【背景】近年、キメラ抗原受容体発現 T 細胞(CART)など腫瘍特異的 T 細胞を用いた養子免疫療
法の研究が進み、徐々に臨床応用されつつある。しかしながら、体外で活性化培養された T 細
胞はしばしば「疲弊」と呼ばれる機能不全に陥り、治療効果を十分発揮できない状況にある。
そこで我々は、T 細胞の疲弊が細胞内の過剰なエネルギー代謝と関連していることから、CD8+ 
T 細胞のグルタミン代謝を抑制して培養して解析したところ、高い抗腫瘍効果が得られること
を見出した。さらに我々は、このグルタミン代謝抑制による CD8+ T 細胞の抗腫瘍活性の増強
メカニズムについて解析した。 
【方法】グルタミン代謝を抑制した CD8+ T 細胞の抗腫瘍活性の解析を、担癌マウスモデルを
用いて行った。疑似腫瘍抗原として卵白アルブミン(OVA)を発現する胸腺腫瘍細胞 E.G7 を接種
した担癌マウスを使用し、活性化培養した OVA 特異的 CD8+ T 細胞をマウスに移入することで抗
腫瘍活性の測定を行った。その際、CD8+ T 細胞のグルタミン代謝を抑制する手段として、活性
化培養する際の培地中のグルタミン濃度を減量する方法、および細胞内のグルタミン代謝を阻
害する薬剤を培地に添加する方法を用いた。さらに、グルタミン代謝を抑制した CD8+ T 細胞
が抗腫瘍活性を発揮する機序を解析するため、通常条件およびグルタミン代謝抑制条件で活性
化培養した CD8+ T 細胞を同数ずつ混合して担癌マウスに移入し、後日腫瘍浸潤 T 細胞(TIL)を
回収してそれぞれの細胞数、および抑制性受容体 PD-1 等の発現から細胞疲弊の解析を行った。
グルタミン代謝抑制が CD8+ T 細胞のエフェクター機能に与える影響を調べるために、in vitro
での腫瘍細胞傷害アッセイを行った。また CD8+ T 細胞のメモリー機能を解析するために、OVA
特異的 CD8+ T 細胞を移入して腫瘍が完全に消失したマウスに、OVA を発現するリステリアを感

染させ、recall response を解析した。通常培養および低グルタミン培養した CD8+ T 細胞のエ
ネルギー代謝状態を明らかにするため、フラックスアナライザーを用いて解析し、さらにエネ
ルギー代謝酵素の遺伝子発現についても定量 PCR を用いて解析した。また、グルタミン代謝産
物であるαケトグルタル酸処理した場合の抗腫瘍活性への影響についても解析した。 
【結果と考察】低グルタミン培養や、機序の異なる複数のグルタミン代謝阻害剤(AOA:aminoox
y-acetic acid hemihydrochloride, L-DON:6-diazo-5-oxo-L-norleucine, EGCG:epigallocate
chin gallate)により細胞内グルタミン代謝を抑制した CD8+ T 細胞では、通常培養に比べて腫
瘍縮小効果、マウス生存率の双方で成績の改善が見られた。また、グルタミン代謝抑制により、
活性化培養した CD8+ T 細胞における PD-1 の発現低下が観察された。通常培養およびグルタミ
ン代謝抑制培養された CD8+ T 細胞を混合して担癌マウスに移入する解析では、コンジェニック
マーカーで識別された移入 T 細胞はグルタミン代謝抑制培養されたものの方が通常培養 T 細胞
と比較して腫瘍内での細胞数が多く、また PD-1 の発現も低下しており、腫瘍内で生存し疲弊
も抑制されていた。腫瘍細胞傷害アッセイでは、通常条件とグルタミン代謝抑制条件で CD8+ T
細胞の傷害活性に差はみられず、グルタミン代謝阻害による抗腫瘍活性の増強は T 細胞の疲弊
の抑制と生存率の向上によるものと推測された。Recall response の解析では、グルタミン代
謝阻害された CD8+ T 細胞の移入マウスの解析では、リステリア感染後の移入 T 細胞の増加が著
明であり、グルタミン代謝阻害によって CD8+ T 細胞におけるメモリー機能の向上を示す結果で
あった。フラックスアナライザーの解析から、低グルタミン培養により解糖能およびミトコン
ドリア呼吸能が上昇することが明らかとなった。この結果と相関して、解糖系およびミトコン
ドリア呼吸系に関連するエネルギー代謝酵素の遺伝子発現の増加が認められた。また、αケト
グルタル酸処理により、CD8+ T 細胞の抗腫瘍活性低下が認められたことから、グルタミン代謝
産物の蓄積が、T 細胞疲弊の原因であると推測された。これらの結果をまとめると、グルタミ
ン代謝抑制された活性化 CD8+ T 細胞では疲弊が起こりにくく、抗腫瘍活性が増強することが示
唆された。本研究より、T 細胞疲弊のメカニズムにおいて、細胞内グルタミン代謝が重要な役
割を担っていることが示され、CD8+ T 細胞内グルタミン代謝を制御することで、養子免疫療法
の治療成績が改善できると期待される。

論  文  名 強制オシレーション法によるスパイロメトリー検査値の予測

【背景と目的】 
 スパイロメトリーは最も一般的な肺機能検査である。慢性閉塞性肺疾患の診断に必須であり、
喘息等の閉塞性疾患や間質性肺炎の病態評価にも活用される。一方、スパイロメトリーでは最大
努力吸気と呼気を伴い、高齢者や呼吸不全患者では施行が困難な場合がある。 
 強制オシレーション法（FOT）は非侵襲的で簡便な肺機能検査法である。FOT は空気振動を胸
郭に与え、振動によって生じた気流量の変動と圧変化を計測し、全呼吸抵抗を推測する。FOT で
は努力換気を必要とせず、小児、呼吸不全患者においても容易に検査を実施できる。現状では、
FOT はスパイロメトリーの補助的な検査である。過去の研究では、喘息、慢性閉塞性肺疾患、間
質性肺炎患者それぞれの疾患群で、FOT で得られた値とスパイロメトリーの測定値との相関を調
べているが、対象者数が少ない上、相関が弱かった。 
 本研究では、FOT の測定値から VC、FVC、FEV1 を予測し、その予測値とスパイロメトリーによ
る VC、FVC、FEV1 の測定値との相関を調べ、その予測値がどの程度測定値を反映しているかを評
価する。 
 
【方法】 
 FOT としてモストグラフ®を用い、R5、R20、X5、Fres、ALX を測定した。スパイロメトリーで
は ATS/ERS のガイドラインに従い、VC、FVC、FEV1 等を測定した。 
 Development study では、愛媛大学医学部附属病院を受診した 17 歳以上の日本人 1089 名を対
象とした。第一に、FOT の測定値とスパイロメトリーの測定値との相関を調べた。第二に、式１
として年齢、性別、身長、体重から VC、FVC、FEV1 を予測する回帰式を線形回帰分析から算出し
た。式２として、年齢、性別、身長、体重に加えて R5、R20、X5、Fres、ALX から VC、FVC、FEV
1 を予測する回帰式を線形回帰分析から算出した。そして、式１と式２で得られた予測値とスパ
イロメトリーの測定値との相関を調べた。 
 Validation study では住友別子病院の 552 名の患者を対象として、式２で得られた予測値と
スパイロメトリーの測定値との相関を調べた。 
 
【結果】 
 Development study において、FOT の測定値とスパイロメトリーの測定値の相関係数の p 値は
一組以外 0.001 未満であり、有意な相関を認めた。式１による予測値と測定値との相関係数はい
ずれも約 0.7 と統計学的に有意であった。式２による予測値と測定値との相関係数はいずれも 0.
85 を超えており、式１より強い相関を認めた。バイアス、平均平方二乗誤差とも小さかった。 
 Validation study では FOT の測定値とスパイロメトリーの測定値の相関係数の p 値は全て 0.0
01 未満であった。式２による予測値と測定値との相関係数はいずれも 0.88 を超えていた。しか
しながら、Development study に比較してバイアス、平均平方二乗誤差とも大きくなった。 
 
【結論】 
 式２で得られた予測値と測定値は強い相関を認めた。しかしながら、Validation studyでは
バイアスを認め、さらなる改善が必要と考えられる。 
 
 本研究は FOT でスパイロメトリーの測定値を予測できる可能性が高いことを示した初めての
報告であり、愛媛発の慢性閉塞性肺疾患の新たな診断基準の作成の可能性等、今後のさらなる発
展が期待される研究成果である。公開審査会は、平成３１年１月２８日に開催され、申請者は、
研究内容を英語で明確に発表し、１）愛大病院と住友別子病院の対象者の特性の違いの影響、
２）肺切除術前後の FOT 測定データ、３）呼吸器疾患のない対象者のみの解析結果、４）最も F
OT が有用となる呼吸器疾患、５）睡眠時無呼吸症における FOT の有用性、６）FOT の周波数につ
いて、７）FOT 測定時の筋緊張の影響、８）握力等の筋力を式に加える可能性、９）身長と体重
の相関について、１０）各呼吸器疾患、喫煙歴を式に加える可能性などに関する多くの質問に対
し日本語で的確に応答した。 
審査委員は、申請者が本論文関連領域に対して学位授与に値する十分な見識と能力を有するこ

Lily — the geophyte is an excellent model for studying the fundamental 
relationship for bulb production in vitro. The in vitro culture techniques of lily bulb 
scales, evolved as an alternative to the conventional method for multiplication of lilies 
because of uniform and healthy bulblets production in a short period of time. Regardless 
of advantages, the major problem with in vitro propagation is small bulblet size and this 
initial size of bulblets not only strongly affects growth and morphogenesis rates but also 
the transition between various vegetative and reproductive phases of lilies during 
development after planting. Therefore, bulblet growth is the most important factor in 
understanding how lily bulblets grow in vitro. Hence, the aim of this study was; i) to 
identify the factors affecting bulblet growth of Lilium sp. via in vitro, ii) modulating the 
links between organogenesis and methodology, and iii) production of high quality (large 
and uniform) bulblets in vitro. With the aim of revealing the effective in vitro factors to 
enhance in vivo performance of lily bulblets, the results showed that bulblet growth 
correlates with the carbon reserves inside explant tissue and in the medium. The internal 
storage of starch plays a vital role and influences regeneration and bulblet growth in both 
in vitro and in vivo. There is a high correlation (R2= 0.9672) between bulb size (weight) 
after the growth season at planting and initial bulb size (weight gained in vitro). The 
correlative assessment of culture media showed that explants grown in liquid medium

formed bulblets that had the best fresh weight, bulblet number, and root:shoot ratio 
compared with explants grown in semi-solid and solid media in vitro. However, this 
performance of bulblets in liquid static culture was influenced by the renewal of medium 
and thus, avoiding nutrient depletion compared with bulblets grown on agar solidified 
medium in vitro. In the pathway, culturing lily scale explants in a high sucrose 
concentration during regeneration of bulblet in vitro to increase bulblet size is not only 
the main point of interest for bulblets production but also the type of medium, nutrient 
status in the medium, explant placement in or on the medium, chlorophyll content in the 
explant tissues and CO2 within the glass vessel affect lily bulb scale explant regeneration 
and organogenesis in vitro. The threshold weight (bulblet size) for ontogenic 
development was about 300 mg at which 100% ontogenic development occurred and 
confirms that avoiding depletion of sucrose in the media is the key to achieving desired 
bulblet growth and ontogenic development in vitro. As well as indicates that the bulblets 
could be sustained and maintained by the elevated amount of medium and re-culture. 
Along with the re-culture in vitro, the course of bulb growth and ontogenic development 
was more rapid (1–2 growing seasons to reach the adult flowering phase) because of their 
bigger initial bulb size and advancement of ontogenic development compare to the 
conventional pathway (3–4 growing seasons). Moreover, this study represents the first 
report of lily bulb production through re-culture with a comparison of the timelines and 
ontogenic development obtained from in vitro versus conventional pathway.

 リゾチームは、魚類や昆虫類から哺乳類に至るまで生物界に広く分布する抗菌
性タンパク質であり、ヒトの涙や唾液中にも多く含まれている。先行研究におい
て、卵白由来リゾチームはリンパ球を活性化し、抗体やサイトカインの産生を促
進することが明らかになっている。一方、リゾチームは抗炎症効果を示すことが
報告されているが、その詳細なメカニズムは明らかにされていない。そこで本研
究では、リゾチームの抗炎症効果とその作用機序の解明を目的とした。  
 まず、炎症応答に深く関与するマクロファージの炎症性サイトカイン産生に対
するリゾチームの効果を検討した。卵白由来リゾチームを10 mM リン酸ナトリウ
ム緩衝液に溶解し、フィルター滅菌したものを実験に用いた。マウス腹腔内マク
ロファージおよびマウスマクロファージ様細胞株RAW264.7細胞に炎症を誘導する
ため、グラム陰性菌の細胞壁外膜構成成分であるリポ多糖（LPS）で1時間刺激し
た。その後、リン酸緩衝生理食塩水で洗浄することでLPSを除去した後、各細胞に
リゾチームを 11時間作用させ、炎症性サイトカインであるインターロイキン
(IL)-6および腫瘍壊死因子(TNF)-αの培養液中の濃度をELISA法で測定した。その
結果、リゾチームはマクロファージの生存率に影響することなく、 IL-6および
TNF-αの産生を有意に抑制した。このことから、リゾチームは炎症状態のマクロ
ファージによる炎症性サイトカイン産生を抑制することで、抗炎症作用を示すこ
とが明らかになった。また、マクロファージは、侵入微生物あるいは変質した自
己細胞の貪食および分解、さらには分解した断片を抗原提示することによる獲得
免疫系の活性化など、免疫応答において重要な役割を果たしている。そこで、リ
ゾチームがマクロファージの貪食活性に及ぼす影響を検討した。その結果、マク
ロファージの貪食活性はリゾチームによって影響されなかった。このことから、
リゾチームはマクロファージの貪食活性を阻害することなく、抗炎症作用を示す
ことが明らかになった。次に、リゾチームの作用機序を明らかにするため、まず、
炎症性サイトカインの遺伝子発現に対する影響を評価した。その結果、リゾチー

ムはIL-6およびTNF-α遺伝子の転写活性を下方制御することにより、サイトカイ
ン産生を抑制することが明らかになった。さらに、マクロファージ活性化に関連
するシグナル経路である NF-κB経路および MAPキナーゼ経路に対する影響を検討
したところ、リゾチームはNF-κBの核内移行には影響しなかった。一方、MAPキナ
ーゼ経路に対しては、ERKおよびp38のリン酸化レベルには影響しなかったものの、
JNKのリン酸化を有意に抑制した。これらの結果から、リゾチームはJNKのリン酸
化の下方制御によるMAPキナーゼ経路の抑制により、LPS誘導性の炎症性サイトカ
イン産生を抑制することが示唆された。  
 続いて、生体内におけるリゾチームの効果を検討するため、LPS誘導全身性炎症
モデルマウスを用いて評価を行った。本実験では、メスBALB/cマウスを非炎症誘
導群（Intact群）、炎症誘導群（Control群）、リゾチーム投与 +炎症誘導群（リ
ゾチーム群）に分けた。リゾチーム群については、投与量別にLow群（4.5 mg/kg 体
重）、Middle群（450 mg/kg 体重）、High群（2,250 mg/kg 体重）を設定し、合
計5群で実験を行った。各マウスに7日間、1日1回規定量のリゾチームを経口投与
し、リゾチームの最終投与後にControl群および各リゾチーム投与群にLPSを腹腔
内投与することにより、全身性炎症を誘導した。その後、血液、脾臓および肝臓
を採取した。その結果、リゾチームの経口投与は、血中の IL-6およびTNF-α量を
抑制することが明らかになった。また、リゾチームは脾臓における IL-6の遺伝子
発現を下方制御し、脾臓中のIL-6量を抑制することが示唆された。さらに、IL-1β
および IL-12の遺伝子発現が有意に抑制された。一方で、リゾチームは脾臓中の
TNF-α量を抑制したものの、TNF-αの遺伝子発現レベルには影響を示さなかった。
これらの結果から、リゾチームはIL-6とは異なるメカニズムで、脾臓中の TNF-α
量を抑制することが示唆された。さらに、肝臓中のIL-6量が抑制されたことから、
リゾチームはLPS誘導全身性炎症モデルマウスにおいて、特にIL-6の遺伝子発現お
よび産生を抑制することで抗炎症効果を示すことが示唆された。 
 本研究により、マクロファージに対するリゾチームの抗炎症効果に関する作用
機序およびLPS誘導全身性炎症に対する緩和効果が明らかになった。これらの結果
から、抗炎症効果が期待できる機能性食品素材として、卵白由来リゾチームの有
効性を示すことができた。

Energy Saving Technology Development for the Treatment of 
Reverse Osmosis Concentrate Using a Rotating Advanced 
Oxidation Contactor （回転円板型促進酸化装置を用いた逆浸
透濃縮排水の省エネルギー型処理技術の開発）

Reverse osmosis (RO) is a well-established and widely used technology for 
wastewater reclamation. With the purification of the treated wastewater, the RO 
process produces a concentrate which reported to be about 15–20 % of the 
influent volume. The RO concentrate contains concentrated levels of rejected 
pollutants originating from the source water. The emerging pollutants persistent 
in wastewater, such as pharmaceuticals and personal care products, have raised 
awareness of the environmental risk of the direct discharge of the RO 
concentrate. In addition, with the increase in global water demand, the scale of 
RO plants has been getting larger for several decades. Therefore, it is necessary 
to develop suitable technology to treat RO concentrates before discharging them 
into receiving water or recycling for other purposes. In this thesis, the removal of 
organic matters especially PPCPs in RO concentrate from a municipal 
wastewater reclamation facility has been researched.  
Firstly in Chapter 2, the synthesized TiO2-high silica zeolite (HSZ-385) 
composites powder was applied for the treatment of crotamiton, a scabicide and 
antipruritic agent persistent during biological treatment processes and 
frequently detected in secondary effluent in Japan. Crotamiton was rapidly 
adsorbed by HSZ-385 from the secondary effluent, and the other components of 
the secondary effluent little affected crotamiton adsorption. Even though the 
TiO2-zeolite composites showed lower adsorption rates than that of HSZ-385, 
similar crotamiton adsorption capacities were revealed using both test materials. 
The photocatalytic decomposition of crotamiton was significantly inhibited by the 
water matrix at low initial concentrations. The TiO 2-zeolite composites rapidly 
adsorbed crotamiton from secondary effluent, and then the crotamiton was 
gradually decomposed under ultraviolet (UV) irradiation. Importantly, when 
using TiO2-zeolite composites, coexisting material in the secondary effluent did 
not markedly inhibit crotamiton removal at low initial crotamiton concentration. 
The behaviors of the main intermediates during treatment demonstrated that the 
main degradation intermediates of crotamiton were also captured by the

composites. This provides a promising mechanism to mitiga te secondary 
pollution caused by the harmful degradation intermediates.  
Secondly in Chapter 3, the two powdered materials TiO2 and HSZ-385 have 
been effectively immobilized into a paper-like composite sheet, which is easy to 
handle and recover the photocatalyst and adsorbent during the treatment process. 
The removal of the antipruritic agent crotamiton from RO concentrate was 
studied using the TiO2/zeolite composite sheet. Excellent adsorption performance 
of crotamiton by the TiO2/zeolite composite sheet has been found without obvious 
inhibition by other components in the RO concentrate. With application of UV 
irradiation, crotamiton was simultaneously removed by adsorption and 
photocatalysis. The photocatalytic decomposition of crotamiton in the RO concentrate was 
significantly inhibited by the water matrix at high initial crotamiton concentrations, whereas 
rapid decomposition was realized at low initial crotamiton concentrations. Similarly, 
the major degradation intermediates of crotamiton were also adsorbed by the 
composite sheet.  
In Chapter 4, a novel rotating advanced oxidation contactor (RAOC) equipped 
with the composite TiO2-zeolite sheet was used to the treatment of RO 
concentrate. The removal of organic matters was studied as well as the variation 
of biodegradability of the RO concentrates. We analyzed the concentration of 53 
PPCPs in the raw RO concentrate and 48 PPCPs were detected at a concentration 
range of N.D.—1560 ng/L. The anti-inflammatory drug ketoprofen showed the 
highest concentration of 1560 ng/L in the RO concentrate, and the concentration 
of crotamiton ranked the third (670 ng/L). Under UV irradiation, the removal 
efficiency of the majority of the PPCPs were removed up to over 80% after 12h 
treatment by the RAOC, even though the adsorption process is not quite effective 
for 8 PPCPs ( ＜ 50%). The adsorption process of the RAOC showed no 
improvement to the biodegradability of the RO concentrate. With UV irradiation 
applied, the RAOC improved the biodegradability of the RO concentrate with the  
degradation of organic matters. And after 6 h treatment by the UV applied RAOC, 
the BOD5/CODcr ratio decreased with the degradation of the produced 
biodegradable products. The TOC and CODc r decreased by 24.5 % and 28.0 % 
after 12 h treatment, respectively. Judging from the improved biodegradability, 
after a short time UV applied RAOC treatment, the residual organic matters in 
the RO concentrate can be easily removed by a post biological process.  
Furthermore in Chapter 5, removing the antipruritic agent cro tamiton from 
RO concentrate with or without a coagulation pretreatment have been studied 
using the RAOC. The polyaluminum chloride (PAC) coagulation of the RO 
concentrate showed higher efficiency at pH 5 in terms of reducing TOC, A 365, and 
A254, and the coagulation efficiency improved with an increase in PAC dosage 
from 0 to 6 mmol Al/L. The RAOC performed well in adsorbing crotamiton

without obvious inhibition by other components in the RO concentrate, and the 
majority of the crotamiton was removed after 6 h, which resulted in the 
purification of the treated solution. With UV irradiation applied to the RAOC, 
crotamiton was simultaneously removed through adsorption and photocatalysis. 
The behavior of the main degradation intermediates of crotamiton during 
treatment showed that the degradation intermediates were also captured by the 
composite sheet. Coagulation followed by the RAOC process was effective in the 
removal of the organic matter in the RO concentrate as  well as the photocatalytic 
degradation of crotamiton.  
In addition in Chapter 6, a solar light irradiated RAOC treatment was used for 
the treatment of RO concentrate with or without the coagulation pretreatment. 
The energy consumption indicated that electric energy per order for TOC removal 
was evaluated with/without PAC coagulation pretreatment applied before the

  未分化幹細胞のもつ多能性の維持とその分化メカニズムの解明は科学的に興味深いのみなら
ず、再生医療への応用という面からも重要である。多能性の維持に関する知見は近年急速に蓄積
され、Nanog、Oct4、Sox2 といった多能性因子と呼ばれる転写因子のネットワークにより多能性
が維持されていることが明らかとなった。従来の考え方では、この多能性因子ネットワークが分
化を抑制することで未分化性が維持されると考えられていた。多能性因子は自らの発現を亢進さ
せることで自己保存的に働くことが多いため、多能性因子のネットワークは一見外部からの刺激
に対して安定な回路を形成しているように思われる。しかし、実際には多能性・未分化性は発生
段階のごく短時間認められる性質であり、胚性幹細胞の由来細胞であるエピブラストは速やかに
多能性を持った未分化状態を脱し、適切な分化を遂げている。このことは単純に多能性因子が分
化を抑制している、とする従来の考え方ではうまく説明できない。これに対して、近年主張され
たのが Precarious balance model である。このモデルでは多能性因子には単純に分化全体を抑
制する機能ではなく、特定の系譜への分化を促進し、相容れない他の系譜への分化を抑制すると
いう Lineage specifier としての機能があり、様々な系譜への親和性をもった多能性因子がバラ
ンスをとることで未分化状態が維持され、逆にこのバランスが崩れることで特定の方向へ分化が
進行する、と考えられている。

我々はこの多能性因子のバランスをとる一つのメカニズムとして Ac/N-end rule タンパク分解
経路が関与するのではないかと考えた。Ac/N-end rule タンパク分解経路とは、Initiator  
methionine の次のアミノ酸が特定のアミノ酸である場合に N 末端がアセチル化を受け、それが
タンパク分解シグナルとして機能する、というタンパク分解経路である。我々はこの経路の主要
な N 末端アセチル化酵素である Naa10 に着目し、マウス胚性幹細胞（mESCs）を初期発生モデル
として利用してこの命題について解析をした。 
mESCs は Mek 阻害剤と GSK3 阻害剤、それに LIF を加えた培地（通称 2iLIF 培地）にて未分化
性の高い均質な集団として維持培養できることが知られている。2iLIF 培地から阻害剤と LIF を
除去することで、mESCs は自発的に分化を始めることが知られており、特定の系譜への誘導を行
わないことから当該細胞株の性質を反映すると考えられている。我々は Naa10 KO mESCs を樹立
し、2iLIF 除去による分化を行ったところ、Naa10 KO mESCs では原始内胚葉への分化が亢進して
いることが種々のマーカーの解析により示された。 
次に、そのメカニズムとして、当初の仮説に基づき「Ac/N-end rule タンパク分解経路の機能
不全により、多能性因子間のバランスが崩れている」かどうか検討した。過去の報告で Lineage
 specifier としての活性があると想定される多能性因子（NANOG, TBX3, OCT4, SALL4, ESRRb, 
KLF4, SOX2）のタンパクの安定性を cycloheximide  chase にて評価したが、Naa10 KO によって
安定性が向上する因子を見出すことはできなかった。 
他のメカニズムを検討するため、公開されている Naa10 KO mESCs の RNA-seq データを用いて
パスウェイ解析を行った。結果、MAPK signaling pathway が優位に亢進していることが示され
た。そこで、我々の Naa10 KO mESCs においても MAPK シグナル経路が活性化していないか 
Western blot にてリン酸化 ERK を定量したところ、KO 株で優位に MAPK シグナル経路が亢進して
いることが確認された。KO 株に Mek 阻害薬を投与したところ、KO 株で認められていた原始内胚
葉マーカーである Gata6 の亢進が認められなくなった。一方で、MAPK シグナル経路の主要な活
性化因子である FGF のレセプターの阻害薬では Gata6 の低下を認めるものの、野生株との有意な
差が解消されなかった。このことから、Naa10 KO mESCs では FGF レセプターより下流、Mek 以上
のレベルで MAPK シグナルが亢進しており、そのため原始内胚葉系譜への分化が亢進しているこ
とが示された。 
まとめると、この研究により、Naa10 は mESCs の初期分化において、MAPK 経路を抑制すること
によって分化傾向が原始内胚葉方向へ傾くことを抑制し、エピブラスト系譜への分化をサポート
していること、Ac/N-end rule タンパク分解経路による多能性因子のバランスの調節は、少なく
とも解析した限りにおいて関与していないことが示された。
