positively charged carbon atoms. As a result, the core electrons are more strongly bound, which is visible in a strong chemical shift in the X-ray photoelectron spectra. The π-electrons also experience a stronger Coulomb interaction with the carbon atoms, which leads to a lowering of the frontier orbital energy levels. Using optical absorption and near-edge X-ray absorption fine structure spectroscopy, it can be shown that the core and frontier orbital energy levels are lowered equally. Indeed, the altered electrostatic potential of the fluorinated molecules influences the intermolecular interactions and thereby changes the packing motif. Thus, in the case of acenes, partial fluorination leads to the breaking of the typical herringbone packing motif and the appearance of planarly stacked molecules. Crystal growth of polar acenes in a polar environment enables the formation of a dipole-parallel arrangement of the acenes, as the dipole moments of the polar solvent molecules compensate for the dipole moments of the polar acenes during growth. In contrast, crystal growth in the absence of polar molecules, for example in non-polar toluene or by vacuum sublimation, leads to the crystallization of polar acenes in the more stable dipole-antiparallel packing. The different solid-state packing of tetracene, which crystallizes in the herringbone packing motif, and fluorinated tetracenes, which exhibit planarly stacked molecules in the solid state, suggest the spectroscopic characterization. The investigation of the solid-state photoluminescence shows that the fluorinated tetracenes exhibit a broadened and red-shifted luminescence compared to the original tetracene and the molecules in solution. This is attributed to excimer formation, i.e. a structural relaxation of the excited states, which can occur for planarly stacked molecules. This example shows impressively that fluorination changes the solid-state packing by modifying the electrostatic potential and thus determines the optoelectronic solid-state properties, such as photoluminescence. Considering that electrostatics significantly influence the packing motif, an analysis of the packing motifs of known PAHs and their functionalized counterparts is carried out. The observed crystal structures are classified into four basic packing motifs and these in turn are correlated with the molecular electrostatics. Based on these considerations, partially fluorinated tetracenes are designed, which have the same degree of fluorination, but for which different packing motifs and thus different optoelectronic solid-state properties are expected. Hence, this work paves the way to specifically modify solid-state properties by varying the fluorination pattern. In general, the knowledge gained in this work expands the understanding of the molecular properties and intermolecular interactions of organic semiconductors. This knowledge allows the optoelectronic properties in the solid state to be specifically tailored by the molecular design. For example, the energy levels, which can be adjusted via the degree of fluorination, influence the injection barriers and charge transport in organic electronic devices. The molecular orientation, which is given by the fluorination pattern, determines the direction of the emitted light in light-emitting diodes or the anisotropic charge transport in organic transistors. Hence this work provides novel strategies to control molecular properties and solid-state packing and thus to improve organic components such as sensors, transistors or light-emitting diodes

    Struktur-basierte Funktions Annotierungen Archaellum regulierender Netzwerk Proteine aus Sulfolobus acidocaldarius

    Der Ursprung des Lebens ist ein komplexes Thema, das weltweit wissenschaftliche Diskussionen befeuert. Im Allgemeinen ist es akzeptiert, dass die Endosymbiose eine zentrale Rolle bei der Eukaryogenese spielt. Für diese Arbeit wurde die inside-out Hypothese, welche sich auf die Asgard Archaeen stützt und ein frühes Endomembransystem voraussetzt, als Ausgangslage angenommen. Da Asgard Archaeen noch nicht kultiviert wurden, stellen die TACK Archaeen mit ihrem archaealen Modellorganismus Sulfolobus acidocaldarius einen der nächsten, verfügbaren Verwandten der Eukaryoten dar. Interessanterweise wurde das Protein ArnB des archaellum regulatory networks (Arn), welches strukturelle Homologien mit dem Sec23/24 Protein des COPII-systems aufweist, in S. acidocaldarius Exosomen gefunden. Entsprechend erweist sich ArnB als vielversprechendes Protein, um den prä-endosymbiotischen Transport in Archaeen zu untersuchen. Diese Arbeit umfasst eine Zusammenfassung der Forschungsergebnisse zu ArnB und seinen assoziierten Proteinen. Hervorzuheben ist dabei die Untersuchung der Interaktion von ArnB mit dem ZnF- und FHA-Domänen enthaltenden ArnA, welche eine sequenzielle, phosphorylierungsabhängige strukturelle Transition mit globalen Änderungen offenbart. Die Untersuchung des ArnAB-Komplexes ergab zum einen neuartigen sequenziellen Phosphorylierungsmechanismus und zeigte die Gegenwart des Sec23/24-Kernmotivs in allen Domänen des Lebens auf. In Zuge dieser Analysen, konnten dem ArnAB-Komplex bzw. dem Sec23/24-Kernmotiv zahlreiche potenzielle Funktionen zugeschrieben werden, die von der Beteiligung am Ubiquitin-Netzwerk bis zur Bildung von COPII-Vesikeln und Assemblierungsrollen reichen. Zudem beinhaltet diese Arbeit die Charakterisierung einer GPN-Loop-GTPase aus S. acidocaldarius, SaGPN, welche ebenfalls mit dem Arn-System assoziiert ist und potentiell mit dem ArnAB-Komplex interagiert. GPN-Loop-GTPasen stellen einen eigenen interessanten GTPase-Typ dar, der durch nukleotidunabhängige Homodimere gekennzeichnet ist. Durch die Kristallstrukturen die in dieser Arbeit gelöst wurden, lassen sich mechanistische Unklarheiten im Zusammenhang mit GPN-Loop-GTPasen lösen und ein Lock-Switch-Rock-Mechanismus für archaeale GPN-Loop-GTPasen ableiten. Zusammenfassend trägt diese Arbeit zum allgemeinen Verständnis der GPN-Loop- GTPasen bei und hebt das ArnAB-Sec23/24-Kernmotiv als eine der häufigsten und funktional vielfältigsten Protein-Faltungen in allen Bereichen des Lebens hervor.The origin of Life is an enigmatic topic that fuels scientific discussion all over the world. In general, it is an accepted concept that endosymbiosis plays a central role within eukaryogenesis. In this context the inside-out theory, which centers on the emerging Asgard Archaea superphylum necessitating an early endomembrane system prior to any endosymbiotic events, holds significant promise as a hypothesis. Since Asgard Archaea had not been cultivatable the sister branch of TACK Archaea with its genetically tractable Sulfolobus acidocaldarius is one of the closest, available model organisms preceding the first eukaryotic common ancestor (FECA). Interestingly, the archaellum regulatory network protein ArnB, which shares structural homology with the membrane curvature protein Sec23/24 of COPII vesicles, was abundantly found in S. acidocaldarius exosomes. Accordingly, ArnB emerges as a promising research target to study pre-endosymbiotic trafficking in Archaea. This work encapsulates a culmination of research concerning ArnB and associated proteins. Of particular note is the exploration of ArnB’s interaction with its ZnF and FHA domain-containing counterpart, ArnA, revealing a sequential, phosphorylationdependent structural transition with far-reaching implications. Investigation of the ArnAB complex not only unveiled a novel sequential phosphorylation mechanism but also underscores the ubiquity of the Sec23/24 core motif across all domains of life. Accordingly, numerous potential functions can be attributed to the ArnAB complex or the Sec23/24 core motif, spanning from involvement in the ubiquitin network to COPII vesicle formation and even to other assembly roles. Additionally, this research delves into the characterization of a GPN-loop GTPase from S. acidocaldarius, known as SaGPN, which is implicated in the archaellum regulatory network as well and potentially interacts with the ArnAB complex. Despite the relative neglect of GPN-loop GTPases within the P-loop NTPases family, they represent a distinct GTPase type characterized by nucleotide-state independent homodimerization. The SaGPN crystal structures resolved mechanistic ambiguities surrounding GPN-loop GTPases, proposing a lock-switch-rock mechanism for SaGPN and its archaeal counterparts. In summary, this work advances the field of GPN-loop GTPases and positions the ArnAB-Sec23/24 core motif as one of the most prevalent and functionally diverse protein folds across all domains of life. Consequently, the presented research opens avenues for further exploration and discoveries in the Archaea-to-Eukarya transition
