    In Silico Evaluation of H1-Antihistamine as Potential Inhibitors of SARS-CoV RNA-Dependent RNA Polymerase: Repurposing Study of COVID-19 Therapy

    Introduction: Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), from the family Coronaviridae, is the seventh known coronavirus to infect humans and cause acute respiratory syndrome. Although vaccination efforts have been conducted against this virus, which emerged in Wuhan, China, in December 2019 and has spread rapidly around the world, the lack of an Food and Drug Administration-approved antiviral agent has made drug repurposing an important approach for emergency response during the COVID-19 pandemic. The aim of this study was to investigate the potential of H1-antihistamines as antiviral agents against SARS-CoV-2 RNA-dependent RNA polymerase enzyme. Materials and Methods: Using molecular docking techniques, we explored the interactions between H1-antihistamines and RNA-dependent RNA polymerase (RdRp), a key enzyme involved in viral replication. The three-dimensional structure of 37 H1-antihistamine molecules was drawn and their energies were minimized using Spartan 0.4. Subsequently, we conducted a docking study with Autodock Vina to assess the binding affinity of these molecules to the target site. The docking scores and conformations were then visualized using Discovery Studio. Results: The results examined showed that the docking scores of the H1-antihistamines were between 5.0 and 8.3 kcal/mol. These findings suggested that among all the analyzed drugs, bilastine, fexofenadine, montelukast, zafirlukast, mizolastine, and rupatadine might bind with the best binding energy (< -7.0 kcal/mol) and inhibit RdRp, potentially halting the replication of the virus. Conclusion: This study highlights the potential of H1-antihistamines in combating COVID-19 and underscores the value of computational approaches in rapid drug discovery and repurposing efforts. Finally, experimental studies are required to measure the potency of H1-antihistamines before their clinical use against COVID-19 as RdRp inhibitors

    Üriner İnkontinans Semptomlarının Multipl Sklerozlu Bireylerde Günlük Yaşam Aktiviteleri ile İlişkisinin Araştırılması

    Amaç: Üriner inkontinans (Üİ), Multipl Skleroz (MS) hastalarında yaygın görülen ve günlük aktiviteleri olumsuz etkileyen bir semptomdur. Bu çalışmada MS tanılı bireylerde Üİ şikayetleri ile günlük yaşam aktivitelerini gerçekleştirme becerisi arasındaki ilişkinin araştırılması amaçlanmıştır. Araçlar ve Yöntem: Çalışmaya MS tanılı ve Üİ’ye sahip, yaş ortalaması 42.06±1.42 olan 45 katılımcı dahil edildi. Katılımcı verileri çevrimiçi bir platform olan Google Forms aracılığıyla toplandı. Uluslararası İdrar Kaçırma Konsültasyon Sorgulaması-Kısa Form (ICIQ-SF) Üİ şikayetlerini, İnkontinans Etki Sorgulaması (IIQ-7) anketi ise günlük yaşam aktiviteleri üzerindeki etkiyi değerlendirmek için kullanıldı. ICIQ-SF ile IIQ-7 fiziksel aktivite, sosyal aktivite, sosyal ilişkiler ve duygusal sağlık alt skorları ve total skoru arasındaki ilişkiyi değerlendirmek için Spearman korelasyon analizi yapıldı. Bulgular: Katılımcıların %26.7'si günde 8 kereden fazla idrara çıkmaktaydı. Mesane günlüğü hakkında bilgi sahibi olmayan bireylerin oranı %75.6 ve işemeye başlamakta zorluk çekenlerin oranı ise %44.4'tü. ICIQ-SF ile IIQ-7 fiziksel aktivite ve sosyal aktivite alt boyutları ile total skoru arasında güçlü ilişki bulurken (sırasıyla rho=0.64, p&lt;0.001; rho=0.53, p&lt;0.001; rho=0.56, p&lt;0.001), sosyal ilişkiler ve duygusal sağlık alt boyutları arasında orta derecede ilişki bulundu (sırasıyla rho=0.45, p=0.002; rho=0.38, p=0.009). Sonuç: Üİ semptomlarının MS'li bireylerin günlük aktiviteleri üzerindeki olumsuz etkileri göz önüne alındığında, Üİ için kapsamlı bir değerlendirme yapılarak MS’lilerin, Üİ’nin şiddetine, tipine ve sıklığına göre uyarlanmış bir rehabilitasyon programına dahil edilmesi önemlidir

    Determining the Effects of Nasopharyngeal Suction With Negative and Positive Pressure: Randomised Controlled Trial

    Objective: The study aims to determine the effects of nasopharyngeal suction with negative and positive pressure on pain level, respiratory parameters, and mucosal irritation in infants. Methods: This study was conducted as a posttest randomized controlled experimental research. The data was collected at G & ouml;ztepe Training and Research Hospital between January and November 2020. While the positive pressure suction method was applied to the experimental group, the negative pressure suction method was applied to the control group. Results: It is determined that the pain level mean scores at the 1 st and 5 th minutes after the suction procedure are lower in the experimental group than in the control group (p=0.01). p =0.01). SpO2 values at the 1st, st , 5th, th , and 15th th minutes after the procedure are significantly higher in the experimental group than in the control group (p=0.01). p =0.01). There is no statistically significant difference in the respiratory rate values between experimental and control groups (p>0.05). p >0.05). Conclusion: It is determined that the post-procedure pain level is lower and the SpO2 value is higher in the experimental group (positive pressure suction) compared to the control group (negative pressure suction).     The Effects of Chard Extract Against Streptozotocin-Induced Erectile Dysfunction in Rats

    Background and Aims: To analyze the potential therapeutic effects of chard against streptozotocin (STZ) -induced erectile dysfunction (ED) and oxidative damage in the corpus cavernousum in rats. Materials and Methods: In this study, Sprague-Dawley rats (250-300g) were allocated into groups as follows: control, diabetic, diabetic + chard, and diabetic + insulin. In order to induce diabetes, rats were given 65 mg/kg intraperitoneal streptozotocin. Chard extract was given orally at a dose 2 g/kg for 45 days beginning on 15 th days. Sixty days after STZ injection, intracavernosal pressure (ICP) was measured and rats were decapitated. Blood samples were obtained for glucose, asymmetric dimethylarginine (ADMA)levels, and lactate dehydrogenase (LDH) activity while cavernous tissues were taken to analyze luminol and lucigenin chemiluminescence (CL), malondialdehyde and glutathione and along with histological analysis. Results: The results revealed that diabetes caused significant decreases in cavernosal tissue glutathione levels, while luminol and lucigenin CL, and malondialdehyde levels were significantly elevated. Plasma glucose, ADMA levels, and LDH activity were also found to be increased in diabetic group. On the other hand, both chard extract and insulin treatment reversed these biochemical parameters significantly. Furthermore, it was found that the ICP value examined for evaluating erectile functions were lower in the diabetic group, but increased in both treatment groups which were similar to the control values. Conclusion: According to our results, chard extract, similar to insulin, reduced diabetes -induced oxidative damage in cavernosal tissue and protected erectile functions. This effects may be attributed its hypoglycemic and antioxidant properties

    A Novel Petox-Based Nanogel Targeting Prostate Cancer Cells for Drug Delivery

This nanogel (referred to as SGK 636/Peptide 563/PEtOx nanogel) is created using hydrophilic poly(2-ethyl-2-oxazoline) (PEtOx) through a combination of living/cationic ring-opening polymerization (CROP) and alkyne-azide cycloaddition (CuAAC) "click" chemical reactions. A fluorescent probe (BODIPY) is also conjugated with the nanogel to monitor drug delivery. The characterizations through 1H-NMR, and FT-IR, SEM, TEM, and DLS confirm the successful production of uniform, and spherical nanogels with controllable sizes (100 to 296 nm) and stability in physiological conditions. The biocompatibility of nanogels is evaluated using MTT cytotoxicity assays, revealing dose-dependent cytotoxicity. Drug-loaded nanogels exhibited significantly higher cytotoxicity against cancer cells in vitro compared to drug-free nanogels. Targeting efficiency is examined using both peptide-conjugated and peptide-free nanogels, with the intracellular uptake of peptide 563-conjugated nanogels by tumor cells being 60-fold higher than that of nanogels without the peptide. The findings suggest that the prepared nanogel holds great potential for various drug delivery applications due to its ease of synthesis, tunable functionality, non-toxicity, and enhanced intracellular uptake in the tumor region. This study emphisizes an innovative PEtOx-based nanogel tailored for targeted drug delivery in prostate cancer. Developed using click chemistry, a valuable technique for chemical synthesis, it exhibits consistent nanogel formation with customizable sizes. In vitro, drug-loaded nanogel exhibits potent cytotoxicity against cancer cells.
