    Investigating the effects of Culturelle probiotics on gut microbiome composition in lab mice

    The gut microbiome plays a crucial role in maintaining host health, and probiotics have been researchered for their potential to modify microbial populations in beneficial ways. This study aimed to explore the impact of Culturelle probiotics on the gut microbiome of laboratory mice within a two-week timeframe. Despite encountering challenges in the initial phase of the study, such as unsuccessful 16S extraction from feces,cecum extraction and investigation will be used from this point forward within this research.     Relationship between restricted ankle dorsiflexion and knee kinematic changes during static and dynamic movements in physically active adults: A critically appraised topic

    Clinical scenario: Knee injuries are prevalent among athletes across a multitude of sports whether it be females or males. Restricted joint motion above or below the knee joint can contribute to the incidence of these injuries during static or dynamic movements.Clinical Question: Can restricted ankle dorsiflexion cause knee kinematics changes during static and dynamic movements in physically active adults?Summary of Key Findings: A literature search was conducted on the relationship between restricted ankle dorsiflexion and knee kinematics. Three studies that measured weight-bearing ankle dorsiflexion and knee kinematics through static and dynamic movements were included. One study demonstrated that a decrease in ankle mobility can lead to knee landing errors. The other two studies indicated that restricted ankle flexion can lead to an increase in knee valgus and internal rotation at the knee.Clinical Bottom Line: There is moderate evidence that supports restricted ankle dorsiflexion could contribute to changes in knee kinematics during static and dynamic motions.Strength of Recommendation: Based on the Physiotherapy Evidence Database, these studies are categorized as a 7 out of 10 in that restricted ankle dorsiflexion can cause changes in knee kinematics during static and dynamic movements

    More than a building with books: Resistance, resilience, and the fight for African American public library access, 1900-1970

    This research examines the intersections between Black public libraries, the public library movement, and twentieth-century civil rights history. Both public library and civil rights historiography has only recently begun to reckon with the history of segregation and the realities of democracy in libraries that were “free for all.” This study utilizes a thematic perspective to examine Black public library development, education, professional membership, community engagement, and civil rights activity. Oral histories, newspaper reports, and publications from Black librarians and the American Library Association are used to examine these subjects from local and institutional lenses. Public libraries currently serve as an important community establishment, but their reputation as a democratic institution has disguised a history of supporting inequality. Histories of Black public libraries must be written to tell the full and true history of public libraries in America. From their development, Black public libraries were a community effort to resist the racist climate around them. Initially developed to gain autonomy over their education and create a place for uplift, libraries also became a place to work, learn, create art, preserve history, and participate in political activism. Black libraries themselves were a form of resistance to the white constructions of American democracy. This history is significant because modern issues within public libraries have roots in the historic issues of institutionalized racism and public library segregation

    Does the somatosensory cortex modulate group Ia afferent spinal circuitry in young and older adults

    The natural progression of aging leads to a decline in the ability to sense the world around us which highly impacts our ability to shape motor output. The relationships between sensory afferents and motor neurons are still being elucidated today. Much of the foundational research relies on the translation from animal models to humans, sometimes which does not carry over well between species. PURPOSE: To determine if the somatosensory cortex (S1) modulates the Ia afferents in the spinal cord of humans. This study also aims to quantify the changes in reflex modulation with aging. We hypothesized transcranial magnetic stimulation (TMS) stimulation of the S1 will lead to a change in the amplitude of the H-reflex that will be either exacerbated or blunted in older adults. METHODS: Twenty-six subjects (20-70 yrs) have completed this study so far (young n = 14, older n = 12). Soleus H-reflex amplitude and latency was measured while holding an isometric contraction 5% of plantarflexion MVIC. TMS pulses were delivered to the premotor, primary motor, and somatosensory cortices calculated to arrive at the L4 level of the spinal cord approximately 5 ms prior to a soleus H-reflex volley evoking an H-reflex 50% of H-max. RESULTS: Mixed factorial ANOVAs revealed no condition × age interaction for amplitude (p = 0.120, η2p= 0.084) and no main effect for age (p= 0.054, η2p= 0.146). However, there was a main effect of modulation (p < 0.001, η2p = 0.665), as the amplitude for the control condition was significantly less than all 3 cortical stimulation conditions. H-reflex latency showed no condition × age interaction (p = 0.376, η2p= 0.042). There was no main effect of condition (p = 0.376, η2p= 0.058). However, there was a main effect of age (p < 0.001, η2p= 0.349), showing greater latency in older adults compared to younger adults for control as well as each condition except S1. CONCLUSION: The findings of this study suggest S1 may modulate the H-reflex. However, there are some concerns with lower body TMS and cortical modulation of spinal cord circuitry thus warranting further investigation to support the findings here

    Encapsulation and targeted delivery of glucose oxidase for prostate cancer therapy

    Glucose oxidase (GOX) is an enzyme that generates reactive oxygen species (ROS) and has been considered a potential therapy for treating cancer. However, to improve the efficacy of the treatment and reduce its systemic toxicity and immunogenicity, a targeted delivery system is needed to retain GOX in the intracellular space of the tumor. In this study, we encapsulated the GOX using a library of poly-l-lysine-grafted-polyethylene glycol (PLL-g-PEG) copolymers to improve its cytotoxicity toward cancer cells, reduce systemic cytotoxicity and delay the clearance by the immune system. The polymeric shell was conjugated with a monoclonal antibody targeting the prostate-specific membrane antigen (PSMA) for a tumor-specific treatment. Two resulting nanoparticles (NPs) with an average size of 30 nm in diameter and neutral ζ-potential showed a high effectiveness in cytotoxicity toward PSMA-expressing prostate cancer cells (~30% cell viability) compared to unmodified GOX (~90% cell viability) in vitro. The efficacy of NPs for a single dose treatment was evaluated in vivo using mice tumor models. The results for the intratumorally injected targeted NPs showed effective suppression of tumor growth and prolonged survival for mice by 4-fold compared to untreated mice and by 1.8-fold compared to free GOX. In addition, toxicity and immunogenicity experiments on this system indicated that the resulting NPs effectively reduced the general toxicity of GOX and showed no major immunogenicity in therapeutic doses. In addition, we conjugated superparamagnetic iron oxide nanoparticles (SPIONs) to GOX for synergistic cytotoxicity of enzymes and made targeted nanoparticles based on SPION-GOX. These anti-PSMA-PLL-g-PEG/SPION-GOX nanoparticles showed enhanced cytotoxicity (~20% cell viability) compared to targeted nanoparticles based on GOX alone. All these results lead us to believe that encapsulation and targeted delivery of GOX and SPION-GOX can provide an efficient and safe therapeutic agent for the treatment of prostate cancer

    New perspective on a classical experiment: Understanding the effects of asymmetry on the double-slit experiment

    The double-slit experiment performed by Thomas Young over two-hundred years ago established that light has wavelike properties. Since then, methods derived from double-slit experiment have become fundamental to numerous precision measurements.Textbook understanding of the double-slit experiment has evolved upon analyzing the wave interference associated with symmetric double-slit. Symmetric double-slit refers to the two slits being identical in terms of their physical dimensions (size and shift from the optical axis) and properties (color) of light passing them. A practical configuration of double-slit will, however, never be exactly symmetric between the two slits. And interpreting the interference patterns associated with asymmetric double-slit by assuming symmetric double-slit may bring in systematic error affecting the precision of measurement that double-slit has to offer.The objective of this research is to theoretically understand how and to what extent the slit-asymmetry affects the wave-interference of double-slit. The theoretical analysis is based on implementing slit-asymmetry into the Fresnel-Fraunhofer treatment of light standard to modeling double-slit interference. The Fresnel-Fraunhofer treatment calculates the complex electrical field of light on an observing plan as the superposition of the light filed originating from the two slits. By adjusting the asymmetry of the complex electrical fields originating from the two slits, it is possible to analytically predict the resulted double-slit interference for comparing against existing results of numerical simulations [Piao, 2022]. Two types of slit-asymmetries will be considered: asymmetry of the geometric dimensions and asymmetry of the light properties.     Development of a sizing and modeling platform for district energy systems with geothermal heat pumps

    Existing tools for community or urban-scale energy system modeling and simulation often have limitations and require expert-level modeling proficiency to develop system models. To address this gap, we propose an integrated sizing and modeling platform for district energy systems with geothermal heat pumps. The proposed platform utilizes both geometric and non-geometric user inputs related to buildings, borefields, and district energy loops. The platform automatically sizes the geothermal heat exchanger, generates a corresponding district energy system model, and conducts an annual simulation. Validation of the borefield component model against EnergyPlus ensures reliable simulation performance. A case study is provided in this paper to demonstrate the workflow and plausibility of simulation results from the proposed platform
