    Heatwave final report: Fire heat flux generator

    Heat flux is critical factor in determining material ignition and spread of fire and assessing structural responses to fire. For a structural component subjected to a fire condition, the method we have used is to apply heat flux to the specimen first and the specimen is subjected to any structural integrity testing later, separately. This method is not realistic as actual structural components are subjected to structural loading and heat at the same time in fire conditions. Using our heat flux simulator, the same heat flux measured from actual fire can be regenerated and applied to any structural building components while they are under various tension, bending, or compression force

    Non-history dependent temporal superposition algorithm for the pint source solution

    Optimal design and operation of borehole fields require accurate prediction of the dynamic heat transfer in the ground. For large borehole systems, current simulation algorithms do not have the scaling properties to enable complex borehole network simulations with both fine source discretizations as well as fine time resolution. This paper is focused on the so-called “non-history dependent” method, a procedure for the acceleration of the time superposition process with promising properties that could be instrumental to improve on current scaling limitations. In this development, we considered the case of the point source and we derived an analytical expression for the scheme. The computation of the temperature involves the integration of an oscillatory function. A scheme for the numerical integration step of the algorithm based on integration techniques for oscillatory integrals is proposed. We study the computational complexity of the algorithm, which is linear in the number of time steps and in the number of integration points. Additionally, refining the discretizations used to perform the numerical integral leads to accuracy close to double precision. As a result, we conclude that the algorithm presents both favorable accuracy and performance features

    Strategic analysis of Wings of Hope Family Crisis Services: Examining organizational effectiveness in the nonprofit sector

    Wings of Hope Family Crisis Services is a 501(c)(3) nonprofit organization in Stillwater, Oklahoma that aims to disrupt domestic violence by providing comprehensive services to the following five Oklahoma counties: Payne, Logan, Pawnee, Noble, and Lincoln. This thesis aims to conduct a holistic business analysis of Wings of Hope, identifying internal and external strengths and weaknesses of the organization reflective of the firm’s strategic positioning within the nonprofit sector and domestic violence services industry. Findings indicated Wings of Hope is an Oklahoma industry leader, cornering the social market on identifying and addressing rural community needs while achieving considerable impact in the lives of those affected by interpersonal violence. The organization has been successful in effectively operating within the parameters of its mission and vision statements. Time spent assessing Wings of Hope identified gaps in the organization’s structure, marketing and community outreach efforts, and fundraising tactics that when developed with a strategic management perspective, have the potential to accomplish further value creation and social impact. This thesis offers objective recommendations to develop the targeted areas, reflecting the firm’s capacity to undertake suggested methods to acquire increased operations efficiency

    Understanding the role of stress-induced cellular transcription factors on human alphaherpesvirus 1 (HSV-1) and bovine alphaherpesvirus 1 (BoHV-1) reactivation from latency

    Herpes simplex virus 1 (HSV-1) and Bovine herpes virus 1 (BoHV-1) establish lifelong latent infections in sensory neurons following acute infection. Periodically, stress factors induce reactivation from latency, leading to virus shedding and transmission. Stress elevates corticosteroid levels, thereby increasing the incidence of reactivation. Dexamethasone (DEX), a synthetic corticosteroid that mimics the effects of stress, induces reactivation. Virion protein tegument VP16, infected cell protein 0 (ICP0), and ICP4 are three viral regulatory genes that are likely to trigger reactivation from latency. My dissertation focuses on the effects of stress-induced cellular transcription factors on activating viral promoters. VP16 is a potent transactivator of HSV-1 immediate early (IE) genes. My studies demonstrated VP16 cis-regulatory module (CRM) is transactivated by glucocorticoid receptor (GR) and Slug, a stress-induced transcription factor that binds enhancer boxes (E-boxes) and were the only stress-induced transcription factors examined that transactivated the VP16 CRM construct. Consequently, I examined which transcription binding sites are important for GR- and Slug-mediated transactivation. Mutations in E-box, 2 ½ GR response elements (1/2 GREs), or NF-kB binding site significantly reduced VP16 CRM transactivation. Notably, silencing Slug expression in Neuro-2A cells significantly reduced viral replication. ICP0 and ICP4 are IE genes. ICP0 is a promiscuous transactivator, while ICP4 is the only viral transcriptional activator essential for productive infection. My studies revealed that the ICP0 promoter and ICP4 CRM, but not VP16 CRM, are transactivated by GR and specificity protein 1 (Sp1) or Sp3 with DEX treatment. Sp1 and Sp3 are expressed ubiquitously in almost all cell types and bind to GC-rich sequences. The effects of Mithramycin A, an anti-tumor drug that preferentially binds GC-rich DNA sequences and displaces Sp1 family transcription factors, was used to examine its effects on ICP0 promoter activity. Mithramycin A significantly reduced GR+Sp1+DEX mediated transactivation in CV-1 cells, but not in Neuro-2A cells. Conversely, GR+Sp3+DEX mediated transactivation was significantly reduced in both Neuro-2A and CV-1 cells. Ex vivo studies revealed Mithramycin A impaired DEX-induced reactivation from latency in male and female mice. These studies revealed Mithramycin A may be a useful antiviral therapy

    Physiological response of agronomic crops to nutrient management and abiotic stress

    In Oklahoma, where resource availability varies throughout the growing season, the growth and development of grain sorghum often depend on tillers influenced by delayed nitrogen (N) application and planting density. However, their combined impact remains insufficiently documented. This study aimed to evaluate the physiological response of sorghum to delayed N application and assess the role of planting density. Field trials were conducted over three years at Chickasha (21,2022), Bixby (2022), and Perkins (2023), testing three N application timings: at planting, 21 days, and 42 days after planting and two planting densities (52,000 and 77,000 plants ha-1). Results indicated no significant differences in growth or yield among N timings or planting densities. However, delayed N application reduced tiller counts and panicle dimensions. These findings suggest growers can delay N application up to 42 days after planting without significantly impacting yield, but further delays could reduce productivity. The second chapter focused on the germination performance and early growth of six cover crops: Barley, Rye (Elbon, Maton, Oklon), Triticale, and Wheat Ok Corral (control) under four temperature regimes (20C, 25C, 20C, and 25C) at 7 and 14 days after sowing. Germination trials measured hypocotyl length, root length, and surface area. Results showed that temperature significantly influenced germination and early growth, with optimal performance at 25C and 30C. rye Maton Exhibited the highest growth metrics, demonstrating strong adaptability, while Triticale and Wheat Ok Corral consistently showed lower performance, especially at extreme temperatures of 20C and 35C. These findings underscore the importance of temperature in early-stage growth and provide insights into selecting cover crops suitable for varying environmental conditions. The last chapter assessed whether outline notes improved note-taking confidence and reduced anxiety, particularly post-COVID-19. A survey among 264 pre-COVID and 277 post-COVID students in an introductory plant and soil systems course (PLNT 1213) revealed that outline notes with 30% blanks helped students stay focused during lectures. Post-COVID, there was a 42.5% decrease in note-taking anxiety, suggesting improved comfort in learning environments. The study highlights that structured course notes reduce anxiety and improve students’ confidence and focus, demonstrating the effectiveness of combining pre-made and handwritten notes
