【目的】   
「糖尿病が強く疑われる人」や「糖尿病の可能性を否定できない人」が急増し
ている今日，糖尿病予防の一助となる食品や食品成分の研究が精力的に行われて
いる。この代表的なものに，α -グルコシダーゼ活性阻害作用で糖の吸収を抑制す
るグァバ葉抽出液，イチョウ葉由来フラボノイドフラクション，桑葉エキスなど
が知られている。一方，核酸あるいは核酸関連物質もα -グルコシダーゼ活性阻害
による血糖値上昇抑制が報告されている。  
そこで，無農薬玄米を，核酸を大量に生産する特殊麹菌で発酵させた後に粉砕・
粉末化した玄米発酵食品（Fermented Brown Rice：FBR）にも血糖上昇抑制効果
があるのではないかと考え，ラットを用いて検討した。  
 
【方法および結果】  
実験 -1：  4週間にわたって，ラットにFBRあるいは玄米粉末（NBR）をそれぞれ2.
5％添加した 20％カゼイン食を摂取させ，空腹時血糖値，インスリン値，HOMA-
R値，グリコアルブミン濃度を比較して検討した。その結果，空腹時血糖値，イン
スリン値，HOMA-R値，グリコアルブミン濃度は，FBR群に比べて対照である20％
カゼイン食群およびNBR群に比べて有意な低値を示した。  
 
実験 -2：ラットでの経口でんぷん液負荷試験の結果，投与後の血糖値およびイン
スリン値の変化は，でんぷん液にNBRを添加した群（BRC群）に比べて，でんぷ
ん液にFBRを添加した群（FBR群）で有意に低かった。また，血糖値，インスリ
ン値の2時間の合計時間曲線下面積（ACU）もBRC群に比べてFBR群で有意に低か
ったが，でんぷん液にFBRと同等レベルの核酸（DNA・RNA）を添加した群（BR
N群）とFBR群に差はなかった。  
 
実験 -3：FBRのヒト唾液アミラーゼ活性に対する見かけ上の阻害作用を調べた。
まず，可溶性でんぷんにFBR，NBR（対照），あるいはNBRと核酸（DNA・RNA）
を混合した基質に，酵素液として希釈ヒト唾液を30分間反応させた後の残存デン
プン量を比較した。その結果，残存デンプン量は，NBR混合を基質とした場合に
比べて，FBR混合およびNBR＋核酸混合を基質とした場合で有意に高値であった
が，FBR混合とNBR＋核酸混合の間に差はなかった。つぎに，アミラーゼ活性を
比較した結果，対照とNBR添加酵素に比べて，FBR添加酵素と核酸添加酵素で有
意に低く，Michaelis定数は有意に高かった。また，Lineweaver-Burkプロットの結
果，これらの阻害様式は拮抗阻害と推定された。FBRおよび核酸のでんぷん消化
に対する影響は，アカルボースやグァバ葉抽出茶と類似していた。

実験 -4： FBR摂取の血糖上昇抑制効果を，Ⅱ型糖尿病モデルのZucker Fattyラット
（ZDF）と対照のZucker leanラットで検討した。ZDFラットにNBRあるいはFBR
添加食を摂取させた群をそれぞれDMC群（糖尿病対照）およびDMF群，Zucker l
eanラットにNBR添加食を摂取させた群をNDM群（正常対照）とした。3週間の飼
育後，2g/kg BWのグルコースで経口糖負荷試験を行った。その結果，NDM群のH
OMA-R値に比べてDMF群とDMC群で有意な高値を示した。血糖値変動について，
DMC群では投与後60分がピークとなったが，DMF群及びNDM群では投与後30分が
ピークだった。また，DMF群の血糖値低下はDMC群に比べて速やかであり，糖負
荷後 60， 90および120分におけるDMF群の血糖値は，DMC群に比べて有意に低か
った。血糖値およびインスリン値のACUは，NDM群に比べてDMF群で有意に高か
ったが，DMC群に比べて有意に低かった。  
 
実験 -5：FBR摂取の血糖上昇抑制効果を，ストレプトゾトシン誘導Ⅰ型糖尿病モ
デルラットで検討した。糖尿病ラットにNBRあるいはFBR添加食を摂取させた群
をそれぞれDMC群（糖尿病対照）およびDMF群，正常ラットにNBR添加食を摂取
させた群をNDM群（正常対照）とした。４週間の飼育後，2g/kg BWのグルコース
で経口糖負荷試験を行った。その結果，空腹時血糖値は，NDM群に比べてDMF群
で有意に高く，DMC群に比べてDMF群でインスリン分泌不全であるにも関わらず
有意に低かった。また，血糖値上昇もDMC群に比べてDMF群で有意に低かった。
さらに，肝・筋グリコーゲン量は，いずれもDMC群に比べてDMF群で有意な高値
を示し，DMF群とNDM群で差はなかった。  
 
【結論】  
 以上の結果から，FBR摂取によって，食後血糖値の上昇抑制ならびにインスリ
ン節約効果が期待できること，その要因として，FBRに含まれる核酸による糖類
分解酵素の活性阻害作用だけでなく，インスリン作用を介さないグリコーゲン合
成促進作用が関与している可能性が示唆された。

Pseudomonas cichorii is a multi-host plant bacterium. P. cichorii strain SPC9018 
(SPC9018) causes rot symptoms in lettuce, though the bacterium does not produce 
pectate lyase, which is the most important plant cell wall-degrading enzyme. The 
development of disease symptoms is closely associated with programmed cell death 
(PCD) following heterochromatin aggregation and laddering of genomic DNA in 
SPC9018-infected lettuce cells. SPC9018 also causes necrotic spots on eggplant distinct 
from the disease symptoms on lettuce. Development of necrotic spot symptoms is 
observed following PCD in eggplant leaves. SPC9018 harbors the hrp genes encoding 
proteins in the type III secretion system. The hrp genes are essential for virulence of 
this bacterium in eggplant but not lettuce, indicating that the hrp genes are implicated in 
the diversity of SPC9018 virulence. The aldH encoding aldehyde dehydrogenase and pat 
encoding N-acetyltransferase are located in the flanking region of the hrp genes. The 
objective of this study is to analyze its virulence diversity on multi-host plants.  
Phylogenetic study of the hrp genes and pat based on the nucleotide sequences 
showed that a pathogenicity island of P. cichorii consists of these genes. The aldH was 
though to conserve among genome of pseudomonad. The aldH-deleted mutant and 
pat-deleted mutant lost their virulence on eggplant but not lettuce. Inoculation into 
Asteraceae species susceptible to SPC9018 showed that the involvement of hrp, pat and 
aldH in SPC9018 virulence is independent of each other and has no relationship with the 
phylogenetic diversity of Asteraceae species based on the nucleotide sequences of ndhF 
and rbcL. These results suggest that not only the hrp genes but also aldH and pat are

implicated in the diversity of SPC9018 virulence on susceptible host plant species. 
Therefore, virulence diversity of P. cichorii might be established after species 
diversification of Asteraceae plants and be responsible for the virulence of respective 
species. 
From the present results, it was thought that SPC9018 has several virulence 
mechanisms. I then analyzed the influence of limited iron acquisition on SPC9018 virulence. High 
performance liquid chromatography and liquid chromatography-mass spectrometry analyses showed 
that SPC9018 produces pyoverdine. Spectrophotometric assay showed that the total concentration 
of pyoverdine is negatively correlated with the bacterial density. The in vitro growth of SPC9018 with 
mugineic acid (MA), a phytosiderophore, was lower than that without MA. When FeCl3 was included 
in the medium, the in vitro growth of SPC9018 in the presence of MA was complemented. Application 
of MA reduced SPC9018 virulence on 15 host species including eggplant, but did not influence 
SPC9018 virulence on five host species including lettuce. Furthermore, MA application led to loss in 
SPC9018 virulence on eight host species. The population of SPC9018 in the intercellular spaces of 
eggplant leaves with MA was significantly lower than that without MA. MA application enhanced 
expression of pyoverdine production-related pvd genes in SPC9018. Furthermore, MA application 
reduced adhesion activity of SPC9018. These results suggests that limited iron acquisition 
plays specific role in adhesion of P. cichorii to the intercellular spaces and its growth in 
the intercellular spaces, involved in its virulence on respective host plants. 
Interestingly, the pat-deleted mutant showed virulence on tested plant species, at 
levels similar to those of SPC9018 applied with MA. The deletion of pat resulted in decreased 
production of pyoverdine at bacterial densities of less than 1.0 × 108 cfu/ml, leading to decreased 
siderophore activity and iron acquisition. Furthermore, at a bacterial density of 2.0  107 cfu/ml, the 
pat deletion enhanced expression of pvdL and pvdR to a similar degree as SPC9018 treated with MA. 
The pat is therefore involved in not only pyoverdine-mediated iron acquisition, 
implicated in SPC9018 virulence on respective host plants. 
In conclusion, SPC9018 implicate not only the hrp genes but also aldH and pat in its 
virulence on respective host species. Iron acquisition by SPC9018, in which pat is 
involved, plays an important role in adhesion ability of this bacterium in the 
intercellular spaces of infected host plants immediately after invasion, establishing its 
virulence diversity on the respective host plants.

    Prosaposin, a 66-kDa glycoprotein, is the precursor of saposins A, B, C, and D, which 
act as cofactors in the hydrolysis of sphingolipids by lysosomal hydrolases. In lysosomes, 
PS is proteolytically processed to generate four sphingolipid activator proteins, known as 
saposins A to D, which are required for hydrolysis of sphingolipids by several lysosomal 
exohydrolases. Many functions have been attributed to secreted PS, which is reportedly a 
trophic factor in the nervous and reproductive systems, being present in milk and 
cerebrospinal and seminal fluids. The PS gene contains 15 exons. It is transcribed into 
several mRNAs, resulting from alternative splicing of the 9-bp exon 8. In situ hybridization 
has shown abundant PS expression in the epithelial cells of the choroid plexus and various 
grey matter areas, including the cortex and hippocampus. Besides its role as the precursor 
protein of saposins, PS is also a neurotrophic factor capable of inducing neural 
differentiation and preventing cell death. A neurotrophic sequence has been identified in 14 
amino acids located in the N-terminal part of saposin C and has been attributed to PS 
neurotrophic activity.  
In order to research the function of PS in central nerve system, we choose two models 
of mice, one is Parkinson disease mice, and the other is mdx mice.  
Parkinson’s disease (PD) is a progressive neurodegenerative disorder with cardinal 
clinical features of bradykinesia (slowness), postural instability (imbalance), rigidity, 
resting tremor, autonomic dysfunction, and psychiatric manifestations. The principal 
neuropathologic feature of PD is selective degeneration of midbrain dopamine-producing 
neurons of the substantia nigra (SN) parscompacta (SNpc) that make up the nigrostriatal 
pathway, which results in depletion of dopamine in the caudate nucleus and putamen 
(collectively termed the striatum). To explore the mechanisms responsible for the effects of 
prosaposin on neuronal survival in PD, we treated dopaminergic neurons with PS18, which 
comprises the hydrophilic sequence of rat saposin C. It was found to resist protease 
cleavage and cross the blood–brain barrier intact when administered systemically. In the 
present study, we used 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) or 
1-methyl-4-phenylpyridinium ion (MPP+)-induced dopaminergic neurotoxicity in 
C57BL/6J mice or SH-SY5Y cells and explored the protective effect and mechanisms of 
PS18 on dopaminergic neurons. Treatment with 2.0 mg/kg prosaposin-derived 18-mer 
peptide (PS18) significantly improved behavioral deficits, enhanced the survival of tyrosine 
hydroxylase-positive neurons, and decreased the activity of astrocytes in the substantia 
nigra and striatum in MPTP-induced PD model mice. In vitro, a Cell Counting Kit-8 assay 
and Hoechst 33258 staining revealed that co-treatment with 300 ng/ml PS18 and 5 mM 
MPP+ protected MPP+-induced nuclear morphological changes and attenuated cell death 
induced by MPP+. We also found that PS18-FAM entered the cells, and the amount of time 
that PS18-FAM was retained in the cytoplasm of MPP+-treated cells was shorter than that 
of untreated cells. In addition, PS18 showed protection from MPP+/MPTP-induced 
apoptosis in the SH-SY5Y cells and dopaminergic neurons in the PD model mouse via 
suppression of the c-Jun N-terminal kinases/c-Jun pathway; upregulation of Bcl-2; 
downregulation of BAX, attenuating mitochondrial damage; and inhibition of caspase-3. 
Duchene muscular dystrophy (DMD) is a fatal genetic disease caused by mutations in 
the DMD gene, leading to dystrophin deficiency. DMD is caused by a mutation in the 
X-linked dystrophin gene; it is a recessive genetic disease characterised by alterations in the 
neuromuscular system, and metabolic and structural disorders of the central nervous system 
(CNS), which cause mental retardation and metabolic damage. While muscle wasting is 
prominent, the CNS is also affected in DMD, with non-progressive intellectual and/or 
cognitive impairment being observed in about one-third of patients with DMD. The 
dystrophin-deficient mdx mouse is a model of human DMD. Recently, we reported low 
levels of PS in muscles in mdx mice compared with C57BL/10 mice. The distribution of PS 
in the brains of juvenile and adult mdx mice was investigated by immunochemistry, 
Western blotting, and in situ hybridization. Immunochemistry revealed lower levels of PS 
in the cytoplasm of neurons of the cerebral cortex, hippocampus, cerebellum, and choroid 
plexus in mdx mice. Western blotting confirmed that PS levels were lower in these brain 
regions in both juveniles and adults. Even with low PS production in the choroids plexus, 
there was no significant PS decrease in cerebrospinal fluid (CSF). This discrepancy may be 
explained by decreased PS intake from the CSF due to decreased expression of PS receptors 
(GRP37, GRP37L1) in mdx mice. In situ hybridization revealed that the primary form of PS 
mRNA in both normal and mdx mice was Pro+9, a secretory-type PS, and the hybridization 
signals for Pro+9 in the above-mentioned brain regions were weaker in mdx mice than in 
normal mice. We also investigated mitogen-activated protein kinase signalling. Stronger 
activation of ERK1/2 was observed in mdx mice, ERK1/2 activity was positively correlated 
with PS activity, and exogenous PS18 stimulated both p-ERK1/2 and PS in SH-SY5Y cells. 
These results suggest that prosaposin may play an important role to protection of

【背景】 
循環器疾患は高血圧の寄与危険度が大きいため、高血圧発症の発症リスクをできるだけ低減
すべきである。そこで、地域住民において高血圧の発症リスクを非侵襲的に計測できるなら、
その結果を用いて、疫学的に高血圧予防への保健指導が期待できる。すでに欧米の地域疫学研
究においては、非侵襲的に計測した心拍変動の低下が高血圧の発症リスクと報告されている。 
しかし、脳卒中の発症病態とそのリスクファクターが従来の日本人と西欧先進諸国では全く
異なっていることを明らかにした小町喜男・小西正光らの秋田における病理・疫学研究が示す
ように、人種・文化の異なる知見をそのまま日本人に援用する拙速は避けるべきである。 
この観点からみると、本邦での地域住民を対象にした心拍変動と血圧との関連について、生
活習慣を含めて検討した研究はまだ無い。 
【目的】 
地域住民を対象とした横断研究により、心拍変動(HRV: Heart Rate Variability)指標と収
縮期血圧と拡張期血圧との関連を、性別・年齢別・疾病・生活習慣等の層別に分けて検討する。
高血圧の発症リスクとなるパラメータを確定し、高血圧発症の予防指導に有効な知見を得る。 
【方法】 
 2009 年～2011 年に愛媛県大洲市で実施された特定健診受診者（40～74 歳）のうち、本研究へ
の同意が得られた男女 3,600 名から、心電図検査において期外収縮、不整脈を認めた者、心拍変
動検査のデータが得られなかった者を除いた 3,458 人を分析対象とした。  
 HRV は、5 分間安静後、5 分間座位で、指尖脈波を Pulse Analyzer Plus(YKC Co.,Inc.,Korea)
で測定した。評価指標は、自律神経の活動性の指標として SDNN ( Standard Deviation of the 
NN[RR] interval)、副交感神経の活動性の指標として rMSSD(root Mean Square of the Succes-
sive Differences)、LnHF(natural logarithm of High Frequency)を使用した。血圧は座位で 5
分間安静の後、右腕で 2 度測定し平均値を分析に用いた。BMI(BodyMass Index)、HbA1c(JDS 値)、
血中脂質、高血圧、糖尿病の治療の有無については、特定健診での身体計測値、検査値および問
診結果を用いた。飲酒、喫煙、身体活動量については自記式質問紙で調査した。 
【結果】 
対象者の平均年齢は男性 63.1±8.6 歳、女性 64.0±8.0 歳、収縮期血圧は男性 132.8±18.3m
mHg、女性 130.0±19.4mmHg、拡張期血圧は 80.0±10.7mmHg、女性では 75.1±10.8mmHg、降圧
薬服用者の割合は、男性 29.4％、女性 27.5％であった。 
 共分散分析により SDNN の 4 分位別の対象者の特徴を見ると、男性では、SDNN が低い群ほど
年齢、心拍数、BMI、飲酒量の平均値が高く、降圧薬服用者と糖尿病の割合が高かった(p<0.0
5)。女性では、SDNN が低い群ほど年齢、心拍数の平均値が高く、降圧薬服用者と糖尿病の割
合が高く、一方で身体活動量、飲酒者の割合は有意に低かった(p<0.05)。 
4 分位別に血圧の年齢調整平均値を見ると、男性では SDNN が低いほど拡張期血圧が有意に
高く(p<0.05)、女性では SDNN が低いほど収縮期・拡張期血圧ともに有意に高かった(p<0.05)。 
年齢、BMI、喫煙、飲酒、身体活動量、降圧薬の服用の有無、糖尿病を調整した線形回帰分
析により HRV 指標と血圧値との関連を検討すると、男女ともに、SDNN、rMSSD、HF は拡張期血
圧値と有意な負の関連を認めた。収縮期血圧は、女性では拡張期血圧と同様に有意であったが、
男性では有意な関連を認めなかった。 
 降圧薬の有無で層別解析を行うと、HRV と血圧値との関連は降圧薬のない群で有意であっ
た。さらに降圧薬治療がない男性を飲酒で層別解析を行うと、非飲酒者において HRV と血圧値
との有意な関連が認められたが、飲酒者においては有意な関連を認めなかった。 
【考察】 
 男性では拡張期血圧が、女性では収縮期・拡張期血圧ともに HRV との負の関連を認めた。こ
れは、欧米における地域住民を対象とした大規模研究の結果を支持するもので、本邦の農村部
地域住民においても、副交感神経機能の活動性低下による、相対的な交感神経系の亢進が血圧
値の上昇と関連している。非飲酒者の男性においては、HRV と収縮期血圧との飲酒の間に有意
な負の関連を認めた。男性において血圧値と HRV との関連が弱いことには、飲酒による効果が
影響している可能性が示された。 
【結論】 
 HRV 値を用いて、地域住民への高血圧発症予防の一助としうる可能性が示唆された。 
 平成 26 年 1 月 20 日に行われた公開審査では、申請者は①性差と飲酒の関係ことにアルコール
脱水素酵素との関係、②国保の集団というバイアスへの配慮、③ストレス指標としての HRV の他
の領域への応用、④先行研究との差違などについて質疑やコメントを受け、それらに明瞭に解答
した。よって本審査委員会は全員一致で本論文を学位授与に値するものと判断した。

   学位論文要旨   
 心房細動は，脳梗塞や一過性脳虚血発作の重要な原因のひとつである．また，心房細動が原
因である場合には抗凝固療法の適応となり，脳梗塞患者の治療方針を決定する上で，心房細動
の有無を早期に診断することは重要である．一方で心房細動には発作性のものがあり，入院時
に心房細動の有無を判断出来ないことも多い．血漿 BNP は，うっ血性心不全の指標として臨床
的に使用されているが，心房細動患者で高値となることが知られている．今回我々は，血漿
BNP が心房細動のバイオマーカーとなり得るか，急性期虚血性脳血管障害患者（脳梗塞または
一過性脳虚血性発作）に対して，心房細動の有無と血漿 BNP 値の関係を検討した． 
 2006 年 3 月から 2007 年 5 月までに来院した発症 24 時間以内の虚血性脳血管障害患者を対象
とした．心筋梗塞，透析，心筋症，弁置換術の既往を有する患者ではこれらにより血漿 BNP が
上昇しうるため，除外した．最終的に 237 名を登録した(一過性脳虚血発作 47 名，女性 91 名，
平均年齢 71.9 歳)．入院時の心電図で心房細動を認めたもの，および心房細動の既往があるも
のを心房細動群とし，それ以外を洞調律群とした．洞調律群はさらに，入院後のモニター心電
図やホルター心電図検査により心房細動の有無を検索し，心房細動が発見された新規心房細動
群と，心房細動を認めなかった非心房細動群に分類した．入院時に血液検査を施行し血漿 BNP
値を測定した． 
 入院時に心房細動を有していた(心房細動群)のは 72 名（30.4％）で，血漿 BNP 値は，洞調
律群の 92.1pg/ml に対して心房細動群では 401.7pg/ml と有意に高値であった（p<0.001）. 洞
調律群と比較して，心房細動群では，女性の頻度が高く(32.7% vs. 51.4%, P = 0.010)，高血
圧(55.2% vs. 70.8%, P = 0.024)や心不全の既往(1.8% vs. 25.0%, P < 0.001)が多く，より
重症で（入院時 NIH stroke scale score：NIHSS score 5.5 ± 6.4 vs. 10.1 ± 8.7, P < 
0.001, 退院時 modifiied Rankin Scale score: mRS score 2.1 ± 1.9 vs. 3.2 ± 2.1, P <

 洞調律群 165 名のうち，入院後に心房細動が判明した新規心房細動群は 13 名で，全体の 5.5
％であった．非心房細動群と比較して，新規心房細動群では，より重症で（入院時 NIHSS 
score 5.1 ± 6.2 vs. 9.5 ± 6.9, P = 0.010, 退院時 mRS score 2.0 ± 1.9 vs. 3.2 ± 1.8, 
P = 0.031）,CTR が大きく(53.8 ± 6.4% vs. 59.7 ± 8.1%, P = 0.007)，D-dimer が高い傾向
を認めた(2.2 ± 7.1 vs. 2.5 ± 3.9 μg/ml, P = 0.079)．血漿 BNP 値は，新規心房細動群で
有意に高値であった(84.1 vs. 184.7 pg/ml, p<0.0001).

用いて検討した．年齢 75 歳以上では感度 69.2％，特異度 67.0％，NIHSS6 以上では感度 61.5
％、特異度 69.7％，d−dimer 0.8μg/ml 以上では感度 69.2％，特異度 60.9％，血漿 BNP 値
85.0pg/ml 以上では感度 83.3％，特異度 76.2 であった．多変量解析では，血漿 BNP 値 85pg/ml
以上のみが新規心房細動に対する有為な因子であった(OR, 7.20; 95% CI, 1.71 to 30.43, P 
= 0.007)． 
 血漿 BNP は，虚血性脳血管障害患者における心房細動の有無を予測する有用なバイオマーカ

Rice MKRN is a member of the makorin RING finger protein gene (MKRN) family, 
which encodes a protein with a characteristic array of zinc-finger motifs conserved in 
various eukaryotes. In the present study, we used non-radioactive in situ hybridization 
technique to investigate the spatio-temporal gene expression pattern of rice MKRN 
during various growth stages of rice (Oryza sativa L. var. Nipponbare). These growth 
stages broadly represented the important life events of rice, such as study at 0-1 to 2-3 
days after pollination (DAP) represented the early ovule development. Also, the 
embryonic development was analyzed at 5-6, 8-9, 11-12, 15-16 and 45 DAP. The impact 
of imbibition on MKRN expression was studied after 1 and 6 day of imbibition, whereas 
early root development was observed 4 and 9 days after germination. The 
post-embryonic organogenesis such as lateral and crown root development was studied 
for MKRN expression at 4, 7 and 9 days after germination. The studies of these 
developmental stages are crucial for the further improvement in grain yield and drought 
resistance in rice. 
 During ovule development, the changes of MKRN expression pattern in cell sap 
conducting vessels of the ovule were observed. During early embryogenesis in rice, the 
apical-basal axis is established at 4-5 DAP. MKRN expression was ubiquitous during 
organogenesis of SAM, stele, coleoptile, coleorhiza, columella, epiblast, scutellum and

the scutellum vascular trace but not in the epidermis and lateral root cap cells in the 
embryo at 5-6 DAP. During the successive stages of embryonic growth such as 8-9 DAP, 
11-12 DAP and 45 DAP, MKRN expression was gradually decreased in the 
differentiating cells including the coleorhiza, epiblast and the scutellum, whereas it was 
obvious in the SAM, RAM, provascular tissues; however it was increased again in the 
differentiating cells during imbibition. According to the positional distance of the 
tissues from the root tip, MKRN expression was variable in the cortex, endodermis, 
pericycle and the provascular tissues. The sections close to the root tip showed stronger 
MKRN expression than the sections distant from the root tip. Tissue-specific and 
position dependent MKRN expression was found during embryonic and post-embryonic 
root and shoot development. 
 During post-germination root development, MKRN expression was spatially 
similar to the embryonic root development, however in fully matured root, MKRN 
expression was localized only in the active companion cells of the phloem and it was 
absent in all other root cells. Also, MKRN was expressed in the various developmental 
stages of lateral root primordia, initiating with the periclinal divisions of the 
presumptive pericycle founder cells to its emergence. Interestingly, expression of MKRN 
was observed in the three endodermis cells adjacent to presumptive pericycle founder 
cells and continued in three consecutive rows of cortex parenchyma cells up to the 
sclerenchyma, suggesting a role of MKRN in the mechanism of lateral root primordia 
emergence. Interestingly, MKRN expression pattern was similar during the development 
of LRP and CRP; however during their emergence it was different. 
 During the vascular pattern development in the embryo, expression of MKRN was 
concentrated in the developing provascular tissues and SAM whereas; it was gradually 
reduced in the surrounding mesophyll cells. MKRN expression was further increased in 
the provascular bundle cells of 1 day imbibed embryo compared to the dry mature 
embryo. A similar expression pattern was also found in the coleoptile collected from the 
6 days imbibed seeds. The MKRN expression pattern during development in various 
growth stages suggests an important role of makorin RING finger protein gene in 
embryonic and post-embryonic organogenesis in both apical–basal and radial 
developmental axes of rice. This study would assist to improve the quantitative and
