    Evaluation of keratin/bacterial cellulose based scaffolds as potential burned wound dressing

    The study presents the preparation and characterization of new scaffolds based on bacterial cellulose and keratin hydrogel which were seeded with adipose stem cells. The bacterial cellulose was obtained by developing an Acetobacter xylinum culture and was visualized using SEM (scanning electron microscopy) and elementally determined through EDAX (dispersive X-ray analysis) tests. Keratin species (β–keratose and -keratose) was extracted by hydrolytic degradation from nondyed human hair. SEM, EDAX and conductometric titration tests were performed for physical– chemical and morphological evaluation. Cytocompatibility tests performed in vitro confirmed the material non-toxic effect on cells. The scaffolds, with and without stem cells, were grafted on the burned wounds on the rabbit’s dorsal region and the grafts were monitored for 21 days after the application on the wounds. The clinical monitoring of the grafts and the histopathological examination demonstrated the regenerative potential of the bacterial cellulose–keratin scaffolds, under the test conditions

    Dinamica metanolului și a acetaldehidei în vinurile albe tratate cu dioxid de sulf și dimetil dicarbonat

    Wine structure is very complex, consequently, the latest researches has been focused on the detailed analysis of this beverage. One of the most important compounds that can be found in wines is methanol. The toxicology of methanol and the admissible limits established by OIV, especially the effect on consumer's health, have to be considered. In this sense, the maximum permissible quantity of methanol and changes in its composition in wines must always be discussed. For this study, forty-five samples were obtained from a blend of Fetească regală and Muscat Ottonel grape varieties at the experimental wine cellar of the Oenology Laboratory of the Faculty of Horticulture from Iași. All variants were treated with 6 % SO2 solution (40, 80, 160 mg/L) and dimethyl dicarbonate liquid solution, in various ratios (0, 100, 200 μL/L). Also, in this experiment, yeasts such as Schizosaccharomyces spp. and Brettanomyces spp. were inoculated separately (S-Schizosaccharomyces pombe, B-Brettanomyces bruxellensis) and the evolution of methanol content and acetaldehyde in wines was recorded following the administration of treatments with dimethyl dicarbonate and sulphur dioxide. The main purpose of this research is to evaluate the methanol content in wine samples using a gas chromatography method and the possible transformation in wine composition (methanol concentrations) produced by DMDC. Moreover, an important compound that can be formed in wines due to SO2 presence is acetaldehyde, so, the quantity and the effect on wine composition were discussed. The results show different amounts of methanol and acetaldehyde due to the reactions of the used treatments with specific compounds of wine. The concentration of methanol and acetaldehyde in the studied wine samples are within permitted limit for white wines, 250 mg/L methanol

    Evaluate some new inbred rice and male sterile varieties under DUS and VCU experiments

    The main objective of study are evaluated some new inbred rice and male sterile varieties under Distinct, Uniform and Stability (DUS) and Value of Cultivated and Used (VCU) Experiments. A number of seven rice varieties were used, Sakha 101, Sakha 102, Sakha 104, Giza 177, Giza 178, Giza 182 and Egyptian yasmine; moreover, GZ 10154 and GZ 8564-Sp 70, as well as EGMS and CMS1 as promising lines to evaluating by DUS and VCU. The data were recorded on morphological and yield characters. The results could be concluded that: regarding to qualitative characteristics (PQ and QL), from 24 PQ and QL characters, the nine rice varieties recorded the same score for 19 characters; moreover, the rice varieties Egyptian jasmine was dissimilar in the score No. 24, 39, and 60, that referred to this variety belong to Indica type, while the rice varieties Sakha 101 and Giza 178 were similar only in score no.11, which belong to Japonica and Indica japonica types, that meaning these varieties were highly uniform and stability in qualitative characters than the other promising line GZ 10154. Regarding to quantities characteristics (QN), from 27 QN characters, nine rice varieties recorded the same score for 20 characters; moreover, the rice varieties Sakha 101, Sakha 102, Sakha 104, Giza 177, Giza 178, Giza 182, Egyptian yasmine and GZ 8564-Sp70 were similar in the score no. 1, 12, 23, 48, 49 and 50, while the promising line GZ 10154 was dissimilar in these scores during the two seasons, meaning that all these varieties were highly uniform and stability than the other promising, line GZ 10154. These results were conformed to VCU results, where the studied varieties recorded the highest grain yield/day. From these results could be concluded that all the varieties, except GZ 10154, accepted as a new rice release variety, but the promising line GZ 10154 required to more recurrent selection to increase their uniform, as well as CMS line was accepted could be evaluated under different conditions, but EGMS should be evaluated under heat stress conditions

    The content of vitamin C in processed horticultural products is influenced by multiple factors. Besides the factorsconcerning the raw material (cultivar, climatic conditions, agricultural practices, harvesting and storage techniques),the processingand preservation technologies are essential. The objective of the present study is to analysethe variation of vitamin C content during the technological processesof obtaining the product "Romanian peppers in vinegar". The samples from5 different steps of the technological flow were supplied by a company for processing vegetables and fruits (S.C. Contec Foods S.R.L. Tecuci),during two consecutive years. The finished product was also analysedafter storage at two temperatures (10°C and 25°C), by using two different methods for vitamin C assessment.The results proved that vitamin C content decreased by 76% during the technological flow. All technological phases decreased the content of ascorbic acid, but the most important reduction was registered after the thermal processing(blanching and pasteurization).The storage of the finished product for 3 months determined the diminution of vitamin C content: at 10 ºC by5% and at 25 ºC by 14%

    In vitro evaluation of probiotic properties of A lactic acid bacteria isolated from piglet

    The aim of this study was to evaluate the probiotic aspects of lactic acid bacteria (LAB) isolated from faeces of a healthy piglet (30-d-old, Topigs). Cultural, morphological and biochemical characteristics by standard API 50 CHL Biomerieux strips of isolated strain was assayed as well. The strain was conserved as Lactobacillus fermentum 1 IBNA 95 and can be found in the collection of INCDBNA. It is a Gram positive, non-spore forming, aerotolerant bacteria. It appears isolated and grew at 37°C in Oxoid MRS broth and MRS agar medium. The functional properties tested were to survive at pH 3.0 and to present resistance at addition of 0.3% bile salts (w/v). The strain was able to survive at a low pH value and to tolerate 0.3% bile salt solution during 3h exposure under according simulated gastrointestinal condition. Lactobacillus fermentum 1 IBNA 95 was presenting some probiotic features, thus can be a promising feed supplement for piglet’s nutrition

    A review on plant genomes of some important traditional chinese fruits and herbs

    Chinese medicinal herbs and fruits have grown rapidly and significantly in recent years and have a positive influence on improving people’s attention to their health and organic life style. According to the advancement of sequencing technologies and reduced costs, the genome sequencing data of medicinal plants are accumulating rapidly. Our aim was to review plant genomes of three important medicinal plants in China. There is an ample genetic diversity of plants with medicinal importance around the globe and this pool of genetic variation serves as the base for selection, as well as for plant improvement. Plant genomes are characterized by large variations of genome size and ploidy level. Comparative genomics provides a method to unravel the relationship between genomes, by describing conserved chromosomes or chromosomal regions between related species. It is also clear that it is possible to use plant genome as a tool for improving breeding strategies. However, certain limitations represent a number of challenges for the generation and utilization of genomic resources in many important medicinal plant species. This review has focused on plant genomes of some important horticultural plants, which are famous in traditional Chinese medicine, namely ginger, ginseng and goji berry. However, more researches are needed to introduce the genome research of medicinal plants

    Tank size and aquaculture management influence on the production of gold fish, Carassius auratus (L.), under tropical conditions

    Growth performance of gold fish Carassius auratus (L.) produced in tanks of different size and treated under different management regimes were compared. Tanks of two different size and make were used (concrete; capacity: 2000 l and earthen; capacity: 59650 l) for four management regimes: (1) live zooplankton fed to fish larvae in concrete (CLF) and earthen tanks (ELF); (2) application of poultry manure in concrete (CPM) and earthen tanks (EPM); (3) application of cow manure in concrete (CCD) and earthen tanks (ECD); and (4) a control treatment for concrete (CC) and earthen tanks (EC). Fish larvae (0.10 ± 0.007 g) were cultured for 11 weeks. Weight gain of gold fish was highest in the ELF treatment. There was a significant difference in the survival of gold fish among the treatments, ranging from 64.83% in CC to 93.40% in ELF. Significantly higher values of pH and dissolved oxygen were obtained in the live food and control treatments (for both concrete and earthen tanks), compared to the manured treatments. The concentration of total alkalinity, BOD, PO4 – P, NO3 – N and specific conductivity were significantly higher in EPM and ECD, compared to other treatments. The results indicate that introduction of live zooplankton into culture units resulted in higher growth of fish larvae. Better growth of cultured fish were obtained in larger earthen tanks compared to smaller concrete tanks through maintenance of superior water quality and greater abundance of plankton
