    CKAP5 enables formation of persistent actin bundles templated by dynamically instable microtubules

Recently, an abundantly present microtubule polymerase, CKAP5 (XMAP215 homolog), has been reported to play a role in mediating crosstalk between microtubules and actin filaments in the neuronal growth cones. However, the molecular mechanism of this process is unknown. Here, we demonstrate, in a reconstituted system, that CKAP5 enables the formation of persistent actin bundles templated by dynamically instable microtubules. We explain the templating by the difference in CKAP5 binding to microtubules and actin filaments. Binding to the microtubule lattice with higher affinity, CKAP5 enables the formation of actin bundles exclusively on the microtubule lattice, at CKAP5 concentrations insufficient to support any actin bundling in the absence of microtubules. Strikingly, when the microtubules depolymerize, actin bundles prevail at the positions predetermined by the microtubules. We propose that the local abundance of available CKAP5-binding sites in actin bundles allows the retention of CKAP5, resulting in persisting actin bundles. In line with our observations, we found that reducing CKAP5 levels in vivo results in a decrease in actin-microtubule co-localization in growth cones and specifically decreases actin intensity at microtubule plus ends. This readily suggests a mechanism explaining how exploratory microtubules set the positions of actin bundles, for example, in cytoskeleton-rich neuronal growth cones

    Pedagogía en Educación Diferencial, Licenciado en EducaciónEl Trastorno del Espectro Autista (TEA) es un trastorno del neurodesarrollo que se caracteriza por dificultades en las interacciones sociales y comunicación. Se sabe las dificultades que puede implicar para las personas que presentan tal condición, no obstante, cómo impacta tal diagnóstico en la familia aún requiere profundización en el contexto local chileno. La presente investigación pretende dar a conocer y comprender las percepciones de las familias sobre el proceso de orientación que se brinda posterior al diagnóstico de TEA, para así indagar en las orientaciones entregadas tanto en el ámbito educativo, como en el ámbito de salud y las barreras que se han presentado en este proceso. Este estudio utiliza un enfoque de investigación cualitativa, bajo el paradigma interpretativo, mediante un estudio de tipo fenomenológico. Se aplicó una entrevista semiestructurada con el fin de profundizar en las diversas experiencias declaradas por seis familias distribuidas en dos establecimientos educacionales de la comuna de Concepción, que corresponden apoderados de un estudiante que presente TEA. Los resultados indican que las percepciones convergen en la importancia de la atención e intervención al diagnóstico de su hijo(a), que las orientaciones indicadas deben ser pertinentes, al igual que la participación de los profesionales y que aún por parte de las familias se presenta un desconocimiento ante el rol a cumplir para brindar el apoyo adecuado a su hijo(a). Además, se concluye que las percepciones de las familias aún están centradas en el antiguo paradigma biomédico, centrado en el diagnóstico, más que en el proceso de inclusión atendiendo las necesidades del niño(a) como lo establece el nuevo paradigma centrado en la neuro divergencia.Autism Spectrum Disorder (ASD) is a neurodevelopmental disorder characterized by difficulties facing social interactions and communication. The hardships that this condition can imply for the individuals who have it, however, the way such a diagnosis may impact the family requires deeper research within Chilean local context. The present investigation aims to promulgate and comprehend the point of view of the families regarding the guidance process provided after the diagnosis of ASD, so it may be possible to inquire about the guidance provided in both the educational and healthcare fields, as well as the barriers that have arisen in this process. This study uses a qualitative research approach, under the interpretative paradigm, through a phenomenological study. A semi-structured interview was applied to delve into the diverse experiences reported by six families distributed in two educational establishments in the commune of Concepción, whose children are diagnosed with ASD. The results indicate that perceptions converge on the importance of attention and intervention following the diagnosis of their child, and that the provided guidance should be relevant, as well as the involvement of professionals and that the various families still lack an understanding regarding the role, they must fulfil to be able to provide the necessary support for their children. Furthermore, it is concluded that the perceptions of the families are still wrapped around the outdated biomedical paradigm, focused on diagnosis rather than the inclusion process that addresses the needs of the child, as established by the new paradigm focused on neurodivergence

    Bacterial biofilm formation on the surface of PEEK and titanium healing abutments

    Objectives . To explore the effect of surface characteristics on the total volume and viability of a bacterial biofilm developed on the surface of PEEK and titanium healing abutments. Methods. Surface parameters S a and S k, as well as the surface energy of PEEK and titanium healing abutments (n=3) were determined using confocal laser scanning microscopy (CLSM) and contact angle, respectively. The total volume and viability of a multispecies bacterial biofilm cultivated for 30 days were determined using CLSM and the LIVE/DEAD BacLight reactive kit. Effect size was determined using Cohen’s d. Results. PEEK healing abutments displayed a higher surface roughness than titanium (S a 0.41 µm vs 0.17 µm), although no differences in surface energy were observed. Despite the higher total volume of the biofilm measured on titanium abutments compared to PEEK (696 µm3 vs 419 µm3), no differences in the live/dead bacterial ratio were observed. Conclusions. Bacterial viability of the biofilm did not show a direct relation to the surface characteristics of PEEK and titanium healing abutments

    El competitivo mundo de las herramientas de inteligencia de negocios ha llevado a las empresas consultoras y proveedoras de estas herramientas a una competencia por aquellos clientes que no tienen desarrollados estos softwares o para los que ya la tienen, mejorar y hacer que sus empresas puedan tomar decisiones basadas en datos de calidad y a tiempo. El principal objetivo de este estudio es identificar las principales causas de la baja de venta de productos y servicios de la empresa y con ello generar mejoras en las estrategias de generación de nuevos negocios, que permitan a Abisko Group ser una empresa reconocida y líder del mercado. Pero para ello debe transformarse, actualizarse, mirar hacia el futuro. Para lograr estos desafíos, se ha revisado el actual escenario de ventas, sus procesos y los actores que participan en él. Se estudia la estrategia actual de ventas para determinar sus falencias, mejoras y nuevas oportunidades, utilizando herramientas como Customer Journey Map, diagrama de Ishikawa, entre otros. Luego de realizar los estudios, se revelan varias posibilidades de mejoras e implementación de nuevas tareas para lograr el aumento de ventas. Se identifican problemas en el proceso de inscripción de las oportunidades, conocimiento del personal, falta de conocimientos para implementar nuevos servicios, y forma de llegar a nuevos clientes. Además, se destaca que es muy importante invertir en estrategias de marketing para lograr un mejor posicionamiento. En resumen, esta investigación permite conocer integralmente el proceso de venta de Abisko Group, identificando las oportunidades que tiene para que pueda aumentar las ventas y así se consolide como una consultora líder en el mercado.The competitive world of business intelligence tools has led consulting companies and providers of these tools to compete for those clients who do not have these softwares developed or for those who already have them, to improve and make their companies able to make decisions based on quality data and in a timely manner. The main objective of this study is to identify the main causes of the drop in sales of the company's products and services and thereby generate improvements in the strategies for generating new business, which allow Abisko Group to be a recognized company and market leader. But to do so, it must transform, update, and look to the future. To meet these challenges, the current sales scenario, its processes, and the actors involved in it have been reviewed. The current sales strategy is studied to determine its shortcomings, improvements, and new opportunities, using tools such as Customer Journey Map, Ishikawa diagram, among others. After carrying out the studies, several possibilities for improvements and implementation of new tasks are revealed to achieve increased sales. Problems are identified in the process of registering opportunities, staff knowledge, lack of knowledge to implement new services, and how to reach new clients. In addition, it is highlighted that it is very important to invest in marketing strategies to achieve better positioning. In summary, this research allows us to fully understand the sales process of Abisko Group, identifying the opportunities it must increase sales and thus consolidate itself as a leading consultancy in the market

    Fonoaudiología, Licenciado en FonoaudiologíaEl desarrollo fonológico de los niños hispanohablantes en edad preescolar es un aspecto crítico de la adquisición del lenguaje que influye significativamente en su comunicación y aprendizaje académico. Este seminario de investigación tiene como objetivo describir las etapas, inventarios sonoros y procesos de simplificación fonológica propios de los niños hispanohablantes en sus años preescolares. Al realizar una investigación secundaria basada en estudios primarios, el análisis explora patrones de desarrollo fonológico, variaciones dialécticas y factores internos y externos que influyen en este proceso. Los hallazgos indican que la trayectoria del desarrollo fonológico está determinada por factores neurológicos, cognitivos y socioculturales, con una disminución gradual de los procesos de simplificación a medida que los niños crecen. Además, la diversidad dialectal y los factores socioeconómicos introducen variaciones que requieren enfoques sensibles al contexto en el diagnóstico y la intervención. El estudio subraya el papel esencial de los logopedas en la detección e intervención tempranas, enfatizando estrategias que integran prácticas culturales y educativas para apoyar el desarrollo fonológico. Además, la investigación aboga por la exploración continua de la adquisición fonológica en diversas comunidades de habla hispana, contribuyendo a mejores prácticas terapéuticas y educativas para niños en edad preescolar.

    Are we cultivating the perfect storm for a human avian influenza pandemic?

    Publisher Copyright: © The Author(s) 2024.The emergence of highly pathogenic avian influenza (HPAI) A H5N1 virus in dairy cattle marks a troubling new chapter in the ongoing battle against zoonotic diseases. Since its initial detection in 1955, the H5N1 virus has primarily been associated with poultry, posing significant threats to both animal and human health. However, recent outbreaks in U.S. dairy herds across nine states have revealed an alarming expansion of the virus, with over 190 herds affected as of September 2024. This unprecedented spread in cattle has sparked intense concern among scientists and health officials, especially with reports indicating that up to 20% of dairy products may contain traces of the virus. The implications of the H5N1 virus establishing itself in cattle populations are profound. This potential endemic presence could transform dairy farms into reservoirs of the virus, facilitating its evolution and increasing the risk of human transmission. Mutations enhancing viral replication in mammals have already been identified, including the notorious PB2 E627K mutation linked to increased virulence. Moreover, the detection of the virus in the central nervous system of infected animals, including cats, underscores the broad tissue tropism and severe pathogenic potential of the H5N1 virus. Current containment efforts include stringent biosecurity measures and financial incentives for enhanced testing and personal protective equipment (PPE) for farmers. Yet, gaps in testing infrastructure and the resurgence of raw milk consumption pose significant challenges. The U.S. Department of Agriculture (USDA) and the Centers for Disease Control and Prevention (CDC) emphasize the critical need for comprehensive testing and pasteurization to mitigate the risk of human infection. As the scientific community races to adapt existing antiviral treatments and develop effective vaccines, the concept of a One Health approach becomes increasingly vital. This holistic strategy calls for coordinated actions across human, animal, and environmental health sectors to preemptively tackle emerging zoonotic threats. Strengthening surveillance, fostering international cooperation, and investing in research are essential steps to prevent the H5N1 virus from igniting the next global health crisis. The current avian influenza outbreak serves as a stark reminder of the delicate balance between human activities and viral evolution. Our collective ability to respond effectively and proactively will determine whether we can avert the perfect storm brewing on the horizon

    The informal production of public spaces in human settlements in Piura (Peru)

Although there are numerous studies that focus on urban informality, the production of public space remains a relatively understudied area. Based on experiences gathered in five human settlements in Piura, Peru, this study aims to investigate the spatial properties of public space produced by its inhabitants, to understand the motivations and initiative behind its production and use. The paper concludes that the spaces produced represent an opportunity to question and redefine the traditional conception of the city.Las actividades no autorizadas o informales han sido parte integral de la vida urbana durante mucho tiempo. Aunque existen numerosos estudios que se centran en la informalidad urbana, la producción del espacio público sigue siendo un área relativamente poco estudiada. Basado en experiencias recopiladas en cinco asentamientos humanos en Piura, Perú, este estudio tiene como objetivo investigar las propiedades espaciales del espacio público producido por sus habitantes, comprender las motivaciones y la iniciativa detrás de su producción y su uso.     Hypoxic peripheral chemoreflex stimulation-dependent cardiorespiratory coupling is decreased in swimmer athletes

Physiological Reports published by Wiley Periodicals LLC on behalf of The Physiological Society and the American Physiological Society.Swimmer athletes showed a decreased ventilatory response and reduced sympathetic activation during peripheral hypoxic chemoreflex stimulation. Based on these observations, we hypothesized that swimmers develop a diminished cardiorespiratory coupling due to their decreased hypoxic peripheral response. To resolve this hypothesis, we conducted a study using coherence time-varying analysis to assess the cardiorespiratory coupling in swimmer athletes. We recruited 12 trained swimmers and 12 control subjects for our research. We employed wavelet time-varying spectral coherence analysis to examine the relationship between the respiratory frequency (Rf) and the heart rate (HR) time series during normoxia and acute chemoreflex activation induced by five consecutive inhalations of 100% N2. Comparing swimmers to control subjects, we observed a significant reduction in the hypoxic ventilatory responses to N2 in swimmers (0.012 ± 0.001 vs. 0.015 ± 0.001 ΔVE/ΔVO2, and 0.365 ± 0.266 vs. 1.430 ± 0.961 ΔVE/ΔVCO2/ΔSpO2, both p < 0.001, swimmers vs. control, respectively). Furthermore, the coherence at the LF cutoff during hypoxia was significantly lower in swimmers compared to control subjects (20.118 ± 3.502 vs. 24.935 ± 3.832 area under curve [AUC], p < 0.012, respectively). Our findings strongly indicate that due to their diminished chemoreflex control, swimmers exhibited a substantial decrease in cardiorespiratory coupling during hypoxic stimulation

    Levantamiento y modelado de los procesos críticos en el área de recepción y cajas del Centro de Salud USS (Odontología)

    El presente proyecto documenta el desarrollo y aplicación de la teoría de gestión de procesos de Juan Bravo en el Centro Médico USS Odontología, con el objetivo de construir un manual de procedimientos que mejore el rendimiento del área de recepción y cajas del Centro de Salud USS (CSUSS). Para la construcción del manual, se contó con la participación del personal involucrado en el área correspondiente, realizando una serie de preguntas detalladas sobre el funcionamiento del centro médico como la recepción de pacientes, manejo de cajas, registro de datos de los pacientes y la atención dental. Como parte de la primera etapa, se elaboró un mapa de procesos con la información obtenida, cuyo propósito fue identificar los procesos involucrados en el área de recepción y cajas, facilitando la observación de los procesos actuales identificados en el centro médico. Además, se desarrolló la Modelación Visual Participativa (MVP) inicial de cada proceso identificado, para observar el comportamiento del flujo de información entre los participantes involucrados. En la segunda etapa, se elaboraron los flujogramas de información (FI) en formato digital, mostrando cada actividad en listas de tareas, el cual permitió detectar las posibles contingencias durante el flujo de información. Completados los FI, en la tercera etapa, los procesos fueron revisados y mejorados por la jefatura del centro médico con la colaboración del personal involucrado, quienes proporcionaron sugerencias de mejora ante las contingencias detectadas anteriormente. Con los procesos mejorados, en la cuarta etapa se construyó el manual de procedimientos, abarcando los procesos identificados en el área de recepción y cajas. Este manual se diseñó como una herramienta de información para los trabajadores, con el objetivo de mejorar el rendimiento operativo del centro médico. Finalmente, se implementó el manual de procedimientos en cada proceso del área de recepción y cajas. La jefatura del centro se hizo cargo de esta etapa, asegurando la correcta implementación del manual y mejorando así el flujo de trabajo entre los involucrados.This project documents the development and application of Juan Bravo's process management theory at the USS Odontology Medical Center, with the objective of constructing a procedures manual to improve the performance of the reception and cashiering area of the USS Health Center (CSUSS). For the construction of the manual, we counted with the participation of the personnel involved in the corresponding area, asking a series of detailed questions about the operation of the medical center such as patient reception, cash register management, patient data registration and dental care. As part of the first stage, a process map was drawn up with the information obtained, the purpose of which was to identify the processes involved in the reception and checkout area, facilitating the observation of the current processes identified in the medical center. In addition, the initial Participatory Visual Modeling (PVM) of each identified process was developed, to observe the behavior of the flow of information among the participants involved. In the second stage, information flowcharts (IF) were developed in digital format, showing each activity in task lists, which made it possible to detect possible contingencies during the flow of information. Once the IFs were completed, in the third stage, the processes were reviewed and improved by the head of the medical center with the collaboration of the personnel involved, who provided suggestions for improvement in response to the contingencies detected earlier. With the improved processes, in the fourth stage the procedures manual was built, covering the processes identified in the reception and cashiering area. This manual was designed as an information tool for workers, with the objective of improving the medical center's operational performance. Finally, the procedures manual was implemented in each process of the reception and cashiering area. The center's management was in charge of this stage, ensuring the correct implementation of the manual and thus improving the workflow among those involved

Publisher Copyright: © 2023 Institut PasteurExtreme acidophiles thrive in acidic environments, confront a multitude of challenges, and demonstrate remarkable adaptability in their metabolism to cope with the ever-changing environmental fluctuations, which encompass variations in temperature, pH levels, and the availability of electron acceptors and donors. The survival and proliferation of members within the Acidithiobacillia class rely on the deployment of transcriptional regulatory systems linked to essential physiological traits. The study of these transcriptional regulatory systems provides valuable insights into critical processes, such as energy metabolism and nutrient assimilation, and how they integrate into major genetic-metabolic circuits. In this study, we examined the transcriptional regulatory repertoires and potential interactions of forty-three Acidithiobacillia complete and draft genomes, encompassing nine species. To investigate the function and diversity of Transcription Factors (TFs) and their DNA Binding Sites (DBSs), we conducted a genome-wide comparative analysis, which allowed us to identify these regulatory elements in representatives of Acidithiobacillia. We classified TFs into gene families and compared their occurrence among all representatives, revealing conservation patterns across the class. The results identified conserved regulators for several pathways, including iron and sulfur oxidation, the main pathways for energy acquisition, providing new evidence for viable regulatory interactions and branch-specific conservation in Acidithiobacillia. The identification of TFs and DBSs not only corroborates existing experimental information for selected species, but also introduces novel candidates for experimental validation. Moreover, these promising candidates have the potential for further extension to new representatives within the class
