【背景と目的】 
 非アルコール性脂肪性肝疾患(NAFLD)は全国で約 1000 万人いると推計され、最も頻度高い
肝疾患であり、そのうち 10%は非アルコール性脂肪肝炎（NASH）を発症して、肝における炎
症、線維化を引き起こし肝硬変，肝細胞癌に進展する。しかし、NAFLD患者から NASH を発
症する背景、機序は明らかではない。申請者らは 13C標識試薬吸収試験を用いて消化管内視
鏡下に投与することにより、正確に脂肪酸吸収動態を経時的に解析できる測定系を構築して
いる。脂肪酸の中でも長鎖脂肪酸は 肝臓における炎症、酸化ストレス、発癌に関与するこ
とから、NASH病因の 1つと想定される。しかし、NASH における長鎖脂肪酸の腸管吸収動態
を評価した報告はない。本研究では NASH患者における長鎖脂肪酸の吸収動態を明らかにす
るとともに、小腸粘膜生検組織を用いて，脂肪酸吸収関連細胞内分子の変化を検討し、NASH
の発症および病態との関連について明らかにすることを研究の目的として解析した。 
 
【方法】 
１、13C標識脂肪酸試薬を用いた 13C呼気試験法 
愛媛大学臨床倫理委員会にて承認を得た後、健常者群 33名、NASH患者 73名（線維化   
stage 1,2：e-NASH 32名、線維化 stage 3,4：a-NASH 41 名）を対象とした。長鎖脂肪酸で
ある[1-13C] sodium palmitateを上部消化管内視鏡を用いて十二指腸水平脚に投与し、呼気
中の 13CO2排出量を経時的に解析した。 
 
２、食餌摂取後の血清 apoB-48の変化 
試験食摂取後、経時的に採血して血清 apoB-48を血清カイロミクロンの動態の指標として
評価した。 
 
３、小腸粘膜組織を用いた脂肪酸吸収関連分子の発現変化と血漿 glucagon-like  
-peptide-2(GLP-2)の定量 
同臨床倫理委員会にて承認を得た後、小腸内視鏡で得た生検検体を用いて、脂肪酸吸収に
関連する細胞内分子の real-time RT-PCRとウエスタンブロットを行った。CD36 を活性化 
(glycosylation)する血漿 GLP-2濃度を各症例で定量した。 
 
４、glycosylated CD36の免疫組織学的評価 
 小腸生検検体を用いて、リンパ管、毛細血管、および脂肪酸輸送蛋白である 
glycosylated CD36 の免疫組織学的評価を行った。  
 
５、脂肪酸吸収量と臨床パラメーター・組織学的肝臓線維化との相関分析 
 血液検査、非侵襲的画像検査から得られた臨床検査値と脂肪酸吸収量との相関を解析した。
さらに、肝生検組織の線維化 stage毎の脂肪酸吸収量を比較した。 
 
【結果と考察】 
① NASH 群において、[1-13C] sodium palmitate (長鎖脂肪酸)投与早期(0-120分)の  13CO2
呼気排出量は有意に増加した(p<0.01)。e-NASH群では投与後期(120-360分)の 13CO2呼気
排出量は増加したが、a-NASH群は低下した。 
② 試験食負荷後の血清 apoB-48濃度は、e-NASH 群で有意に高値であった(p<0.05)。 
③ CD36、MTTP の mRNA、glycosylated CD36、MTTPのタンパク発現は NASH群で有意に増加
した。また、血漿中の GLP2は NASH群で有意に増加しており、NASH 患者における 
glycosylated CD36増加への関与が考えられた。  
④ NASH 群では血管上皮と絨毛上皮における glycosylated CD36 の増加が見られており、 
NASH では glycosylated CD36を介して門脈血管へ直接脂肪酸が流入する経路が想定され
た。 
⑤ e-NASH 群では投与早期の 13CO2排出量と肝脂肪化・線維化に正の相関がみられ、線維化 s
tage 1 と stage 2に分けると、投与早期の 13CO2排出量は stage 2 で有意に増加してい
た。また投与後期の 13CO2排出量はインスリン抵抗性に正の相関がみられた。 
 これらの結果から、e-NASHでは長鎖脂肪酸の吸収が増加し、その機序として MTTP 増加を
伴うカイロミクロン合成増加と glycosylated CD36 の発現増加の関与が考えられた。特に投
与早期の長鎖脂肪酸吸収は肝線維化にともない増加し、吸収経路の変化の影響が考えられた。 
 
【結論】 
 NASH、とりわけ病初期の NASHにおいて、長鎖脂肪酸の吸収が亢進しており、特に投与早
期の亢進が顕著であった。この吸収動態に腸細胞内分子の発現変化、活性型 CD36 の増加が
関与し、また、投与早期(120分以内)と後期(120-360 分)での吸収動態変化は、肝脂肪化、
肝線維化、インスリン抵抗性と関連していた。NASH の発症、病態形成に長鎖脂肪酸吸収変
化が関与している可能性が考えられた。

【背景】 
 心房細動(AF)は心房筋の電気的リモデリングによる伝導遅延をもたらし、発作の頻度と持続時
間が増すとともに、構造的リモデリングによる収縮不全をおこし、心不全、心房内血栓症に伴う
血栓塞栓症などの重篤な心血管イベントに繋がる。 
 経胸壁心エコー検査 (TTE) における組織ドプラ法は優れた時間分解能で局所心筋の動態を簡
便に評価できる。近年、心電図の P 波開始点から組織ドプラ法で求めた僧帽弁輪側壁部における
心房収縮期僧帽弁輪部移動速度波形 (a’) までの時間（EMT）は心房内伝導遅延と関連し、新規
AF の発症予測や電気的除細動後の再発予測の指標として報告されている。しかし心房拡大を有
した AF 症例において、組織ドプラ法を用いた僧帽弁輪側壁部における心房収縮期僧帽弁輪部移
動速度波形 (a’) が二峰性に分かれた、すなわち ‘double peak sign （DP-sign）’を有す
る症例が散見され、EMT の計測に難渋する。 
 一方で speckle tracking 法 (STE)は局所心筋の「収縮拡張動態」が評価可能であり、心房筋
における peak late diastolic longitudinal strain は長軸方向の収縮の終点と考えられる。P
波開始点から左房側壁基部 peak late diastolic longitudinal strain までの心房内伝導−収縮
時間(EMT-ε)は心房内収縮時間であり、この時間が心房筋の電気的、構造的リモデリングを表す
指標を考えられるが、検討した研究は少ない。 
【方法】 
 当院で心房細動アブレーションを行った連続 46 例（平均年齢 62 歳、女性 26%、発作性 31 例、
持続性 15 例）に対して術後 24 時間以内と 6 ヶ月後に TTE (Toshiba 社製 Artida) を行った。左
室駆出分画が 50%以下の左心機能低下例、有意な心臓弁膜症を有する例を除外した。左室四腔断
面像から組織ドプラ法を用いて P 波開始点から僧帽弁輪側壁部心房収縮期速度 (a’)までの時間
を EMT-a'と定義した。一方で speckle tracking 法 (STE)は局所心筋の「収縮拡張動態」が評価
可能であり、心房筋における peak late diastolic longitudinal strain は長軸方向の収縮の終
点と考えられる。P 波開始点から左房側壁基部 peak late diastolic longitudinal strain まで
の心房内伝導−収縮時間(EMT-ε)を測定し、組織ドプラ指標と併せて電気生理学的指標の比較検
討を行った。心房細動アブレーション術後 24 時間以内と 6 ヶ月後の TTE において EMT-εを測定
し、組織ドプラ指標と併せて比較検討を行った。 
 
【結果】 
 10 例の control 群に比べ、心房細動群において LAVI は有意に拡大し、EMT-a'、EMT-εは有意
に延長していた。心房細動アブレーション術直後の EMT-εは EAT (R2=0.35, P<0.001) と強い相
関を認め、左房容量係数とも相関 (R2=0.18, P<0.01) していた。術後 6 ヶ月後において EMT-a'
は有意な改善を認めなかったものの、左房容量係数の縮小とともに EMT-εは有意に改善した。 
 
【結語】 
 Speckle tracking 法による心房内伝導−収縮時間(EMT-ε)は心房内伝導−収縮遅延を示唆する
簡便で有用な指標であり、将来慢性期のリモデリングの評価に有用な指標と思われた。

The term of allelopathy is derived from the Greek’ root words allelon, 'of each other', and pathos, 'to suffer', 
hence it means, the injurious effect of one upon another. As a sub-discipline of chemical ecology, the study of 
allelopathy is concerned with the influence of biochemicals produced by one organism on the growth, survival, 
development, reproduction and distribution of the other organism in an ecosystem. In this case, the biochemicals, or 
known as allelochemicals may have beneficial or detrimental effects on the target organisms (Cheng & Cheng, 
2015). The term of allelopathy to refer to any process, which involving secondary metabolites produced by plants, 
microorganisms, viruses and fungi that influence the growth and development of other biological systems, in 
addition, the donor and receiver of allelopathic should include animals (Rice, 1984). Plants are biochemical 
factories. Biochemicals with allelopathic potential may existent in all part of plant tissues, including leaves, stems, 
roots, rhizomes, flowers, fruits, and seeds. Allelochemicals may be released from plant tissues in a variety of ways, 
including volatilization, root exudation, leaching, and decomposition of the plant residues (Putnam, 1985). 
As noted by Blum (2014), both organic and inorganic compounds are released from plants to the soil by 
active and passive processes (Figure 1). Active processes include secretion by glands and hairs, secretion and 
exudation of high molecular compounds by cells particularly root cells, and the active transport of organic 
compounds across cell membranes. Passive release includes exudation of low molecular weight compounds by 
cells, sloughing and lysis of cells, particularly root cells, abscission of vegetative and reproductive tissues and 
organs, leaching of living, dying, or dead tissues such as leaves, stems, roots, flowers, and fruits by condensates or 
rain events, and loss of volatile compounds by living and dead vegetative and reproductive tissues. As described by 
Rizvi, Haque, Singh, & Rizvi, (1992), the mode of action of allelochemicals can be divided into indirect and direct 
action. Indirect action may include effects through alteration of soil property, its nutritional status and altered

population and/or activity of harmful/beneficial organisms like microorganisms, insects, nematodes, etc. Whereas, 
the direct mode of action, which includes effects of allelochemicals on various aspects of plant growth and 
metabolism. The following are some important processes known to be influenced by allelochemicals, including (1) 
cytology and ultrastructure, (2) phytohormones and their balance, (3) membrane and its permeability, (4) 
germination of pollens/spores, (5) mineral uptake, (6) stomatal movement, pigment synthesis and photosynthesis, 
(7) respiration, (8) protein synthesis, (9) leghaemoglobin synthesis and nitrogen fixation, (10) specific enzyme 
activity, (11) conducting tissue, (12) water relations of plants, and (13) genetic material.

The allelochemicals with allelopathic effects are being explored intensively to reduce the dependency on 
synthetic pesticides in plant pest and diseases management. The present study examined allelopathic effects of sea 
mango (Cerbera manghas), ferntree (Filicium decipiens), and Madeira vine (Anredera cordifolia) from Indonesia 
(Figure 2). P-hydroxybenzaldehyde, benzamide, n-hexadecane acid monoglyceride, loliolide, β-sitosterol, cerberin, 
neriifolin, cerleaside A, and daucosterol were identified in sea mango (Zhang, Pei, Liu, Kang, & Zhang, 2010). 
Sitosterol  -D-glucoside, 3-O--D-glucopyranosyl kaempferol, 3-O--D- glucopyranosylquercetin and 
3-O--L-rhamnopyranosyl(1  2)--D-glucopyranosylkaempferol were identified in ferntree (Jayasinghe, 
Balasooriya, Bandara, & Fujimoto, 2004). Saponins, alkaloids, polyphenols, flavonoid and mono polysaccharide 
including L-arabinose, D-galaktose, L-rhamnose and D-glucose were identified in Madeira vine (Astuti, Sakinah 
A.M, Andayani B.M, & Risch, 2011). The three plants are known as medicinal plants locally and the allelopathic 
potential of the plants are unidentified to date. The experiments used leaves as plants material because the leaf is one 
of main sources of allelopathic compounds (Khalid, Ahmad, & Shad, 2002; Rice, 1984). Whereas, fresh fruit of sea 
mango was used for the experiment of R. argentiventer repellent due to local experience of Indonesian farmers. 
The results the experiments demonstrated that the aqueous methanol extracts of sea mango, ferntree and 
Madeira vine leaves had inhibitory effects on seedlings growth of test species, including monocot species (barnyard 
grass, foxtail fescue, Italian ryegrass, and timothy) and dicot species (alfalfa, garden cress, lettuce and rapeseed) 
under laboratory conditions. The obtained results indicated that the seedling elongation varied depending on test 
species and also the extract concentrations, whereas the sensitivities of shoots and roots were different among 
species, and the inhibition of seedling elongation increased along with the increase of the extract concentrations in 
all test species, in both case of shoots and roots. At some lower extract concentration, growth promotion was 
observed at root of lettuce and shoot of barnyard grass and foxtail fescue. The similar phenomenon was reported in 
case of the extract of Cymbopogon nardus (Suwitchayanon, 2013), Leucas aspera (Islam & Kato-noguchi, 2012) 
and Aglaia odorata (Kato-Noguchi et al., 2016). Dose response phenomenon characterized by a low dose 
stimulation and a high dose inhibition are known as hormetic responses. The phenomenon of phytotchemical 
phytotoxins and hormesis were also reviewed by Duke, whose generated a special issue dealing with dose-response 
aspects of allelochemicals (Duke, 2011). In Conclusion, these results suggested that sea mango, ferntree and 
Madeira vine may possess allelopathic substances. The crude extract of these plants leaves could be recommended 
to apply directly as natural herbicide or the residue also could be applied as mulch cover for weed control in farming 
practices, for practical purposes.

The results of experiment of the sea mango also suggested that sea mango fruit can be used as a natural 
repellent for R. argentiventer. Symptom of stress detected in the behaviour of R. argentiventer. Stress causes various 
physiological process disorders in mammals, including sleep–wake cycles, hormonal and nervous activity, body 
temperature, and exhibiting rhythmic changes over the course of 24h (Barker, Bobrovskaya, Howarth, & Whittaker, 
2017; Hurtubise & Howland, 2016; Tahara, Aoyama, & Shibata, 2016). In these experiments, the comparison of 
metabolic parameters of R. argentiventer between with and without sea mango fruit treatments indicated that sea 
mango caused metabolic disorders. Sea mango fruit treatment decreased food and beverage intake by R. 
argentiventer and decreased its body weight significantly. Comparing the daily activities between with and without 
sea mango fruit treatments, R. argentiventer spent more time for locomotion and less time for foraging and resting. 
The results indicated that sea mango fruit has a potential as repellent for R. argentiventer. To date, sea mango fruit 
has no economic value yet in Indonesia. The fruits have not been utilised for any purposes. Farmers can find the 
fruits under their trees freely and easily. For practical purposes, farmers can put the sea mango fruit directly at any 
places where R. argentiventer are found.

In this study, 3-hydroxy-α-ionone was isolated from Madeira vine leaf (Figure 3). The C13H2102 was 
determined as molecular formula of the allelopathic substance, which was characterized by HRESIMS at m/z 
209.1530 [M+H]+ (calcd for C13H2102, 209.1542, Δ = -1.2 mmu). The 
1H NMR spectrum (400 MHz, CDCl3 as 
internal standard) showed δH 6.53 (dd, J= 15.9, 10.3 Hz, I H, H-7), 6.10(d,J= 15.9, I H, H-8), 5.63 (br s, I H, H-4), 
4.27 (br s, I H, H-3), 2.50 (d,J= 10.3, I H, H-6), 2.26 (s, 3 H, H-10), 1.84 (dd, J = 13.9, 6.1, I H, H-2), 1.62 (d, J = 0.7, 
3 H, H-13), 1.40 (dd, J= 13.9, 6.7, I H, H-2'), 1.03 (s, 3 H, H-11), 0.89 (s, 3 H, H-12). The 13C NMR spectrum (100 
MHz, CDCl3 as internal standard) showed δc 198.3 (C-9), 147.3 (C-7), 135.6 (C-5), 133.8 (C-8), 125.9 (C-4), 65.6 
(C-3), 54.4 (C-6), 44.0 (C-2), 34.0 (C-1), 29.4 (C-11), 27.3 (C-10), 24.8 (C-12), 22.8 (C-13). The specific rotation of 
the compound was  (c 0.10, CH2Cl2). The active substance was determined as 3-hydroxy-α-ionone by 
referring the data compared with previous report (D’Abrosca et al., 2004).

The ionone and its derivatives involves in the terpenoids metabolism as essential intermediates (Lutz-Wahl 
et al., 1998). The substance have been isolated from various sources (Pabst, Barron, Sémon, & Schreier, 1992). In 
this research, the substance showed the ability to inhibit the seedling development of garden cress and barnyard 
grass (Figure 4). A similar substance, 3-hydroxy-β-ionone was isolated from moss Rhynchostegium pallidifolium 
(Kato-Noguchi & Seki, 2010). The substance was reported as a growth inhibitor in garden cress bioassay. 
Comparing 3-hydroxy-α-ionone and 3-hydroxy-β-ionone in garden cress bioassay, the IC50 values were 35.60 µM; 
38.03 µM (shoot; root) and 14.90 µM; 16.3 µM (shoot; root), respectively. This result suggested that 
3-hydroxy-α-ionone may be less strong in activity than 3-hydroxy-β-ionone as allelopathic substance. Another 
ionone derivate, 3-hydroxy-5,6-epoxy-β-ionone was isolated from Athyriurn yokoscense. The substance was 
reported as inhibitor in lettuce seed germination (Kurokawa et al., 1998). In conclusion, the isolated compound, 
3-hydroxy-α-ionone was one of the active substance that responsible for the allelopathic effect.

背景：びまん性大細胞型 B 細胞性リンパ腫(DLBCL)患者の予後は、Rituximab（R; 抗 CD20 モノ
クローナル抗体）を用いた R-CHOP 療法が導入後顕著に改善している。しかしながら、80 歳以上
の超高齢患者に対する治療の詳細はほとんど報告されていない。本報告では、80 歳以上の超高
齢者患者と一般年齢層患者とを比較し、おのおのの予後因子と死亡リスクとの関係を明らかにす
ることで、超高齢者 DLBCL に対する治療を考察した。 
方法：愛媛大学医学部附属病院で、2006 年から 2014 年までに、びまん性大細胞型 B 細胞性リ
ンパ腫(DLBCL)と診断された（80 歳以上の症例 33 例を含む）141 症例を後ろ向きに集計した。
International Prognostic Index (IPI)の構成因子である、年齢、病期(II 期までと III 期以
上)、Performance status Score(PS：1 以下と 2 以上)、乳酸脱水素酵素値(LDH：正常範囲内と
正常範囲超)、リンパ節外病変数(1 個以下と 2 個以上)に加え、性別、標準的治療の有無(標準治
療である R-CHOP/R-THP-COP 療法を行っている群とそれ以外の治療群)、modified International 
Prognostic Index (mIPI) score(年齢は 79 歳以下と 80 歳以上、score が 2 以下と 3 以上)、を
変数として年齢・性別調整死亡率を検討した。 
結果：全年齢層（n=141）の解析では、患者年齢が 80 歳以上(HR:2.68, 95%CI:1.56-4.61)、PS
が 2 以上(3.15, 1.88-5.57)、mIPI score が 3 以上(2.71, 1.60-4.60)であること、は有意に死
亡率を上昇させた。一方、病期、LDH 値、節外病変数は患者予後に影響を与えなかった。興味深
い事に、女性患者であることは全年齢層において有意(0.56, 0.32-0.97)に患者の予後を改善し
た。更に患者を 80 歳未満(n=108)と 80 歳以上(n=33)の 2 群に分け、各々の集団で性差と死亡率
を解析したところ、80 歳以上の女性患者で有意に死亡率が低下し(0.35, 0.13-0.94)、80 歳未満 
では性差は予後に影響を与えなかった(0.72, 0.36-1.43)。また PS が 2 以上、および mIPI

score が 3 以上であることは 80 歳以上の患者(PS≥2; 3.90, 1.36−11.2)（mIPI≥3; 3.23, 
1.22−8.55）、80 歳未満の患者(PS≥2; 2.93, 1.44−5.95)（mIPI≥3; 2.56, 1.33−4.91）共に有意
に死亡率を上昇させ、その影響は 80 歳以上の患者でより有意な傾向にあった。また 80 歳未満の
患者では、標準治療未施行群で有意に予後が不良(2.96, 1.34−6.54)であるものの、80 歳以上の
患者では標準治療の有無は予後に有意な影響を与えなかった(1.74, 0.65−4.67)。 
考察：今回我々が行った解析では、従来 DLBCL 患者で言われているように、①（modified）
IPI は患者予後を全年齢層において推測する予後予測因子であった。更に、② 全身状態が不良
であることは標準治療の選択を断念させ、全年齢層ならびに 80 歳未満の一般治療年齢層におい
て死亡率に影響する予後不良因子であると言える。一方、③ 超高齢者 DLBCL 患者においても、
依然全身状態の悪化は予後不良因子であるものの、標準治療の有無は大きく患者の予後には影響
しなかった。このことと、本症例のほぼ全てが標準治療の有無にかかわらず低毒性の Rituximab
の投与がなされていることを鑑みれば、従来報告のある、④ 高齢女性での Rituximab のクリア
ランスの低下が、超高齢者女性の予後に影響を与えている可能性が推測される。 
 結語：DLBCL 患者において、全身状態が不良であるが為に標準治療から逸脱することは、患者
の予後を悪化させる誘因である。その一方で、超高齢者女性では全身状態が許せるならば、
Rituximab 治療を行うことは、患者予後を改善する可能性がある。この事は、個々の患者におい
て最適な Rituximab 投与量が異なる事を示唆しているのかも知れない。

Yacon (Smallanthus sonchifolius), belongs to a perennial plant in the Asteraceae family, is 
native to the Andes or Andean Mountains. It has been cultivated in many countries in Europe, 
Americas, and Asia countries such as China and Japan. Yacon tubers contain fructo-oligosaccharide 
that has potential for prebiotic effect. Instead, the leaf contains a various kind of metabolites such as 
polyphenols, kaurene-type diterpenenoides, and melampolide-type sesquiterpene lactones (SLs). In 
this study, we isolated a novel dimer SL, uvedafolin (1) which has the great cytotoxicity against 
HeLa (cervical cancer cells) cells, being potent anticancer drug. Finally, anticancer activity of 
uvedafolin (1) was clearly attributed to the apoptosis through mitochondria pathway. 
 
In Chapter 2, dimer SLs, uvedafolin (1) and enhydrofolin (2) which are composed of enhydrin 
(4) + sonchifolin (6) for 1, and uvedalin (5) + sonchifolin (6) for 2 were isolated from yacon leaf and 
elucidated the chemical structures by spectroscopic methods such as IR, NMR and MS. After that we 
further carried out quantitation analysis of the valuable dimer SLs in eight varieties of yacon leaves 
that cultivated and hybridized in the Western Region Agricultural Research Center at Zentsuji city 
(National Agriculture and Food Research Organization). It was found that the newly identified dimer, 
1, was found in only 'Sarada otome' (0.064 mg/g fresh weight) and 'Sarada okame' (0.045 mg/g fresh 
weight) cultivars and the related varieties. The cytotoxicity of two dimer SLs was determined against 
typical cancer cell lines such as HeLa (cervical cancer), HL-60 (leukemia) and B16-F10 melanoma 
(skin melanoma) cells. The dimers, 1 and 2, had the most effective IC50 values against the three tumor 
cell lines in a range from 0.2 to 1.9 µM, compared with those of monomers, 3-6 (0.7 - 9.9 µM). 
Moreover, 1:1 mixtures of enhydrin (4) + sonchifolin (6), and uvedalin (5) + sonchifolin (6) did not 
show high cytotoxicity as 1 and 2 did. Therefore, a cross-linkage of both monomer sesquiterpene

lactones can be concluded as an important factor for the cytotoxicity, which may be due to higher 
lipophilicity and disorder of functional structure of the cell membranes. 
 
In Chapter 3, in vitro cytotoxic mechanism of dimer SL, uvedafolin (1) and the related 
monomer SLs was studied on the HeLa cells by referring to the cytotoxicity on NIH/3T3 cell line 
(used as a normal cell line). Apoptotic pathway by compound 1 was investigated by various kinds of 
assay systems such as cell cycle analysis, DAPI stain, caspase activity, JC-1 stain and cytochrome c 
release. IC50 of compound 1 against NIH/3T3 cell line at 24 h was 4.98 µM, meaning that it is 
1.68-times safer than that of HeLa cells. Cell cycle analysis after treatment of compound 1 showed a 
significant increase in the percentage of G2/M phase time-dependently, increasing the percentage of 
sub-G1 peak, a parameter of fragmented DNA caused by apoptosis. Morphological change in nuclear 
caused by apoptosis was observed by using DAPI. The increases of caspase activities by compound 1, 
were observed in the following order, caspase-9 > caspase-3/7 > caspase-8, meaning mitochondria 
pathway was expected to be the major apoptotic pathway. Mitochondria membrane potential change 
and cytochrome c releasing were observed after treatment of 1. Therefore, 1 has the potential to induce 
apoptosis via mitochondria pathway more than cell death receptor pathway. 
 
In Chapter 4, sonchifolinic acid (7) was isolated from the yacon leaf and then esterified in 
order to understand the effect of lipophilicity of the esterified derivatives. Sonchifolinic acid (7) and 
its synthetic ester derivatives such as propyl, butyl and pentyl esters were examined for the 
cytotoxicity and structure-activity relationship (SAR). Consequently, sonchifolinic acid (7) did not 
show great activity (IC50 47.12 µM) whereas, alkylation of the carboxylic acid group at C-10 position 
in 7 was enhanced the cytotoxic activity more than 2-5 times (7a - 7d, IC50 5.29 - 21.07 µM). This 
result shows sonchifolinic acid (7) could be valuable for the modification of active substances and 
making potent lead compounds through semi-synthetic strategy. 
 
In conclusion, a novel dimer SL, uvedafolin (1) newly isolated from the yacon leaf could have 
potential for anti-cancer drug and lead compound for further investigation, contributing to cancer drug 
designing.

炭素繊維強化プラスチック (Carbonfiber reinforced plastics; CFRP) は軽量高
強度材料として航空機の構造部材として使用されている。 CFRP積層板は運用中あるい
はメンテナンス過程において異物衝突を受けることがある。そこで異物衝突による損
傷の進展を抑制するために，層間をじん性の高い熱可塑性樹脂粒子で強化した層間強
化型 CFRPが航空機用途には使用されている。本論文は層間強化型 CFRPの低速衝撃お
よび高速衝撃による損傷挙動を実験及び数値解析によって解明することを目的として
いる。
第 1章は序論であり，研究の背景，目的，概要について述べている。
第 2章では実験方法が説明されている。層間強化型 CFRPクロスプライ積層板試験片
について，落錘衝撃試験装置による低速衝撃試験，および，飛朔体衝突試験装置を用
いた高速衝撃試験の方法が述べられている。次に，光学顕微鏡および軟X線撮影装置
を用いた衝撃後試験片の表面・裏面および内部の微視的損傷の観察方法が説明されて
いる。
第 3章は数値解析手法について述べており，有限要素法を用いて，衝撃荷重を受け
る試験片の損傷過程を再現する手法を説明している。炭素繊維と樹脂からなる基材層
と層間樹脂層に固体要素を適用し，繊維破断，樹脂き裂を再現 している。 一方，層内
および層間のはく離を模擬するために，基材の中央層および基材と層間樹脂層の層聞
に結合力要素が導入されている。損傷の発生 ・進展のクライテリオンと して，前者で
は応力クライテリオン，後者ではエネノレギクライテリオンが適用されている。
第 4章は低速衝撃及び高速衝撃による微視的損傷のキャラクタリゼーションが行わ
れており，以下の実験的知見が得られている。低速，高速衝撃後の表面にはそれぞれ
デント(へこみ)，クレークーが発生する。低速衝撃においては高速衝撃と比べて衝撃
時の変形がグローパルであるために，デント周辺部に座屈による繊維破断が見られる。
一方，高速衝撃では局所変形であり，クレーター近傍に多重の樹脂割れが発生する。
裏面は低速衝撃・高速衝撃ともに曲げによる樹脂割れと繊維のピールオフ(剥がれ)
が発生する。内部にはコーンクラックと呼ばれる樹脂割れと層間はく離が見られるが p
層間強化型 CFRP積層板では層間のじん性が高いために，通常の CFRP積層板では見ら
れない層内はく離も発生する。低速衝撃・高速衝撃ともに，層間樹脂層は基材層か ら
の割れの進展を停留し，層聞はく離の進展を抑制する役割を果た している。
第 5章では，低速衝撃試験について，第 3章で述べた手法で損傷進展解析を行った
結果を第 4章で示された微視的損傷観察結果と比較検討している。表面のデントと繊
維破断および裏面の樹脂割れと繊維剥がれが実験と同様の形態と寸法で再現されてい
る。また内部損傷として 3 層間はく離だけでなく，層内はく離の発生も 再現されてい
る。さらに，層内はく離を考慮しない解析結果と比較して，層内はく離を考慮した解
析結果のほうが衝撃時の荷重一変位線図の傾きがよりよく実験結果と一致している。
以上より，本解析手法の妥当性が示されている。
第 6章は結論であり，全体を総括して以下の知見が得られている。 1)層間強化型
CFRPにおける低速衝撃と高速衝撃による損傷モードは類似しているが，損傷規模は高
速衝撃のほうが大きい。低速衝撃ではグローバル変形によりエネルギ吸収されるため，
損傷は緩和されるε2)層間樹脂層は低速衝撃 ・高速衝撃ともにき裂を停留し，層間
はく離を抑制する。 3)本数値解析手法は低速衝撃による損傷のモー ドとサイズを再
現している。 4)層間強化型 CFRPでは層間樹脂層が高じん性であるために，層内はく
離が発生するという特徴を有する。この損傷は数値解析において再現できる。
以上のように本論文は層間強化型 CFRP積層板の衝撃損傷過程を実験的かつ数値的
に明らかにしたものであり，航空機構造の安全性確保に貢献するだけでなく，新規
CFRP材料の開発に役立つ有益な知見を与えている。よって本論文は博士(工学)の学
位論文に値すると認められる。
