Multi-modal refinement of the human heart atlas during the first gestational trimester
No full textInternational audienceForty first-trimester human hearts were studied to lay groundwork for further studies of principles underlying congenital heart defects. We first sampled 49,227 cardiac nuclei from three fetuses at 8.6, 9.0, and 10.7 post-conceptional weeks (pcw) for single-nucleus RNA sequencing, enabling distinction of six classes comprising 21 cell types. Improved resolution led to identification of novel cardiomyocytes and minority autonomic and lymphatic endothelial transcriptomes, among others. After integration with 5-7 pcw heart single-cell RNAseq, we identified a human cardiomyofibroblast progenitor preceding diversification of cardiomyocyte and stromal lineages. Analysis of six Visium sections from two additional hearts was aided by deconvolution, and key spatial markers validated on sectioned and whole hearts in two- and three-dimensional space and over time. Altogether, anatomical-positional features including innervation, conduction and subdomains of the atrioventricular septum translate latent molecular identity into specialized cardiac functions. This atlas adds unprecedented spatial and temporal resolution to the characterization of human-specific aspects of early heart formation
Au lieu du lien : quand la blessure narcissique organise les couples et les familles
No full textInternational audienceIn this paper, using two clinical entry points relating to the moment of diagnosis and self-diagnosis with families and couples, we will show how one of the markers of contemporaneity (post-hypermodernity) could be the mutual and reciprocal narcissistic wounding between members of a family, a couple but also in the therapeutic or educational relationship. We will examine how the experience of a latent feeling of loss, a source of anxiety, opens up a singular motif of ambivalence to the place of connection (family, coupal, educational and therapeutic). Between dependence and loss, bonding and rejection, subjects with narcissistic wounds illusorily relieve themselves by seeking out various forms of compensation as attempts to make up for their feelings of helplessness. This mobilises a type of cleaved relationship to the objects of libidinal investment that blocks the subjects' access to the radical otherness of others. It is in this respect that we will argue that the failure to grasp the essentiality of the other stems above all from a failure to grasp the essentiality of the self. This is a formula that we will detail clinically and theoretically.Dans cette communication, nous montrerons, à partir de deux points d'entrée cliniques relatifs au moment diagnostic et autodiagnostic auprès de familles et de couples, comment un des marqueurs de la contemporanéité (post-hypermodernité) pourrait être la blessure narcissique mutuelle et réciproque entre membres d'une famille, d'un couple mais aussi dans la relation thérapeutique ou éducative. Nous interrogerons en quoi l'éprouvé d'un sentiment larvé de perte, source d'angoisse, ouvre au lieu du lien (familial, coupal, éducatif et thérapeutique) un motif singulier d'ambivalence. Entre dépendance et perte, collage et rejet, les sujets blessés narcissiquement se soulagent illusoirement en sollicitant des formes variées de compensation comme autant de tentatives de combler leur sentiment d'esseulement. Cela mobilise un type de rapport clivé aux objets d'investissement libidinaux qui vient barrer l'accès des sujets à l'altérité radicale d'autrui. C'est en cela que nous argumenterons que le raté de l'essentiel de l'autre procède avant tout d'un raté de l'essentiel de soi. Formule que nous détaillerons cliniquement et théoriquement
Gut microbiota restoration with oral pooled fecal microbiotherapy after intensive chemotherapy: the phase Ib CIMON trial
No full textInternational audienceIntensive chemotherapy (IC) combined with broad-spectrum antibiotics for acute myeloid leukemia (AML) leads to gut microbiota dysbiosis, promoting pathological conditions and an increased incidence of complications, possibly limiting eligibility to allogenic hematopoietic cell transplantation (alloHCT). The purpose of this dose-ranging phase I study (CIMON) was to evaluate the first-in-man use of MaaT033, a pooled, allogeneic, lyophilized, and standardized fecal microbiotherapeutic product, formulated as a delayed-release capsule for oral administration. Primary objectives of the study were to evaluate the maximum tolerable dose of MaaT033 in 21 AML patients having undergone IC and antibiotics. Secondary objectives were to assess MaaT033 safety, its efficacy in restoring the patients' gut microbiome using shotgun sequencing in order to evaluate the recommended dose regimen, and patient compliance (ClinicalTrials.gov number: NCT04150393). MaaT033 was shown to be safe and effective for gut microbiota restoration in AML patients receiving IC and antibiotics, with an excellent gut microbiota reconstruction based on diversity indices at the species level, and restoration of microbial communities close to the composition of the drug product. Moreover, inflammatory markers (C-reactive protein, interleukin-6) decrease with treatment, while short-chain fatty acids increase over time. Life on the dry side: a roadmap to understanding desiccation tolerance and accelerating translational applications
No full textInternational audienceAbstract To thrive in extreme conditions, organisms have evolved a diverse arsenal of adaptations that confer resilience. These species, their traits, and the mechanisms underlying them comprise a valuable resource that can be mined for numerous conceptual insights and applied objectives. One of the most dramatic adaptations to water limitation is desiccation tolerance. Understanding the mechanisms underlying desiccation tolerance has important potential implications for medicine, biotechnology, agriculture, and conservation. However, progress has been hindered by a lack of standardization across sub-disciplines, complicating the integration of data and slowing the translation of basic discoveries into practical applications. Here, we synthesize current knowledge on desiccation tolerance across evolutionary, ecological, physiological, and cellular scales to provide a roadmap for advancing desiccation tolerance research. We also address critical gaps and technical roadblocks, highlighting the need for standardized experimental practices, improved taxonomic sampling, and the development of new tools for studying biology in a dry state. We hope that this perspective can serve as a roadmap to accelerating research breakthroughs and unlocking the potential of desiccation tolerance to address global challenges related to climate change, food security, and health
High‐grade intraepithelial lesions of the anus—patience: A road to wisdom
No full textInternational audienceAim: The progression of high-grade squamous intraepithelial lesions (HSILs) to anal cancer is feared. Data on the remission of HSILs are scarce, and factors associated with remission have not been identified. The aims of this study were to determine the rate of remission of HSILs and to identify factors associated with remission.Method: Consecutive patients with HSILs referred to a tertiary proctology unit for monitoring were identified in a prospective database. Clinical data, as well as cytological, histological and treatment data, were recorded. The remission of HSILs was defined by the normality of the macroscopic examination and the normal histology and/or cytology of at least two consecutive samples.Results: Overall, 144 patients with HSILs were followed for 4.1 (2.1-6.5) years. The cumulative probabilities of remission of HSILs were 9.7% (6.2%-14.9%), 14.6% (10.1%-20.7%) and 23.8% (17.3%-31.8%) at 2, 3 and 5 years, respectively. Human papillomavirus 16 (HPV16) clearance was achieved in 19/84 (22.6%) patients. Multivariate analysis revealed that three main factors were associated with remission of HSILs: absence of HPV16 at referral [hazard ratio (HR) 0.3, 95% CI 0.1-0.9], location (absence of endoanal lesion) of HSILs (HR 0.1, 95% CI 0.01-0.08) and excision of HSILs (HR 3.2, 95% CI 1.1-9.0).Conclusion: The remission of HSILs was not a frequent event in this long-term cohort study. HPV16 is a pejorative factor, while other high-risk HPVs are not. Excision of the HSIL, when possible, could be a preferred treatment to allow better remission

Analyse transversale de 28 démarches de filières agrialimentaires : des dynamiques d'innovation variables au travers de leur engagement dans l'agroécologie
No full textLe secteur agroalimentaire fait l’objet d’une segmentation croissante du marché avec la multiplication de démarches dites de qualité, publique ou privée, qui engagent les filières dans la construction de pratiques de production, de transformation ou de distribution différenciées. Nous connaissons peu de choses sur la manière dont ces démarches inscrivent leurs pratiques dans l’agroécologie, et en particulier pour les filières ayant une mention valorisante autre que l’agriculture biologique. De 2022 à 2023, la Direction Scientifique Agriculture INRAE a engagé un chantier exploratoire, avec 11 groupes filières INRAE, pour analyser comment se construisent des démarches de filières différenciées sur le marché et leur engagement en faveur de l’agroécologie.National audienceSince the 2014 law dedicated to the development of agroecology, French plant and animal supply chains have mobilize agroecological principles in various ways. Twenty-eight case studies of initiatives claiming to differentiate themselves through practices related to agroecological principles have been analyzed across different production domains from animals and plants. The analytical framework used is based on innovation functions implemented by value chains : these different actions taken within the value chains can feed into a process of collective learning in the transition towards agroecology. The presented results show a variable transformative capacity of the value chains, but allow for the identification of several regularities on which the agroecological transition of the sectors in France seems to be taking place. The article builds suggestions for collective actions to support the agroecological transition in France.Depuis de la loi de 2014 dédiée au développement de l’agroécologie, les filières végétales et animales françaises mobilisent des principes de l’agroécologie de façon variée. Cet article synthétise l’analyse transversale de 28 cas de démarches de filières déclarant se différencier par des pratiques reliées à des principes de l’agroécologie, dans différents domaines de production de l’animal ou du végétal. Le cadre d’analyse mobilisé est basé sur des fonctions d’innovation mises en œuvre par ces filières. Ces différentes actions conduites dans la filière peuvent alimenter un processus d’apprentissage collectif dans la transition vers l’agroécologie. Les résultats présentés montrent une capacité transformative variable des filières, mais permettent d’avancer un ensemble de régularités sur lesquelles semble reposer la transition agroécologique des filières en France. L’article conclut par des pistes d’actions collectives pour soutenir la transition agroécologique en France

No full textInternational audienceLes sports équestres entretiennent une relation singulière et complexe aux Jeux Olympiques. Le cheval était, en effet, au cœur des épreuves antiques, au travers d’activités toutefois très différentes d’aujourd’hui, privilégiant les courses de char et la voltige. Ce n’est, néanmoins, qu’à l’aube de la Grande Guerre, aux Jeux de Stockholm en 1912, que l’équitation intègre définitivement l’olympisme moderne. Si le cheval trouve ainsi progressivement sa place aux Jeux, son accès demeure très limité. Héritier d’une pratique pluriséculaire aristocratique et martiale, il se voit l’apanage exclusive des militaires jusqu’à 1948 et des hommes jusqu’à 1952. L’ouverture des sports équestres aux civils et aux femmes a aussi favorisé sa diffusion internationale : le nombre de nations participantes a été multiplié par 5 en un siècle. Le cheval permet aussi une inclusion, les sports équestres étant les seules épreuves mixtes aux Jeux Olympiques, où femmes et hommes s'affrontent dans les mêmes classements. L'intégration des femmes a toutefois était progressive en fonction des disciplines -dressage en 1952; obstacle en 1956; concours complet en 1962- les femmes devant conquérir une acceptation sociale de leur pratique. La mixte d'aujourd'hui ne signifie néanmoins toujours pas l'égalité de genre, les femmes ne représentant que 35% des participantes, avec de fortes différences, en fonction des disciplines

La réponse peut être positive à condition de bien entendre le concept de droit naturel qui ne signifie pas la nécessité d'un droit positif figé dans l'éternité. La procédure demeure le moyen organisé d’une fin : attribuer à chacun son dû, le plus souvent en situation de conflit mais aussi parfois de manière gracieuse. Comme tout moyen il doit être le plus possible ordonné à sa fin, ce qui nécessite toujours des adaptations liées aux sociétés, aux hommes, aux contraintes économiques et matérielles
