    Blockade of the pro‐fibrotic reaction mediated by the miR‐143/‐145 cluster enhances the responses to targeted therapy in melanoma

    International audienceLineage dedifferentiation toward a mesenchymal-like state displaying myofibroblast and fibrotic features is a common mechanism of adaptive and acquired resistance to targeted therapy in melanoma. Here, we show that the anti-fibrotic drug nintedanib is active to normalize the fibrous ECM network, enhance the efficacy of MAPK-targeted therapy, and delay tumor relapse in a preclinical model of melanoma. Acquisition of this resistant phenotype and its reversion by nintedanib pointed to miR-143/-145 pro-fibrotic cluster as a driver of this mesenchymal-like phenotype. Upregulation of the miR-143/-145 cluster under BRAFi/MAPKi therapy was observed in melanoma cells in vitro and in vivo and was associated with an invasive/undifferentiated profile. The 2 mature miRNAs generated from this cluster, miR-143-3p and miR-145-5p, collaborated to mediate transition toward a drug-resistant undifferentiated mesenchymal-like state by targeting Fascin actin-bundling protein 1 (FSCN1), modulating the dynamic crosstalk between the actin cytoskeleton and the ECM through the regulation of focal adhesion dynamics and mechanotransduction pathways. Our study brings insights into a novel miRNA-mediated regulatory network that contributes to non-genetic adaptive drug resistance and provides proof of principle that preventing MAPKi-induced pro-fibrotic stromal response is a viable therapeutic opportunity for patients on targeted therapy

    From cultivar mixtures to allelic mixtures: opposite effects of allelic richness between genotypes and genotype richness in wheat

    International audienceAgroecosystem diversification through increased crop genetic diversity could provide multiple services such as improved disease control or increased productivity. However, we still poorly understand how genetic diversity affects agronomic performance. We grew 179 inbred lines of durum wheat in pure stands and in 202 binary mixtures in field conditions. We then tested the effect of allelic richness between genotypes and genotype richness on grain yield and Septoria tritici blotch disease. Allelic richness was tested at 19K single nucleotide polymorphisms distributed along the durum wheat genome. Both genotype richness and allelic richness could be equal to 1 or 2. Mixtures were overall more productive and less diseased than their pure stand components. Yet, we identified one locus at which allelic richness between genotypes was associated with increased disease severity and decreased grain yield. The effect of allelic richness at this locus was stronger than the effect of genotype richness on grain yield (−7.6% vs +5.7%). Our results suggest that positive effects of crop diversity can be reversed by unfavourable allelic associations. This highlights the need to integrate genomic data into crop diversification strategies. More generally, investigating plant-plant interactions at the genomic level is promising to better understand biodiversity-ecosystem functioning relationships

    Nonlinear elliptic equations with measure valued absorption potentials

    International audienceWe study the semilinear elliptic equation −∆u + g(u)σ = µ with Dirichlet boundary conditions in a smooth bounded domain where σ is a nonnegative Radon measure, µ a Radon measure and g is an absorbing nonlinearity. We show that the problem is well posed if we assume that σ belongs to some Morrey class. Under this condition we give a general existence result for any bounded measure provided g satisfies a subcritical integral assumption.     Impact of Private Observation in the Bayesian Persuasion Game

    International audienceIn the Bayesian persuasion setting, the sender aims at persuading the decision maker, so called the decoder, to choose a certain action among a set of possible actions. This paper considers two Bayesian persuasion games: one that involves the observation of a private signal by the decoder in addition to the signal transmitted by the encoder, and another version where no private signal is accessible by the decoder. Our goal is to examine the impact of this private signal on the encoder's optimal utility. In order to do so, we investigate an example involving a binary state, a binary private signal and a binary receiver's actions set. We identify the optimal splitting of the decoder's beliefs satisfying the information constraint imposed by the restricted communication channel, and we compute the encoder's optimal utility value, with and without private signal. Varying the parameters such as the prior belief, the precision of the private signal and the channel capacity, we aim at determining which of the two settings is more favorable to the encoder

For instance, it would be interesting to analyse, compare and understand whether the BJP-ruled Indian government passed the National Educational Policy (NEP; 2020) so as further privatize the nation’s educational system. What multidimensional impacts could the New Education Policy (NEP) have on the future of India’s educational system? We would also like to investigate how it affects institutional autonomy, premier public universities, cultural dynamics, and intellectual diversity. This paper investigates how the NEP will reshape education in India. The broad conclusion is that said education is witnessing deep shifts and changes which will not only lead to a deepening of social inequalities and affect learning but will also burden poor and middle-class families with added financial pressure
