Borászati laboratóriumi gyakorlat
a Kémia Tanszéken

A 2002/2003-as tanévben indult a főiskolán a biológus laboratóriumi
operátor képzés három szakiránnyal, köztük a borászati szakirány. A szaki-
rány egyik kurzusa a laboratóriumi gyakorlat, ahol a hallgatók az alapvető
laboratóriumi műveleteket és méréseket sajátítják el három félév alatt .
A gyakorlat célja olyan emberek képzése, akik a saját szakterületükön
— borászati laborokban — az alapvető laboratóriumi mérések előkészítésé-
ben, szükséges eszközök, műszerek kezelésében jártasak, azokkal mérni tud-
nak, valamint a borászati laborokban folyó kutatások komplexebb méréseibe
is be tudnak kapcsolódni, s ezzel a minőségi bor előállításában tevékenyen
részt vesznek.
A hároméves főiskolai képzés harmadik-negyedik félévében két féléves
gyakorlati képzés során heti 2 órában, majd a hatodik félévében heti 8 órá-
ban foglakoznak a hallgatók a borászatban alkalmazott mérésekkel. A gya-
korlaton kívül a külső — nyári gyakorlatok alkalmával — az üzemekben
is gyakorolhatnak, szélesíthetik azt a tudást, amelyet a gyakorlatok során
megszereztek.
A hallgatók részére az oktatás harmadik félévében a borászathoz kap-
csolódó alapmérések elvégzése és megtanulása a célja a gyakorlatoknak. Eb-
ben a félévben elvégzik és megtanulják azokat a méréseket, amelyeket a 
leggyengébb felszereltségi szinten lévő laborokban is el kell tudni végezni.
Ezek a következő mérések:
— Borok titrálható savtartalmának meghatározása indikátoros titrálással,
pH mérése.
— Borok cukor t art almának meghatározása Rebelein-módszerrel.
— Alkoholtartalom meghatározása desztillálással.
— Borok illósavtartalmának meghatározása.
— Borok Össz-, illetve szabadkénessav tartalmának meghatározása.
— Extr akttart alom meghatározása.
— Derítési igény meghatározása.
— Összes polifenol meghatározása fotometriás módszerrel.
— Kereskedelemben kapható gyorstesztek használata.

— Számonkérés: A félévben tanult egy boranalitikai vizsgálat egyéni el-
végzése, borok ismeretlen paramétereinek meghatározása.
Az alapmérések közé tartozik azoknak a paramétereknek a meghatá-
rozása, amelyek a borról vagy mustról — mint komplex élő anyagnak a 
fejlődéséről — a borásznak jellemző és informatív adatokat adnak.
A borászati kémia meglehetősen analitikaigényes és meglehetősen össze-
tett . A hallgatók az analitikai mérések nagy részével nem foglalkoznak, mivel
a rendelkezésre álló idő erre nem elég, de a gyakorlat során az alapmérések-
kel kötelezően megismerkednek. Az alapvető méréseket két nagy csoportra
oszthatjuk, amelyek tükrözik az első félévben tartandó gyakorlat fo nyom-
vonalát is. Az egyik nagy csoport a térfogatos elemzésen alapuló mérések
(titrálások), a másik nagy csoport a műszeres mérések. (A felosztás megle-
hetősen önkényes.)
Az első gyakorlati órák során a hallgatók a klasszikus laboratóriumi
műveletekkel — tömegmérés, oldatkoncentráció számítás, oldatkészítés — 
ismerkednek meg. Ezek után olyan mérésekkel folytatják a gyakorlatsor me-
netét, amelynek keretén belül a titrálható savtartalmat, cukortartalmat Re-
belein módszerével, valamint az összes-, illetve a szabad kénes sav tartalmat
határozzák meg. A titrálási művelet nagy pontosságot, odafigyelést és jó ér-
zéket követel meg, és megfelelő elsajátítása a további gyakorlatok folyamán
a minőségi munkavégzést segíti. Az első félévben megismerkednek még az al-
koholtartalom desztillálással történő meghatározásával, az illósav-, valamint
az extrakttartalom meghatározásával.
Ebben a félévben helyet kap még az összes polifenoltartalom meghatá-
rozása, ami fotometriás műszeres módszerrel történik. Amint az előbb em-
lítettem, a harmadik félévben szinte csak titrálással foglalkoznak, így ezen
mérés átmenetet jelent a következő félévre.
A polifenolok borászati szempontból az egyik legjelentősebb vegyület-
csoportot alkotják. Jellegükből adódóan oxidációjuk során a borok bámulá-
sát, barnatörését és más kiválással járó folyamatot okozhatnak, másrészt — 
főleg a vörösborok esetén — a borjelleg kialakításában van jelentős szere-
pük. A polifenol elnevezés a fenolos-hidroxil csoportok számára utal (poli = 
sok). A szőlő fenolos vegyületei jelentős élettani hatást kölcsönöznek a bor-
nak, főleg a vörösbornak. Régebben a baktericidhatást, illetve a P-vitamin-
aktivitást emelték ki, napjainkban azonban a kutatások révén nagy szerepet
kaptak ezek a vegyületek a szív- és érrendszeri betegségekre gyakorolt ha-
tásuk miatt, kiemelve a procianidineket, valamint a rezveratrolt. (A borok
rezveratroltartalma főleg a technológiai feldolgozás függvénye.)

Az oktatás során a negyedik szemeszterben az egyszerűbb műszeres mé-
rések kerülnek túlsúlyba a gyakorlatokon. Ezek a következő mérések: keres-
kedelemben kapható borászati gyorsteszt használata, mérés a gyorsteszttel.
— Cukormentes extrakt tart alom meghatározása.
— Gyorstesztek és a klasszikus borászati analitikai módszerek eredménye-
inek összehasonlítása.
— Derítési próbák.
— Színintenzitás meghatározása fotometriás an.
— Labilis vas (ü)-tartalom meghatározása spektrofotometriásán.
— Összes almasavtartalom elválasztása ioncserélő gyantán, meghatáro-
zása fotometriásan.
— Borok pH- és savtartalmának meghatározás, összefüggések keresése.
— K- és Na-tartalom meghatározása lángfotometriásan.
— Aromakomponensek meghatározása SPME szállal, gázkromatográfiás
módszerrel.
— Borok savösszetételének meghatározása HPLC módszerrel (FVM-
SZBKI — Eger nagyműszeres mérés megtekintése.)
Ebben a félévben is végeznek térfogatos elemzést a hallgatók, felhasz-
nálják az előző félévben megtanultakat. Összehasonlítás céljából önállóan
készítik el azokat a méréseket, amelyeket korábban megtanultak. Ilyen mé-
réssorozat a kereskedelemben kapható gyorstesztek és a klasszikus mérések
közti összehasonlítás is, azaz a hallgatók a kereskedelmi teszt segítségével
kapott adatokat vetik össze a klasszikus mérési adatokkal. Ezek közül pél-
dául a szabadkénes sav meghatározása meglepően jó eredményt ad a valós
értékhez viszonyítva, viszont a többi gyorstesztes mérésnek nagy a hiba-
százaléka. A költségek részéről nézve a gyorsteszttel történő meghatározás
sokkal drágább.
Tanulják még a hallgatók a cukormentes extraktt art alom, vörösborok
színintenzitásának fotometriás meghatározásával, labilis vas(II)-tartalom fo-
tometriás meghat ár oz ás ával, almasavt art alom vizsgálata a pH- és a sav-
tartalom összefüggésének mérésével, valamit kálium- és nátriumtartalom
lángfotometriás meghatározása is szerepel a gyakorlatokon. Záróakkordként
pedig egy üzemlátogatás keretén belül a nagyműszeres mérésekbe nyernek

betekintést, mint pl.: szilárd fázisú mikroextrakciós szállal való mérés gáz-
kromatográf— tömegspektrométer (SPME-GC/MS) segítségével, valamint
borok savtartalmának elválasztása és kvantitatív meghatározása nagy haté-
konyságú folyadékkromatográf segítségével (HPLC).
A gyakorlat heti óraszámát tekintve — amely heti 2-2 óra — átfogó
képet kapnak a hallgatók azokról a mérésekről, amelyek egy átlag borászati
laboratóriumban előfordulhatnak. A mérések további elmélyítésére és begya-
korlására, valamint a nagyműszeres mérések kellő elsajátítására a harmadik
év második félévében a heti 8 órás gyakorlat biztosít megfelelő időt. Ez
már elegendő arra hogy a bonyolultabb mérésekhez is hozzá lehessen fogni.
A nagyműszeres mérések közül a gázkromatográfFal, folyadékkromatográf-
fal mérni tudó ember napjainkban nagy előnyt tudhat magáénak, ezért a 
következő félévek anyagaiban ez a mérési lehetőség is helyet kap.
Örülök, hogy ebbe az oktatási formába aktívan bekapcsolódhattam,
és szívesen végzem a gyakorlatorientált feladatomat. Szeretném, ha azok az
hallgatók, akik ezen a kurzuson végeznek, további munkáik során a minőségi
bor előállításában hasznos és tevékeny szakemberként vennének részt. Ezt a 
célt a tanszék- és a gyakorlatvezetők részéről minden formában támogatjuk.

Abstract. Respiration rates in the re-wetted desiccated thalli of a 
desiccation-tolerant lichen Cladonia convoluta were measured during
rehydration and revival. Treatment with bromoxynil as uncoupler of
oxidative phosphorylation showed that the high intensity of resaturation
respiration rate is not entirely caused by the uncoupled mitochondrial
membrane. Treatments with KCN or absence of 0 2 obtained by applying N2
resulted in the same effect indicating the possible role of the pentose-
phosphate shunt in the boosted C02 efflux of the rehydrating and reviving
desiccated lichen thalli.

Introduction
Desiccation tolerant plants can survive the loss of 80-95% of their cell
water, so that the plants appear completely dry and no liquid phase remains
in their cells; after a shorter or longer period in the desiccated state, they
revive and resume normal metabolism when they are re-moistened (Proctor
et al., 2002). In a dehydrated desiccation tolerant plant an intense C02

emission can be observed in the very first minutes after re-wetting, which is
considered to be a physical process; i.e. water replaces C02 molecules
absorbed on the cell walls (Brown et al., 1983). However, this wetting burst
is followed for a few hours by a respiration which has been termed
resaturation respiration and which has a higher intensity than that of
constantly metabolically active lichens (Smith & Molesworth, 1973). This
elevated respiration does not serve the replenishment of the ATP pool
because the latter is already completed within 5-10 minutes after rewetting
(Krochko et al., 1979). Gordy and Hendrix (1982) considered the
resaturation respiration of mitochondrial origin because they could eliminate
completely the 02 uptake during this phase with the simultaneous usage of
cyanide and salicylhydroxamic acid. The increasing evidence of membrane
destruction in desiccated lichens led to the conclusion that resaturation
respiration is a result of the uncoupled inner mitochondrial membrane
(Farrar & Smith, 1976).
The initially high rehydration respiration rate is also occurring in the
higher/flower desiccation-tolerant higher flowering plants, like in the
monocot Xerophyta scabrida where it lasts up to 30 hours (Tuba et al.,
1994). In the lichens and mosses the resaturation respiration lasts only from
few minutes up to 5 h (e.g. Smith & Molesworth 1973; Bewley et al. 1978).
In order to reveal the biochemical background of the resaturation
respiration, we treated a highly desiccation tolerant cryptogamic species, the
lichen Cladonia convoluta (see: Tuba et al., 1996) with uncouplers, with
potassium cyanide (KCN) and bromoxynil (Kerr and Wain, 1964 and Hock
et al., 1995) or we deprived them of oxygen by applying inert N2 gas. Our
aim was to identify the possible cause of the boosted C0 2 efflux during the
rehydration of the desiccated C. convoluta lichen thallus.
Materials and Methods
The foliose green algae lichen thallus Cladonia convoluta (Lam.) P.
Cout. were used in the experiment after being collected from a semi-desert
sand grassland (Fekete at al., 1997) near Fülöpháza (Hungary, Great Plain).
The samples were cleaned by rinsing in deionized water and placed on wet
filter paper. The upper green part of the moss and the separated healthy lobes
of lichen thallus were used. Prior to desiccation plants spent 24 hours in a 
metabolically active state (20 °C, 100% Rh, 200 jiM m"2 sec 1 PhAR for 14
hours). Slow desiccation was carried out on the next day when Rh was
lowered to 70%. For rehydration deionized water (control, N2-treatment), 10"
3 M KCN, and 10~3 M bromoxynil (4-hydroxy-3,5 dibromo benzonitrile)
were applied. Rehydration was conducted in small Plexiglas chambers
(20 °C, 100% Rh, 200 pM m"2 sec"1 PhAR). C02 gas exchange
measurements were carried out according to Tuba et al. (1996) in five

replications with a ADC LCA2 (Hoddesdon, UK) infrared gas analyser in
the first 30 minutes after rewetting.
Results and Discussion
The basic respiration rate of the continuously wet lichen C. convoluta 
was 2.6±0.29. However, the resaturation respiration rateof the wetted lichen
was three times higher (Fig. 1.). The high intensity rehydration
('resaturation') respiration was terminated within 30 minutes and during the
remaining 90 min of the experiment the respiration declined to a steady level
(data not shown).
Treatment with bromoxynil, an uncoupler of oxidative phosphorylation,
increased this already high C02 emission by 66% indicating that the
extraordinarily massive C02 efflux does not come entirely from the
mitochondria, which has lost adenylate control due to uncoupled inner
membranes. Nitrogen exposure (anaerobic conditions) and KCN treatment
resulted in a surprising coincidence. They both decreased C02 emission rate
by 30% in the lichen. This fact suggests that there is a component, which is
non-sensitive to both cyanide and also oxygen deprivation. The only
biochemical process, which can produce a significant amount of C02 in not
senescing plant cells under anaerobic conditions, is the pentose-phosphate
pathway. This pathway can play an essential part in the restoration of the
plant's redox status by producing NADPH (Kranner & Grill, 1994).
This observation also infers that the alternative mitochondrial pathway is
not engaged after rehydration of the desiccated tissues, because C02
emission was not decreased more by the lacking of 0 2 than by cyanide,
which clogs cytochrome pathway selectively. Due to the Bahr-Bonner model
(Lambers, 1982), which considers the alternative oxidative pathway as an
overflow, operating the alternative pathway would not make too much sense
in desiccation tolerant plants spending its resources for recovery from the
desiccated stage.
Based on the above findings we would like to emphasise that the
preponderance of resaturation respiration of the desiccated lichen thalli
probably originates from the pentose-phosphate pathway.

A Ligustrum vulgare L. gyökérrendszerének

Abstract 
Structure of root system of Ligustrum vulgare in Síkfőkút oak forest, 
Hungari. The author studied the roots of Ligustrum vulgare species in the oak 
forest (Quercetum petraeae-cerris, at Síkfőkút, northern Hungary). The soil of 
the forest at Síkfőkút is a significantly acidified version (pH=5,3-5,9) of the 
clay-containing brown wood soil. In the past few years the pH value has been 
reduced to some extent. Throughout the investigations the root structure of 12 
Ligustrum of different ages (6-15 year-old) has been revealed by means of grad-
ual grubbing method. When revealing horizontal and vertical rhyzogram has 
been taken of the roots, on which each root-item of over 1 mm diameter has 
been indicated, he observed its root-system is not typically a tap-root system. 
The Ligustrum vulgare might be classified into the plants with finger-like tap-
root capable of secondary thickening in the system of Krasilnikov (1968) (group 
1., subgroup 2., type 2.). 
Bevezetés  
A cserjék gyökérrendszerének szisztematikus kutatása hazánkban a „Sík-
főkút Project” komplex környezetbiológiai kutatóprogram részeként 1979-ben 
kezdődött. Az azt megelőző időkből csupán Faragó (1961) tanulmánya foglalko-
zik néhány faj gyökérzetének elemzésével (Kárász, 1986). A síkfőkúti tölgyes 
domináns cserjefajairól már számos tanulmány megjelent (Kárász–Juhar 1982, 
Kárász 1984, 1984a, 1984b, 1988, 1991, 2006). Jelen közleményben az erdő 
cserjeszintjében egyik leggyakoribb fajnak, a közönséges fagyalnak (Ligustrum 
vulgare) gyökérrendszerére vonatkozó eredményeket foglaltuk össze. 
Anyag és módszer 
A Ligustrum vulgare európai mediterrán-szubatlanti jellegű flóraelem. Síksá-
gi-dombvidéki részben örökzöld cserje. Tölgyes-bükkös (Querco-Fagetea) elem-

ként gyakorlatilag valamennyi hegy- és dombvidéki erdőtársulásban él, helyen-
ként tömegesen. Meglehetősen társulásközömbös cserjefaj, így valamennyi al-
földi erdőtársulásunkban is jelen van a sziki tölgyesektől a homoki erdőkig és a 
folyóparti ligeterdőkig. Ökológiai toleranciájának szélét a nyírláp (Salici 
cinereae – Betuletum pubescentis) jelzi. 
Fényigényes, de különösen fiatalon jó árnytűrő, melegkedvelő faj, az idő-
szakos szárazságot is jól bírja. Inkább mészkedvelő, de savanyú talajon sem 
ritka. A talaj tápanyagtartalmával szemben igényes, jelenléte az alföldi erdőkben 
az erdőtenyészet optimumát jelzi. Gyorsan nő és jól sarjadzik. Erdőgazdaságilag 
mint talajvédő és elgyomosodott területek visszaerdősítésének pionirja játszik 
szerepet (Csapody et al. 1966). Ökológiai értékszámai a következők: T5a, W4, 
R3, természetvédelmi érték besorolása szerint társulásalkotó faj (Simon 1992). 
A síkfőkúti cseres-tölgyesben a gyökérvizsgálatok idején (Kovács 1983) a 
leggyakoribb faj volt, hektáronként több mint 15 ezer talaj feletti hajtásával (Ká-
rász et al. 1987). Elsősorban vegetatív módon szaporodik. 
A síkfőkúti erdő talaja Stefanovits (1985) vizsgálatai szerint az anyagbemo-
sódásos barna erdőtalaj jelentősen savanyú (pH 5,3 – 5-9). A vizsgálatok során 
12  cserje egyed gyökérzetét tártuk fel fokozatos kiásás módszerrel. Közülük 6 
db átlagos méretűnek tekinthető, a másik 6 db pedig különböző korú és méretű 
és különböző lombborítású helyen élő egyed volt. A feltárás során a gyökérzet-
ről horizontális és vertikális rizogramot készítettünk. A mintacserjék adatait az 
1. táblázatban foglaltuk össze.

Eredmények 
A fagyal talajbani részeinek tömegét tekintve jelentős hányadot tesznek ki 
az avarszintben, a talaj felületén futó száreredetű sztólók (1. fénykép). Színük 
különböző árnyalatú szürke vagy szürkésbarna. Kérgükön finom, hosszanti lefu-
tású bordázat és 2-5 mm-enként kb. 1 mm átmérőjű világosabb színű para-
szemölcsök láthatók. A járulékos és valódi gyökerek egyaránt barnásszürke szí-
nűek. 
A feltárt mintacserjék gyökerekkel kapcsolatos mérési eredményeket a 2-3. 
táblázatok tartalmazzák. A gyökérrendszer fiziognómiai szerkezetét a 1-24. áb-
rák szemléltetik. 
A Ligustrum vulgare gyökérrendszere kevés valódi gyökérből és sok a ta-
lajjal érintkező sztólókból fejlődő járulékos gyökérből áll. A sztólók rácsszerűen 
behálózzák a talaj felszínét és belőlük sok hajtás fejlődik, így egy-egy gyökér-
rendszerhez esetenként 25-30 különböző korú hajtásból álló, szabálytalan alakú 
polikormon tartozik. A sztólók többsége 3-6 mm átmérőjű és a főágai kb. derék-
szögben ágaznak el. Egyenes, iránytartó lefutásúak. Egyes részeik az avarszint 
fölé emelkednek ezeken járulékos gyökér nem fejlődik (1. fénykép). Továbbfut-
va ismét érintkeznek a talajjal és gyökereket fejlesztenek. A járulékos gyökerek 
általában 1 mm-nél vékonyabbak, rendkívül gazdagon elágaznak. 
A gyökérrendszer horizontális kiterjedése a lombkorona vetülethez viszo-
nyítva igen nagy, átlagosan annak hatszorosa. Karógyökere nincs. A gyökérzet 
vertikális kiterjedése kicsi, a gyökerek többsége az avarszintben és a talaj felső 5 
cm-es rétegében helyezkedik el. Az anyanövény egy-egy valódi gyökere eseten-
ként a mélyebb talajrétegekbe nyúlik, ilyenkor 40-60 cm-es mélységbe is beha-
tol. (pl. I., II., IV. és VI. sz. minta). Hasonló megfigyelésekről számol be Faragó 
(1961), aki egy 8 éves 1 m magas egyedet tárt fel homoktalajon. Megállapítása 
szerint a Ligustrum vulgare gyökerei sekélyen futottak a laza humuszos talajban, 
karógyökér nem volt. 
Kraszilnyikov (1968) osztályozása szerint a Ligustrum vulgare a „másodla-
gos vastagodásra képes, járulékos gyökérrendszerű” növények közé sorolható (1. 
csoport 2. alcsoport, 2. típus). 
A Ligustrum e sajátos avar-talaj átmeneti rétegben való elhelyezkedése és 
intenzív járulékos gyökérképző képessége lehetővé teszi az avar és a legfelső 
talajréteg vízkészletének együttes felhasználását, beleértve a napi hőingás követ-
keztében lecsapódó harmatot is. Rendkívüli sarjadó képessége pedig maximali-
zálja a gyors térfoglalást és forrás kihasználást. Mindezek hozzájárulnak a faj 
széles ökológiai toleranciájához.

Összefoglaló 
Morfinszármazékok savkatalizált átrendeződése mikrohullámú aktivá-
lással. A morfin, kodein és tebain savkatalizált átrendeződési reakcióját vizsgál-
tuk mikrohullámú aktiválással. A hagyományos melegítési technikákkal össze-
hasonlítva minden esetben sikerült az átrendeződés hozamán jelentősen javítani. 
A gyógyászati szempontból is jelentős, dopamin agonista hatású R(-)-apomorfin 
szintézisét 75%-os hozammal sikerült megvalósítanunk morfinból kiindulva. 
 
 
A key step in the synthesis of many aporphines is the acid-catalyzed rear-
rangement of the corresponding morphinan. It is well known, that morphine (1) 
and its congeners rearrange with concentrated acids to aporphines
1-4
 (Scheme 
1.). However the yields of the conversion of morphine (1) to apomorphine (4)
1,2
 
and codeine (2) to apocodeine (5)
3
 are low, using a variety of acids. Granchelli et 
al.
4
 investigated the acid-catalyzed rearrangement of thebaine (3) with methane-
sulfonic acid, resulting in the formation of 2,10-dimethoxy-11-hydroxy-
aporphine (6) in 60% yield. The yield of this rearrangement can be improved
5
 by 
carrying out the rearrangement with methanesulfonic acid, in the presence of 
methanol. Recently we reported the investigation of the acid catalyzed rear-
rangement of a series of morphinans
6
 using conventional thermal techniques. 
The results of our study on the microwave-assisted acid-catalyzed rearrangement 
of a series of morphinans are reported here.

Automated and focused microwave flash heating has recently proven to be 
very effective in accelerating organic transformations and has been widely ap-
plied in parallel syntheses and in drug discovery processes. Numerous successful 
reactions with great efficiency and dramatically enhanced reaction rates have 
been described
7, 8, 9
. 
The conditions of the microwave-promoted rearrangements with methane-
sulfonic acid and hydrochloric acid were optimized in the case of the morphine 
(1) to apomorphine (4) rearrangement (Table 1.).

The change in temperature and microwave power did not affect the yield of 
the microwave assisted rearrangement. The optimal conditions were found to be 
in the case of the rearrangement with methanesulfonic acid at 90
o
C, with 5 min 
stirring. In the case of the rearrangement with hydrochloric acid at 150
o
C, with 5 
min stirring, under pressure. Methanesulfonic acid was shown to be an excellent 
solvent to mediate microwave energy, thus lower microwave power was suffi-
cient to achieve full conversion. 
In the course of our study we found a significant improvement in the yields 
in the acid-catalyzed rearrangements of morphinans using microwave heating, 
compared to the conventional heating techniques (Table 2.). Another advantage

to the use of microwave heating was, the shorter reaction time required, thus the 
products obtained did not need any further purification.

The morphine (1) to apomorphine (4) rearrangement (Scheme 1.) was car-
ried out with methanesulfonic acid in 75% yield using microwave-assisted heat-
ing at 90 
o
C, for 5 min. This rearrangement was also attempted in concentrated 
hydrochloric acid using microwave-assisted heating at 150 
o
C, for 5 min, under 
pressure. This reaction resulted in apomorphine (4) in 70% yield, a considerable 
improvement compared to the classical method using traditional heating tech-
niques
1
. In the case of codeine (2) and thebaine (3) the optimized conditions 
were applied for the microwave-promoted rearrangement with methanesulfonic 
acid. After heating at 90 
o
C, for 5 min in a microwave reactor with methanesul-
fonic acid, codeine (2) afforded apocodeine (5) in 78% yield. 2,10-dimethoxy-
11-hydroxyaporphine (6) was obtained from thebaine (3) with methanesulfonic 
acid in 70% yield using similar conditions. Morphothebaine (7) was also iso-
lated, whose formation is the result of the water content of the methanesulfonic 
acid

. 95% yield was achieved by carrying out the microwave-assisted rear-
rangement of thebaine (3) with methanesulfonic acid, in the presence of 16% 
methanol. Under these conditions morphothebaine (7) was not isolated. 
In conclusion, the microwave-promoted synthesis was successfully applied 
for the acid-catalyzed rearrangement of morphinans. R(-)-apomorphine (4) was

synthesized from morphine (1) with methanesulfonic acid in 75% isolated yield 
using microwave heating. 
Experimental: Microwave irradiation was carried out with a CEM Dis-
cover microwave instrument. Melting points were measured with a Thomas 
Hoover Capillary Melting Point Apparatus, and are uncorrected.

H NMR spec-
tra were obtained on Varian 300 spectrometer, chemical shifts are reported in 
ppm (δ) from internal TMS and coupling constants (J) are measured in Hz. Thin 
layer chromatography was performed on precoated Merck 5554 Silica gel 40 
F

 foils, the spots were visualized with Dragendorff's reagent. 
General procedure for the microwave-assisted rearrangement of 
morphinans 
The morphinan (0.3mmol) was dissolved in the appropriate acid (1.5mL) in 
a 10mL glass tube, under nitrogen, with ice-cooling. The vessel was sealed with 
a septum and placed into the microwave cavity. The reaction mixture was stirred 
for 5 min, at 90
o
C in microwave reactor, then after cooling to room temperature 
the reaction mixture was added to ice-water (20mL). The pH was adjusted to 9 
by adding ammonia, with ice-cooling. The mixture was extracted with ethyl 
acetate (3x10mL), the organic layer was washed with brine (20mL), dried with 
sodium sulfate, filtered and evaporated in vacuo to afford the appropriate apor-
phines.  
R(-)-apomorphine hydrochloride (4): a) Starting from morphine hydrate 
(1) using methanesulfonic acid the extraction was carried out with chloroform 
(5x10mL) and after drying with sodium sulfate to the filtered extract HCl-ether 
was added to afford the hydrochloride salt. After evaporation the solid pure HCl-
salt was filtered from anhydrous ether (68mg, 75%), mp: 210
