Role of Mir-452-5p Overexpression in Epithelial–Mesenchymal Transition (EMT) in Early-stage Colorectal Cancer
香川大学Kagawa University博士(医学)Background/aim: The microRNA miR-452-5p holds a critical role in the progression of multiple tumor formations, but there is limited understanding regarding the epithelial-mesenchymal transition (EMT) progression and its underlying mechanisms in the early-stage colorectal cancer (CRC). We aimed to explore the change in miRNA expression in early-stage CRC and examine the role of these miRNAs in CRC.Materials and methods: The expression levels of miR-452-5p in tissues and cells of early-stage CRC were determined by real-time quantitative polymerase chain reaction. Additionally, the biological effects of miR-452-5p on CRC were investigated by in vitro functional experiments.Results: The expression levels of miR-452-5p were found increased in early-stage CRC tissue. We found that miR-452-5p promoted CRC cell proliferation but inhibited epithelial-mesenchymal transition. Furthermore, miR-452-5p promoted cell proliferation through activation of the extracellular signal-regulated kinase pathway, and inhibited cell invasion through suppression of Slug (Snail2) expression and up-regulation of E-cadherin expression.Conclusion: The expression of miR-452-5p is up-regulated in early CRC and suppresses epithelial-mesenchymal transition in CRC. These discoveries suggest that miR-452-5p has the potential to serve as a viable therapeutic target for CRC.doctoral thesi
Importance of preconception care for a pregnant woman with suspected osteogenesis imperfecta
妊娠中に母体の一過性大腿骨頭萎縮症による疼痛・歩行障害と分娩後発症の大腿骨頸部骨折をきたし、のちに骨形成不全症を疑った症例を経験した。妊娠前にPreconception care(PCC)を受けていれば骨折を回避できた可能性もあり、改めてPCCの重要性を認識したので報告する。症例は31歳、身長146cm、非妊時体重39.5k、軽度の青色強膜あり。初経13歳、月経整順、1妊0産。既往歴なし、ダイエット歴無し。実母が50歳代から骨粗鬆症にて加療中。自然妊娠成立後、妊娠初期にて当科を受診した。産婦人科の受診は初めてであった。妊娠34週、左股関節痛が出現したが単純X線検査では異常は認められなかった。妊娠38週3日、疼痛のため歩行困難となり選択的帝王切開術を施行した。手術直後の単純X線検査も異常は認められなかった。術後5日目になっても疼痛による歩行困難が改善せず、単純X線検査による再検査で、左大腿骨頸部骨折を認めた。MRIにて大腿骨頸部はT1強調画像で低信号、脂肪抑制のT2強調画像で高信号を示し、一過性大腿骨頭萎縮症と診断した。術後12日目に左大腿骨人工骨頭置換術を施行した。術中所見にて骨の強度の脆弱性が判明した。DEXA法にて骨密度測定を施行し、腰椎YAM59%、右大腿骨頸部YAM52%と骨粗鬆症の所見を認めた。臨床症状と合わせて骨形成不全症が疑われた。テリパラチド投与を開始したが通院中にも胸椎の多発性圧迫骨折を発症した。もし妊娠前にPCCを行い、青色強膜や低身長、母体の若年期骨粗鬆症に注目し、骨系統疾患を疑うことができれば妊娠前に適切な対応ができていた可能性は考えられる。PCC施行時に低身長を認める女性では、青色強膜の現症や骨痛の既往、骨粗鬆症の家族歴等、骨系統疾患の可能性についても注意深く検討する必要があることが示唆された。journal articl
A pilot study on the efficacy of a diabetic diet containing the rare sugar d-allulose in patients with type 2 diabetes mellitus : A prospective, randomized, single-blind, crossover study
入院病院食に希少糖含有糖尿病治療食を開発し、通常の糖尿病食と比較し持続血糖測定器を用いて詳細に解析した世界で初めての報告である。希少糖含有治療食では有意に2型糖尿病患者の食後血糖値を改善できた。High sugar consumption increases the risk of diabetes, obesity, and cardiovascular diseases. Regarding the diet of patients with diabetes, artificial sweeteners are considered a safe alternative to sugar; however, there is also a risk that artificial sweeteners exacerbate glucose metabolism. D-allulose (C-3 isomer of d-fructose), which is a rare sugar, has been reported to have antidiabetic and antiobesity effects. In this study, the efficacy of a diabetic diet containing D-allulose was investigated in patients with type 2 diabetes using an intermittently scanned continuous glucose monitoring system (isCGM). This study was a validated, prospective, single-blind, randomized, crossover comparative study. Comparison of peak postprandial blood glucose (PPG) levels after consumption of a standard diabetic diet and a diabetic diet containing 8.5 g of D-allulose was the primary endpoint. A D-allulose-containing diabetic diet improved PPG levels in type two diabetes patients compared with a strictly energy-controlled diabetic diet. The results also showed a protective effect on endogenous pancreatic insulin secretory capacity owing to reduced insulin requirement. In patients with type two diabetes mellitus, diabetic diets containing 8.5 g D-allulose were effective in improving PPG levels.journal articl
An analysis of the JICA Japan-Mexico Training Program for the Strategic Global Partnership: a case of the course “Startup Creation and Innovation Ecosystem Development”
The central goal of our research is to understand how to foster an innovation ecosystem through entrepreneurship education and create a cooperative system between Japan and Latin America. To do this, in 2015, we began to look into academic exchanges between Hiroshima University and several Latin American universities. As a result, in 2018, an international entrepreneurship training platform was created, the Global 1st Penguin Club (G1PEC). Likewise, to promote international cooperation, in 2017, the course "Startup Creation and Innovation Ecosystem Development" started. Sponsored by the Japan International Cooperation Agency(JICA) and initially hosted at Hiroshima University, then the program has been continuously implemented at Kagawa University since 2021 as a successor. Since then, sixteen Mexican professional trainees have come to Japan to study the current situation of the ecosystem, and during the second half of the course, they developed practical research projects. By working in collaboration with the trainees, we are trying to explain the mechanisms that lead to the formation of the innovation ecosystem from different points of view. Identification of substances related to ochratoxin A produced by coffee roasting and investigation of it's contamination of commercially available roasted coffee
The chromatogram of ochratoxin A (OTA) analysis of roasted coffee shows an unknown peak that appears about 2 minutes after OTA. This unknown peak was identified as (14R)-ochratoxin A ((14R)-OTA) based on the similarity of the formation pattern of thermal decomposition of OTA. As a result of (14R)-OTA contamination investigation of 29 commercially available roasted coffee samples, the contamination concentration of (14R)-OTA was 0.19±0.27 μg/kg, and the contamination concentration of OTA was 1.99±1.98 μg/kg. (14R)-OTA was about 1/10 of OTA. This suggests that the risk of (14R)-OTA in roasted coffee is not so high. The correlation coefficient between OTA and (14R)-OTA contamination levels was R²=0.3941,showing a weak correlation, but no strong correlation. It was thought that this was because the amount of OTA decreased and the amount of (14R)-OTA produced differed depending on the coffee roasting temperature and time.焙煎コーヒーのオクラトキシンA(OTA)分析のクロマトグラムでは、OTAより約2分遅れて出現する不明なピークが出現する。この不明なピークは、OTAの加熱分解の生成パターンの類似性などから(14R)-Ochratoxin A(14R)-OTAと同定した。29検体の市販焙煎コーヒーの(14R)-OTAの汚染調査をおこなった結果、14-(R)-OTAの自然汚染濃度は0.19±0.27μg/kgで、OTAの自然汚染濃度1.99±1.98μg/kgの約1/10であった。このことから、焙煎コーヒーの(14R)-OTAのリスクはそれほど高くないと考えられた。また、OTAと14-(R)-OTAの汚染濃度の相関係数はR²=0.3941で、弱い相関性は認められたが、強い相関性はなかった。これは、コーヒーの焙煎温度と時間によって、OTAの減少量と(14R)-OTAの生成量が異なるためと考えられた。departmental bulletin pape
Producing of monoclonal antibodies against nivalenol
Nivalenol (NIV) and deoxynivalenol (DON) are typical B-type trichothecene mycotoxins, which frequently contaminate domestic wheat and barley, causing health hazards to humans and livestock. Many monoclonal antibodies (mAbs) against DON have been developed, and screening methods for DON in cereals by ELISA have been reported using these mAbs. However, there are no reports of the production of a specific antibody mAb against NIV. Therefore, NIV-oxime was prepared by oxime of carbonyl at the 8-position and bound to cationized bovine serum albumin (cBSA).We performed cell fusion of spleen cells from the mouse, which was immunized four times with NIV-oxime-cBSA, and succeeded in establishing NIV specific mAb producing hybridomas for the first time. Our NIV.9 mAb did not cross-reacted with NIV related compounds. NIV. 10, 12, and 13 mAbs weakly cross-reacted with15-acetly DON. NIV.11 mAb cross-reacted with 15-acetly DON, and weakly cross-reacted with DON and 3-acetly DON. When NIV.10 and 11 mAbs with the highest NIV reactivity were used, 1 ng/mL of NIV could be detected in the optimized indirect competitive ELISA. We expect that a simple screening method for NIV in crops will be established by ELISA using these antibodies.NIV-オキシム化蛋白質結合体を作製し、国内産の小麦や大麦を高頻度に汚染しているニバレノール(nivalenol,NIV)に対するモノクローナル抗体(mAb)は初めて作製した。NIV-オキシム-cBSAで免疫したマウス脾臓細胞を細胞融合し、19クローンの抗NIVmAb産生ハイブリドーマを得た。NIV.9 抗体はNIVに特異的に反応した。NIV.10、12と13 抗体は15-acetly DONに弱い交差反応を示した。NIV.11抗体は15-acetly DONに強い交差反応を示し、15-acetly DONとDONに弱い交差反応を示した。NIV.9とNIV.11抗体を用いた競合ELISAでは、1 ng/mLまでのNIVの検出が可能であった。departmental bulletin pape
Study of nursing student's process in spiritual care with trajectory equifinality model (TEM)
目的: 看護学生が患者に行ったスピリチュアルケアの実施に至る過程を明らかにすることを目的とした。研究方法: 研究対象は、看護学実習生が作成した臨地実習終了後レポート5件であった。データは、患者の価値観に関連した主観的および客観的記述、患者へのスピリチュアルケアの看護計画とその実施、自己評価の記述であった。分析方法は、TEMを用い、抽出したテキストを時系列とし、等至点、分岐点、必須通過点を検出して、看護学生のスピリチュアルケアの実施過程を整理した。結果: TEMによる分析の結果、看護学生のスピリチュアルケア過程には、等至点【目標達成に近づく】、必須通過点【患者の全人的苦痛のアセスメント】、分岐点【患者の苦しみの焦点化】、【患者の変化を実感する】があった。また、スピリチュアルケア過程は、患者に関する情報収集、アセスメント、看護計画の立案、実施、評価という標準的な看護過程に沿うものであった。考察: 看護学生のスピリチュアルケア過程は、情報収集、アセスメント、看護計画の立案、実施と評価という標準的な看護過程が基盤にあった。スピリチュアルケア過程は、アセスメントの段階でスピリチュアルな観点で考えることが必須であり、また、スピリチュアルケアの知識により現象を感覚的に捉える体験が分岐点になると考えられる。患者の苦しみを焦点化させる分岐点では、看護学生は、患者の苦悩を焦点化し、それを記述したことによって、スピリチュアルケアとなりうる行動を明確にしたと考えられる。結論: 本研究は、スピリチュアルケア過程が従来の看護過程で説明可能であることを示した。さらに、スピリチュアルケアとして成立するための分岐点を初めて明らかにした。Aim: This study was designed to clarify the actual situation of processes in spiritual care that nursing students provided to patients.Research methods: Five post-clinical training reports written by nursing students were examined. The data included subjective and objective descriptions related to patients' values, description of the spiritual care interventions in the nursing care plan, and description of the implementation and evaluation of spiritual care provided to the patients. For the analysis, TEM was used; the extracted texts were chronologically analyzed; and the implementation process of spiritual care by the nursing students was examined with respect to the equifinality, obligatory passage, and bifurcation points.Results: In the TEM analysis, the spiritual care process had the equifinality point of “approaching goal achievement,” the obligatory passage point “assessment of the patient's holistic distress,” and the bifurcation points “focusing on the patient's suffering” and “feeling changes in the patient.” The spiritual care process was a basic nursing process that involved patient information collection and assessment, along with nursing care plan development, practice, and evaluation.Discussion: The spiritual care process practiced by nursing students is based on information gathering and assessment, followed by planning, implementation, and evaluation of the nursing process. In the spiritual care process, thinking from a spiritual viewpoint at the assessment stage is essential, and the experience of sensibly grasping phenomena based on the knowledge of spiritual care will be a turning point. At the bifurcation point of focusing on the patient's suffering, the nursing students focused on the patient's suffering and described it, thereby clarifying behaviors that could be spiritual care.Conclusions: This study showed that the spiritual care process can be explained by the conventional nursing process. Effects of jiritsu katsudo combined group learning and individual learning in a school for the intellectually disabled
知的障害特別支援学校の自立活動について集団学習と個別学習を併用させた実践を行い、効果を検討した。知的障害特別支援学校の小学部に在籍する6名の児童に集団学習と個別学習を行った。指導者あるいは友達への要求を標的行動とした。プロンプトがあれば全児童が要求できるようになり、児童間の相互作用も確認できた。児童間で相互作用が見られたことから、集団を活かした学習活動を展開できることが示された。departmental bulletin pape
