    Osetljivost sivog pacova (Rattus norvegicus) na kombinacije antikoagulanata prve i druge generacije

    Primenom testa bez izbora hrane u trajanju od četiri dana prema metodi EPPO standarda (2004) jedinkama sivog pacova (Rattus norvegicus) ponuđeni su mamci antikoagulanata prve (hlorofacinon) i druge generacije (bromadiolon, difenakum i brodifakum) koji su međusobno kombinovani. Dobijene kombinacije hlorofacinona i antikoagulanata druge generacije kao i kombinacije antikoagulanata druge generacije bile su višestruko niže (10 ppm i 8 ppm) u odnosu na komercijalno dostupne mamce (50 ppm). Smrtnost ispitivanih jedinki (100 %) postignuta je za sve kombinacije u kojima je bio prisutan difenakum (difenakum+hlorofacinon, difenakum+bromadiolon i difenakum+brodifakum) kao i primenom kombinacije bromadiolon+brodifakum. Pri spamenjom sadržaju aktivnih supstanci (8 ppm) zabeležena je smrtnost od 100 % u kombinacijama u kojima je bio prisutan brodifakum (difenakum+brodifakum i bromadiolon+brodifakum), dok je u kombinacijama difenakum+hlorofacinon i difenakum+bromadiolon smrtnost bila 66 %, odnosno 83 %. Smrtnost od 33 % i 50 %, zabeležena je za mamce na bazi hlorofacinona, u kombinaciji sa bromadiolonom ili brodifakumom. Na osnovu dobijenih rezultata može se zaključiti da se kombinovanjem određenih antikoagulanta pri koncentraciji višestruko nižoj od komercijalno dostupnih postiže smrtnost sivog pacova od 100 %. Njihovom primenom značajno se unapredjuje program zaštite od štetnih vrsta glodara, naročito sa stanovišta zaštite životne sredine i neciljnih organizama

    Rubbery taproot disease of sugar beet in Serbia associated with 'Candidatus phytoplasma solani'

    Rubbery taproot disease (RTD) of sugar beet was observed in Serbia for the first time in the 1960s. The disease was already described in neighboring Bulgaria and Romania at the time but it was associated with abiotic factors. In this study on RTD of sugar beet in its main growing area of Serbia, we provide evidence of the association between 'Candidatus Phytoplasma solani' (stolbur phytoplasma) infection and the occurrence of typical RTD symptomatology. 'Ca. P. solani' was identified by PCR and the sequence analyses of 16S ribosomal RNA, tuf, secY, and stamp genes. In contrast, the causative agent of the syndrome "basses richesses" of sugar beet-namely, 'Ca. Arsenophonus phytopathogenicus'-was not detected. Sequence analysis of the stolbur strain's tuf gene confirmed a previously reported and a new, distinct tuf stolbur genotype (named 'tuf d') that is prevalent in sugar beet. The sequence signatures of the tuf gene as well as the one of stamp both correlate with the epidemiological cycle and reservoir plant host. This study provides knowledge that, for the first time, enables the differentiation of stolbur strains associated with RTD of sugar beet from closely related strains, thereby providing necessary information for further epidemiological work seeking to identify insect vectors and reservoir plant hosts. The results of this study indicate that there are differences in hybrid susceptibility. Clarifying the etiology of RTD as a long-known and economically important disease is certainly the first step toward disease management in Serbia and neighboring countries

    Prouzrokovači truleži plodova breskve nakon berbe

    Po obimu proizvodnje breskve (Prunus persica (L.) Batsch) Srbija se nalazi na petom mestu u Evropi. U Srbiji, breskva je takođe na petom mestu, posle jabuke, kruške, šljive i višnje, sa ukupnom zastupljenošću od oko 5% i obimom proizvodnje koji varira od 53 894 tone u 2012. godini do 130 808 tona u 2015. godini. Breskva se gaji u rejonu Podunavlja, centralne Šumadije i delimično Srema, severne Bačke i južnog Banata, sa najvećim površinama u Grockoj, Smederevu, Topoli, Mladenovcu i Inđiji. Proizvodnju ove voćarske vrste ugrožava veliki broj patogena, kako tokom perioda vegetacije, tako i tokom perioda skladištenja, transporta i prodaje. U godinama sa povoljnim vremenskim uslovima za razvoj patogena, poseban značaj imaju prouzrokovači truleži koji se javljaju posle berbe i koji mogu naneti ekonomske gubitke i do 80%. Cilj ovog istraživanja bio je da se prouči pojava simptoma truleži na plodovima breskve posle berbe, da se izoluju i identifikuju prouzrokovači oboljenja. Tokom 2021. godine, iz okoline Smedereva i Topole, odabran je po jedan reprezentativni zasad breskve u kome su primenjene sve preporučene mere zaštite. Iz svakog zasada prikupljeno je po 300 ujednačenih plodova (sorte Red Haven i Royal Glory) u fazi tehnološke zrelosti. Plodovi su po dopremanju u laboratoriju pojedinačno upakovani, raspoređeni u gajbice po slučajnom blok sistemu i inkubirani na sobnoj temperaturi u periodu od deset dana. Pojava simptoma truleži na plodovima breskve praćena je svakodnevno. Nakon pojave simptoma, iz plodova je vršena izolacija patogena primenom standardnih fitopatoloških metoda. Po dobijanju čistih kultura, patogenost izolata proverena je na zrelim plodovima breskve, a identifikacija patogena izvršena je na osnovu makroskopskih i mikroskopskih morfoloških karakteristika izolata. Identifikacija reprezentativnih predstavnika morfološki identičnih izolata potvrđena je sekvenciranjem PCR produkata ITS (Internal transcribed spacer) rDNA regiona, dobijenog korišćenjem prajmera ITS1/ITS4. Rezultati istraživanja pokazali su da su 2021. godine prouzrokovači truleži bili visoko zastupljeni na plodovima sa oba proučavana lokaliteta; njihovo prisustvo detektovano je na 44,3% plodova breskve sa lokaliteta u Topoli i 70% sa lokaliteta u Smederevu. Osim toga, utvrđen je i veliki diverzitet patogena koji su razvrstani u preko deset rodova fitopatogenih gljiva, među kojima su najznačajniji sledeći: Rhizopus, Aspergillus, Monilinia, Alternaria, Penicillium, Trichothecium, Fusarium, Botryospheria, Mucor i Epiccocum. Na oba lokaliteta najdestruktivnije su bile vrste roda Rhizopus koje su prouzrokovale trulež plodova već nakon inkubacije od tri dana, kao i vrste roda Monilinia sa pojavom karakterističnih simptoma nakon sedam i deset dana inkubacije. Monilinia fructicola, najdestruktivniji patogen breskve, pronađen je na oba ispitivana lokaliteta. Dalja istraživanja će pokazati da li su detektovani patogeni prouzrokovači latentnih infekcija koji izazivaju truleži tek nakon berbe, ili su patogeni na plodovima prisutni epifitno, a infekciju ostvaruju nakon berbe

    Occurrence and Identification of Pectobacterium carotovorum subsp. brasiliensis and Dickeya dianthicola Causing Blackleg in Some Potato Fields in Serbia

    Blackleg outbreaks were noticed on three fields (about 100 ha total) in 2 consecutive years (2018, 2019) in one of the main potato growing areas in Serbia (Backa region, Vojvodina). The percentage of infected plants reached 40 to 70%, with 10.5 to 44.7% yield reductions. From the three fields, out of 90 samples Pectobacterium carotovorum subsp. brasiliensis was most frequently identified and diagnosed as causal agent of potato blackleg in Serbia for the first time (29 isolates). Dickeya dianthicola was a less frequently causative bacterium, which was also noticed for the first time (nine isolates). A total of 38 isolates were characterized based on their phenotypic and genetic features, including a pathogenicity test on potato. The repetitive element PCR (rep-PCR) using BOX, REP, and ERIC primer pairs differentiated five genetic profiles among 38 tested isolates. Multilocus sequence analysis (MLSA) of four housekeeping genes, acnA, gapA, icdA, and mdh, revealed the presence of three so far unknown P. c. subsp. brasiliensis multilocus genotypes and confirmed clustering into two main genetic clades as determined in other studies. MLSA also revealed the presence of a new genotype of D. dianthicola in Serbia

    Otkriće prisustva istočnoazijske cikade Arboridia kakogawana (Hemiptera: Auchenorrhyncha: Cicadellidae: Typhlocybinae) u Srbiji

    The most recent findings on allochthonous Auchenorrhyncha species in Serbia have reported two Typhlocybinae (fam. Cicadellidae) species new to the local entomofauna: Tautoneura polymitusa Oh & Jung, 2016 and Erasmoneura vulnerata (Fitch, 1851) (Kosovac et al., 2020; Šćiban & Kosovac, 2020). A further survey in 2020 revealed the presence of another alien typhlocybin species, Arboridia kakogawana (Matsumura, 1932). This East Asian species, known as the Japanese grape leafhopper, was recorded on the European continent for the first time in the Krasnodar area of the southern Russia in the early 2000s (Sugonyaev et al., 2004; Gnezdilov et al., 2008a). Soon after, it was detected on the Crimean Peninsula (Radionovskaya & Didenko, 2014; Radionovskaya & Didenko, 2015), while its first occurrence on EU territory was recently reported in Romania (Chireceanu et al., 2019). Each finding portrayed A. kakogawana as a phytosanitary risk for local grapevine production due to the intense feeding of its nymphs and adults on leaf mesophyll, consequently affecting grape maturation (Gnezdilov et al., 2008b; Chireceanu et al., 2019).Do sada su u Srbiji detektovane samo dve invazivne vrste cikada iz podfamilije Typhlocybinae - Tautoneura polymitusa Oh & Jung, 2016 i Erasmoneura vulnerata (Fitch, 1851). Istočnoazijska vrsta, Arboridia kakogawana, je u Evropi prvi put evidentirana početkom 2000-ih u evropskom delu Ruske Federacije, dok je 2019. u Rumuniji prijavljen prvi nalaz za teritoriju EU. Tokom jula i avgusta 2020. otkriveno je prisustvo populacija A. kakogawana na četiri lokaliteta u severnom delu Srbije – Novi Sad, Srbobran, Beograd (Zemun) i Crvenka. Na svim lokalitetima A. kakogawana je pronađena u asocijaciji sa vinovom lozom; uzorkovano je i genitalizovano više primeraka, dok su u Srbobranu i Beogradu zabeležene i vinove loze sa specifičnim simptomima hlorotičnih pega na listovima koji ukazuju na prisustvo ove alohtone cikade. Arboridia kakogawana je alohtona cikada, najverovatnije prisutna u Srbiji već nekoliko godina, i uspešno se širi prema zapadu evropskog kontinenta

    Model za simulaciju procene rezerve semena korovskih biljaka u zemljištu

    Rezerve semena korovskih biljaka u zemljištu sa jedne strane predstavljaju stalni izvor zakorovljenosti, dok sa druge imaju ulogu u održavanju stabilnosti ekosistema i biodiverziteta. Malobrojna istraživanja na ovu temu su pokazala veliku varijabilnost u rezultatima, u sličnim eksperimentalnim uslovima, ali pri korišćenju različitih metoda. Stoga ciljevi u ovom istraživanja su bili odabir adekvatne i precizne metode za procenu rezerve semena korovskih biljaka i pravljenje pouzdanog modela za prognoziranje dinamike pojave korovskih populacija. Istraživanja su izvedena na oglednom polju Instituta za ratarstvo i povrtarstvo u Novom Sadu (N 45°19', E 19°50') u okviru dugogodišnjeg stacioniranog ogleda „Plodoredi”, koji se smatra jednim od najstarijih eksperimenata ovog tipa na prostoru jugoistočne Evrope. U ogledu se ispitivao uticaj sistema gajenja useva na sastav rezervi semena korovskih biljaka, a potom se upoređivao trоpoljni plodored (ozima pšenica-soja-kukuruz) sa monokulturom ozime pšenice. U obe varijante primenjen je isti sistem đubrenja i to 100 kg/ha N (50 kg/ha u jesen pred osnovnu obradu i 50 kg/ha u proleće u prihrani). Za uzorkovanje zemljišta primenjen je „metod kvadrata”, kao pouzdaniji u odnosu na šahovski tip i uzorkovanje po dijagonali koji se u praksi češće koriste. Procena rezerve semena korovskih biljaka urađena je primenom dva metodološka postupka: fizička ekstrakcija semena i naklijavanje zemljišnih uzoraka. Fizička ekstrakcija, uprkos dugotrajnosti i fizičkoj iscrpnosti, pokazala je bolje rezultate i dala uvid u celokupnu rezervu semena, pa je u ukupnom zemljišnom profilu (0-40 cm) u monokulturi zabeleženo 12 korovskih vrsta, odnosno 21.575 semena/m2, dok je u plodoredu izdvojeno 25 korovskih vrsta i 16.300 semena/m2. Metoda naklijavanja, iako jednostavnija za izvođenje dala je drugačiju sliku potencijalne zakorovljenosti parcela, pa je u monokulturi pšenice zabeleženo samo 5 korovskih vrsta i 8.500 semena/m2, a u plodoredu 6 vrsta i 4.500 semena/m2. Dobijeni rezultati ukazuju da nije cela rezerva semena u zemljištu aktivna, odnosno da neka semena nisu sposobna da klijaju, dok su druga u stanju mirovanja. Uvid u brojnost semena i njegovu vertikalnu distribuciju u zemljištu pomogao je oko izbora i pravljenja modela. Iako se modeliranje najčešće radi za pojedinačne vrste kroz jednogodišnja ispitivanja, izabran je „Artificial Neural Network” model (ANN model) koji je pokazao pouzdanost kod dugoročnih ekoloških istraživanja. Model je uzeo u obzir bazu podataka procenjene brojnosti semena u zemljištu i pokazao se kao veoma pouzdan. Naime, sa stepenom sigurnosti od 93% simulirao je zakorovljenost na parcelama i predvideo pojavu potencijalnih korovskih vrsta koje bi mogle predstavljati problem u narednom period

    Which methods are the most reliable for predict weed seed bank?

    The primary aim of the study was to estimate the weed seed bank in different management systems. Research on this topic is modestly and there are great variability in the results with similarly set experimental conditions, because of using different methods. Therefore, it was decided to use square method and to compare two methods for estimating weed seed bank: physical extraction of seed and seedling emergence method. The research was performed at the stationary long-term experiment "Plodoredi" of the Institute of Field and Vegetables Crops in Novi Sad. During three years and 6 assessment and 2-year crop rotation (winter wheat-maize) with standard application of mineral fertilizer 100 kg ha-1 N was monitored. During the first year (2014), method physical extraction of seed gave an insight that 20.100 seeds per m- 2, which belong to 18 weed species, persist in the entire examined soil layer (0-40 cm). Using the seedling emergence method, it was estimated only 4.625 seeds per m-2, which originate from 5 weed species. During the last assessment (2017), the physical extraction, although more complicated, indicated that there were 27.075 seeds per m- 2 in the examined layer, i.e. 20 weed species. Also, rhizomes Convolvulus arvensis and Sorghum halepense were observed. The second method for determine weed seed bank gave a different picture of potential weeding, and only 6 weed species and 3.000 seeds per m-2 were recorded. This indicates that the entire weed seed bank isn’t active, i.e. that some seeds are not able to germinate, others are dormant, but they present a potential danger. The estimated number of seeds per m-2 by physical extraction as more efficient method was used in further research for the create a „Artificial Neural Network’’ model for predicting the dynamics of weed emergence

    Production of fructooligosaccharides by Aspergillus welwitschiae inulinase enzyme complex, obtained on natural substrate

    Fructooligosaccharides (FOS) are used in the human diet thanks to their prebiotic effect and they are generally regarded as safe [1]. The increase in the functional food that contains prebiotics, has been tremendous over the last two decades. The most suitable process for FOS synthesis is by enzymes [2], particularly the one produced by filamentous fungi from the genus Aspergillus [2,3]. Aspergillus welwitschiae FAW1 strain, isolated from the environment, has proven to be non-toxigenic and safe for use in food production. Moreover, the absence of ochratoxins and fumonisins production capability was molecularly confirmed by the absence of complete or critical parts of biosynthetic gene clusters. Growing fungi on the natural substrate, triticale (Triticosecale sp), led to the production of various enzymes from inulinase complex (InuA, InuE, FTase, FFase). Production of FOS has been tested in two possible ways with the obtained inulinase enzyme complex: (1) synthesis from sucrose – fructosyltransferase activity and (2) hydrolysis of inulin – endoinulinase and exoinulinase activity. The obtained FOS were detected by TLC and HPLC methods and characterized by examination of antioxidant capacity. Produced FOS showed significant antioxidant potential according to ABTS and ORAC which classifies them as important additives in functional food. These findings open up opportunities for an easy approach for FOS production by fungal inulinase enzymes, without their prior separation and purification

    Mogućnosti primene hlađenja u zaštiti uskladištenog žita od štetnih insekata

    Gubici prouzrokovani aktivnošću insekata u skladištima na godišnjem nivou iznose oko 10% u razvijenim zemljama, dok u zemljama u razvoju mogu da budu i preko 40%. Globalna potreba za hranom bez ostataka pesticida kao i promena osetljivosti skladišnih insekata na insekticide utiče na promene dosadašnjih programa zaštite uskladištenih proizvoda. Sa tog aspekta, hlađenje ima niz prednosti zbog kojih se izdvaja kao jedna od prihvatljivih mera suzbijanja. Štetni insekti koji su prilagođeni životu u skladištu, oko 250 vrsta, su uglavnom tropskog i suptropskog porekla. Optimalne temperature za razvoj i razviće većine ovih insekata, u okviru kojih se ceo životni ciklus (jaje-imago) završava u najkraćem periodu, kreću se između 25 i 33⁰C, dok su minimalne temperature u rasponu od 13-24⁰C, a maksimalne od 33-37⁰C. Imajući u vidu da skladišni insekti na temperaturama od 5-10⁰C prestaju da se hrane i razmnožavaju, ove temperature se smatraju letalnim i mogu da se primenjuju u cilju sprečavanja i/ili usporavanja razvoja skladišnih insekata, s tim da je neophodna duža ekspozicija (u danima i nedeljama). Efektivnost primene hladnog vazduha (niskih temperatura, 0-10⁰C), zavisi od više faktora, kao što su: vrsta i stadijum razvića, način primene, dužina izlaganja ili konstrukcija skladišta. Takođe, aklimatizacija skladišnih insekata može značajno da utiče na efekte primene hladnog vazduha, a uskladištena zrnena masa ima nisku toplotnu provodljivost zbog čega dolazi do spore promene temperature kada se primenjuje tretman hladnim vazduhom. Polako hlađenje zrnene mase prouzrokuje prelazak insekata u toplije delove, što u zavisnosti od vremena potrebnog za postizanje željene temperature u svim delovima skladišta, može dovesti do aklimatizacije. Istraživanja pokazuju da insekti mogu da se aklimatizuju već na 15⁰C i da budu dva do deset puta tolerantniji na hladan vazduh temperature od 0-10⁰C od neaklimatizovanih jedinki. Brojna saznanja o efektima niskih temperatura na skladišne insekte pokazuju da je ova oblast istraživanja vrlo aktuelna, a nove informacije o hlađenju značajno doprinose daljem unapređivanju programa zaštite uskladištenog žita i drugih biljnih proizvoda od štetnih insekata

    Molecular variability and host distribution of 'Candidatus Phytoplasma solani' strains from different geographic origins

    The knowledge of phytoplasma genetic variability is a tool to study their epidemiology and to implement an effective monitoring and management of their associated diseases. 'Candidatus Phytoplasma solani' is associated with "bois noir" disease in grapevines, and yellowing and decline symptoms in many plant species, causing serious damages during the epidemic outbreaks. The epidemiology of the diseases associated with this phytoplasma is complex and related to numerous factors, such as interactions of the host plant and insect vectors and spreading through infected plant propagation material. The genetic variability of 'Ca. P. solani' strains in different host species and in different geographic areas during the last two decades was studied by RFLP analyses coupled with sequencing on vmp1, stamp, and tuf genes. A total of 119 strains were examined, 25 molecular variants were identified, and the variability of the studied genes was linked to both geographic distribution and year of infection. The crucial question in 'Ca. P. solani' epidemiology is to trace back the epidemic cycle of the infections. This study presents some relevant features about differential strain distribution useful for disease monitoring and forecasting, illustrating and comparing the phytoplasma molecular variants identified in various regions, host species, and time periods
