魚類エドワジエラ症は細胞内寄生細菌による感染症であるため、抗菌剤の投薬
治療が可能である一方で、慢性化による治療が難しくなることが多く、日本の海
面養殖、特にヒラメとマダイにおいて深刻な問題となっている。これまで原因菌
は両魚種ともにEdwardsiella tardaとされてきたが、Oguroら（2014）のドラフトゲ
ノム解析の成果から、ヒラメ由来株はAbaynehらにより報告されたEdwardsiella 
piscicida（Abayneh et al., 2013）、マダイ由来株はShaoらにより報告された  
Edwardsiella anguillarum（Shao et al., 2015）とそれぞれ別種であることが示され
た。本研究はこれら種間の差異に着目して、両エドワジエラ属細菌の病原機構を
明らかにし、今後の実用的なエドワジエラ症対策の確立に資する知見を得ること
を目的とした。  
 まず、両エドワジエラ属細菌を用いてヒラメ、マダイ、およびブリの3魚種に交
差感染試験を実施した。その結果、E. piscicida感染区ではヒラメのみで死亡率が
高く、マダイ由来E. anguillarum感染区ではマダイのみで死亡率が高い結果となっ
た。このことからE. piscicidaとE. anguillarumはそれぞれ特定の宿主に対して特異
的な病原性を示すことを明確にできた。  
 次に両エドワジエラ属細菌を遺伝学的、性状学的に比較して相違点を抽出した。
遺伝子に関しては次世代シーケンサーを用いて、両エドワジエラ属細菌のドラフ
トゲノム解析を行い、性状に関しては、Api50CHテストに供して炭水化物代謝の比
較を行った。その結果、両エドワジエラ属細菌間にはL-アラビノースとD-マンノ
ースの代謝に遺伝子的、性状的な違いがあることが判明し、これは両種を明確に
分けるための有効な判別マーカーになることを提唱した。  
 両エドワジエラ属細菌の性状の差異を検証する上で、既成培地では栄養成分を
調整することが不可能となる。そのため、新規な合成培地を開発して成功した。
この培地はエドワジエラ属細菌の細菌学的研究の今後の基盤となるとともに、ワ
クチン製造での培地ロット差のリスクを減らせることや、コーシャやハラルとい
った宗教問題にもきちんと対応できるものである。またこの開発の中で、両エド
ワジエラ属細菌間にはアミノ酸要求性に違いが見つかり、E. piscicidaはTyrとAla
を、E. anguillarumはThrと Ileをそれぞれ多く要求した。さらに、宿主側の血清中の
アミノ酸含量はヒラメ血清でTyrとAla、マダイ血清でThrと Ileがそれぞれ多く、両
エドワジエラ属細菌の要求するアミノ酸と宿主血清含有のアミノ酸が一致する結
果となった。このことは、両エドワジエラ属細菌が宿主の体内へと侵入した後、
速やかな増殖に必要な栄養素が身近に存在する場合においてのみ感染成立すると
いった病原性機構の仮説を立てた。  
 以上より、本研究において実施した両エドワジエラ属細菌の宿主特異性の証明、
菌種の判別法確立、合成培地の開発、新たな切り口からの病原機構の提唱を通じ

て、将来のエドワジエラ症対策の確立に向けた取り組みが大きく前進するものと
期待される。

Co n ve n t i on a l  wee d  ma na g em e nt  me t ho d s  o f t e n  r e l y  on  t h e  a pp l i c a t i o n  o f   
synthetic herbicides, which can have adverse environmental and  health impacts.   
As  a  r e s u l t ,  t h e r e  i s  a  p r e s s i n g  n e ed  f o r  a l t e r n a t i v e  a p p r o a c he s  t h a t  a r e   
sus t a ina b l e ,  eco nomica l ly  v i ab l e ,  and  ec o log i ca l l y  ben ign .  F or  sus t a ina b l e   
agriculture,  exploit ing allelochemicals  from dif ferent  groups of plants  to treat   
noxious weeds is increasing. Therefore, this research focused on the evaluation  
of the allelopathic potential of Stephania japonica  (Thunb.), Piper longum  (Linn),
Annona ret iculata  (Linn) ,  Trewia  nudi f lora  (Linn) ,  and Glinus  opposi t i fol ius   
(Linn) extracts against  the growth of  three dicot and three monocot test  plant   
species and the identification of active allelopathic substances from the extracts . 
Aqueous methanol extracts of these plant s showed significant growth inhibitory  
e f fec t s  on  the  seedl ings '  growth  of  a l fa l fa ,  c re ss ,  l e t tuce ,  I t a l i an  ryegrass ,   
barnyard grass, timothy, and foxtail fescue. The inhibition was dependent on the 
concentration and the treated plant species.  
The I50  values (concentration required for 50% shoot and root growth inhibition) 
of S. japonica , P. longum , A. reticulata, T. nudiflora , and G. oppositifolius  were
0.003–0.045,  0 .003–0.070,  0.003–0.057,  0 .0003–0.040,  and 0 .002–0.082 g DW 
equivalent  extracts  per  mL,  respectively.  Based on these values ,  T. nudi f lora   
showed the strongest  inhibitory activity, followed by  A. reticulata ,  S.  japonica ,   
P. longum , and  G. oppositifolius, despite the differences in susceptibility of the  
t es t  p lants  to  the  d i f fe ren t  ex t rac t s .  Thus ,  these  resul t s  indica te  the  potent   
al lelopathic activity of  the five plant  species,  which also implies the presence  
of allelopathic substances in the extracts.  
Aqueous methanol extracts of A. reticulata ,  T. nudiflora ,  and G. oppositifol ius   
were purified through a series of bioassay-guided chromatographic steps to isolate
t h e  g r o wt h  i n h i b i t o r y  s u b s t a n c e s .  T h e  c h e m i c a l  s t r u c t u r e  o f  t h e  i s o l a t e d

substances was identified by HRESIMS analysis . The 1H NMR spectroscopic data
was collected using an optical rotation of 500 MHz. Purification of A. reticulata 
leaf  extract  resul ted  in the isolat ion of  three act ive al le lopathic  substances :   
loliolide, 5-hydroxy-3,4-dimethyl-5-pentylfuran-2(5H)-one, and 3,4-dihydroxyphenyl
ethanol. Six active substances, loliolide, 6,7,8-trimethoxycoumarin, methyl gallate,
pinoresinol, protocatechuic acid, and gallic acid were isolated from the leaf  
extracts of T. nudiflora .  
The bioactivity of the seven identified compounds was determined against the growth of 
cress, timothy (for loliolide, 5-hydroxy-3,4-dimethyl-5-pentylfuran-2(5H)-one, and 
3,4-dihydroxyphenylethanol), lettuce (for loliolide, 6,7,8-trimethoxycoumarin), and 
alfalfa and barnyard grass (for methyl gallate and pinoresinol) , and cress and Italian 
ryegrass (for protocatechuic acid and gallic acid) . The growth-inhibitory activity of the 
compounds increased with the increase in their concentrations. The co ncentrations 
required for 50% cress and timothy growth inhibition ranged from 0.015–0.060, 
0.013–0.120, and 0.028–0.100 mM for loliolide, 
5-hydroxy-3,4-dimethyl-5-pentylfuran-2(5H)-one, and 3,4-dihydroxyphenylethanol. On 
the other hand, lettuce shoot and root growth were decreased by 50% with 0.043  and 
0.128 mM of loliolide, respectively, which were 1.5 and 4 times higher t han 
6,7,8-trimethoxycoumarin, indicating that the lettuce was more sens itive to 
6,7,8-trimethoxycoumarin than loliolide. The I50 values of methyl gallate ranged 
between 0.137–2.051 mM for alfalfa and barnyard grass and 0.065–0.216 mM for 
pinoresinol, and the results inferred that pinoresinol exhibited stronger inhibition than 
methyl gallate for both alfalfa and barnyard grass.  The 50% cress and Italian ryegrass 
growth was decreased by 2.07–4.50 mM of gallic acid and 1.50–10.18 mM of 
protocatechuic acid, indicating that the treated plants were more susceptible to gallic 
acid than protocatechuic acid. The root growth of cress and Italian ryegrass was 7, 1, 
and 2 times higher than the shoot growth by protocatechuic and gallic acid, except for 
the Italian ryegrass root  by gallic acid. 
 
In conclusion, aqueous methanol extracts of five plant s significantly inhibited the 
growth of the corresponding test plants, indica ting their potential allelopathic activity, 
which may be due to the allelopathic compounds in the extracts. Nine allelopathic 
compounds were isolated from A. reticulata  and T. nudiflora leaf extracts. The growth 
inhibitory activity of these compounds may be responsible for the allelopathy of A. 
reticulata and T. nudiflora . Therefore, the incorporation of the five plant extracts as 
well as the identified allelopathic compounds (lol iolide, 
5-hydroxy-3,4-dimethyl-5-pentylfuran-2(5H)-one, 3,4-dihydroxyphenylethanol, 
6,7,8-trimethoxycoumarin methyl gallate, pinoresinol, protocatechuic acid and gallic 
acid) in weed control has the potential to decrease the reliance on synthetic herbicides 
by promoting the production of bioherbicides.
