    Tunable Resistive Pulse Sensing results from the isolation of extracellular microvesicles in comparison with the theoretical model

    The data include the size distribution and concentration of EVs were measured by TRPS technology using the qNano system (Izon Science Ltd.,. Oxford,. UK). The instrument was calibrated using CPC100 and CPC200 calibration particles (Izon Science) according to the manufacturer’s instructions. Exosomes were measured using NP100 nanopores (analysis range 50-330 nm; Izon Science) at a pressure of 5 mbar. Ectosomes were measured using NP200 nanopores (analysis range 85-500 nm; Izon Science) with a pressure of 1 mbar. Voltage and stretch were adjusted to obtain a stable current between 100-120 nA. The samples were analyzed for 3 minutes or until the measurement was performed on 500 objects. Data processing and analysis were prepared on the Izon Control Suite v2.2 software (Izon Science).||4 - Łączna liczba autorów publikacjiExtracellular vesicles (EVs) are heterogeneous 30-5000 nm membrane vesicles that transport proteins, non-coding RNAs (miRNAs), lipids and metabolites. Major populations include exosomes, ectosomes and apoptotic bodies. The aim of this work was to compare the distribution of EVs obtained under different conditions of differential centrifugation, including ultracentrifugation, with the results developed on the basis of a theoretical model.     Polycyclic aromatic hydrocarbons (PAHs) are a group of organic compounds derived mostly from the incomplete combustion of fossil fuels and biomass. Human skin can absorb PAHs and the uptake increases with their molar mass and lipophilicity. Benzopyrene is high-molecular-weight PAH frequently appearing in ambient pollution. It exists in two isomeric forms: benzo[a]pyrene (BaP) and benzo[e]pyrene (BeP), which exhibit different biological activity. Although certain properties of benzopyrenes suggested photoreactivity of the compounds, no direct measurements were previously conducted to characterize their photochemical activity. In this study, quantum yield and action spectra of singlet oxygen photogeneration by BaP and BeP were measured by time-resolved near-infrared phosphorescence and the ability of both compounds to photogenerate superoxide anion was assessed by electron paramagnetic resonance (EPR) spin-trapping. The measurements revealed high efficiency of benzopyrenes to photogenerate singlet oxygen and their ability to photogenerate superoxide anion. Using HaCaT cells as single-layer skin model, we demonstrated concentration-dependent and light-dependent cytotoxicity of BaP and BeP. The compounds induced damage to the cell mitochondria and elevated the levels of intracellular reactive oxygen species
