審査結果の要旨 
地球で最強の海流の一つである黒潮は低緯度海域から中緯度海域まで流れるが、熱以外に、
植物プランクトンの成長に必要な栄養塩も日本周辺海域に運んでくる。しかしながら、その
栄養塩の大部分は光の届く表層（有光層）ではなくて、光の届かない中層と深層にある。一
方、黒潮流域は魚類の良い産卵場と生育場として知られている。そこで、栄養塩濃度と植物
プランクトン濃度の低い黒潮流域でなぜ高い魚類生産が存在するのかという黒潮パラドック
スが提出され、その解釈を求める研究が様々の角度で展開されている。
解釈の一つとして黒潮と地形の相互作用で生じる栄養塩の鉛直輸送が挙げられる。なぜな
ら、黒潮が島と海山を通過する際に、その下流側に渦が形成され、その渦が剥離する現象が
衛星データからすでに報告されているからである。そこで、近年、現場観測、衛星データ解
析、数値モデルなどの手段を駆使して、この現象が海洋生態系に与える影響が調べてられて
いる。
Gao Jie 氏は、本学位論文において、数値モデルを用い黒潮が島を通過する際に有光層に供
給される栄養塩の量とそれに対する植物プランクトンの応答を調べた。続いて、島ではなく
海山を黒潮が通過する際に有光層へ供給される栄養塩の量を調べた。最後に、島と海山が多
数存在するトカラ海峡を対象として、島と海山を無くした仮想地形を用いた計算と実地形を
用いた計算を行い、黒潮流域における島と海山の効果を評価した。それぞれの部分について、
詳細は以下の通りである。
（１）島の効果
島の効果について、水深方向に流速が変化しない順圧流を用いた先行研究があるが、Gao
Jie 氏は黒潮の流動構造により近い傾圧流（表層で 1m/s、水深の増加に伴い徐々に弱くなる）
を用いて数値モデル実験を行った。標準ケースでは島を半径 10 ㎞の円柱で代表し、黒潮の
中心が島を通過するように設定した。使用した生態系モデルでは、栄養塩以外に、4 種類の
植物プランクトンと 4 種類の動物プランクトンを計算対象とした。 
計算結果では、島の下流側の表層に栄養塩と植物プランクトンの濃度増加が見られた。

また、4 種類の植物プランクトンの中でピコプランクトンの増殖が一番多いかった。さらに、
生物過程を無くしたトレーサー計算の結果と比較すると、黒潮流域の表層で増加した植物プ
ランクトンの５割は、湧昇と鉛直混合によって亜表層から表層へ輸送されたもので説明でき
ることが分かった。逆に言えば、亜表層から供給された栄養塩を利用して光合成により増加
した植物プランクトンは残りの 5 割であることが分かる。そして、モデル結果から動物プラ
ンクトンの増殖は島の下流側では顕著ではなく、さらに遠い（>300 ㎞）場所で徐々に顕著
になることも分かった。 
（２）海山の効果 
順圧流が海山を通過する際に二つの流動形態を生じることは先行研究で報告されている。1
つは等深線に沿って海山の両側から回る（flow-around）形態である。もう一つは等深線を横
切って海山の頂上まで登ってから下る（flow-over）形態である。海面流速を 1m/s とし、水深
の増加に伴い流速が徐々に弱くなる現実的な黒潮を、高さの異なる海山付近を通過する計算
では黒潮がほとんど flow-around 形態を示すことが分かった。そして、有光層（<110m）への
栄養塩供給は、海山の水深に強く依存することが分かった。400m より深い海山による栄養塩
供給は有光層へ届かないが、400m より浅い海山では有光層への栄養塩供給量は海山の水深に
反比例して変化する。つまり、海山が浅ければ浅いほど、栄養塩の供給量が大きくなる。し
かしながら、同じサイズの島と海山を比較すると、海山による栄養塩供給量は島によるもの
より小さいことも分かった。島を水深ゼロの海山として例えれば、島と海山による栄養塩の
供給量の違いは容易に理解できる。 
（３）トカラ海峡の島と海山の効果 
上記の単独な島と海山に関する計算結果を現実海洋に適用させるため、トカラ海峡周辺海
域を対象とする黒潮の数値シミュレーションを行った。この計算では、島と海山を有する現
実的な地形を使用したケースと、島と海山を人為的に無くしたケースの実験を実施し、両ケ
ースの比較からトカラ海峡における島と海山がその下流域（例えば四国沖黒潮）の栄養塩濃
度を高める効果を有することを確認した。また、この結果は上記の単独な島と海山に関する
計算結果および人工衛星データに見られているトカラ周辺海域の植物プランクトン濃度のパ
ターンに整合的であった。 
 
提出された学位論文の研究成果の一部は、国際学術雑誌に 1 編の論文が公表されている。
海山とトカラ海峡における研究成果は投稿準備中である。 
 本学位論文の公聴会は令和 5 年 2 月 10 日にオンライン形式で開催し、約 45 分の英語によ
る論文発表と 25 分の質疑応答が行われた。引き続いて、学位論文審査委員会を開き、本論文
の内容を厳正に審議した結果、審査委員が全員一致で、博士（理学）の学位を授与するのに
値するものと判定した。

【背景・目的】  
レジスチンは、インスリン抵抗性を惹起するサイトカインである。ヒトでは、主に単
球、マクロファージに発現し、インスリン抵抗性、炎症、動脈硬化と関連する。  
当研究室では、レジスチン遺伝子転写調節領域の SNP-420(rs1862513)C>G の G/G 型が
2 型糖尿病感受性と関連することを見出した。機序として、SNP-420 が G の場合、Sp1/3
転写因子がその DNA 配列に特異的に結合してレジスチン遺伝子の転写活性を高め、レ
ジスチンの単球 mRNA、及び血中濃度を上昇させ、インスリン抵抗性を介して 2 型糖尿
病のリスクを高める機序が想定された。  
さらに当研究室は、一般住民において、SNP-420 の G が血中レジスチンを高めるため
には、SNP-358(rs3219175)G>A の A が必要であり、SNP-420/-358 のハプロタイプが G-A
の場合に血中レジスチンが最も高いことを報告した。  
血中レジスチンは上記の 2 つのプロモーターSNP によって強く規定されているが、環
境因子の影響は明らかでない。そこで、インスリン抵抗性及び 2 型糖尿病との関連が報
告されている喫煙に着目し、喫煙と血中レジスチンとの関連、及びその関連に G-A ハプ
ロタイプが及す影響を解析した。  
 
【方法】  
東温ゲノムスタディは、東温市の一般住民を対象とした観察型疫学研究である。健診
受診者 2032 人のうち、SNP-420 と -358 の両遺伝子型を決定し得た 1975 人について横断
的に解析した。喫煙の有無と本数は、自記式質問票を用いて調査した。血中レジスチン
は ELISA 法で定量した。SNP は TaqMan 法、及びパイロシークエンス法で解析した。レ
ジスチン mRNA は、全血細胞から RNA を抽出し、リアルタイム PCR により定量した。
統計解析には JMP13.2 を使用した。  
この研究は愛媛大学臨床研究審査委員会によって承認されている。

【結果】  
まず、喫煙状況（never、 former、current）により血中レジスチンを比較した。その結
果、血中レジスチンは、current smokers で最も高かった（ANCOVA P＜0.0001）。さら
に、1 日の喫煙本数を中央値（15 本）で分け、非喫煙者  (never＋ former)、喫煙本数 1～
15 本、16 本以上の 3 群で血中レジスチンを比較した。喫煙本数が多いほど、血中レジス
チンはより高かった（P for trend＜0.0001）。次に、G-A ハプロタイプをホモで有する群
（G-A/G-A）、ヘテロで有する群（G-A/*）、有さない群（*/*）に分けて血中レジスチ
ンと喫煙との関連を解析した。全ての群において、血中レジスチンは喫煙者で高かっ
た。喫煙による血中レジスチンの上昇幅は、*/*＜G-A/*＜G-A/G-A の順に大きく、交互
作用を認めた（ two-way ANCOVA; haplotype: P＜0.0001、smoking: P＝0.0003、交互作用 : 
P＜0.0001）。最後に、G-A/G-A の喫煙者 7 名に、年齢、性、BMI をマッチさせた G-
A/G-A の非喫煙者、C-G/C-G の喫煙者、非喫煙者の各 7 名ずつについて、全血細胞にお
けるレジスチン mRNA を比較した。G-A/G-A の喫煙者は、非喫煙者に比し、mRNA は
1.4 倍高かった（Student’s t-test, P＝0.022）。C-G/C-G では、喫煙と mRNA は関連しなか
った。  
  
【考察】  
レジスチン SNP-420/-358 の G-A ハプロタイプと喫煙は、相互に血中レジスチンを高め
た。SNP-420 が G の場合、転写因子 Sp1/3 がその DNA 配列を特異的に認識する。別の遺
伝子において、喫煙やニコチンの刺激で、Sp1 配列への核タンパクの結合が増強される
ことが報告がされている。したがって、喫煙が Sp1 の結合を増強することによりレジス
チン遺伝子転写活性を高める可能性が想定される。また、SNP-358 が G の場合、転写抑
制因子である NFκB p50/p50 がその DNA 配列に結合し、レジスチン転写活性を抑制する
可能性が報告されている。G-G ハプロタイプの場合、SNP-358 の G の配列に抑制因子が
結合することにより、喫煙と SNP-420 の G によるレジスチン転写活性の上昇が低く抑え
られる可能性がある。すなわち、喫煙が血中レジスチンを高めるためには、SNP-420 が
G の配列に Sp1 が結合し、SNP-358 が A の配列に抑制因子が結合しないことが必要と考
えられる。これは、G-A ハプロタイプをホモで有する場合に、喫煙と血中レジスチンが
最も強く関連するという遺伝子・環境因子相互作用の機序を説明しうる。レジスチン
SNP-420/-358 を標的とした高精度医療として、遺伝的リスクの高い G-A ハプロタイプを
ホモで有する場合に、禁煙による 2 型糖尿病や心血管疾患の発症や進展予防が期待され
る。  
 
【結論】  
レジスチン SNP-420/-358 の G-A ハプロタイプと喫煙は、ともに血中レジスチンを高
め、遺伝子・環境因子相互作用を認めた。

ANJUM FERDOUS ONA は 2019 年 4 月に入学、2022 年 3 月に単位取得退学、2022 年 4 月から現在まで特
定研究員として在籍し、上記のテーマで研究を行った。
温帯生地生シンビジウムのシュンラン（Cymbidium goeringii）、スルガラン（Cymbidium encifolium）、カ
ンラン（Cymbidium kanran）‘室戸錦’および熱帯生着生シンビジウムの品種を用いて in vitro での器官形
成に影響を与える様々な環境因子を調査した。その結果、以下の知見が得られた。
シンビジウム属植物の根茎培養における器官形成に及ぼす、修正 MS 培地中の様々な植物成長調節物質
（PGR）およびエリシターの影響について検討した。その結果、根茎切片に培養において、これまでベン
ジルアデニン（BAP）添加では難しかったシュート形成の誘導が、カンランおよびシュンランはアデニン
硫酸塩を培地に添加することで可能であることを見出した。チジアズロン(TDZ)を培地に添加したところ、
両シンビジウム種ともプロトコーム様シュート(PLS)が発生し、器官形成が抑制された。また、これまで低
分子のヒアルロン酸ナトリウム（HA9 資生堂製）が器官形成を促進することは分かっていたが、さらに低
分子のマイクロヒアルロン酸ナトリウム(FCHおよび FCH-SU; キッコーマンバイオケミファ製)の作用につ
いて検討し、これらのマイクロヒアルロン酸ナトリウムは、同様に低濃度で培地に添加することでシュン
ラン根茎のシュート形成を誘導できることも確認した。
次に、シンビジウムの器官形成に及ぼす光質の影響について検討した。カンランの根茎および Cymbidium
Sweet ‘Waffle’のプロトコーム様球体（PLB）のシュート形成は赤色 LED 照明で促進され、Cymbidium 
Sweet ‘Waffle’のプロトコーム様球体（PLB）の増殖は緑色 LED 照明が促進的に作用することを明らかに
した。また、緑色 LED と赤色 LED を交互に照射すると、Cymbidium Sweet ‘Waffle’の PLB 増殖、シュー
ト形成、新鮮重が促進された。シンビジウムの組織培養における緑色光と赤色光の重要性について確認し
た。 
続いて、種々のランプの照明下、培地に添加した塩化アルミニウムおが PLB および根茎の器官形成に及
ぼす影響について検討した。塩化アルミニウム 1.0 mg/l 添加区では、Cymbidium Sweet ‘Waffle’の PLB
の増殖は緑色 LED 照明下促進され、赤色 LED 照明下でシュート形成およびシュートからの発根が促進され

た。また、カンランの根茎では塩化アルミニウム 1 mg/l 添加区でシュート形成およびシュートの発根が促
進された。 
養蜂業において需要が高いキンリョウヘンは培養組織による増殖中の PLB の褐変が問題である。そこで本
試験ではキンリョウヘンの PLB を供試し、酸化防止剤であるクエン酸添加の影響を検討した。キンリョウ
ヘンの PLB の褐変はクエン酸を 0.1 g/l の濃度で培地に添加することで著しく抑制できることを明らかに
した。また、PLB からのシュート形成および発根も有意に促進されることを確認した。 
以上は、シンビジウムの組織培養における器官形成制御方法を培地の添加物質および光条件を中心
に検討した貴重な知見である。 
 本論文に関する公開審査会は、令和 5 年 8 月 5 日に香川大学農学部で開催され、論文発表と質疑
応答が行われた。引き続いて行われた学位論文審査委員会で、本論文の内容を慎重に審議した結果、
審査委員全員一致して博士（農学）の学位を授与するものと判定した。

Soil is one of the most important and widely used building materials in the world. It 
has been effectively used for a variety of purposes, such as building social infrastructures, 
growing agricultural products, and performing several other important activities that can be 
useful for human life. The nature of soil varies from place to place due to its physical, 
chemical, and mechanical properties. Expansive soil is one of the different types of soils 
known as geotechnically problematic soils, and it represents a significant challenge for both 
civil engineering and geotechnical applications. Its fundamental problem is mainly related to 
the variation of moisture content, and it has shrinkage and swelling behavior during dry and 
wet seasons. It causes enormous damage to the infrastructure such as buildings, roads, 
pipelines, and other facilities that are built on the expansive soil. Figure 1 shows that the 
cobbled pavement failed due to the failure of the subgrade.  
 
 
 
 
 
 
 
 
 
This study investigated the existing and new mechanisms to mitigate the problem of

has been an important technique used for many years. The purpose of soil stabilization is to 
improve bearing capacity, reduce permeability, reduce compressibility, and improve the soil's 
overall physical and mechanical performance. Despite environmental concerns and cost issues, 
traditional soil stabilizers such as cement, lime, fly ash, bitumen, etc. have been effectively 
used to improve the engineering properties of soil. Nowadays, using sustainable and 
environmentally friendly materials for soil stabilization is attracting a lot of attention 
worldwide. The use of agricultural waste resources for stabilizing problematic soils is one of 
the areas of interest for geotechnical engineers, researchers, and practitioners. Agricultural 
waste by-products, such as cellulose-based fiber additives, are sustainable, environmentally 
friendly, and locally available. They are also cost-effective materials. It is clear that not all 
agricultural by-products are suitable for soil stabilization, but there are some criteria to 
consider from selection to application as soil stabilizers.  
This study had three main objectives. First, the selection of agricultural waste 
by-products as cellulose-based fiber additives for soil stabilization was carried out. Many 
attempts have been made to select suitable cellulose-based fibers from agricultural waste 
resources. In the selection of cellulose-based additives, their strength, water absorbency, 
chemical composition, and cellulose content are considered. Therefore, three suitable 
candidates such as bamboo fiber, rice husk fiber, and wheat straw fiber were selected based on 
the test results. The result shows that bamboo fiber has high cellulose content and high-water 
absorption capacity, while wheat straw has high water absorbency with relatively low 
cellulose content. Rice husk has low water absorbency and moderate cellulose content. Based 
on these parameters, it was proved that bamboo fiber, rice husk fiber, and wheat straw fiber 
are suitable for the stabilization of expansive soils. The preparation process of the 
cellulose-based fiber additives from agricultural waste is shown in Figure 2 (a), and Figures 2 
(b) and (c) describe the water absorption test and the evaluation of the cellulose content for 
the cellulose-based fiber additives.  
 
The second objective of the study was to create a pseudo-expansive soil (PES) from a 
combination of clay soils like Kunigel V1, Kasaoka clay, and Tochi clay soils. Various types 
of geotechnical test methods were used to experimentally evaluate the replication of the 
expansive soil using the combination of the different soil types. The proportion by weight of 
KunigelV1 (21.8%), Kasaoka clay (39.1%) and Tochi clay (39.1%) was used for the 
replication of the expansive soil with the aim of achieving similar properties to the original

expansive black cotton soil. The presence of expansive soil in Japan is rare, and the possibility 
of obtaining the expected properties of expansive soil is limited.  Therefore, creating a 
pseudo-expansive soil by combining clay soils is an essential part of the study. Engineering 
properties such as Atterberg limits, free swell ratio (FSR), maximum dry density (MDD), 
optimum moisture content (OMC), and UCS values were compared with expansive soil 
properties. Recorded test results indicated a 101.2% liquid limit, 1.70 free swelling ratio, 
1.41g/cm3 MDD, 26 percent OMC, and 110kPa UCS for the pseudo-expansive soil, and a 
106.8 percent liquid limit, 1.94 free swelling ratio, 1.38g/cm3 MDD, 29.5% OMC, and 84kPa 
UCS for the original expansive black cotton soil. Figure 3 (a), (b), and (c) shows the 
comparison of some geotechnical and microstructural properties for the pseudo-expansive soil 
and the original expansive soil. 
 
Third, the pseudo-expansive soil was stabilized with cellulosic fiber additives to 
further improve the engineering properties of the soil. Bamboo fiber, rice husk fiber, and 
wheat straw fiber were used to stabilize the pseudo-expansive soil composed of kunigelV1, 
Kasaoka clay, and Tochi clay soils. This part of the study presents a comprehensive study 
aimed at optimizing the physical and strength performance of cellulose-based fiber additives, 
namely bamboo fiber (BF), rice husk fiber (RHF), and wheat straw fiber (WSF), for 
stabilizing expansive soils. Various factors of cellulose-based fibers, such as cellulose content, 
water absorbency, fiber content and size, and fiber reinforcement mechanism, were studied to 
investigate their suitability as soil stabilizers. In this experiment, fiber dosages of 5%, 10%, 
and 15% and fiber sizes of 75 µm, 150 µm, and 300 µm were used in different combinations 
to achieve the optimum condition. The experiment was designed using the Taguchi method of 
signal-to-noise ratio approach to optimize the experimental conditions in terms of the 
Atterberg limit test, free swell ratio, linear shrinkage, and unconfined compressive strength as 
the main response factors. The result indicated that the RHF fiber dosages of 5% and 15% 
with a fiber size of 300 µm, as well as the WSF fiber dosage of 15% with fiber sizes of 75 µm 
and 150 µm, significantly improved the physical performance of the stabilized soil, while the 
BF fiber dosage of 15% with a fiber size of 300 µm was found to be an effective 
cellulose-based fiber additive for achieving the desired mechanical performance. Figure 4 and 
Figure 5 show some of the improved engineering properties such as Atterberg limit and UCS 
values of the stabilized soil by using cellulose-based fiber additives.

In addition, the microscopic analysis was carried out by SEM-EDX to observe the 
fiber reinforcement interaction with the stabilized soil. The SEM image shows that bamboo 
fiber is uniformly mixed with soil and strongly attached to the stabilized soil surface. A 
similar phenomenon was observed for the soil stabilized with wheat straw fiber. In the case of 
the rice husk-stabilized soil, the rice husk was completely mixed with the soil, and an ash-like 
surface was observed. This is because the length of rice husk fiber is much smaller than 
bamboo and wheat straw fiber materials. Besides surface morphology, chemical element 
composition was also observed by SEM-EDX image analysis. The main elements identified 
for pseudo-expansive soil were C, O, Si, and Al. In each case, liquid nitrogen was used for 
EDX observation and detector cooling, hence the high amount of N detected at 0.4 keV, as 
observed in all EDX analyses. The elements C and O were observed in the fiber materials 
since C and O are the main constituents of the cellulose fibers (C6H10O5). As shown in Figure 
6, the SEM-EDX analysis for the soil stabilized with cellulosic fibers is presented in the form
