    Distribution of ciliary adaptor proteins tubby and TULP3 in the organ of Corti

    Tubby-like proteins are membrane-associated adaptors that mediate directional trafficking into primary cilia. In inner ear sensory epithelia, cilia—including the hair cell’s kinocilium—play important roles as organizers of polarity, tissue architecture and cellular function. However, auditory dysfunction in tubby mutant mice was recently found to be related to a non-ciliary function of tubby, the organization of a protein complex in sensory hair bundles of auditory outer hair cells (OHCs). Targeting of signaling components into cilia in the cochlea might therefore rather rely on closely related tubby-like proteins (TULPs). In this study, we compared cellular and subcellular localization of tubby and TULP3 in the mouse inner ear sensory organs. Immunofluorescence microscopy confirmed the previously reported highly selective localization of tubby in the stereocilia tips of OHCs and revealed a previously unnoticed transient localization to kinocilia during early postnatal development. TULP3 was detected in the organ of Corti and vestibular sensory epithelium, where it displayed a complex spatiotemporal pattern. TULP3 localized to kinocilia of cochlear and vestibular hair cells in early postnatal development but disappeared subsequently before the onset of hearing. This pattern suggested a role in targeting ciliary components into kinocilia, possibly related to the developmental processes that shape the sensory epithelia. Concurrent with loss from kinocilia, pronounced TULP3 immunolabeling progressively appeared at microtubule bundles in non-sensory Pillar (PCs) and Deiters cells (DC).     Is a Secondary Currency Essential? – On the Welfare Effects of a New Currency

    The coexistence of cash and digital currencies constitutes a system of parallel currencies. This paper tackles the question whether a new (digital) currency is essential: Does a new currency allow for a better resource allocation even if a fully accepted currency is in circulation and still remains in circulation? Using the dual currency search model of Kiyotaki and Wright (1993), we show how the introduction of a secondary currency affects average utility. There is some scope for a welfare improvement, the welfare effect depends on differences in returns and costs, and, in particular, the fraction of cash traders who will be replaced by digital money traders

    Media content’s role in the making of a democrat: Evidence from East Germany

    This paper explores the causal influence of access to Western television programming on voting behavior. We exploit a natural experiment involving access to West German TV within the German Democratic Republic in which only geography and topography determined the allocation of individuals to treatment and control groups. Focusing on both the shares of extremist parties and voter turnout, we find that in the post-reunification decade in which TV content was harmonized, regions that already had access to Western TV broadcasts before reunification experience lower vote shares of extremist parties and higher voter turnout

    Die Bedeutung von MAGED2 für die Regulation von HIF-1α

    Mutationen des MAGED2-Gens, welches für das gleichnamige Protein MAGED2 kodiert, verursachen bei Betroffenen das transiente antenatale Bartter-Syndrom, welches sich phänotypisch mit schwersten renalen Salzverlusten präsentiert, die zu massiver Polyurie und somit zum ausgeprägten Polyhydramnion führen. Die besondere Schwere dieser Erkrankung wird durch die hohe Frühgeburtlichkeitsrate und die hohe Mortalität deutlich. Im Unterschied zu den anderen Formen des Bartter-Syndroms zeigt diese Erkrankung einen transienten Charakter und die Symptome verschwinden einige Wochen nach der Geburt bzw. ab der 30. Schwangerschaftswoche. Der Zeitpunkt des Symptomendes überschneidet sich mit dem Zeitpunkt, ab dem HIF-1α, welches als Indikator für hypoxische Bedingungen gilt, nicht mehr in fetalen Nieren nachweisbar ist und somit von einem ausreichenden Sauerstoffangebot auszugehen ist. Es ist bekannt, dass MAGE-Proteine Zellen vor unterschiedlichen Stressoren schützen. Auf Basis dieses Wissens wurde folgende Hypothese zur Pathophysiologie des transienten antenatalen Bartter-Syndroms verfasst: MAGED2 ist für den Schutz und die Funktion der renalen Salztransportmechanismen unter hypoxischen Bedingungen essentiell. Die Beobachtungen, dass bei Mutationen von MAGED2 ein schnellerer Abbau von NKCC2 stattfindet und auch die Menge von NCC an der apikalen Tubulusmembran reduziert ist, unterstützen diese These. Der zugrundeliegende, detaillierte Pathomechanismus ist bisher nicht geklärt und die weitere Untersuchung Bestandteil dieser Arbeit. Aufgrund der Beobachtung, dass wt-MAGED2 mit Gαs interagiert, während diese Interaktion zwischen Gαs und mt-MAGED2 nicht nachweisbar ist, ist die Vermutung, dass diese Wechselwirkung wesentlich für die Adaptation der Zellen an hypoxische Bedingungen ist. Gαs wirkt stimulierend auf den cAMP/PKA-Signalweg, der wiederum für den Erhalt der Salztransportprozesse unter hypoxischen Bedingungen notwendig ist, da dieser unter anderem stabilisierend auf den Transkriptionsfaktor HIF-1α wirkt. Die Ergebnisse der Untersuchungen bestätigten diese Annahme und machten eine reziproke Beeinflussung zwischen MAGED2, dem cAMP/PKA-Signalweg und HIF-1α deutlich, wobei Gαs als gemeinsame Schnittstelle agiert. Dabei kommt MAGED2 eine Schlüsselrolle zu, die insbesondere in Situationen, in denen die Zellen Hypoxie ausgesetzt sind, zum Tragen kommt. Durch die fehlende Interaktion von MAGED2 und Gαs bei Depletion bzw. Funktionsverlust von MAGED2 kommt es zur fehlregulierten Internalisierung von Gαs. Dies führt zu einer gestörten Aktivierung des cAMP/PKA-Signalwegs sowie einer unzureichenden Stabilisierung von HIF-1α. Um die Bedeutung des cAMP/PKA-Signalwegs und MAGED2 für die hypoxische Induktion von HIF-1α genauer zu untersuchen, wurde in weiteren Experimenten versucht, die beeinträchtigte HIF-1α-Induktion wiederherzustellen. Dabei kamen Isoproterenol und Forskolin zum Einsatz, wobei Ersteres upstream und Letzteres downstream von Gαs wirkt. Forskolin war in der Lage, die HIF-1α-Expression in den MAGED2-depletierten Zellen deutlich über die Kontrollwerte zu steigern, während die Exposition mit Isoproterenol keine erkennbare Wirkung zeigte. Um die beteiligte PKA-Isoform der beschriebenen Prozesse zu identifizieren, wurde der Einfluss zweier PKA-Inhibitoren untersucht. Die Ergebnisse machten deutlich, dass PKA-Isoform II an der Regulation von HIF-1α beteiligt ist. Interessanterweise konnte außerdem eine Erhöhung der MAGED2-mRNA und -Proteinexpression durch den Einfluss des cAMP-Mimetikums Forskolin unter Hypoxie beobachtet werden. Außerdem zeigte sich die Abhängigkeit der MAGED2-Proteinexpression von HIF-1α, was zusätzlich die Beobachtung der wechselseitigen Regulation von MAGED2 und HIF-1α unter hypoxischen Bedingungen unterstützt. Die dargestellten Ergebnisse erklären in Teilen die unzureichende Anpassung der MAGED2-depletierten Zellen an hypoxische Bedingungen. Bei Patienten mit der transienten Form des antenatalen Bartter-Syndroms macht sich die unzureichende Anpassung an hypoxische Bedingungen anhand des Zusammenbruchs der Salztransportprozesse während der hypoxischen Phase in der Fetalperiode bemerkbar, was zumindest in Teilen eine Erklärung für den transienten Charakter ist. Auf Grundlage der Ergebnisse ist es denkbar, dass Betroffene im Verlauf ihres Lebens erneut Phasen erleben, in denen die Salztransportmechanismen aufgrund von Hypoxie, beispielsweise bei Aufenthalt in großen Höhen oder bei entzündlichen Prozessen, nicht aufrechterhalten werden können. Die Schwere des Phänotyps lässt sich vermutlich nicht allein auf Grundlage der oben dargestellten Mechanismen erklären und bedarf ebenso wie die Erforschung möglicher Therapieoptionen beispielweise mit Forskolin weitere Untersuchungen. Neben der bedeutenden Rolle von MAGED2 für die beschriebene Erkrankung sollte MAGED2 auch im Zusammenhang mit anderen Krankheiten nicht vernachlässigt werden. So wird MAGED2 in vielen Tumoren überexprimiert. Im Zusammenhang mit unseren Ergebnissen ist eine Überlegung, dass MAGED2 in der Tumorgenese eine bedeutende Rolle für die Anpassung der Tumorzellen an hypoxische Bedingungen hat und somit zum Progress und Wachstum von Tumoren beiträgt. Somit sollte ebenfalls die Rolle von MAGED2 für die Tumorgenese erforscht werden und MAGED2 als Angriffspunkt neuer Therapieansätze in Betracht gezogen werden

    Skin-infiltrating T cells display distinct inflammatory signatures in lichen planus, bullous pemphigoid and pemphigus vulgaris

    Introduction: We here thought to dissect the inflammatory signature in lesions of three skin disorders, which show a common adaptive immune response against autoantigens of the skin but are characterized by diverging clinical phenotypes. Pemphigus vulgaris (PV) and bullous pemphigoid (BP) are type-2-dependent, IgG autoantibody-driven blistering disorders of mucous membranes and skin, which target desmoglein (Dsg)3 and bullous pemphigoid (BP)180, respectively. In contrast, lichen planus (LP) is a common chronic inflammatory disease of the skin and mucous membranes with a pronounced dermal T cell infiltrate. We previously identified peripheral type 1 and 17 T cell responses against Dsg3 and BP180 in a cohort of LP patients strongly suggesting that the underlying inflammatory T cell signature may drive the evolving phenotype. Methods: Paraffin-embedded skin biopsies from well-characterized patients with LP (n=31), BP (n=19), PV (n=9), and pemphigus foliaceus (PF) (n=2) were analysed. Areas with the most prominent inflammatory infiltrate were excised with punch biopsies and tissue microarrays (TMA) containing multiple biopsies were created. Using multicolor immunofluorescence, the inflammatory infiltrate was stained with antibodies against multiple cellular markers, i. e. CD3ϵ, CD4, CD15, TCR-δ, the cytokine IL-17A, and the transcription factors, T-bet and GATA-3. Results: In LP, there was a higher number of CD4+ T cells expressing T-bet compared to GATA-3. In contrast, CD4+ T cells in PV and BP skin lesions more frequently expressed GATA-3 than T-bet. IL-17A+ cells and IL-17A+ T cells were found to a similar extent in all the three disorders. IL-17A+ granulocytes were more predominant in BP than in LP or PV. Of note, the majority of IL-17A+ cells in LP were neither T cells nor granulocytes. Discussion: Our findings in inflammatory skin infiltrates clearly show a predominant type 1 signature in LP in contrast to a preponderance of type 2 T cells in PV and BP. In contrast to LP, granulocytes and to a much lesser extent CD3+ T cells were a cellular source of IL-17A in BP and PV. These data strongly suggest that different inflammatory cell signatures drive evolving clinically diverse phenotypes of LP, PV and BP despite common target antigens of the skin.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg

    Public attention to environmental issues and stock market returns

    This paper empirically examines the effect of public attention to climate change and pollution on the weekly returns on US sustainability stock indices (i.e. the DJSI US and the FTSE4Good USA Index) in comparison to their conventional counterparts (i.e. the S&P 500 Index and the FTSE USA). In addition to unexpected global climate-related natural weather disasters, we consider two complementary measures of public attention to these environmen-tal issues: (i) US media attention to climate change and pollution and (ii) the US Google Search Volume Index for these two keywords. Robust to several sensitivity analyses, our econometric analysis for the period from 2004 to 2018 reveals that public attention to envi-ronmental issues has a significantly positive (negative) effect on the returns on US sustaina-bility (conventional) stock indices. A possible explanation of this result is that high public attention to environmental issues may drive traditionally sustainable investors, neo-sustainable, and opportunistic self-interested investors to favor stocks of sustainable firms. The insights from our empirical study are important for private and institutional investors, managers of firms, and public policy

    Wachstumsdynamik und Morphologie von GaP Nanodrähten untersucht mittels Transmissionselektronenmikroskopie

    In this work, the growth of GaP nanowires is demonstrated. Self-catalyzed nanowires were grown in a conventional MOVPE reactor by predeposition of Ga droplets by group III precursor preflow. Several growth series demonstrate the control over the morphology of the nanowires. Nanowire diameter can be controlled by temperature as well as TMGa preflow time, while the nanowire length is controlled by growth time. The conicity of the nanowires can be changed by a variation of the applied V/III precursor ratio. These achievements form a starting point for further growth experiments such as the growth of core/shell structures with GaP nanowires as cores. After finding the optimal growth parameters, also a transfer of the self-catalyzed growth to the in-situ setup is possible. The nanowires, presented in this work, which were grown in the in-situ setup, are grown with gold nanoparticles as catalysts. The grown nanowires show a linear dependence of their growth rate with the interface ratio. By this, the incorporation of the growth material in the catalyst droplet could be identified as the growth rate limiting step in VLS mechanism. Furthermore, investigations of the growth rates in dependence of the applied V/III ratio show that at a V/III ratio of 6.2±0.9, a transition from group V limited to group III limited growth takes place. This finding is in agreement to those in conventional MOVPE reactors, demonstrating the comparability of the in-situ setup with these MOVPE reactors. In a last step of investigation, the grown nanowires were investigated post-growth. Especially, the formation mechanism of the dominant kinks could be identified. Orientation mapping via SPED showed that a twin boundary of first order can initiate 70.5° and 109.5° kinks of the nanowires. The also frequent 123.7° kinks can be explained by second order twins. The relative occurrence of this kinks can presumably be explained a change in the elemental termination of the growth surface at 109.5° kinks. The twinning of the second-order by two subsequent 109.5° kinks could prevent this change in termination and thus explain the suppression of the 109.5° kink angles as well as the high occurrence of 123.7° kinks. It is expected that the formation of nanowire kinks is a thermodynamic effect and could be suppressed by the change in growth parameters, like the growth temperature.In der vorliegenden Arbeit wird das Wachstum von GaP Nanodrähten demonstriert. Dabei wurden selbstkatalysierte Nanodrähte in einem konventionellen MOVPE Reaktor gewachsen, indem Ga Tropfen durch einen Fluss von Gruppe III Präkursor vor dem Wachstum deponiert wurden. Verschiede Wachstumsserien demonstrieren dabei die Kontrolle über die Morphologie der Nanodrähte. Dabei kann der Nanodraht-Durchmesser durch die angelegte Temperatur und die TMGa Vorflusszeit geregelt werden. Die Länge der Nanodrähte lässt sich über die Wachstumsdauer kontrollieren. Weiterhin kann die Konizität der Nanodrähte mit dem angelegten V/III Verhältnis verändert werden. Diese Erkenntnisse bilden einen Starpunkt für weitere Wachstumsexperimente, wie das Wachstum von Kern/Hülle Nanodrähten mit den präsentierten GaP Nanodrähten als Kern. Nachdem die optimalen Wachstumsbedingungen gefunden wurden, kann das Wachstum von solch selbstkatalysierten Nanodrähten auch in das in-situ System übertragen werden. Die Nanodrähte, welche im Rahmen der vorliegenden Arbeit im in-situ System gewachsen wurden, wurden mit Goldnanopartikeln als Katalysator gewachsen. Diese Nanodrähte zeigen einen linearen Zusammenhang der Wachstumsrate zu dem vorliegenden Oberflächenverhältnis des Nanodrahtsystems. Aus dieser Erkenntnis konnte der Einbau von Wachstumsmaterial in den Katalysatortropfen als wachstums¬raten¬limitierenden Schritt im VLS-Mechanismus identifiziert werden. Weiterhin zeigten Untersuchungen der Wachstumsraten in Bezug auf Änderungen im angelegten V/III Verhältnis, dass bei einem V/III Verhältnis von 6,2±0,9 ein Übergang von Gruppe V Limitierung der Wachstumsrate hin zu einer Gruppe III Limitierung stattfindet. Dieser Wert ist im Einklang mit dem in konventionellen MOVPE Reaktoren, was die Vergleichbarkeit des verwendeten in-situ Aufbaus mit diesen MOVPE Reaktoren zeigt. In einem letzten Untersuchungsschritt wurden die gewachsenen Nanodrähte nach dem Wachstum vermessen. Im Speziellen, konnte der Mechanismus identifiziert werden, welcher zur Entstehung von häufigen Knickwinkeln der Nanodrähte führt. Dafür wurden Kristallorientierungen in den Nanodrähten durch SPED gemessen. Es zeigt sich, dass Mikrozwillinge erster Ordnung Nanodrahtknicke von 70,5° und 109,5° hervorrufen können. 123,7° Knicke, welche auch häufig vorkommen, können durch Mikrozwillinge zweiter Ordnung erklärt werden. Das relative Auftreten der verschiedenen Knickwinkel kann vermutlich durch einen Wechsel der elementaren Terminierung der Wachstumsfront bei den 109,5° Knicken erklärt werden. Dadurch entsteht ein Zwilling zweiter Ordnung durch zwei kurz hintereinander folgende 109,5° Knicke, welche dadurch die entstandene Terminierungsänderung wieder aufheben. Dies erklärt das relativ seltene Auftreten der 109,5° Knicke und die hohe Häufigkeit der 123,7° Knicke. Da erwartet wird, dass die Entstehung von Knicken ein thermodynamischer Prozess ist, kann diese voraussichtlich durch Änderung der Wachstumsparameter, wie der Temperatur, unterdrückt werden

    The histone acetyltransferase KAT6A is recruited to unmethylated CpG islands via a DNA binding winged helix domain

    The lysine acetyltransferase KAT6A (MOZ, MYST3) belongs to the MYST family of chromatin regulators, facilitating histone acetylation. Dysregulation of KAT6A has been implicated in developmental syndromes and the onset of acute myeloid leukemia (AML). Previous work suggests that KAT6A is recruited to its genomic targets by a combinatorial function of histone binding PHD fingers, transcription factors and chromatin binding interaction partners. Here, we demonstrate that a winged helix (WH) domain at the very N-terminus of KAT6A specifically interacts with unmethylated CpG motifs. This DNA binding function leads to the association of KAT6A with unmethylated CpG islands (CGIs) genome-wide. Mutation of the essential amino acids for DNA binding completely abrogates the enrichment of KAT6A at CGIs. In contrast, deletion of a second WH domain or the histone tail binding PHD fingers only subtly influences the binding of KAT6A to CGIs. Overexpression of a KAT6A WH1 mutant has a dominant negative effect on H3K9 histone acetylation, which is comparable to the effects upon overexpression of a KAT6A HAT domain mutant. Taken together, our work revealed a previously unrecognized chromatin recruitment mechanism of KAT6A, offering a new perspective on the role of KAT6A in gene regulation and human diseases.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg
