アカマツのアレロケミカルに関する研究  
  
      審査結果の要旨 
 
アレロパシーとは植物が生産，放出する化学物質が他の植物の生長を抑制あるいは促進する現象のこ
とであり，この原因となる物質をアレロパシー物質という．アカマツ(Pinus densiflora Sieb. et Zucc.)はマ
ツ科マツ属の常緑針葉樹であり，日本においては最も一般的なマツである．アカマツ林の樹下には光が
十分に当たるにもかかわらず，下草がまばらにしか生えていないことが知られている．その原因として
アカマツのアレロパシーが挙げられており，アカマツの葉に含まれるアレロパシー候補物質として 9,13-
epidioxyabiet-8(14)-en-18-oic acid，abscisic acid β-D-glucopyranosyl ester，1-mono (16-hydroxyhexadecanoyl) 
glycerol，フェノール類などが単離されている．森林生態系におけるアレロパシーでは，土壌有機層から
供給される物質も重要であると考えられている．また，葉や落葉から放出されるアレロパシー物質が土
壌に蓄積することによって，周囲の植物の生長に影響を与えていると考えられるが，充分に明らかにさ
れていない．  
 
本研究では，まずアカマツの葉，落葉，土壌のアレロパシー活性を明らかにし，1 年間定期的にサン
プリングしたアカマツの葉のアレロパシー活性を測定した．更に，落葉と土壌の含水メタノール抽出物
に含まれるアレロパシー物質を単離，同定を試みた．アカマツの葉，落葉，土壌の含水メタノール抽出
物のアレロパシー活性を測定したところ，落葉もアレロパシー放出源の一つであり，葉や落葉から放出
されたアレロパシー物質が樹下の土壌に蓄積していることが示唆された． 
 
落葉抽出物を精製し，最終的に 4 つのアレロパシー候補物質を単離した．そのうち 2 つは 9,13-
epidioxyabiet-8(14)-en-18-oic acid，abscisic acid β-D-glucopyranosyl ester であった．クレスを用いた生理活
性試験の結果，9,13-epidioxyabiet-8(14)-en-18-oic acid がクレスの生長を 50 %に抑制するために必要な濃
度（I50値）は約 500 μM であり，abscisic acid-β-D-glucopyranosyl ester の I50値は約 1.5 μM であった．こ
 
 
れらのアレロパシー候補物質はアカマツの葉からも単離されていることから，葉に含まれるアレロパシ
ー候補物質が落葉の状態になっても保持されており，落葉からも放出されていることが示唆された． 
また，アレロパシー候補物質 A は 10.0 g D.W. equivalent extract/mL で、クレスの下胚軸の生長を対照区
と比較して 22.5 %に、根の生長を 17.9 %に抑制した。 アレロパシー候補物質 D は 8.0 g D.W. equivalent 
extract/mL でクレスの下胚軸の生長を 52.9 %に、根の生長を 25.5 %に抑制した。 一方，アレロパシー候
補物質 A 及び D の化学構造は葉のアレロパシー候補物質のスクリーニングでは検出されなかったこと
から，落葉の分解過程で生じるアレロパシー候補物質である可能性が示唆された． 
 
更に，土壌で直接周囲に作用するアレロパシー物質を明らかにするため，土壌の含水メタノール抽出
物からアレロパシー候補物質の単離を行った． 最終的に土壌に含まれる主要なアレロパシー候補物質
として methyl 15-hydroxy-7-oxo- dehydroabietate と 7-oxodehydroabietic acid を単離した．クレスに対する
methyl 15-hydroxy-7-oxo-dehydroabietat の I50値は約 1000 μM であり，7-oxodehydroabietic acid の I50値は約
200 μM であった． 更に，アカマツ林の土壌に含まれるアレロパシー候補物質の類縁体として 15,16-
bisnor-13-oxolabda- 8(17),11E- dien-19-oic acid と methyl 8,11,13,15- abietatetraen-18-oate を単離した． これ
らの物質は土壌に樹脂酸が蓄積する際の主要な成分であることが示唆された. 
 
また，アカマツの葉，落葉，土壌のアレロパシー候補物質を比較すると，落葉と土壌にはアカマツの
葉とは異なる活性物質が含まれており，アカマツのアレロパシーには分解による影響が大きいことが示
唆された． アカマツから単離された活性物質のクレスに対する I50 値をもとに比較すると，1-mono(16-
hydroxyhexadecanoyl) glycerol が最も強く，abscisic acid β-D-glucopyranosyl ester，7-oxodehydroabietic acid，
9,13-epidioxyabiet-8(14)-en- 18-oic acidmethyl 15-hydroxy-7-oxo- dehydroabietate の順に強い活性を有してい
た． 土壌で検出されなかった活性物質もアカマツのアレロパシー活性に部分的に貢献している可能性
があるが，アレロパシー物質は植物体に含まれているだけでなく，環境に放出されて作用している必要
がある． 従って，物質自体の活性が強い 1-mono(16-hydroxyhexadecanoyl) glycerol よりも，土壌に蓄積
していると考えられる methyl 15-hydroxy-7-oxo- dehydroabietate や 7-oxodehydroabietic acid はアカマツの
アレロパシーへの寄与がより大きいと考えられる．アカマツ植物体から環境へ大量に供給され，土壌微
細物の働きによって活性態に異性化されるアビエタンジテルペンの化合物群はアカマツのアレロパシー
において重要な役割を果たすことが示唆された． 
 
本論文に関する公開審査会は，平成２７年２月７日に愛媛大学農学部において開催され，論文発表と
質疑応答が行われた．引き続いて行われた学位論文審査委員会で本論文の内容を慎重に審査した結果，
審査委員全員一致して博士（農学）の学位を授与するに値するものと判定した．

 本研究では、水資源に乏しく、灌漑のための水利用が盛んな新川で引き起こさ
れている深刻な有機物汚濁について、そのメカニズムを明らかにすることを目的
とする。以下の 3 つの小題に示した観点から、主に灌漑による水利用によって形
成される流域の水・物質循環を解析し、河川の有機物汚濁のメカニズムを明らか
にした。 
 
流域の水利用が河川の有機物汚濁に及ぼす影響 
 香川県の新川流域において、水中の水安定同位体比（δD, δ18O）および各種物
質量を広域的に調べ、物質マップを作成することにより、有機物の起源を解析し、
新川中下流における有機物汚濁の原因を明らかにした。 
 新川中下流における POC/Chla は 38 (n=30)で、富栄養化した湖沼と同等かそれ以
下で、有機物の大部分が藻類であると考えられる。2010 年 6 月における POC は源
流 0.2 mg/L、上流 1.1 mg/L、中下流 5.1mg/L、中下流域のため池 2.5 mg / L で、中
下流およびその流域のため池で高かった。δ 18O も同様に中下流域で高かった。非
灌漑期の 2010 年 10 月の中下流は、δ 18O と POC の値が、6 月と比べて特に高かっ
た。2011 年 7 月は、河川流域にかなりの降水量があったが、POC はこれまでと同
様に中下流での濃度増加が見られた。ただし、POC/Chla は 82 と高く、森林や水
田由来の高等植物の割合が増えていると考えられる。以上のことから、新川流域
の有機物濃度は、δ18O と同様の変化を示しており、ため池で増殖した藻類が河川
に供給されたと考えられる。この傾向は、流域の水が少ない非灌漑期において顕
著で、雨量の多い時期は有機物中の藻類の割合が減少する。  
 
水資源に乏しい河川の有機物汚濁に対する植物プランクトンの影響 
 先の研究から河川の有機物汚濁の原因がため池起源の藻類であることが示され
た。日本の河川は短く、植物プランクトンが増殖しにくいため、存在する藻類は
主に付着藻であるとされてきた。一方で、新川では水柱に多くの藻類が恒常的に
存在しており、これを河床から剥離した付着藻で説明することは難しい。そこで、
本研究では、新川流域における植物プランクトンの空間分布と動態の解明を行う
（第３号様式）(Form No. 3) 
ことを目的に研究を行った。   
 新川中下流において、種組成のほとんどが Pseudanabaena sp.のような、富栄養
湖で増殖する植物プランクトンが占めた。 河川本流の細胞数は最大規模のため池
の流入点の下流から増加した。δ18O も同様に合流点より下流で、値が高くなった
ことから、中下流での細胞数の増加と種組成の変化は、ため池から排出された水
を反映していると考えられる。また、中下流の細胞数は 2010 年 6 月より、非灌漑
期の 2010 年 10 月が多かった。2011 年 7 月は、香川用水の供給や雨量が多く、灌
漑期であり、流域のため池から水が絶えず河川に供給されていたが、細胞数は比
較少なかった。  
 
有機物汚濁が著しい香川県の河川における浮遊藻類の一次生産 
 新川の有機物はため池の植物プランクトンが起源であることがわかったが、春
先に濃度が増える等、河川内の堰における増殖の影響も重要だと考えられる。本
研究では、河川内の内部生産と有機物に対する寄与率を明らかにした。 
 新川河口堰止水域での純生産速度は、冬から春にかけて上昇し、降水量の多い
初夏には減少するが、夏には最大値の約 9mgC/L/day（2013 年）となった。また、
総生産と Chl a 及び光合成活性は、まとまった雨が降った時に減少した。総生産
と Chl a は総生産が高い夏期に一致しなかった。河川水量が多く、河川水の滞留
時間が短い夏には、河川内での植物プランクトンの増殖は POC に反映されないこ
とがわかった。 
 POC への総生産の寄与率は多くの時期で 50％以下であり、外部負荷の影響が大
きいことが分かった。しかし、非灌漑期など、水量の低下する時期については、
河川水の滞留時間が長くなるため、最大で総生産の POC への寄与率は 90％に達す
ることがわかった。これが、冬期から春期に新川の POC 濃度が高い原因であると
いえる。  
新川では河川水量が減少する非灌漑期には植物プランクトンはため池から供給
されるだけでなく、河川の止水域において、滞留、増殖しており、これが深刻な
有機物汚濁を引き起こすことがわかった。春期の水温の上昇がこれを促進してい
ると考えられる。  
 
まとめ 
持続的な水や生物資源の利用のためには、良好な水・物質循環の存在が不可欠
である。本研究では水利用が効率的に行われている流域における河川の有機物汚
濁のメカニズムを解析した。本研究の成果は、水利用が、河川〜海域の物質循環、
生態系にあたえる影響を解明するもので、今後も研究成果を蓄積することで、流
域における適正な貯水池の規模や運用等、環境負荷の少ない水利用の提言を行う
ことが出来るであろう。

The antitermite and antifungal activities of several Indonesian wood biomass were 
investigated against the subterranean termite, Coptotermes formosanus and wood rotting 
fungi, a white rot fungus (Trametes versicolor) and a brown rot fungus (Fomitopsis 
palustris). The objective of this study was to isolate the chemical components that 
responsible as antitermite and antifungal properties of those wood biomass samples. 
 
The heartwood mill was extracted with acetone and methanol to give their extracts, 
each of which were fractionated successively using n-hexane, ethyl acetate, and water to 
afford their fractions.  No-choice and two-choice tests were employed to assess the 
antitermite activity of the wood biomass samples. To examined the antifungal activity, 
Potato Dextrose Agar medium that contain the obtained consentration of the extracts 
tested was used to inoculate the fungal mycelium. 
Gofasa (Vitex cofassus Reinw.) is one of the wood species that potential to explore 
its bioactive compounds, since this wood species showed high resistance to wood 
deteriorating organisms without any chemical preservatives. The yield of the extracts 
and fractions suggested that the extractives of V. cofassus heartwood tend to be polar.  
It was demonstrated that the methanol extract itself, the ethyl acetate fraction and the 
n-hexane fraction from the methanol extract, and the ethyl acetate fraction from the 
acetone extract had high antifeedant activity against C. formosanus. Further 
investigation showed that there were four active sub-fractions that recently obtained 
from the ethyl acetate fractions of the acetone and the methanol extracts. Furthermore, 
all the extracts and fractions were shown to have strong-moderate activity against the 
wood rotting fungi, where the aqueous fraction of the acetone extract had the strongest 
antifungal activity against T. versicolor, while almost these extracts and fractions had 
moderate-weak activity against F. palustris. 
Sandoricum koetjape Merr. (Meliaceae), also known as wild mangosteen, is one

kind of trees that produce fruits, and various parts of the tree have medicinal properties. 
The yield of the extract and fraction indicated that the extractives of S. koetjape tend to 
be non-polar. Antitermite test revealed that the acetone extract and its fractions had the 
strongest antifeedant activity classified into III-IV of the antifeedant class, and that the 
methanol extract and its fractions had lower antitermite activity mostly belonging to I-II 
of the antifeedant class. Furthermore, the antifungal test indicated that against        
T. versicolor the n-hexane fraction of the methanol extract had the strong antifungal 
ability belonging to the strong category according to the antifungal activity class, while 
against F. palustris almost these extracts and fractions had the weak activity.  
Albizzia falcataria one of the member of Family Fabaceae, is most widely 
developed and utilized tree species for forestry development program in Indonesia. The 
extraction and fractionation indicated that the extracts of A. falcataria waste wood and 
A. falcataria bark tend to be polar.  Antitermite test revealed that Albizzia falcataria 
bark classified into class II – III, while the A. falcataria  waste wood had revealed the 
antitermite activity that classified into class I – II of the antifeedancy classification. 
From the antifungal test results confirmed that the extract of A. falcataria  waste wood 
were classified into moderate-weak class of antifungal activity class against T. 
versicolor. On the contrary, all these extracts and fractions only classified into the 
weak-not active antifungal activity against F. palustris. Furthermore, the extracts and 
fractions of A. falcataria  bark demonstrated the moderate-weak antifungal activity 
against T. versicolor.  Conversely, all these extracts and fractions revealed classified 
into weak-not active antifungal classification against F. palustris. 
As the conclusions were among those Indonesian wood biomass extracts, the S. 
koetjape heartwood extracts revealed the higher antitermite activities compared to V. 
cofassus and A. falcataria waste wood and A. falcataria bark.  Moreover, the wood 
biomass extracts showed the strong-moderate antifungal activities against T. versicolor, 
while almost the extracts were indicated the weak-not active against F. palustris fungi.
