Mega Tiber, Pinar/0000-0003-0819-0702; Atlihan, Irem/0000-0003-0351-0279(R,S)-Etodolac [1,8-diethyl-1,3,4,9-tetrahydrapyrano(3,4-b)indole-1-acetic acid] is a nonsteroidal anti-inflammatory drug that contains carboxylic acid group with the structure of pyrano[3,4-h]indole. In this study, a series of novel (R,S)-Etodolac derivatives (3a-1) bearing hydrazide-hydrazone moiety were synthesized. The structures of these compounds were characterized by spectral (H-1-NMR and FT-IR analyses) methods. All synthesized compounds were screened for anticancer activity against androgen-independent prostate adenocarcinoma (PC-3, DU-145) and androgen-dependent prostate adenocarcinoma (LNCaP) cell lines by using WST-8 colorimetric method. This method was used for cell viability and cytotoxicity analysis. Compound 3b (SGK-720) [2-(1,8-diethyl-1,3,4,9-tetrahydropyrano[3,4-b]indole-1-yl)acetic acid[(2,6-dichlorophenyl)methylene]hydrazides] showed 10.36, 5.24, 15.53 mu M anticancer activity against PC3, DU145, LNCaP cancer cell lines, respectively. According to JC-1 mitochondrial membrane potential test and Annexin V/PI staining, 3b was found to have apoptotic effect on these cancer cells. It is concluded that compound 3b containing 2,6-dichloro substituents may be one of the candidate molecules to cope with prostate cancer

/0000-0002-0479-6937; Caloglu Buyukselcuk, Elif/0000-0002-5976-6727In today???s rapidly changing business environment, green and innovative (greenovative) activities have become indispensable elements of sustainable supply network management. Realization of this fact obliges firms to consider greenovative as well as traditional criteria in determining their supplier. This study provides a new greenovative systematic approach to supplier selection for small and medium-sized enterprises. Fuzzy multi-criteria decision-making (FMCDM)-based techniques were used to determine the most appropriate supplier with the proposed model. To show the usability of the model, an application was carried out on an automotive supply company. Analytic Hierarchy Process (AHP) and Fuzzy Analytic Hierarchy Process (FAHP) approaches were used to calculate the weights of the supplier selection criteria. After determining criteria weights, different multi-criteria decision-making (MCDM) techniques that are often encountered in the literature were used to identify the best supplier.Science Citation Index Expande

This study aims to investigate the possible effects of myrtle leaf extract (Myrtus communis L.) on, heart, kidney and aorta tissues in the experimental renovascular hypertension (RVH). 32 male Wistar Albino rats were divided into four groups as control, hypertension, hypertension+ramipril, and hypertension+myrtle leaf extract treatment groups. RVH model was induced by Goldblatt's 2-kidney 1-clip method. 12 weeks later than the treatment blood pressures were recorded and oxidant and antioxidant parameters, tissue factor activity, and histological analysis were determined in the kidney, heart, and aortic tissues. The blood pressure levels of the hypertension group significantly increased compared to the control group. Administration of myrtle leaf extract and ramipril significantly decreased the increased blood pressure. In the hypertension group, oxidative damage increased in the kidney, heart, and aorta tissues. In the histological evaluation of tissues in RVH, heart muscle fibres degenerated. Bowman capsule and glomeruli dilated and tubules damaged in the kidney. Myrtle leaf extract administration regenerated the damages and degenerations. The administration of myrtle leaf extract restored the impaired oxidant-antioxidant balance in the heart, kidney and aorta tissues of hypertensive rats. Myrtle leaf extract can be considered as an alternative antihypertensive treatment target in the prevention of oxidative stress-induced damage in renovascular hypertension.Marmara University Scientific Research Project Department [SAG-C-YLP-170419-0142]This study was funded by a grant from Marmara University Scientific Research Project Department (Project ID: SAG-C-YLP-170419-0142).Science Citation Index Expande

Materials and Methods: 30 Sprague Dawley rats (200-300 g) were divided into four groups: Control, control + whey, MTX, and MTX+whey. MTX was administered at 20 mg/kg (single dose) intraperitoneally to the MTX group rats, and 2 mg/kg of whey protein were administered by oral gavage for 10 days to the whey groups. Lipid peroxidation, glutathione, and nitric oxide (NO) levels, as well as glutathione-Stransferase and superoxide dismutase activities were measured in the brain and small intestine. SDS-polyacrylamide gel electrophoresis of the brain and intestine tissues were also carried out. Results: While MTX treatment caused oxidative damage in the brain and small intestine, whey protein administration ameliorated MTXinduced oxidative stress. MTX administration did not change the brain's NO level, while an increase in intestinal NO level was detected. Conclusion: MTX induced oxidative stress in the brain and small intestine changed the protein metabolism in these tissues regardless of reduced food intake. Consecutive 10-day administration of whey proteins has shown its therapeutic effect on MTX-induced brain and small intestine oxidative damage.Emerging Sources Citation Inde

Universal Design Approach To Analysis of Physical Environment for Users With Multiple Sclerosis

    No full text
The concept of Universal Design aims for the utilization of every product and environment for every individual without the need for further adaptation. The frequent occurrence of Multiple Sclerosis (MS) in societies has necessitated a more innovative and exploratory approach to the field of design. Many of the products and built environments encountered by individuals with MS in their daily lives fall short in terms of meeting safety and accessibility needs. It is essential to establish the correct design approach so that users with MS can meet their needs easily and safely without depending on others. In this research, the problems encountered by users with MS in their residences were evaluated within the framework of Universal Design principles, and design recommendations were stated using a sample case in Turkey. Throughout the study, accessibility challenges, mobility, fatigue and balance problems, vision and lighting conditions, climatization and noise sensitivity, and psychological issues were primarily considered. The residence of the sample user with MS was examined in detail, and an in-depth interview was conducted during the research. The difficulties and life routines of the user with MS were observed and identified. Home design solutions associated with Universal Design principles were represented so that the users with MS have safe and accessible housing regarding their physiological and psychological conditions. The highlighted proposals may contribute to architectural design to make modifications for comfortable and safe housing in related cases having similar symptoms with MS. Future research can benefit from these findings and proposals to provide innovative approaches, develop policies, and conduct interdisciplinary studies on related design issues. © 2022, Universitat Politecnica de Catalunya. However, the significant side effects of MTX limits its use. In this study, we aim to assess the hepatoprotective properties of Momordica charantia (MC) against MTX-induced liver damaged in rats.MATERIALS AND METHODS: Following one dose of MTX (20 mg/kg), the rats were given either distilled water or MC extract (300 mg/kg, po) for 5 days. After the dissection of the rats, the liver was removed to analyse tumour necrosis factor -a (TNF-a), interleukin-113 (IL-113), transforming growth factor 13 (TGF-13) and 8-hydroxy-2'-deoxy-guanosine (8-OhdG) levels and superoxide dismutase (SOD), catalase (CAT), and caspase-3 activities. The tissues were also examined histopathologically.RESULTS: The hepatic TNF-a, IL-113, TGF-13, 8-OhdG levels, and Caspase-3 activity in the MTX group were found to be significantly increased compared to the control group. However, MC extract was able to significantly decrease TNF-a, TGF-13, 8-OhdG levels, and Caspase-3 activity. Also, both the SOD and CAT activity of the MTX group decreased compared to the control group. Although only the SOD levels elevated significantly with MC treatment, the SOD and CAT activities of the MC treated group were similar to the control group. Supporting these biochemical parameters, MTX-induced histologic alterations in the liver were also ameliorated via MC treatment.CONCLUSION: Our results demonstrated that MC has a protective role against MTX-induced hepatic tissue injury by reducing apoptosis, oxidative damage, and the expression of pro-inflammatory cytokines.Emerging Sources Citation Inde

Sener, Goksel/0000-0001-7444-6193; Tunali-Akbay, Tugba/0000-0002-2091-9298; DURSUN, Ercan/0000-0001-6025-9565; Tufan, Elif/0000-0003-0684-3693This study aims to investigate the effects of whey proteins on SARS CoV-2 in methotrexate-induced lung tissue damage in rats. To determine the possible effects, rats were divided into four groups as control, control + whey, methotrexate (20 mg/kg, i.p.) and methotrexate + whey. Whey protein concentrate (2 g/kg, oral gavage) was administered for 10 days. Cytokine levels were measured and protein electrophoresis was carried out in serum samples. Lipid peroxidation, nitric oxide and glutathione level, and superoxide dismutase and glutathione S transferase activities were determined in lung samples. Inhibition of SARS CoV-2-targeted lung furin activity and SARS CoV-2 spike protein-angiotensin converting enzyme binding with whey protein concentrate were also measured in each group. In conclusion, whey protein concentrate improved methotrexate-induced lung damage and inhibited lung furin activity targeting SARS-CoV-2 S1/S2 site cleavage and SARS CoV-2 spike protein-angiotensin converting enzyme binding. Whey proteins are potential protective candidates that inhibit SARS CoV-2-related interactions, even in methotrexate-induced lung injury. Practical applications Whey proteins have anticarcinogenic, antihypertensive, antioxidant, antibacterial, antiviral, and immunomodulating properties due to the protein, bioactive peptide, and essential amino acid content. Methotrexate is a folate antagonist and inhibits cell proliferation and purine synthesis. The combined use of whey protein concentrate and methotrexate may be an alternative in the development of new strategies to the treatment approaches against COVID-19. In addition, according to the results of this study, it is thought that the protective effect of whey proteins in healthy conditions before encountering the SARS CoV-2 may be higher than those who have never used it.Science Citation Index Expande
