    No full text
Objective: We aimed to investigate the differences in the immune response to body fat content between the genetically mutant obese BFMI860 (BFMI) mouse strain and the lean C57BL/6Ncrl (B6) mouse strain as a control and the effects of obesity on gene expression on inflammation-related pathways in epididymal adipose tissue. Materials and Methods: Six males from each strain were maintained on a standard maintenance diet (SMD) or a high-fat diet (HFD). At the age of 10 weeks, serum and epididymal adipose tissue samples were collected for cytokine and gene expression analyses. RNA samples from epididymal adipose tissue were hybridized using the microarray technique to study the quantitative transcript amounts of genes. Results: Pathway analysis of gene expression data revealed no considerable development of inflammatory state in BFMI and B6 on SMD. Both strains responded to HFD distinctly; the inflammatory state was more prominent in the obese BFMI group than in the lean B6 group. Several genes, such as Adipoq, NFkbia, Plaur, F2r, C3ar1, and Nfatc4 in pathways involved in the immune system have been found to be differentially regulated in BFMI mice. Under the condition of obesity in BFMI mice, the induction of inflammation-related pathways indicates an increased risk of insulin resistance, atherosclerosis, and cardiovascular disease. Conclusion: This study identified distinct expression patterns of genes involved in inflammatory pathways, particularly those associated with the adipocytokine signaling pathway and complement and coagulation cascades, in the epididymal adipose tissue of BFMI and B6 mice. The BFMI strain is a valuable and promising model for clarifying the mechanisms underlying obesity and the activation of inflammation in adipose tissue

The growing integration of the Internet of Things (IoT) in smart organizations is increasing the vulnerability of cyber threats, necessitating advanced frameworks for effective threat detection and risk assessment. Existing works provide achievable results but lack effective solutions, such as detecting Social Engineering Attacks (SEA). Using Deep Learning (DL) and Machine Learning (ML) methods whereas they are limited to validating user behaviors. Like high false positive rates, attack reoccurrence, and increases in numerous attacks. To overcome this problem, we use explainable (DL) techniques to increase cyber security in an IoT-enabled smart organization environment. This paper firstly, implements Capsule Network (CapsNet) to process employee fingerprints and blink patterns. Secondly, the Quantum Key Secure Communication Protocol (QKSCP) was also used to decrease communication channel vulnerabilities like Man In The Middle (MITM) and reply attacks. After Dual Q Network-based Asynchronous Advantage Actor-Critic algorithm DQN-A3C algorithm detects and prevents attacks. Thirdly, employed the explainable DQN-A3C model and the Siamese Inter Lingual Transformer (SILT) transformer for natural language explanations to boost social engineering security by ensuring the Artificial Intelligence (AI) model and human trustworthiness. After, we built a Hopping Intrusion Detection & Prevention System (IDS/IPS) using an explainable Harmonized Google Net (HGN) model with SHAP and SILT explanations to appropriately categorize dangerous external traffic flows. Finally, to improve global, cyberattack comprehension, we created a Federated Learning (FL)-based knowledge-sharing mechanism between Cyber Threat Repository (CTR) and cloud servers, known as global risk assessment. To evaluate the suggested approach, the new method is compared to the ones that already exist in terms of malicious traffic (65 bytes/sec), detection rate (97%), false positive rate (45%), prevention accuracy (98%), end-to-end response time (97 s), recall (96%), false negative rate (42%) and resource consumption (41). Our strategy's performance is examined using numerical analysis, and the results demonstrate that it outperforms other methods in all metrics

Antimicrobial peptides (AMPs) may mitigate the danger of increasing antimicrobial resistance. We aimed to determine the activities of catestatin, temporin A, nisin and cecropin A against Bacteroides fragilis ATCC 25285, Prevotella melaninogenica ATCC 25845, Cutibacterium acnes ATCC 6919, Peptostreptococcus anaerobius ATCC 27337 and Peptostreptococcus stomatis DSM 17678. strains. The susceptibility of all anaerobic bacteria was determined by Kirby-Bauer disc diffusion method, agar dilution and broth microdilution method, recommended by CLSI. By broth microdilution the MIC of temporin A for P. anaerobius was 500µg/mL, and MBC >500µg/mL. The MIC of nisin for P. melaninogenica was 200µg/mL, with a MBC of 400µg/mL, for C. acnes, P. anaerobius, and P. stomatis, MIC were 40mg/mL. The MIC of cecropin A for B. fragilis was 50µg/mL, MBC was 500µg/mL. For C. acnes, the MIC was 4µg/mL, MBC was 8µg/mL. The MIC for P. melaninogenica, P. anaerobius and P. stomatis were 8µg/mL, with corresponding MBC values of 16, 32 and 50µg/mL, respectively. Conversely, catestatin proved ineffective against all strains. In conclusion, our study, demonstrated that cecropin A and nisin showed promising results against anaerobic standard strains. We believe thatfurther research conducted to explore those AMPs could hold promise as a treatment option for anaerobic bacterial infections

Thermal and mineral springs are natural geological formations where groundwater emerges from the Earth's crust at higher temperatures than the surrounding environment. These springs are often associated with regions of geological activity, such as volcanic areas, fault lines, or areas with deep underground reservoirs. Ispendere Mineral and Thermal Water Spring, located in Malatya province, serves as a vital mineral water source and a significant balneotherapy center for the region, boasting recognized health benefits. Despite extensive geochemical analyses of the waters, microbiological studies have been notably absent. The healing waters of Ispendere are identified by their bicarbonate and sulphur content, forming a blend of mineralized waters. Hydrochemical and isotopic analyses conducted in the region indicate that the thermal and mineralized waters in the vicinity align with the characteristics of MgHCO3-type waters, whereas the cold waters exhibit properties akin to the CaHCO3 type. Despite the thorough chemical assessments that underscore the meticulous attention given to the chemical composition of Ispendere Springs, no microbial study of the region has been reported. This thesis aimed to address this gap by employing a sequence-based metagenomic approach to assess the microbial diversity of the region for the first time. Samples were taken from different parts of the region with sterile spatula into sterile containers. DNA isolation was performed using HigherPurity Soil DNA isolation kit from samples taken from different regions. In order to determine the microbial population present in the samples, sequence analysis was performed as a service procurement. Illumina MiSeq platform was used in the sequencing performed by Eurofins Genomics company within the scope of InView Microbiome Profiling package. The samples were sequenced by targeting the 16S RNA V3V5 variable region. While aquatic microorganisms were prevalent, as anticipated based on existing literature, soil-associated microorganisms were also identified. Notably, members of the genus Planktothrix constituted the most abundant bacterial community, comprising 37.1% of the total microbial composition. The Naviculales team is the second most represented group, comprising 21.6% of the sequences. At the species level, the sequences bear the closest resemblance to Navicula sp. C20, Phaeodactylum tricornutum, and Pinnularia sp. U-strain. Rhodobacteria, constituting 4.1% of the sequenced samples, are purple photosynthetic bacteria. Parasegetibacter terrae representing 1.2% of the fraction, is a Gram-negative, flagellated, yellow-pigmented aerobic bacterium. Furthermore, Terrimonas ginsenosidimutans accounting for 1.3% of the fraction, was isolated from soil and noted for its ability to produce glycoside hydrolase and convert minor-major ginsenosides.

Wireless Sensor Networks (WSNs) are increasingly being used in a variety of applications such as environmental monitoring, healthcare, and industrial automation. However, the widespread deployment of WSNs has raised concerns about their security, as these networks are vulnerable to a wide range of attacks. Cybersecurity in WSNs involves protecting the confidentiality, integrity, and availability of data, as well as ensuring the authenticity of the communication between the nodes. One of the key challenges in WSN security is the limited resources of the sensor nodes, such as low processing power, memory, and energy. As a result, traditional security solutions may not be suitable for WSNs, and new lightweight security mechanisms must be developed to provide efficient protection against attacks.Several techniques have been proposed to enhance the security of WSNs, including encryption, authentication, access control, intrusion detection, and secure routing protocols. These techniques aim to prevent unauthorized access to the network, detect and respond to attacks, and ensure the privacy of data transmission. Therefore, this paper provides a comprehensive examination of security risks against WSN , as well as solutions for preventing, detecting, and mitigating such attacks. By being familiar with the mechanics of these attacks, the relevant defense mechanisms can aid in the development of a secure IoT expansion and widespread knowledge. The goal of this research is to discuss and show the impact of security issues on WSNs from the perspective of IoT and thus its applications. A description of relevant attacks and threats against these requirements was also included in the study performed for this work

The Effect of Concrete Cover Thickness for Reactive Powder Concrete on the Column Resistance Against Fire

    No full text
Structures made of reinforced concrete are often exposed to fire of varying intensity. If the building does not burn down, a post-fire study of the building's structural integrity is necessary before determining the building's destiny. It can forecast the remaining loading capacity of the columns after a fire by knowing the temp of the fire, the thickness of the cover of concrete, the concrete's residual strength, and the tensile strength of the embedded strengthening. Over the past few years, there has been significant progress in the construction of urban cities. This progress has led to improvements in the building process, particularly in the requirements of structural design for extremely robust building products. One important factor that has been given more attention in building design is the length of building elements. Since its exceptional strength and durability, reactive powder concrete (RPC) has gained significant attention in the field of building construction in recent times. Therefore, incorporating enhancements can greatly contribute to preserving the optimal performance of concrete in various scenarios, including fire exposure. The current study seeks to examine the variations in strength of reactive powder concrete once subjected to different burning temperatures (200, 400, and 600℃) through experimental testing. The experiment involved three sets of specimens; each set exposed to different burning temperatures for one hr. Within each set, the specimens were further split into four subgroups to observe the effects on cover of concrete at depths of 15mm and 30mm, with and without secondary strengthening. The findings gained indicate that greater burning temperatures, ranging from 200-600, lead to a decrease in the compressive, tensile, and flexural strength of reactive powder concrete. In addition, the decision for increment the cover of concrete from 15 to 30 mm has proven to be effective in enhancing the fire resistance of the concrete column. Furthermore, the utilization of secondary strengthening with a similar cover of concrete has resulted in a significant increment in the overall fire resistance of the concrete structure. © (2024) The authors. Insights from CD71 presentation and serum lipid peroxidation in myasthenia gravis - A small cohort study

    No full text
Myasthenia gravis (MG) is a multifaceted autoimmune disorder affecting the postsynaptic neuromuscular junction. In this study, we examined CD4+ and CD8+ T lymphocyte levels and ratios within peripheral blood mononuclear cells (PBMCs) in MG patients. Additionally, we assessed lymphocytes for the expression of CD71, which functions as a transferrin receptor mediating the uptake of iron into the cells. Building on recent discussions regarding CD20 depletion treatments in MG, we also scrutinized lymphocytes for CD20 expression. Comparative analyses were conducted among healthy controls, newly diagnosed MG patients, those undergoing pyridostigmine treatment alone, and MG patients receiving combination therapies. In the patients, the ratio of CD3+CD4+ T lymphocytes to CD3+ T lymphocytes was found to be decreased compared to the healthy controls, while the ratio of CD3+CD8+ cells to CD3+CD4+ cells increased. An increase in the percentage of CD71-expressing lymphocytes was observed in MG patients compared to the healthy control group, while CD20+ lymphocytes exhibited no statistical changes. Moreover, heightened serum lipid peroxidation levels were found in MG patients. These results suggest a possible relationship between iron metabolism, levels of CD71-expressing cells, and lipid peroxidation in MG. Conversely, pyridostigmine treatment reduced the levels of CD71-expressing cells and lipid peroxidation, suggesting potential immunomodulatory and antioxidant impacts of pyridostigmine in MG, either directly or indirectly

Enhancing self-care prediction in children with impairments: a novel framework for addressing imbalance and high dimensionality

    No full text
Addressing the challenges in diagnosing and classifying self-care difficulties in exceptional children's healthcare systems is crucial. The conventional diagnostic process, reliant on professional healthcare personnel, is time-consuming and costly. This study introduces an intelligent approach employing expert systems built on artificial intelligence technologies, specifically random forest, decision tree, support vector machine, and bagging classifier. The focus is on binary and multi-label SCADI datasets. To enhance model performance, we implemented resampling and data shuffling methods to tackle data imbalance and generalization issues, respectively. Additionally, a hyper framework feature selection strategy was applied, using mutual-information statistics and random forest recursive feature elimination (RF-RFE) based on a forward elimination method. Prediction performance and feature significance experiments, employing Shapley value explanation (SHAP), demonstrated the effectiveness of the proposed model. The framework achieved a remarkable overall accuracy of 99% for both datasets used with the fewest number of unique features reported in contemporary literature. The use of hyperparameter tuning for RF modeling further contributed to this significant improvement, suggesting its potential utility in diagnosing self-care issues within the medical industry
