Falabella is one of the leading retail chains in the country, founded in 1889 as a family business by Salvatore Falabella as an “affordable tailor shop” located in the central area of the capital, specifically on Ahumada Street, one of the city's main commercial hubs to this day. Over the decades, thanks to the vision of its descendants, the brand has successfully diversified its business approach. The addition of new partners who shared the same vision helped Falabella evolve from a simple family-run tailoring business into one of the most important department stores in Chile and South America. With 46 branches across the country and a presence in other South American countries such as Peru and Colombia, Falabella has positioned itself as one of the leading companies in the industry. With over 120 years of history, the company has demonstrated its ability to adapt to the constant changes in the market over time, particularly with the arrival of the digital era and the new approach to retail service through e-commerce. This transformation has led to an increasingly competitive retail environment, both physically and online, with the latter gaining greater importance due to its accessibility. Online platforms have empowered customers with more information before making purchasing decisions, making it essential for Falabella to meet customer expectations, ensure high satisfaction levels, and build customer loyalty. The goal is to remain the preferred choice for consumers when addressing their needs and concerns. However, challenges in store coordination can sometimes arise, affecting the final service delivered to customers and leading to dissatisfaction. This, in turn, could result in a loss of market share to competitors. One of the main issues is the lack of clarity regarding product availability, as stock levels are not always the same across all branches or online. This uncertainty affects both customers, who struggle to get a clear answer about whether an item is available, and store employees, who lack the necessary tools to provide accurate information. In the short term, this could lead to a decline in sales, putting the achievement of management's goals at risk. This situation could create a domino effect that ultimately impacts the store’s collaborating partners in terms of commissions and other sales-related benefit

OXGR1-Dependent (Pro)Renin Receptor Upregulation in Collecting Ducts of the Clipped Kidney Contributes to Na+ Balance in Goldblatt Hypertensive Mice

The reduced RBF and oxygen bio-ability causes the accumulation of the tricarboxylic cycle intermediary, α-ketoglutarate, which activates the oxoglutarate receptor-1 (OXGR1). In the kidney, OXGR1 is abundantly expressed in intercalated cells (ICs) of the collecting duct (CD), thus contributing to sodium transport and electrolyte balance. The (pro)renin receptor (PRR), a member of the renin–angiotensin system (RAS), is a key regulator of sodium reabsorption and blood pressure (BP) that is expressed in ICs. The PRR is upregulated in 2K1C rats. Here, we tested the hypothesis that chronic reduction in RBF in the CK leads to OXGR1-dependent PRR upregulation in the CD and alters sodium balance and BP in 2K1C mice. To determine the role of OXGR1 in regulating the PRR in the CDs during renovascular hypertension, we performed 2K1C Goldblatt surgery (clip = 0.13 mm internal gap, 14 days) in two groups of male mice: (1) mice treated with Montelukast (OXGR1 antagonist; 5 mg/Kg/day); (2) OXGR1−/− knockout mice. Wild-type and sham-operated mice were used as controls. After 14 days, 2K1C mice showed increased systolic BP (SBP) (108 ± 11 vs. control 82 ± 5 mmHg, p < 0.01) and a lower natriuretic response after the saline challenge test. The CK group showed upregulation of erythropoietin, augmented α-ketoglutarate, and increased PRR expression in the renal medulla. The CK of OXGR1 knockout mice and mice subjected to the OXGR1 antagonist elicited impaired PRR upregulation, attenuated SBP, and better natriuretic responses. In 2K1C mice, the effect of reduced RBF on the OXGR1-dependent PRR upregulation in the CK may contribute to the anti-natriuretic and increased SBP responses. The two-kidney, one-clip (2K1C) Goldblatt rodent model elicits a reduction in renal blood flow (RBF) in the clipped kidney (CK). According to the World Health Organization (WHO), Cancer means \"a group of diseases that can originate in almost any organ or tissue of the body when abnormal cells grow uncontrollably\". According to WHO 2020 statistics, cancer is the second leading cause of death worldwide with 16.3%. Due to the different alterations that cancer generates, one of these being psychological, studies warn that between 40% and 60% of patients have presented some \"unpleasant emotion or state\", while between 12% and 47% suffers from a mental disorder, which results in a higher mortality rate. A psychological approach in the treatment of patients with cancer has not been explored in the country yet. However, the national plan for cancer for the years 2022-2027, is emphatic in stating that care must be comprehensive, emphasizing psychological support. At Clínica Dávila, out of a total of 670 patients treated, 440 do not receive psychological support, which is the reason for implementing a continuous improvement program focused on detecting and providing tools for dealing with psychological discomfort to all patients and their families who are undergoing active treatment, to provide constant and specific interventions that favor the transition and adaptation to the disease

Publisher Copyright: © 2024Gold(Ⅰ) organometallics of the type alkynylgold(Ⅰ)(NHC) (where NHC = N-heterocyclic carbene) 5a – 5f and 6a/b were prepared and evaluated as prospective anticancer agents by computational and medicinal chemistry methods. DFT calculations showed similar stability for the series, with differing dipole moments, depending on the substituents on the alkynyl and the NHC ligands. Studies in cancer cell lines indicated in general that the complexes with the largest dipole moments exhibited the highest antiproliferative activity in comparison with structurally closely related complexes. Thus, complexes 5f, (with 4-fluoroethynylbenzene as alkynyl precursor), and 6b, (with mestranol as alkynyl precursor) containing the same methoxy‑substituted NHC ligand showed in general the strongest biological activity. Protein binding studies confirmed a high affinity to albumin (around 90 %) and cellular uptake studies indicated an increased uptake of the most active complex 5f

Publisher Copyright: © 2024 The Authors. Addiction published by John Wiley & Sons Ltd on behalf of Society for the Study of Addiction. Background and aims: Substance use treatment (SUT) has shown to be effective in reducing self-reported offending; however, the association between SUT completion and criminal justice system (CJS) contact has been underexplored, especially in Latin America. This study aimed to estimate the association between SUT completion status and (1) any subsequent CJS contact and (2) CJS contact leading to imprisonment, at 1, 3 and 5 years post-discharge, in Chile. Design: Retrospective cohort study using multivariable survival analysis based on linked administrative data from 2010 to 2019. Setting: This study took place in Chile, where SUT is available at no cost through Chile's publicly funded health-care, and is provided in outpatient and inpatient modalities in public and private centres. Participants: A total of 70 854 individuals received their first SUT from 2010 to 2019. They were mainly males (76.3%), and their main substance used at admission was cocaine paste (39.2%). Measurements: SUT completion status included completion, late dropout (≥ 3 months) and early dropout (< 3 months). Outcomes were (1) any CJS contact and (2) CJS contact leading to imprisonment after baseline treatment. We estimated the association between treatment completion and CJS contact through flexible parametric Royston−Parmar models while adjusting for several covariates. Findings: Those who completed SUT (27.2%) were less likely to have any CJS contact at 5 years post-SUT compared with those who dropped out late [with a gap of −9.5%, 95% confidence interval (CI) = −8.7, −10.3] and early (−11.2%, 95% CI = −10.1, −12.3). Also, those who completed SUT were less likely to have CJS contact leading to imprisonment at 5 years post-SUT compared with those who dropped out late (−2.6%, 95% CI = −2.2, −3.1) and early (−4.0%, 95% CI = −3.3, −4.6). These differences were also observed at 1 and 3 years post-SUT for each outcome. Conclusions: In Chile, completion of substance use treatment appears to be associated with lower probabilities of both any criminal justice system contact and contact leading to imprisonment. Background and aims: Substance use treatment (SUT) has shown to be effective in reducing self-reported offending; however, the association between SUT completion and criminal justice system (CJS) contact has been underexplored, especially in Latin America. When bone injuries exceed the body’s natural healing capacity, they become significant obstacles. The global rise in the aging population and the escalating obesity pandemic are anticipated to lead to a notable increase in acute bone injuries in the coming years. Our research developed a novel DLP resin for 3D printing, utilizing poly(ethylene glycol diacrylate) (PEGDA) and various monomers through the PET-RAFT polymerization method. To enhance the performance of bone scaffolds, triply periodic minimal surfaces (TPMS) were incorporated into the printed structure, promoting porosity and pore interconnectivity without reducing the mechanical resistance of the printed piece. The gyroid TPMS structure was the one that showed the highest mechanical resistance (0.94 ± 0.117 and 1.66 ± 0.240 MPa) for both variants of resin composition. Additionally, bioactive particles were introduced to enhance the material’s biocompatibility, showcasing the potential for incorporating active compounds for specific applications. The inclusion of bioceramic particles produces an increase of 13% in bioactivity signal for osteogenic differentiation (alkaline phosphatase essay) compared to that of control resins. Our findings highlight the substantial improvement in printing precision and resolution achieved by including the photoabsorber, Rose Bengal, in the synthesized resin. This enhancement allows for creating intricately detailed and accurately defined 3D-printed parts. Furthermore, the TPMS gyroid structure significantly enhances the material’s mechanical resistance, while including bioactive compounds significantly boosts the polymeric resin’s biocompatibility and bioactivity (osteogenic differentiation)

This research addresses the knowledge that teachers have about trans culture within the classroom, aiming for inclusion in the school context, with the aim of generating pedagogical proposals. The persistence of the hidden gender curriculum in education and its influence on the pressure exerted on study programs is pointed out. The 19th century marked a change in the conceptualization of sexuality, being seen as a behavior to an identity. However, the fight for the recognition of gender identity as a personal and unitary process continues even in the 21st century. Pedagogy becomes a crucial aspect to understand and live trans culture. It advocates eliminating the regularization of bodies and beliefs in education and giving teachers the necessary tools to approach pedagogy from an inclusive perspective. The general objective of the research is to analyze the knowledge of teachers who belong to a school located in the city of Rancagua in the VI Region about Trans culture during the period 2023, to establish a pedagogical proposal that facilitates the process of inclusion within said establishment. A socio-critical paradigm will be used as a search to promote changes in society, having a non-probabilistic and intentional sampling, since the selection of these has in common essential characteristics and criteria in the research, such as being teachers in subjects, in total of 52 teachers of which the research sample is 10 secondary education teachers. To respond to the research objectives, a type of semi-structured interview instrument is carried out, with a focus group method for each specific research objective. All of this focuses on the need to analyze teachers' knowledge of trans culture, especially in rural contexts with a strong heterocisnormative culture. The alarming statistics of homophobic comments and lack of intervention by teachers in cases of hatred towards trans students are mentioned. The importance of representing and understanding trans culture in educational institutions so that each student has trusted references, recognizing the fundamental role of teachers in this process. The research concludes that there is knowledge on the part of the teachers about the Trans culture, which is present, given that there are basic terms integrated into the discourse that the professionals deliver, in addition to knowing and adapting to the various forms of learning and inclusion processes of trans students to improve educational quality for all

A conserved NR5A1-responsive enhancer regulates SRY in testis-determination

Publisher Copyright: © The Author(s) 2024.The Y-linked SRY gene initiates mammalian testis-determination. However, how the expression of SRY is regulated remains elusive. Here, we demonstrate that a conserved steroidogenic factor-1 (SF-1)/NR5A1 binding enhancer is required for appropriate SRY expression to initiate testis-determination in humans. Comparative sequence analysis of SRY 5’ regions in mammals identified an evolutionary conserved SF-1/NR5A1-binding motif within a 250 bp region of open chromatin located 5 kilobases upstream of the SRY transcription start site. Genomic analysis of 46,XY individuals with disrupted testis-determination, including a large multigenerational family, identified unique single-base substitutions of highly conserved residues within the SF-1/NR5A1-binding element. In silico modelling and in vitro assays demonstrate the enhancer properties of the NR5A1 motif. Deletion of this hemizygous element by genome-editing, in a novel in vitro cellular model recapitulating human Sertoli cell formation, resulted in a significant reduction in expression of SRY. Therefore, human NR5A1 acts as a regulatory switch between testis and ovary development by upregulating SRY expression, a role that may predate the eutherian radiation. We show that disruption of an enhancer can phenocopy variants in the coding regions of SRY that cause human testis dysgenesis. Since disease causing variants in enhancers are currently rare, the regulation of gene expression in testis-determination offers a paradigm to define enhancer activity in a key developmental process

Exact oscillations and chaos on a non-Abelian coil

Publisher Copyright: © 2024 The Author(s)We construct new exact solutions of the Georgi-Glashow model in 3+1 dimensions. These configurations are periodic in time but lead to a stationary energy density and no energy flux. Nevertheless, they possess a characteristic frequency which manifests itself through non-trivial resonances on test fields. This allows us to interpret them as non-Abelian self sustained coils. We show that for larger energies a transition to chaotic behavior takes place, which we characterize by Poincaré sections, Fourier spectra and exponential growth of the geodesic deviation in an effective Jacobi metric, the latter triggered by parametric resonances
