    Antigen-independent, autonomous B cell receptor signaling drives activated B cell DLBCL

    This study describes and characterizes antigen-independent, autonomous signaling of the clonal B cell receptor as an intrinsic oncogenic driver in activated B cell type DLBCL. This long-sought non-genetic lymphomagenic mechanism has profound implications for development of effective novel therapies. Diffuse large B cell lymphoma of activated B cell type (ABC-DLBCL), a major cell-of-origin DLBCL subtype, is characterized by chronic active B cell receptor (BCR) signaling and NF-kappa B activation, which can be explained by activating mutations of the BCR signaling cascade in a minority of cases. We demonstrate that autonomous BCR signaling, akin to its essential pathogenetic role in chronic lymphocytic leukemia (CLL), can explain chronic active BCR signaling in ABC-DLBCL. 13 of 18 tested DLBCL-derived BCR, including 12 cases selected for expression of IgM, induced spontaneous calcium flux and increased phosphorylation of the BCR signaling cascade in murine triple knockout pre-B cells without antigenic stimulation or external BCR crosslinking. Autonomous BCR signaling was associated with IgM isotype, dependent on somatic BCR mutations and individual HCDR3 sequences, and largely restricted to non-GCB DLBCL. Autonomous BCR signaling represents a novel immunological oncogenic driver mechanism in DLBCL originating from individual BCR sequences and adds a new dimension to currently proposed genetics- and transcriptomics-based DLBCL classifications

    Cancer genetic counseling in Chile: Addressing barriers, confronting challenges, and seizing opportunities in an underserved Latin American Community

    Purpose: Despite the rapid advancements in genomics and the enactment of a new cancer law in Chile, the implementation of cancer genetic counseling continues to face significant challenges because of limited resources and infrastructure. Methods: We conducted a survey targeting health care providers who offer genetic counseling to patients with cancer and possess training in genetics and counseling. Additionally, we distributed a separate survey to high-risk patients associated with an advocacy group to gather insights on their perceptions of and experiences with cancer genetic counseling. Results: Among the surveyed providers, 21% were nonmedical professionals who developed their skills through postgraduate continuing education programs. Germline testing was not performed in 47% of cases. Among the participants, 37% considered genetic counseling important for understanding the cause of their cancer, 25% valued knowing their risk of developing future tumors, and 33% believed it would benefit their current cancer treatment. Just over half of the patients (54%) had access to genetic counseling. Among those that received genetic counseling, 85% found it beneficial. Conclusion: In Chile, barriers to genetic counseling persist, particularly in rural areas and because of a shortage of trained professionals. Public policies recognizing genetic counseling's importance are crucial, along with expanding training and infrastructure. Understanding patient perceptions and increasing the number of trained genetic counseling into cancer care, educating clinicians, and advocating for increased access are key steps for enhancing cancer treatment effectiveness in Chile

    Sphagnum mosses, the impact of disturbances and anthropogenic management actions on their ecological role in CO2 fluxes generated in peatland ecosystems

    Mosses of the genus Sphagnum are the dominant vegetation in most pristine peatlands in temperate and high-latitude regions. They play a crucial role in carbon sequestration, being responsible for ca. 50% of carbon accumulation through their active participation in peat formation. They have a significant influence on the dynamics of CO2 emissions due to an efficient maximum potential photosynthetic rate, lower respiration rates, and the production of a recalcitrant litter whose decomposition is gradual. However, various anthropogenic disturbances and land use management actions that favor its reestablishment have the potential to modify the dynamics of these CO2 emissions. Therefore, the objective of this review is to discuss the role of Sphagnum in CO2 emissions generated in peatland ecosystems, and to understand the impacts of anthropogenic practices favorable and detrimental to Sphagnum on these emissions. Based on our review, increased Sphagnum cover reduces CO2 emissions and fosters C sequestration, but drainage transforms peatlands dominated by Sphagnum into a persistent source of CO2 due to lower gross primary productivity of the moss and increased respiration rates. Sites with moss removal used as donor material for peatland restoration emit twice as much CO2 as adjacent undisturbed natural sites, and those with commercial Sphagnum extraction generate almost neutral CO2 emissions, yet both can recover their sink status in the short term. The reintroduction of fragments and natural recolonization of Sphagnum in transitional peatlands, can reduce emissions, recover, or increase the CO2 sink function in the short and medium term. Furthermore, Sphagnum paludiculture is seen as a sustainable alternative for the use of transitional peatlands, allowing moss production strips to become CO2 sink, however, it is necessary to quantify the emissions of all the components of the field of production (ditches, causeway), and the biomass harvested from the moss to establish a final closing balance of C

    En el presente escrito se comenta, muestra y justifica la importancia de la lectura y de la relectura de clásicos de la educación y la pedagogía por parte de maestros, profesores y educadores en su formación inicial y durante el ejercicio de la docencia. Se valora la lectura de estos clásicos tanto como fuentes de información y de conocimiento, como elementos que coadyuven a la construcción de un conjunto sólido de referentes y fundamentos donde se apoyen las intervenciones educativas, así como su revisión y renovación. Para ello se seleccionan diez autores clásicos de la educación y la pedagogía y se sugiere el conocimiento de sus aportaciones y obras a la educación. Asimismo, se aporta un conjunto de recomendaciones para la integración de su lectura en las actividades docentes, con una extensión a la lectura literaria

    Resumen: El presente trabajo expone los resultados del análisis y caracterización individual de los restos bioantropológicos correspondientes a un rescate arqueológico realizado en el año 2019 en el sector La Cordonada, comuna de Coyhaique, región de Aysén, Chile. Este estudio comprende observaciones macroscópicas y caracterización bioantropológica de un individuo femenino de entre 30-35 años de edad biológica. Además, se precisó una cronología radiocarbónica de 390 años cal AP y la existencia de una dieta diversificada a través del análisis de isótopos estables del individuo. Se discuten tanto estos resultados, en función de otras muestras bioarqueológicas previamente descritas en la región y de cronología similar, como la posibilidad de estar frente a un tipo de inhumación diferente al tipo chenque, comúnmente descrita en Patagonia

    Transcranial Direct Current Stimulation Associated with Visual and Auditory Cueing during Gait Training: A Case Study with a Parkinson's Disease Patient

    Parkinson's Disease (PD) is a neurodegenerative disorder that leads to motor dysfunctions and restricts walking independence. Approaches such as transcranial direct current stimulation (tDCS) and cueing gait are potential rehabilitation strategies that have proven efficient in improving motor function in individuals with PD in preliminary and separate studies. This case study with a single PD patient explores the feasibility and potential effects of combining anodal tDCS (a-tDCS) with visual and auditory cues during 15 gait training sessions. The patient was assessed before the first intervention and after completing all training weeks regarding kinematic parameters of gait, neuronal activation, muscle strength, and functional skills. Results showed a beneficial impact in significantly increasing the participant's speed, stride length, step length, and muscle strength. Furthermore, the decrease in beta brain activity and increase in the delta were associated with a reduction in the effects of the disease, improved motor performance, and shorter functional task times. This study suggests the a-tDCS strategy with double cueing gait can lead to therapeutic potential in relieving the disease's motor symptoms

    Field environmental philosophy: concepts and practices to resolve global society's blindness to biological and cultural diversity

    A great diversity of living beings and human values are invisible to the prevailing global culture, which associates the word biodiversity with large organisms such as trees and mammals. Paradoxically, most biodiversity is made up of small organisms that remain invisible to global society and are under-represented in philosophy, science, and education. To help resolve this "biocultural blindness" about biological and value diversity, in this article we present concepts and practices of field environmental philosophy. We focus on the activity "ecotourism with a hand-lens" to illustrate "conceptual lenses" that amplify the appreciation of biological and cultural diversity. This activity integrates philosophy, sciences, and arts to appreciate the beauty and relevance of small organisms and the multiplicity of languages, forms of knowledge, and values that different cultural traditions give to biodiversity. Participants practice forms of analogical thinking to co-create knowledge and biocultural expressions of their own, instead of being passive recipients of information. Finally, this field environmental philosophy activity combines bodily and sensory activities with theoretical readings that provide conceptual foundations for cultivating a biocultural ethic that guides forms of co-inhabitation with small and large co-inhabitants (humans and others-than-humans) with whom we share both our local habitats and the global biosphere

    Habitability as a historical category for interpreting the Anthropocene

    The article examines the development of a new discourse on habitability in the sixteenth century, which breaks with the ancient notion that distinguished between habitable and uninhabitable spaces according to their climate and location. In it, a new conception of the world as completely habitable and exploitable is articulated, and the European ideal of a temperate climate as a reference to characterize the territories and inhabitants of various latitudes is put forward. I considered more than sixty texts related to cosmography, and a hundred maps and diagrams that were published and circulated as Europe expanded around the globe in the early modern period, and that exhibit new semantics and meanings about habitability. This study, through the analysis of these specific sources, aims to contribute to the debate on the origins of the Anthropocene and the political and imaginative logic that enables its development. The author suggests that the discursive pillars that shaped modern thinking about habitability were crucial in giving life to the ideas of extractivism, speculation, and unlimited connectivity that structures the Anthropocene.
