    3D melanoma spheroid model for the development of positronium biomarker

    Melanoma spheroids (lWM 115 WM 266-4)were irradiated by positrons emitted from the 22Na radionuclide (with activity of about 1 MBq) placed in Kapton foil. A dedicated aluminum chamber was used as a container for spheroids and a holder which was connected to a heater to keep cells at 370C. Exemplary experimental lifetime spectrum (black histogram) with superimposed histograms resulting from the fit of the sum of the exponential function convoluted with the detector resolution, performed by means of the PALS Avalanche program. First component (yellow line) shows the contribution from p-Ps (mean lifetime: 0.125 ns), second component (green line) originates from annihilations in source (Kapton foil) (0.374 ns), third component (light blue) shows free annihilation lifetime (0.395 ns), forth component (dark blue) illustrates the contribution from o-Ps. A sum of all contributions resulting from the fit is shown as a red curve

    The human skin is exposed to various environmental factors including solar radiation and ambient air pollutants. Although, due to its physical and biological properties, the skin efficiently protects the body against the harm of environmental factors, their excessive levels and possible synergisticaction may lead to harmful effects. Among particulate matter present in ambient air pollutants,PM2.5 is of particular importance for it can penetrate both disrupted and intact skin, causing adverseeffects to skin tissue. Although certain components of PM2.5 can exhibit photochemical activity, only a limited amount of data regarding the interaction of PM2.5 with light and its effect on skin tissue are available. This study focused on light-induced toxicity in cultured human keratinocytes, which was mediated by PM2.5 obtained in different seasons. Dynamic Light Scattering (DLS) and Atomic Force Microscopy (AFM) were employed to determine sizes of the particles. The ability of PM2.5 to photogenerate free radicals and singlet oxygen was studied using EPR spin-trapping and time-resolved singlet oxygen phosphorescence, respectively. Solar simulator with selected filters was used as light source for cell treatment to model environmental lightning conditions. Cytotoxicity of photoexcited PM2.5 was analyzed using MTT assay, PI staining and flow cytometry, and the apoptotic pathway was further examined using Caspase-3/7 assay and RT-PCR. Iodometric assay and JC-10 assay were used to investigate damage to cell lipids and mitochondria. Light-excited PM2.5 were found to generate free radicals and singlet oxygen in season-dependent manner. HaCaT cells containing PM2.5 and irradiated with UV-Vis exhibited oxidative stress features–increased peroxidation of intracellular lipids, decrease of mitochondrial membrane potential, enhanced expression of oxidative stress related genes and apoptotic cell death. The data indicate that sunlight can significantly increase PM2.5-mediated toxicity in skin cells

    The data relating to the commercially available zeolites HY and its desilicated analogues. Various spectroscopic techniques (in situ probe molecules adsorption and time-resolved propane transformation FT-IR studies, XAS, 51V MAS NMR, and 2D COS UV-vis) were employed to study speciation, local coordination, and reducibility of the vanadium species introduced into the hierarchical faujasite zeolite. The acid-based redox properties of V centres were linked to catalytic activity in the oxidative dehydrogenation of propane. The modification of zeolite via caustic treatment is an effective method of adjusting its basicity—a parameter that plays an important role in the ODH process. The Al-rich shell was also beneficial for anchoring V species and thus their reducibility. The operando UV-vis experiments revealed higher reactivity of the bridging oxygens V-O-V over the oxo-group V=O. The (SiO)3V=O species were found to be ineffective in propane oxidation when temperature does not exceed 400 °C
