    Sensory threshold neuromuscular electrical stimulation fosters motor imagery performance

    Motor imagery (MI) has been largely studied as a way to enhance motor learning and to restore motor functions. Although it is agreed that users should emphasize kinesthetic imagery during MI, recordings of MI brain patterns are not sufficiently reliable for many subjects. It has been suggested that the usage of somatosensory feedback would be more suitable than standardly used visual feedback to enhance MI brain patterns. However, somatosensory feed-back should not interfere with the recorded MI brain pattern. In this study we propose a novel feedback modality to guide subjects during MI based on sensory threshold neuromuscular electrical stimulation (St-NMES). St-NMES depolarizes sensory and motor axons without eliciting any muscular contraction. We hypothesize that St-NMES does not induce detectable ERD brain patterns and fosters MI performance. Twelve novice subjects were included in a cross-over design study. We recorded their EEG, comparing St-NMES with visual feed-back during MI or resting tasks. We found that St-NMES not only induced significantly larger desynchronization over sensorimotor areas (p<0.05) but also significantly enhanced MI brain connectivity patterns. Moreover, classification accuracy and stability were significantly higher with St-NMES. Importantly, St-NMES alone did not induce detectable artifacts, but rather the changes in the detected patterns were due to an increased MI performance.     The transcriptional landscape of hematopoietic stem cell ontogeny

    Transcriptome analysis of adult hematopoietic stem cells (HSCs) and their progeny has revealed mechanisms of blood differentiation and leukemogenesis, but a similar analysis of HSC development is lacking. Here, we acquired the transcriptomes of developing HSCs purified from >2,500 murine embryos and adult mice. We found that embryonic hematopoietic elements clustered into three distinct transcriptional states characteristic of the definitive yolk sac, HSCs undergoing specification, and definitive HSCs. We applied a network-biology-based analysis to reconstruct the gene regulatory networks of sequential stages of HSC development and functionally validated candidate transcriptional regulators of HSC ontogeny by morpholino-mediated knockdown in zebrafish embryos. Moreover, we found that HSCs from in vitro differentiated embryonic stem cells closely resemble definitive HSCs, yet lack a Notch-signaling signature, likely accounting for their defective lymphopoiesis. Our analysis and web resource will enhance efforts to identify regulators of HSC ontogeny and facilitate the engineering of hematopoietic specification.GR-NAVEIRA

    In-Network Packet Scheduling and Rate Allocation: A Content Delivery Perspective

    We investigate two important problems in media delivery via active network agents. First, we consider streaming multiple video assets over a shared backbone network through an intermediate proxy-server to a set of receiving clients. The proxy is located at the junction of the backbone network and the last hop to each of the clients and coordinates the delivery of the videos from the origin media server to the clients. We propose an optimization framework that enables the proxy to coordinate the streaming process such that the overall end-to-end performance of the video streams is maximized for the given data rate resources on the backbone and the last hop links. Prospective video quality requirements for the associated media sessions are also taken into consideration in the analysis. Through experiments, we study in detail the operation of the framework and the influence of the various constraints that it considers. Furthermore, we measure its performance gains relative to a sender-driven system where the media server controls the delivery of the data with no assistance from an intervening proxy. We establish an analytical relationship between the relative improvement of the proxy-based system, the network conditions on the backbone and the last hops, and the number of streams served. The gains of the proxy-driven system measured in our experiments closely match their expected values predicted by this relationship.LTS
