【背景】 
高血圧、高血糖、脂質異常は強力な動脈硬化の危険因子であるが、これらのコントロールは薬剤
の開発により容易になってきた。現在、残存リスクのコントロールの重要性が叫ばれており、そ
の 1 つが高ホモシステイン血症である。ホモシステインはメチオニンからシステインが生合成さ
れる際の必須中間生成物である。遺伝性ホモシステイン尿症が若年で脳卒中や心筋梗塞を発症す
ること、血中ホモシステイン濃度が高値であるほど脳卒中や心筋梗塞等の動脈硬化疾患の発症が
増加することが報告されている。しかし葉酸や各種ビタミンによる血清ホモシステイン低下療法
が心血管病の発症を抑制することは証明されておらず、また高ホモシステイン血症が心血管病の
発症リスクを高める機序は依然不明である。最近、ホモシステインが末梢血単球数を増やし単球
が血管壁に侵入し活性酸素負荷や炎症を誘導し、動脈硬化性疾患の進行に重要な役割を演じるこ
とが報告されている。 
動脈硬化の評価に頸動脈エコーが汎用されている。総頸動脈の intima media thickness(IMT）は
粥状動脈硬化を、Doppler エコー法を用いた血流速度(flow velocity：V)波形からは arterial s
tiffness の評価が可能である。 
 
【目的】 
本態性高血圧患者を対象に血中ホモシステイン濃度と頸動脈エコーで評価した動脈硬化性変化と
の関連を明らかにし、ホモシステインが動脈硬化を促進する機序を炎症に焦点を絞って明らかに
することである。 
 
【対象者および方法】 
対象者は当院に通院している本態性高血圧患者 67 例。外来受診時、坐位・安静時の血圧測定によ
って、収縮期血圧 140mmHg 以上または拡張期血圧 90mmHg 以上の患者、または降圧薬を内服中の患
者を高血圧と診断した。V 測定値に影響を及ぼす可能性のある冠動脈疾患、弁膜症、脳血管障害、
甲状腺疾患を有する患者および二次性高血圧の患者は除外した。本研究に同意の得られた患者に
対して安静時採血を施行し、従来の動脈硬化危険因子としてホモシステイン、中性脂肪、HDL-C、
LDL-C、推定糸球体濾過量(eGFR)、HbA1ｃ、および炎症性マーカーとして TNF-、IL-6、CRP、末
梢血白血球数およびその分画を測定した。ホモシステイン、TNF-、IL-6 および CRP は正規分布
を来さないため自然対数(ln)で表記した。頸動脈エコーで総頸動脈 IMT および同部で Doppler 法
により最大血流速度（maxV）、最小血流速度（minV）および平均血流速度（meanV）を測定し、末
梢血管抵抗の指標である Pulsatility Index(PI)＝（maxV-minV）/meanV）を算出した。2 群間の
比較および関連はそれぞれ Student T 検定および Peason’s correlation coefficient、PI およ
び末梢血単球数の独立した規定因子の同定は Stepwise regression 法を用いた。 
 
【結果】 
対象の平均年齢は 62.3 歳で、27 例が男性であった。平均収縮期および拡張期血圧は 154.8/92.4
mmHg、Body Mass Index (BMI)は 25.1kg/m2であった。平均の eGFR は 76mL/min/1.73m2であった。
平均 PIは 0.64であった。PI値により対象を 2群に分類した。PI高値群と低値群の比較では BMI、
IMT、末梢血単球数、ln ホモシステインが PI 高値群で有意に高かった。PI に相関する因子は ln
ホモシステイン、末梢血単球数、IMT、年齢、IL-6、BMI であった。また末梢血単球数、性別、年
齢、IL-6、BMI を独立変数とし PI について多変量解析を行ったところ、末梢血単球数、年齢が P
I の独立した規定因子であった。末梢血単球数と相関する因子は ln ホモシステイン、BMI、男性、
HDL-C、および lnTNF-であった。これらの因子を独立変数とし末梢血単球数について多変量解
析を行ったところ、HDL-C と ln ホモシステインが末梢血単球数の独立した規定因子であった。IM
T と相関する因子は eGFR および年齢であった。 
 
【考察】 
本研究において末梢血管抵抗・atrial stiffness の指標である PI の独立した規定因子は末梢血
単球数および年齢であった。一方末梢血単球の独立した規定因子は HDL-C とホモシステインであ
った。基礎研究でホモシステインは炎症や酸化ストレスを介して末梢血単球を増加させることが
報告されている。以上の結果から、本態性高血圧患者においてホモシステインは末梢血単球数の
増加を介して arterial stiffness の増加に関連していることが示唆された。葉酸やビタミンの摂
取を増やして血清ホモシステイン濃度を低下させることは、動脈硬化進展予防に重要であること
があらためて示唆された。

    学位論文の要約 
<はじめに> 
骨髄単核球細胞は、血管内皮前駆細胞や骨髄間質細胞を含む。これらの細胞は様々な虚血性疾
患で局所における血管新生を促進し、組織の修復作用があることが知られている。血管内皮前駆
細胞は、血管内皮細胞への分化、血管新生因子の分泌により、虚血局所での血管新生を促す。ま
た、骨髄間質細胞も様々な血管新生因子を分泌する。 
突発性難聴の原因はいまだ不明であるが、ウィルス説とともに内耳虚血説が有力である。今回
われわれは、突発性難聴治療に対する臨床応用に向けて、この骨髄単核球細胞を用いた内耳障害
防御効果を検討した。 
<対象および方法> 
1． 骨髄単核球細胞の分離抽出法：スナネズミ大腿骨および脛骨から骨髄細胞を取り出し、比重
遠心分離法にて抽出した。最終的に一匹あたり 4～5×10６個の細胞を得た。 
2． 一過性内耳虚血負荷：実験動物には雄スナネズミ（8～12 週齢、体重 60～80g）を用いた。
前頚部皮膚切開を行い、両側の椎骨動脈を剥離し、絹糸を掛けて牽引することで血流を遮断
した。虚血負荷 15 分後に牽引を解除し、血流を再開通させた。 
3． 骨髄単核球細胞の静脈内投与：内耳への虚血負荷 15 分後、血流を再開通させた直後に、大
腿静脈にカテーテルを留置し、骨髄単核球細胞（2～3×10６個/ml in PBS）、phosphate buf
fered saline(PBS)のいずれかを 2ml/㎏ずつ投与した。 
4． 骨髄単核球細胞の局在の評価：分離抽出した骨髄単核球細胞を、蛍光色素 PKH67 にて染色し
た。PKH67 陽性細胞を静脈内投与し、虚血負荷 4 日目に全身潅流固定を行い、蝸牛を採取し
た。採取標本で凍結切片を作製し、蛍光顕微鏡にて観察した。 
5． 聴力評価：聴力の評価は聴性脳幹反応（以下、ABR と略）にて行った。虚血前、虚血 1、4、
7 日後に沈静下に記録した。刺激には 8000、16000、32000Hz の tone burst 音を用いた。 
6． 組織学的評価：虚血 7 日後に蝸牛を採取した。surface preparation 法にてコルチ器を取り
出した。基底回転を摘出し、Rhodamine-phalloidin、Hoechst33342 による二重染色を行い、
蛍光顕微鏡で観察し、有毛細胞の脱落割合を算出した。 
7． ウエスタンブロット解析：サンプルは摘出した蝸牛を浸漬、細片化し、超音波破砕を加えホ
モジナイズした後、遠心分離で得られた上清の可溶性蛋白質を用いた。一次抗体として抗 G
DNF、抗 BDNF、抗 VEGF、抗 Ang1、抗 FGF2、抗 NT3 抗体と反応させ、二次抗体以後は ECL We
stern blotting Kit を用いて施行し、神経栄養因子の発現量を比較検討した。 
8． 統計学的検討：ABR 閾値変化および内有毛細胞脱落割合の統計学的検討には Mann-Whitney U
 test を、ウエスタンブロット解析には one-way analysis of variance を用いて有意差検定
を行った後、Bonferroni’s multiple comparison test にて多変量解析を行った。得られた
結果は、p<0.05 を有意差ありと判定した。 
<結果> 
1． 骨髄単核球細胞の局在：PKH67 陽性細胞は迷路動脈やらせん動脈の内腔にみられたが、コルチ
器や周辺組織への侵入像や融合像はみられず、有毛細胞と置換や融合することはなかった。 
2． ABR 閾値変化：虚血前の ABR 閾値を 0dB として、ABR 閾値変化について比較検討した。
8000、16000Hz においては、1、4、7 日後いずれにおいても骨髄単核球細胞投与群では control
群と比較して、ABR 閾値の上昇を有意に抑制していた。 
3． 内有毛細胞の脱落割合：骨髄単核球細胞投与群と control 群において、内有毛細胞の核の消
失と聴毛の脱落所見を観察したところ、control 群と比較して細胞投与群では内有毛細胞の脱
落割合が有意に抑制されていた。 
4． 神経栄養因子蛋白の発現：骨髄単核球細胞投与群と control 群において、6 種類（GDNF、
BDNF、VEGF、Ang1、FGF2、NT3）の神経栄養因子の発現を比較したところ、GDNF と
NT3 の発現が、細胞投与群では有意に上昇していた。 
<考察> 
今回の実験では、ABR 閾値変化や内有毛細胞脱落割合の結果から、細胞投与群は control 群よ
りも内耳障害が軽微であり、虚血性内耳障害を保護する効果をもつことが確認された。 
骨髄単核球細胞による内耳保護作用効果のメカニズムは明らかではない。静脈内投与した細胞
はらせん動脈内には確認されたがコルチ器には観察されず、この結果からは投与した細胞がコル
チ器内へ沈着し、有毛細胞への分化や細胞融合による保護効果を示した可能性は低いと考えられ
る。神経栄養因子の発現が内耳において上昇していたことを考えると、骨髄単核球細胞が栄養因
子を分泌し、これが障害部位に作用することによって内耳障害を抑制したという機序が最も考え
られる。骨髄単核球細胞は神経栄養因子を活性化し、内耳機能保護効果に働いた可能性が推察さ
れた。

学位論文要旨 
多相性の腹部造影CTは慢性肝炎や肝硬変患者、HCCが疑われる患者におけるCTスクリーニング
検査として重要である。動脈相は多血性腫瘍を描出するのに重要であるが、同様に門脈相は高分
化肝細胞癌や転移性腫瘍といった乏血性腫瘍を描出するために重要である。このためには、十分
な肝実質の造影効果（50HU以上）が必要とされ、最適な肝実質造影に関する多くの研究がなされ
てきた。しかし、これらの研究では各時代における旧式のCTを用いたdynamic CTの撮像プロトコ
ールであったり、患者背景が各々で異なっていた。肝実質の造影に関しては、造影剤の投与量や
濃度、投与速度、撮影タイミングといった多くの要因が影響するが、とりわけ、造影剤の投与量
が最も重要な要素である。従来は総体重によって造影剤投与量は規定されてきた。近年、造影効
果のばらつきを軽減するため除脂肪体重や体表面積により造影剤投与量を規定する研究が報告さ
れている。その中で、体表面積法による造影剤投与量を補正する方法は、きわめて簡便に造影の
ばらつきを少なくすることが可能である。一方、患者の体格以外の要素として、慢性肝障害患者
では肝の造影効果が低下すると報告されてはいるが、その程度について十分な検討はなされてい
なかった。現在の多列CTを用いた標準的な造影プロトコールを用いて、造影剤の投与量を体表面
積で補正することにより、肝実質の造影の程度をより正確に評価する事が可能となった。慢性肝
障害患者では、しばしば必要とされる至適な造影効果が得られないことが多く、その肝実質の造
影の程度に関して正確に評価することが必要である。 
本研究の目的は造影剤投与量を体表面積で規定し、標準的なdynamic CTのプロトコールを用
いて、正常肝と慢性肝疾患患者の肝実質造影効果の差異をより正確に評価することである。 
対象は2012年8月から2012年12月までに、同一のプロトコールでdynamic CTを施行された連続
525症例である。この内、肝の造影に影響を及ぼす術後症例など196例を除外した328症例を対象 
氏名 小岩原 元 
した。これらを正常肝（n = 125）、慢性肝炎（以下CH）（n = 92）、Child-Pugh分類grade Aの
肝硬変（以下LC-A）（n = 78）、Child-Pugh分類grade Bの肝硬変（以下LC-B）（n = 33）の4群
に分類した。造影剤は非イオン性ヨード造影剤を使用した。投与量はDu Boisの計算式にて算出
した体表面積法により規定し、体表面積あたり22gI/m2 の造影剤を30秒の固定注入で投与した。
撮像タイミングについてはボーラストリガーテクニックを用い、動脈相、門脈相、平衡相を撮像
した。5mm厚の画像データから、各時相の門脈本幹、腹部大動脈、肝実質に関心領域を置きCT値
を計測した。造影効果については、造影後のCT値から造影前のCT値を差し引いた値（ΔHU）で評
価した。4群間で、各時相において、門脈本幹、腹部大動脈、肝実質のΔHUに有意差があるか否
かについて、一般化推定方程式を用いて検定した。また、門脈相における至適造影（ΔHU > 
50HU)の割合についても検討を加えた。 
門脈相での肝実質のΔHUは正常肝、CH、LC-A、LC-Bで、それぞれ55.5 ± 11.8 HU, 55.2 ± 
12.5 HU, 50.0 ± 12.9 HU, and 43.0 ± 12.7 HUであった。門脈相での肝実質のΔHUは4群間で
有意差があり（p < 0.001）、肝障害が重度になるにしたがい肝実質のΔHUは低下していた。ま
た、動脈相での門脈本幹のΔHUも正常肝、CH、LC-A、LC-B群で、それぞれ肝障害が重度になるに
したがって低下していた（p < 0.001）。他の時相では、肝実質、大動脈、門脈のΔHUに4群間で
有意差を認めなかった。門脈相における肝実質の至適造影とされる50HU以上に達したものの割合
は、LC-A群では55.1％、LC-B群では36.4％で、正常群の66.4%、CH群の66.3％よりも低い割合と
なった。 
肝障害度の進行により肝実質の造影効果が低下する理由として、門脈圧亢進症をきたし門脈血
流が低下するためと考えられた。慢性肝疾患患者、特に肝硬変患者では、門脈相で肝実質の有意
な低下をきたし、しばしば至適な造影効果が得られないことがわかった。至適な造影効果を得る
ためには、造影剤の投与量を増やす方法があるが、造影剤の腎毒性と医療費増大の観点から、こ
れは望ましい手段とは言えない。最新の MDCT 装置では、低電圧撮像により造影能のコントラス
トを上昇させ、逐次近似法によりノイズを低減することで、造影剤の投与量を増加させることな
く、最適な肝実質の造影効果を得ることができ、肝障害度に応じた電圧を選択することで十分な
造影効果を得ることが可能となると思われる。

         審査結果の要旨 
 
植物病害で多く見られる葉の退緑・黄化は光合成を行う葉緑体の障害によって生じため，植物の生産性
低下に直結しており，これを軽減することは作物保護の一戦略となり得る．それゆえ，それら病徴発現
の分子機構を明らかにすることは，新たな耐病性作物の開発の基盤として重要である．カリフラワーモ
ザイクウイルス（CaMV）がコードする多機能性タンパク質，Transactivator/viroplasmin（Tav）について
は，これを発現する形質転換タバコが病徴様の表現型を示すことが明らかにされていた．本研究は，人
為的に発現制御可能な Tav 遺伝子を用い，形質転換タバコにおける病原性機能を解析することによって，
植物ウイルス病における退緑・黄化病徴の発現機構の詳細を解明する端緒を得ることを目的として行わ
れたものである． 
 
１．人為的に発現誘導可能な Tav 遺伝子を持つ形質転換タバコの作出 
CaMV Tav を発現する形質転換タバコが病徴様の表現型を示すことが報告され，この植物を用いるこ
とによって退緑・黄化病徴の発現機構の詳細な研究が可能になると期待された．しかし，Tav の発現を
構成的なプロモーターの制御下で行った場合，病徴様の表現型は植物の生育につれて発現され，病徴発
現に至る分子生物学的な変化を継時的に解析することは困難である．また，病徴様の表現型は子葉にお
いても認められ，その発現に至るシグナル伝達を解析することも困難である．そこで，人為的に発現誘
導の可能なプロモーターの制御下で Tav を発現させることを着想し，デキサメサゾン（Dex）誘導性の
GVG プロモーターの支配下に Tav のコード領域を挿入した形質転換タバコ（iTav タバコ）を作出した．
iTav タバコでは，Dex 処理７日後に肉眼で確認可能な退緑・黄化が観察され，クロロフィル量を継時的
に計測したところ，Dex 処理２日後以降に統計的に有意なクロロフィル量の減少を検出することができ
た．また，光合成関連遺伝子の発現が種々のウイルス感染植物において低下することが知られているこ
とから，iTav タバコにおいて Dex 処理後の光合成関連遺伝子の発現を継時的に検討した．その結果，異
なる iTav タバコ系統において，Dex 処理２～５日後に明確な発現レベルの低下を観察した．これらの結
果は，iTav タバコにおける退緑・黄化病徴様の表現型が種々のウイルス感染植物と共通した分子生物学
 
 
的変化に基いて起こっていることを示唆する． 
Tav を構成的に発現する形質転換タバコにおいて病原性関連タンパク質 PR1a の発現が誘導されるこ
とが報告されている．一方，Tav を一過性に発現させた場合には，病原体関連分子パターン，およびサ
リチル酸（SA）によって誘導される PR1a 遺伝子の発現を抑制することが報告されている．これらのこ
とから，形質転換タバコにおける Tav による PR1a 遺伝子の発現誘導は，病徴様表現型の発現による，
二次的な反応である可能性が考えられた．しかし，iTav タバコにおいて PR1a 遺伝子の発現は Dex 処理
の 1 日後に認められた．この結果は，Tav に対する反応がその発現様式および宿主によって異なること
を示唆する． 
 
２．iTav タバコにおける退緑・黄化病徴様表現型と Tav の病原性機能の関連性についての解析 
上述のように iTav タバコにおいて PR1a 遺伝子の発現が Dex 処理後，速やかに誘導されたことから，
病徴様表現型の発現が宿主の防御応答関連遺伝子の活性化を伴うことが示唆された．そこで，他の病原
性関連タンパク質遺伝子の発現を解析したところ，iTav タバコにおいて Dex 処理後，SA 誘導性および
エチレン誘導性防御関連遺伝子が誘導されることが分かった．これらの応答と Tav の病原性因子機能と
の関連を明らかにする目的で，Tav の欠失変異体を用いて iTav タバコと同様な形質転換タバコを作出し，
退緑・黄化病徴様表現型の発現と PR1a 遺伝子の発現を検討した．その結果，病徴様表現型の発現，お
よび PR1a 遺伝子の活性化が，Tav の病原性因子機能（RNA サイレンシングを抑制する機能，およびト
マト矮化病ウイルス P19 タンパク質によって誘導される細胞死を抑制する機能）と同じドメインによっ
て担われていることが明らかとなった．一方，SA 分解酵素遺伝子の一過性発現によって，病徴様表現
型の発現，および PR1a 遺伝子の活性化が，SA 非依存的に起こることが示唆された．SA 非依存的な
PR1a 遺伝子の活性化はエフェクター誘導性免疫（ETI）においても起こることが報告されていることか
ら，iTav タバコにおける退緑・黄化病徴様の表現型発現は，Tav の病原性因子機能に対する宿主応答の
結果であり，その応答は SA 非依存的な ETI に類似のメカニズムによるものであることが示唆された． 
 
以上のように得られた成果は，ウイルス病徴の発現機構の理解に向けて非常に有用な実験系を提供す
るとともに，病徴発現機構の解明に端緒を与える新規性の高いものである．今後，本研究で確立された
実験系を用いたさらなる研究を通してウイルス病徴発現機構の解明に大きく寄与するとともに，糸状菌
病，細菌病，線虫病および虫害に対する耐性植物を開発しようとする分子育種の分野にも影響を与える
ものと考えられ，評価される． 
 
本論文の公開審査会は平成 27 年 2 月 7 日に愛媛大学農学部において開催された．申請者が論文内容
について発表した後，質疑応答が行われた．同日引き続いて論文審査委員会をあらためて開催し，審査
を行った．これらの結果から本論文は博士（農学）の学位を授与するに値すると審査委員会全員一致し
て判定した．

Cereal grains are generally ground for poultry diets. There has recently been an 
increasing interest in using whole grain in diet in order to decrease feed cost by 
removing the grinding step. Moreover, feeding whole grains to poultry also meets 
consumer demands for a natural feeding system. In Japan, corn has been the major 
ingredient for farmer. However, this became expensive as corn was imported and used 
extensively by other livestock and humans. This situation has led to development of 
whole-grain paddy rice (WPR) as a new source of local poultry feed ingredients.  
Considering the above points, the research in this thesis focused on the 
utilization of local alternative feed ingredients is likely to offer reducing poultry feed 
supply problems as well as to evaluate how alterations would be induced in the growth 
performance and intestinal histology of chickens after feeding WPR. 
At first, the chemical composition, nutrient digestibility, and the metabolizable 
energy (ME) of WPR were determined in order to confirm nutritional values of WPR 
and to provide information on accurate formulation of poultry diets (Experiment 1). 
The results reviewed that the chemical composition of WPR had values close to those of 
corn. The apparent ME and true ME of WPR were 2.79 and 3.02 kcal/g air-dried, 
respectively.  
Dietary dilution is one of management practice to reduce poultry feed cost for 
farmer. Therefore, this experiment was carried out to study the growth performance and 
intestinal histology in chickens fed diets diluted with WPR (Experiment 2). The growth 
performance, the relative length of intestines and relative weight of visceral organs did 
not change except that the weight of gizzard increased in both the 20 and 40%WPR 
groups. In addition, the surface of the villus tip was not damaged after feeding WPR. 
This suggests that chickens can be fed a basal diet diluted with WPR up to a level of

40% without negative effects on growth performance or intestinal histology.  
Experiment 1 suggested that WPR can be used as a good alternative cereal grain 
source in poultry diets, and can replace corn. However, the use of WPR seems to cause 
severe problems due to its high crude fiber and non-starch polysaccharides (NSP). This 
problem may be overcome by dietary enzyme supplementation. Therefore, the effects of 
WPR replacement with or without enzyme addition on broiler performance and 
intestinal morphology were determined (Experiment 3). The finding that broilers fed on 
diets replacing corn with WPR without enzyme showed hypertrophy in duodenal and 
ileal villi and in duodenal and jejunal epithelial cells, especially in the 50WPR group, 
without negatively affecting growth performance. These findings suggest that WPR can 
replace corn up to a level of 50% (283.0 g/kg, starter, and 250.0 g/kg, finisher) in 
broiler diets without enzyme supplementation.  
Based on the results of Experiment 3 that enzyme addition is not beneficial to 
the WPR-based diets, Experiment 4 was carried out to study the effect of replacing corn 
in laying hen diets with WPR up to a level of 100% on egg production performance and 
quality without enzyme supplementation. It reveals that WPR can totally replace corn in 
laying hen diets without harming egg production performance and quality.  
In the above experiments, the rations were made isocaloric by increasing the 
level of animal fat to elevate the WPR level in the diets, resulting in oily diet. Therefore, 
whole-grain brown rice (BR) was supplemented to reduce animal fat in the diet. The aim 
of this study was to investigate replacing corn in the diets with different levels of WPR 
and its effect on growth performance and intestinal histological structures of broiler 
chickens (Experiment 5). The results showed that replacing corn in broiler diets with 
WPR and BR led to the increased size of gizzard in addition to few changes in intestinal 
morphology. These findings suggest that WPR and BR can totally replace corn in broiler 
diets without negatively affecting growth performance. 
Finally, the results from this thesis demonstrate that WPR can use as a feed 
ingredient in poultry diets. This development of WPR as a new alternative for poultry 
feed ingredients would be highly evaluated for poultry industry.

希少糖とは，自然界に存在量が少ない単糖およびその誘導体と定義されている．この希少糖の一つ
である D-プシコースは，自然界に存在量が少ないことから，これまで栄養学の研究対象とならなか
った．しかし，近年，D-フルクトースを希少糖 D-プシコースに変換する新規酵素 D-タガトース 3-エ
ピメラーゼが発見され， D-プシコースの生理作用に関する研究成果が報告がされるようになった． 
D-プシコースを甘味料として利用することを考えた場合，嗜好飲料や調味料など様々な食品に利用
されて，ほぼ生涯にわたり摂取されることを想定する必要がある．また，これまで実施された D-プ
シコースが糖代謝に与える影響を調べた研究は，主に若年から成熟ラットを用いた動物実験での，D-
プシコースの摂取期間が数週から長くても数ヶ月間である場合がほとんどである．このため，D-プシ
コースを甘味料として普及させるためには，さらに長期間の糖代謝への影響を検討する必要性が考え
られる．  
そこで本研究では，肥満・糖尿病を発症しやすい高脂肪食に D-プシコースを添加し，ラットに生
涯摂取させることによる生体への影響を調べることを目的とした．そのために，まず，ラットへの
D-プシコースの投与量を決定した(第 2章)．さらに，D-プシコースが生存期間に及ぼす影響(第 3章)
および糖代謝に及ぼす影響(第 4章)を調べた．特に加齢に伴う糖代謝の低下を抑制できるか否かを検
討することで，生涯にわたる安全性と有効性を明らかにすることを目指した． 
ラットへの D-プシコースの投与量について(第 2章)は，3週齢の Wistar系雄ラットに D-プシコー
スを市販飼料に 0.3％，1.0％，3.0％の割合で添加した食餌を摂取させる群を設定し，対照群には市
販飼料を摂取させ，6ヶ月(24週)間長期摂食試験を実施した．その結果，体重増加量，摂取エネルギ
ー量および摂取エネルギー効率には各群間に有意差は認められなかった．小腸相対重量は対照群に比
べて各 D-プシコース添加群で有意に低値を示し，肝臓および腎臓相対重量は 3.0％ D-プシコース添
加群で有意に高値を示したが，関連する血清生化学検査結果には各群間で差を認めなかった．以上の
結果から，長期摂食試験において飼料の 3.0％の D-プシコース添加量が妥当であること，および D-
プシコースの 6ヶ月(24週)間長期摂取における安全性が示された． 
D-プシコースがラットの生存期間に及ぼす影響について(第 3 章)は，第 2 章で決定した 3%D-プシ
コース添加飼料をラットに生涯摂取させ，寿命への影響を調べた．Wistar系雄ラット 61匹を，高炭
水化物食(HC)群，高脂肪食(HF)群，3％D-プシコース添加高脂肪食群(HFP)群に分け，4週齢から死亡
するまでの生存期間を比較した．その結果，飼育 24ヶ月(96週)までの体重は，HC群に比べて HF群，
HFP群で有意に高値であったが，30ヶ月(120週)における血清生化学検査値に有意な差は見られなか

った．平均生存期間には各群間に有意な差を認めなかったが，HFおよび HFP群に比べて HC群で短い
傾向にあった．以上の結果から，少量の D-プシコースを生涯にわたって摂取しても安全性に問題が
ないことが示唆された． 
ラットの糖代謝に及ぼす影響について(第4章)は，3週齢のWistar系雄ラット61匹を3群に分け，
それぞれ高炭水化物食(HC)，高脂肪食(HF)および 3%D-プシコース添加高脂肪食(HFP) を自由に与え，
全てのラットが死亡するまで飼育した．実験開始 6ヶ月(24週), 18, 24ヶ月(72，96週)後に経口糖
負荷試験(OGTT)を, 12ヶ月(48週), 18ヶ月(72週), 24ヶ月(96週), 30ヶ月(120週)後に血液生化
学試験を実施した．24ヶ月(96週)までの体重は，HC群に比べて HF群および HFP群で有意に高値で
あったが，HFP 群と HF 群の間には差を認めず，D-プシコース添加による抗肥満作用は認められなか
った．OGTTにおいて，血糖値および血漿インスリン濃度の経時的変動，血糖値時間曲線下面積(AUC)
には各群間に差は見られなかったが，インスリン AUC およびインスリン抵抗性の指標である HOMA-R
値は，HFP，HF，HC群の順に低い傾向にあった．また血液生化学検査において，検査時期に関わらず
糖代謝および脂質代謝指標には，HF 群および HFP 群の間に有意な差を認めなかった．以上の結果か
ら，非常に長期にわたり D-プシコース添加高脂肪食をラットに摂取させた場合，D-プシコースによ
る糖代謝改善作用を確認できないことが示された． 
本研究により，D-プシコースを生涯にわたり摂取させた場合の安全性が確認されたものの，寿命延

マメ科植物は根粒菌が感染すると根に根粒を形成する。根粒菌は植物細胞内へ侵入するとバクテロイ
ドとして分化し、宿主マメ科植物と協調して効率よく窒素固定を行っている。老化に従い、窒素固定能
は減少し多くの遺伝子発現の挙動が観察される。本研究ではマメ科植物ミヤコグサを利用し、根粒老化
のメカニズムを解明することを目的とした。 
本論文の概要は以下の通りである。 
１）共生窒素固定で生成したアンモニアは植物側へ輸送され、グルタミン合成酵素（GS）によってグル
タミン酸をグルタミンに変換することで有機化合物として同化される。本研究では根粒菌の GS に
着目しその機能について調べた。根粒菌の GS は isogene を構成している。STM（Signature Tagged 
mutagenesis）変異体のなかからそれぞれの GS 遺伝子に変異がある GS 変異株を検索し、それら変
異体の共生窒素固定能について調べた。その結果、GSI 変異体をミヤコグサに感染させると着生し
た根粒は緑色で小さく、窒素固定活性が低くなりその結果葉も黄化が観察された。この結果から根
粒菌 GSI 酵素が欠損すると早期老化現象が確認され、根粒菌 GSI は共生に重要であることが明らか
となった。 
２）根粒老化について詳細に調べるために、マイクロアレー解析を行い網羅的な発現を調べた。共生窒
素固定活性が高い根粒菌感染後２週目および４週目の根粒と、老化が確認できる８週目の根粒から
RNA を抽出して解析を行った。RNA-seq の解析から、根粒菌感染後２週目で発現する 20,165 個の遺
伝子を選抜した。１遺伝子に対し 60mer の２プローブ作成後、アジレント社の 4×44K プレートを
カスタムアレイにより作成した。その結果、根粒菌感染後 8 週目に 2 倍以上発現が誘導する遺伝子
641 個を決定した。NCBI の euKaryotic Orthologous Groups (KOGs)に応じて機能カテゴリー別に分
類した結果、55.4%の遺伝子が既知の遺伝子と相同性を示した。この遺伝子群には老化誘導性遺伝
子、NAC domain containing protein, 老化誘導型 heat shock protein、cysteine protease など
が含まれることが明らかとなった。一方、老化時に発現が２倍以上減少する遺伝子として 416 個同
 
 
定した。この遺伝子群には共生窒素固定に必要な遺伝子が同定できた。得られた結果の信頼性を確
認するために３２個の遺伝子については RT-PCR により確認を行った。 
３）マイクロアレー解析の結果からフェリチン遺伝子に着目した。フェリチンは鉄含有タンパク質とし
て知られているが老化により発現が増加する遺伝子として同定した。根粒の鉄含量を測定した結果、
老化に伴い鉄含量が減少していることが明らかとなった。レグヘモグロビンタンパク質は、根粒全
タンパク質の 10-30%を占めるヘムタンパク質である。しかし老化時に鉄が遊離しフェリチンとして
地上部に転流しているのではないかと考えた。根粒を鉄イオン、また、NO ドナーである SNP を含む
培地で育てるとフェリチン遺伝子発現が増加し、また、高濃度の鉄イオンを含む培地で育てた根粒
は NO が発生している事が明らかとなった。以上の結果より鉄イオンは NO シグナルを介してフェリ
チン遺伝子発現を誘導することが明らかとなった。老化により必要なくなったレグヘモグロビンは
鉄を遊離し、フェリチンとして地上部へ転流していることを示唆する結果となった。 
以上のように、本論文ではマメ科植物ミヤコグサを用いて根粒老化により誘導する遺伝子を明らかにし
ており、今後の研究につながる価値ある新しい知見を得ている。 
本論文に関する公開審査会は2015年2月7日、愛媛大学農学部で開催され、申請者の論文発表と質疑応答が
行われた。引き続いて開催された学位論文審査委員会において慎重な審議を行った結果、審査委員全員一致
して本論文が博士（農学）の学位を授与するに値するものと判定した。

  Obesity and diabetes mellitus are two major lifestyle diseases in the world not only in leading 
countries but also in developing countries. Type 2 diabetes mellitus, a metabolic disorder 
characterized by insulin resistance or low insulin levels or both, is linked to inflammation and 
oxidative stress, and is a leading cause of cancer, cardiovascular disease and other diseases. A 
therapeutic approach for postprandial hyperglycemia is to control the blood glucose level by retarding 
carbohydrate digestion into glucose with α-amylase and α-glucosidase inhibitors. Additional, the 
prevalence of TypeⅠallergy, such as pollinosis, allergic rhinitis and allergic asthma has been 
increasing in the recent decades in the leading countries. Japanese cedar pollinosis occurs in spring 
causing the typical symptoms of seasonal allergic rhinitis, such as sneezing, rhinorrhea, nasal 
obstruction, nasal itching, and itching of the eyes. Allergic diseases such as rhinitis, which with the 
high incidence, affects social life, sleep, school and work performance, is also likely to cause a 
substantial economic burden on society. 
   Perilla frutescens (L.) Britt. (Lamiaceae) is an edible plant frequently used in some Asian 
countries such as China, Korea and Japan. Perilla leaves are used as a fresh vegetable and a food 
colorant in Japan and China, because of its bright red color, due to the presence of major anthocyanins, 
malonylshisonin and shisonin, and other related anthocyanins compounds. Natural resources of 
anthocyanins are urgently required for safe colorants and additives in the food industry. Moreover, 
phenolic compounds in Perilla leaves such as rosmarinic acid (RA), luteolin, apigenin, and α-linolenic 
acid have an antiallergic, antiinflammatory, and antioxidant properties, have a neuroprotective effect, 
act against tumors and suppress diabetes mellitus. RA, a typical phenolic acid in the Lamiaceae family,

is potentially valuable for improvement of diabetes mellitus and obesity by inhibiting digestive 
enzyme.  
Supramolecular pigments, found in blue flower petals, such as Commelina communis, Centaurea 
cyanus, Salvia patens, Salvia uliginosa, and Nemophila menziesii, are the complex of anthocyanins, 
flavonoids and metals by intermolecular hydrophobic association and metal complexation of their 
anthocyanins. Recently, Asada et al. applied aluminum complex formation, based on supramolecular 
formation, to selectively purify bilberry anthocyanin 3-glycosides from the crude pigments in order to 
get biological active anthocyanins.  
In this study, we tend to isolate some active fractions/compounds for the treatment of obesity, 
allergy, and other biological activities using the novel technique of supramolecular, and to improve the 
application value of Perilla leaves.     
In chapter 2, we successfully applied supramolecular technology in RA extract separation from 
Perilla leaf extract. By the supramolecular complex which was formed from flavocommelin and 
Perilla leaf extract as initial materials, the supernatant containing rosmarinic acid was isolated. 
Rosmarinic acid extract (62.9% purity) was further and partly purified by partitioning ethyl acetate 
and water (Figure 1).  
RA extract isolated from Perilla leaf extract using the supramolecular technique showed the greatest 
Total phenolics (TP), 433.9 ± 58.6 mg/g gallic acid equivalent (GAE) among the supramolecular 
products (Figure 1). The TP of standard RA (711.8 ± 5.3 mg/g GAE) was higher than RA extract. The 
TP of RA extract was calculated to be 61.0% of standard RA. Therefore TP of RA extract was 
interpreted by the purity of RA (62.9 ± 4.5%) in the extract. In addition, RA extract exhibited the 
highest DPPH radical scavenging activity (DRSA) with an SC50 value of 5.5 ± 0.2 μg/mL, which was 
more than two times higher DRSA than Perilla leaf extract (SC50 value, 10.8 ± 0.1 μg/mL).

For antiallergic activity, RA extract (52.9 ± 6.7 μg/mL) suppressed β-hexosaminidase release 3.9 
times and 2.6 times more than Perilla leaf extract and standard RA, respectively, and exhibited the 
highest antiallergic activity. Also, it is suggested that other compounds as well as RA in RA extract 
may contribute to antiallergic activity. Rosmarinic acid methyl ester (RA-me) was found as a minor 
component in RA extract suppressed β-hexosaminidase release with IC50 of 9.9 ± 0.8 μg/mL. At IC50 
of RA extract (52.9 μg/mL), it was calculated as 33.3 μg/mL for RA and 3.7 μg/mL for RA-me, 
respectively, which contributed to 43% β-hexosaminidase release inhibition as an additive effect. 
Therefore, RA-me, as a component in RA extract, contributed to high β-hexosaminidase release 
inhibitory activity besides RA. We found the greatest activity of RA-me in RA extract and supernatant 
as a minor component. 
For α-glucosidase inhibitory activity, RA extract inhibited 4.1 times and 1.8 times higher than 
standard RA (IC50 value, 0.95 ± 0.01 mg/mL) and Perilla leaf extract (IC50 value, 0.42 ± 0.01 mg/mL), 
respectively, although RA is reported to be a better research target on α-glucosidase inhibitory activity. 
It is found that IC50 values of caffeic acid and RA-me as minor components in RA extract were 3.46 ± 
0.18 mg/mL and 0.44 ± 0.01 mg/mL, respectively. It showed that RA-me exhibited greater 
α-glucosidase inhibitory activity than standard RA. We concluded that RA extract containing RA-me 
and other minor components as unknown compounds besides RA contributed to high α-glucosidase 
inhibitory activity, especially unknown compounds might highly contribute to the activity.  
Through supramolecular formation, RA-containing supernatant was excluded by supramolecular 
complex and RA extract (62.9 ± 4.5% purity) with potent biological activities was obtained. The 
obtained RA extract had DPPH radical scavenging activity, antiallergic activity and α-glucosidase 
inhibitory activity. RA extract exhibited higher antiallergic activity and α-glucosidase inhibitory 
activity than standard RA. This study shows that RA and other minor components in RA extract 
contributed antiallergic activity and α-glucosidase inhibitory activity. We will discuss molecular 
mechanisms of the activities in detail by linking with the interaction of biologically important 
chemicals after identification of these active compounds. In conclusion, the RA extract isolated from 
Perilla leaf extract by supramolecular formation and solvent extraction has potential applications in 
managing diabetes mellitus and other chronic and degenerative diseases.  
  As rosmarinic acid-rich extract with high α-glucosidase inhibitory activity was isolated on a large

scale using a supramolecular separation method and the contribution of rosmarinic acid methyl ester, a 
minor component in the extract contributed to α-glucosidase inhibitory activity more than double that 
of rosmarinic acid, in Chapter 3, we investigated the esterification of rosmarinic acid and its related 
compounds as an approach to optimize α-glucosidase inhibitory activity as a leading compound to 
explore potentially attractive therapeutic reagents for diabetes mellitus. Methyl, propyl and hexyl 
esters of rosmarinic acid (RA), caffeic acid (CA) and p-coumaric acid (Cou) synthesized were 
assessed α-glucosidase inhibitory activities.  
Rosmarinic, caffeic and p-coumaric acids esterified with methyl, propyl and hexyl chains showed 
great α-glucosidase inhibitory activity. Rosmarinic acid hexyl ester exhibited the greatest 
α-glucosidase inhibitory activity with IC50 values of 0.05 mM for B. stearothermophilus source, and 
0.61 mM for S. cerevisiae source. The α-glucosidase inhibitory activity of these esters was greatly 
related to alkyl chain length of each ester series, and the correlation between LogP (octanol-water 
partition coefficient) and pIC50 values (−logIC50) of all these compounds was observed as positive 
correlation coefficients, 0.80 for α-glucosidase of B. stearothermophilus source (p<0.001) and 0.46 for 
S. cerevisiae source (p<0.05). Furthermore, the inhibition kinetics and interaction mechanism of 
rosmarinic acid hexyl ester was investigated. It was found that rosmarinic acid hexyl ester was a 
mixed inhibition type, with the inhibition constants, Ki of 0.24 mM (for the binding with the free 
enzyme), and Ki’ of 0.80 mM (for the binding with the substrate-enzyme complex). It is suggested that 
rosmarinic acid hexyl ester has a strong binding affinity with the free enzyme. Additionally, the 
molecular docking result shows that rosmarinic acid hexyl ester was highly fitting to the active site 
pocket by the hydrophobic interaction and hydrogen bonding of 3,4-dihydroxyphenyl group with 
amino acids surrounded.  
Elongation of alky chain of rosmarinic acid, caffeic acid and p-coumaric acid esters in accompany 
with an increase of hydrophobicity, enhanced α-glucosidase inhibitory activity. Rosmarinic acid hexyl 
ester with great inhibitory activity is supposed to be a high binding affinity with the enzyme pocket. 
The molecular docking shows that hydroxyl groups forming hydrogen bonding with the important 
amino acid residues, surrounding by the hydrophobic amino acids, fitting to the active site pocket of 
α-glucosidase, which may lead to great inhibitory activity of rosmarinic acid hexyl ester on 
α-glucosidase. The inhibition kinetics and molecular docking of rosmarinic acid hexyl ester facilitate

its inhibition mechanism evaluation. The further study of rosmarinic acid hexyl ester on its toxicity, in 
vivo test and food/medicinal application should be deeply investigated.  
In chapter 4, methyl, propyl and hexyl esters of rosmarinic, caffeic and p-coumaric acids were 
tested for antiallergic activity using β-hexosaminidase release suppression. Among all free acids and 
esters, rosmarinic acid propyl ester (RA-pro, IC50, 23.7 μM) exhibited the greatest suppression on 
β-hexosaminidase release, followed by rosmarinic acid hexyl ester (RA-hex, IC50, 34.5 μM), caffeic 
acid propyl ester (CA-pro, IC50, 55.3 μM) and rosmarinic acid methyl ester (RA-me, IC50, 57.3 
μM).The activity of rosmarinic acid propyl ester was only 9.1 times lower than luteolin (IC50, 2.6 μM), 
a flavonoid with high antiallergic activity that was tested in a parallel experiment of our laboratory, but 
increased 55.4 times compared to the activity of rosmarinic acid. 
Furthermore, strong quadratic correlations were observed between LogP and pIC50 (−logIC50) for 
each of the acid-esters series, with correlation coefficients equal to 0.94 (p < 0.05) for the rosmarinic 
acid series, 0.98 (p < 0.01) for the caffeic acid series and 1.00 (p < 0.01) for the p-coumaric acid ester 
series. The quadratic equations reflect the lowering of antiallergic effects of hexyl esters after the very 
strong effects of shorter esters. Our results showed that the hydrophobic propyl esters of RA, CA and 
Cou strongly suppressed β-hexosaminidase release, thus it is possible that propyl esters have higher 
affinity to the cross-linked IgE/FcεRI receptor while the bulky hexyl esters would bind less efficiently. 
Esterification of rosmarinic, caffeic and p-coumaric acids with short alkyl chains influences their 
antiallergic activity. Propyl esters exhibited the highest antiallergic activity. Hydrophobicity and steric 
bulkiness of the alkyl chain are likely to be modulating the interaction between phenolic acid/esters 
and the binding sites in the IgE/FcεRI receptor. Furthermore, our data indicates that the 
3,4-dihydroxyphenyl moieties in phenolic ester backbone could be essential for the inhibitor-binding 
site interaction. The elucidation of mechanism underlying the interactions between phenolic 
acids/esters warrants further investigations. 
In conclusion, rosmarinic acid extract with high biological activities including DPPH radical 
scavenging activity, antiallergic activity and α-glucosidase inhibitory activity, can be easily extracted 
from Perilla leaves using a supramolecular technique with a newly-established isolation method. 
Rosmarinic acid extract shows high potential for diabetes mellitus and allergy treatments by inhibiting 
α-glucosidase activity and measuring β-hexosaminidase, related to life-style disease. Esters of

rosmarinic acid and its related compounds exhibited enhanced α-glucosidase inhibitory activity and 
antiallergic activity. Elongation of alky chain of rosmarinic acid, caffeic acid and p-coumaric acid 
esters in accompany with an increase of hydrophobicity, enhanced α-glucosidase inhibitory activity. 
Rosmarinic acid hexyl ester with great inhibitory activity is supposed to be a high binding affinity with 
the enzyme pocket. The antiallergic activity of these compounds is modulated by their hydrophobicity 
and bulkiness of alkyl chain and rosmarinic acid propyl ester exhibited the greatest β-hexosaminidase 
release suppression. The further study of the active rosmarinic acid esters on its toxicity, in vivo test
