    Six Hours of Hydrogen Gas Inhalation Has a Neuroprotective Effect Even in Piglets with Delayed Functional Recovery

    香川大学Kagawa University博士(医学)Introduction: Combined therapeutic hypothermia (TH) and 24-h hydrogen (H2) gas inhalation reduces seizure burden in piglets in the latent phase of hypoxic-ischemic (HI) injury versus TH alone. Nevertheless, the effects of H2 gas in the earliest phase following resuscitation were unclear.Methods: After HI insult, 17 piglets ≤24 h old were divided into a HI insult group (HI, n = 8) and a HI and H2 gas group (HI-H2, 2.1%-2.7% H2 gas, 6 h, n = 9). Time to recovery to a normal amplitude-integrated electroencephalogram background (RT-aEEG) was examined for 6 h after HI insult and undamaged neurons were counted.Results: The duration of low-amplitude (<5 μV) EEG after insult was not different between the two groups. Undamaged neuron numbers were significantly higher in the HI-H2 group than in the HI group (p < 0.01), although RT-aEEG was not different.Conclusion: Six hours of H2 gas inhalation initiated from resuscitation significantly increased the number of undamaged neurons compared to the untreated group, although there was no difference in RT-aEEG.     子どもの登校渋りに葛藤を抱える母親との面接過程

    本稿は,子どもの登校渋りに悩む母親との面接過程を通して,母親の不安や葛藤を共に抱える臨床的意義を検討した。セラピストはクライエントを受容し,共感的な態度で語りを傾聴した。その中でクライエントは,子ども達の気持ちを尊重したいと語りながらも子ども達に過干渉に関わる時期を経て(第Ⅰ期),自己に注意を向け始め(第Ⅱ期),自己と子ども達に向き合い(第Ⅲ期),気持ちの揺らぎを自覚して自己や家族をありのまま受容するという変化が生じた(第Ⅳ期)。この経過から,「共感的なセラピストの内在化」,「登校渋りへの認識と親子関係の変化」,「自身のケアと社会との繋がり」の三点から母親の不安や葛藤を抱える臨床的意義を考察した。departmental bulletin pape

    Occurrence and risk assessment of deoxynivalenol in wheat flour, bread, and pasta commercially available in Japan

    Deoxynivalenol(DON), produced by Fusarium graminearum, is a pathogen of red mold of wheat. We investigated DON contamination in domestic commercial wheat flour, bread and pasta using an immunoaffinity column-HPLC method. DON was detected in 11(29.7%)of 37 commercial wheat flours. The average value for all samples was 0.051 ppm, and both the DON contamination rate and concentration were significantly higher in the domestic wheat flour. DON spiked wheat was prepared, and bread was baked in a home bakery to investigate the behavior of DON. The results showed that DON did not decrease during the baking process. Therefore, we conducted a DON contamination survey of commercial bread. We detected DON in 17(56.7%)out of 30 breads. The average DON concentration for all samples was 0.040 ppm, with domestic flour having a significantly higher DON concentration. The intake of DON per body weight was 0.22 μg/kg body weight/day, which was 22% of the TDI, when two slices(120 g)of bread contaminated with DON at the highest concentration of 0.109 ppm were consumed daily.  A survey of commercially available dried pasta for DON contamination revealed that DON was detected in 11(35.5%)out of 31 samples. The average value for all samples was 0.049 ppm. Pasta contaminated with the highest concentration of DON(0.357 ppm)was boiled, and the transfer of DON to the boiling water was examined. As the result, DON in the noodles decreased to 0.185 ppm dry pasta equivalent at the recommended optimum boiling time of 9 minutes, and 0.190 ppm DON dry pasta equivalent was detected in the boiling water. The intake of DON per serving of 200 g of this noodles consumed daily was 0.67 μg/kg body weight/day, which was 67% of the TDI.ムギ類の赤カビ病の病原菌であるFusarium graminearumなどによって産生されるデオキシニバレノール(DON)は,国内産の小麦や大麦を高頻度汚染している.そこで,国内市販の小麦粉,食パンとパスタ中のDONの汚染調査をイムノアフィニティーカラム-HPLC法で行った.市販小麦粉37検体中11検体(29.7%)から最大0.368 ppmのDONを検出した.全検体の平均値は0.051ppmであり,有意に国内産小麦粉の方がDON汚染率と濃度ともに高かった.DON人工汚染小麦を調製し,ホームベーカリーでパンを作製し,DONの挙動を調べた.その結果,DONは製パン工程で減少しなかった.そこで,国内市販の食パンのDON汚染調査を行った.その結果,30検体中17検体(56.7%)から最大0.109 ppmのDONを検出した.全検体の平均値は0.040 ppmであり,有意に国内産小麦粉の方がDON汚染濃度は高かった.市販食パンのリスク評価を行った.最高濃度0.109 ppmのDONに汚染していた食パンを毎日2枚(120 g)摂取した場合のDONの摂取量は体重あたりのDONの摂取量は0.22 μg/kg体重/日で,TDIの22%であった.  国内市販の乾燥パスタの汚染調査を行った結果,31検体中11検体(35.5%)から最大0.357 ppmのDONが検出され,全検体の平均値は0.049 ppmであった. 最高濃度のDON(0.357 ppm)に汚染されていたパスタを茹で,茹で汁へのDONの移行について調べた.その結果,推奨最適茹で時間の9分で麺中DONは乾燥パスタ換算で0.185ppmに減少し,茹で汁には乾燥パスタ換算で0.190 ppmのDONが検出された。最大汚染の乾燥パスタを推奨時間茹でた場合の麺中のDON濃度は0.185 ppmで,このパスタを毎日1人前200 gを摂取した場合のDONの摂取量体重あたりのDONの摂取量は0.67 μg/kg体重/日で,TDIの67%であった.departmental bulletin pape

    Production of monoclonal antibodies against ergothioneine and thiourocanic acid for a simple analytical method for ergothioneine.

    Ergothioneineine(EGT)is an amino acid derivative with strong antioxidant activity and is also called longevity vitamin. We succeeded in producing a monoclonal antibody(EGT. 1-1)against EGT for the first time. However, the sensitivity of competitive ELISA using EGT. 1-1 antibody is 30 μg/mL, which is not sensitive enough to measure EGT in human whole blood. Therefore, we were performed. monoclonal antibodies against thiourocanic acid(TU),which is quantitatively produced when EGT is treated with ergothionase. As a result, we succeeded in producing three monoclonal antibodies. The best of these antibodies could measure 20 ng/mL of TU in a competitive ELISA. In the future, we plan to establish a simple method for measuring EGT in human whole blood using this TU. 1-1 antibody.エルゴチオネイン(EGT)は強力な抗酸化作用があるアミノ酸誘導体で長寿ビタミンとも呼ばれている.このEGTに対するモノクローナル抗体(EGT. 1-1)の作製に初めて成功した.しかし,EGT. 1-1を用いた競合ELISAの検出感度は30μg/mLであるり,ヒト全血中のEGTの測定には充分な感度ではなかった.そこで,EGTをエルゴチオナーゼ処理すると定量的に生成されるチオウロカン酸(TU)に対するモノクローナル抗体を試みた.その結果,3つのモノクローナル抗体の作製に成功した.その中で最も優れた抗体を用いた競合ELISAでは,20ng/mLのTUの測定が可能であった.今後,このTU. 1-1抗体を用いたヒト全血中のEGTの簡易測定法の確立を行う予定である.departmental bulletin pape

    Occurrence of aflatoxins in commercial peanut butter and almonds in Japan

    Peanut butter and almonds commercially available in Japan in 2023 were collected and investigated for contamination with aflatoxins(AFs)by immunoaffinity column-HPLC method. Total AFs(0.20-3.47 μg/kg)were detected in 13(58.5%)of 23 peanut butter samples. AFB1 was detected in 12(52.2%)samples in the range of 0.17-2.99 μg/kg, with a mean value of 0.35 μg/kg for all samples. Total AFs(0.01-0.44 μg/kg)were detected in 15(55.5%)of 27 almond samples. 0.17 μg/kg of AFB1 was detected in one sample(3.7%). These results were similar to those obtained 18 years ago by the Ministry of Health, Labour and Welfare in 2006, suggesting that there was no significant change in AFs contamination in peanut butter and almonds commercially available in Japan.2023年に国内で市販されていたピーナッツバターとアーモンドを収集し,IAC-HPLC法でAFsの汚染調査を行った.ピーナッツバター23検体中13検体(58.5%)から総AF(0.20〜3.47 μg/kg)が検出された.全検体平均値は0.53 μg/kgであった.AFB1は12検体(52.2%)から0.17〜2.99 μg/kgの範囲で検出され,全検体平均値は0.35μg/kgであった.アーモンド27検体中15検体(55.5%)から総AFs(0.01〜0.44 μg/kg)が検出された. 全検体平均値は0.11 μg/kgであった.AFB1は1検体(3.7%)から0.17 μg/kg検出された.これの結果は,18年前の厚生労働省2006の結果概ね同じであり,こららのAFs汚染には大きな変化はなかったと考えられた.departmental bulletin pape
