Determination of the tumor suppressor properties of INI-1 in a human cancer cell line
Rhabdoid tumors have historically been characterized by their histological phenotype, but genetic investigations have revealed a common characteristic: the loss or mutation of the INI-1 gene. INI-1 (SMARCB1) is a tumor suppressor gene located at 22q11.2 which encodes a subunit of the SWI/SNF chromatin remodeling complex. Originally identified in the Atypical Teratoid Rhabdoid Tumor (AT/RT) found in pediatric patients, they have been implicated in the pathogenesis of numerous tumor types. However, few, if any, studies have investigated the actual tumor suppressing properties of this gene. The goal of this study is to investigate and determine if the deletion of the INI-1 gene within the human genome leads to a tumorigenic phenotype which can be identified in future studies using assays measuring cell proliferation and growth. This report details the creation of INI-1 deficient cell lines that can be used in future studies. These experiments aim to establish a causal relationship between INI-1 loss and tumor progression and eventually assist with the development of therapies to treat INI-1 deficient tumors
Autonomy, competence, and relatedness support for virtual charter school teachers: A case study
This qualitative case study explores teacher support and persistence in a large virtual charter school in the midwestern part of the United States. Using self-determination theory as a theoretical lens, this study examines how the psychological needs of autonomy, competence, and relatedness of teachers in a virtual charter school are supported or thwarted. Findings suggest that teachers’ needs for autonomy and competence can be satisfied in a virtual charter context, while relatedness is a more difficult need for virtual charter schools to meet. This study also provides support for the utility of self-determination theory as lens for the further exploration of teacher support and retention in virtual schooling
Does economic inequality really hurt democracy?
Scholars have shown great interest in the relationship between democracy and economic inequality for a long time. Based on the literature currently available, political scientists, economists, and academics from several disciplines concur that excessive economic inequality is detrimental to democracy. Scholars have generally depended on survey data or specific democratic metrics, which consistently demonstrate that as economic inequality increases, the quality of democratic institutions and public satisfaction with democracy decreases. To learn more about whether inequality hurts democracy as the current consensus suggests is my main objective for this paper. To do this, I am using survey data from the World Value Survey (WVS) and the core measurements of democracy from the V-Dem indices in conjunction with the disposable Gini inequality index from SWIID (Standardized World Income Inequality Data) to determine the potentially negative relationship between economic inequality and democracy. According to my research, while not all democratic procedures or institutions are harmed, an increase in economic inequality appears to negatively impact institutional democracy partially. Meanwhile, a growth in economic inequality has little effect on citizens' satisfaction with democracy. However, my findings mostly go counter to the body of existing research, and therefore, more research is necessary to comprehend the potentially detrimental consequences of inequality on democracy fully. Meanwhile, my research is the first of its kind given that I try to show economic inequality’s impacts on democracy through both democratic institutions and individual citizens’ assessment. Also, I take a broader account of democracy by using all five V-Dem core democracy indices which will help us to understand inequality’s impacts on democracy much more comprehensively
Influence of oxygen scavenger technology and tray composition on retail stability of beef strip steaks and skirt steaks
The objective of this study was to compare the efficacy of two different oxygen scavenger strengths in master packages of beef strip and skirt steaks, in two different trays: foam and fiber compostable. Two master bags of each treatment were randomly selected for opening after 10, 15, and 18 d of dark storage and the trays were placed in the display case for 5 d of retail storage. Objective color measurements and subjective visual color panel data were collected on d 1 – 5 of retail display. No interaction was observed (P > 0.05) between scavenger × tray type combinations and pull day for any measurement taken. Regardless of time in dark storage, muscle color and surface discoloration increased (darkened and discolored), and overall acceptability decreased with retail day for both strip steaks and skirt steaks. All treatments for each pull day and retail display day remained below the discriminatory threshold for all visual color analysis factors. All total plate counts were well below any indicator for bacterial spoilage for all steaks. All scavenger/tray type combinations performed similarly throughout the study
Mechanical response and failure of bi-modulus and transversely-isotropic materials at different strain rates
This study investigates the response of bi-modulus and transversely-isotropic shale material with respect to elastic and mechanical properties. Primarily, these properties are characterized in to explore the effects of presence of bedding. In materials with well-defined bedding, the influence of bedding planes (Parallel and Perpendicular to loading direction) and stress state (Tensile or Compressive) on material properties are investigated. Several distinct shale materials from Oklahoma and Utah are subjected to quasi-static and dynamic loading resulting in strain rates of 3.28 × 10⁻⁵ s⁻¹ and 1500 s⁻¹ to characterize their elastic constants, mechanical strength, and strain rate dependence. A novel experimental--numerical technique based on full-field measurements of stresses, strains, and linear-least squares regression analysis is developed to quantify these properties. Full-field strains due to applied load are measured on the deforming specimen surface using Digital Image Correlation (DIC). Full-field stresses are numerically calculated using Finite Element Method (FEM) from experimental measured load. This technique is successfully utilized to characterize isotropic, transversely-isotropic, and bi-modulus materials.
The unique advantage of field measurements allows to characterize bulk material response. Shale materials with distinct microstructure exhibited different responses; isotropic bi-modulus, transversely isotropic bi-modulus, and transversely-isotropic with no bi-modulus effect in quasi-static tests with varying failure strengths in tension and compression. Dynamic loading experiments are performed using split-Hopkinson pressure bar (SHPB) on same shale materials to evaluate rate dependence of material properties. The anisotropic damage distribution and bi-elastic properties exhibited by shale materials in different directions and stress states promises beneficial for optimizing hydraulic fracturing technology and enhancing efficiency
Inflammatory sources: Exploring Google searches for gastritis
Introduction/Objectives: Gastritis is a general form of upper abdominal pain. With its high frequency and the copious non-surgical and surgical treatment options, we believe patients are likely searching the internet for questions applicable to gastritis. No investigation has ever been completed into gastritis Google searches, therefore we sought to classify these questions as well as assess their levels of quality and transparency using Google’s Frequently Asked Questions (FAQs).Methods: We searched Google using questions on gastritis and extracted the sources of a minimum of 200 questions for evaluation. Information transparency was assessed using the Journal of the American Medical Association (JAMA) Benchmark, classification was defined using the Rothwell Classification of Questions, and information quality was assessed using Brief DISCERN.Results: Our Google search returned 228 unique FAQs after removing duplicates and unrelated FAQs. The majority were classified as fact-based questions (103/228, 45.2%), followed by value (76/228, 33.3%), and policy questions (49/228, 21.5%). Most FAQs pertained to surgical treatment (127/228, 55.7%), followed by non-surgical treatment (66/228, 28.9%), and then disease process (35/228, 15.4%). The one-way analysis of variance revealed a significant difference in median quality scores among the 5 source types (H(4) = 18.97, P <.001) with medical practices (12/30) scoring the lowest compared to government (16/30), academic (16/30), and media outlet (22/30) sources which were found to have the highest.Conclusions: To increase the transparency and quality of online information regarding gastritis treatment, sources should refer to accepted standards such as JAMA Benchmark and Brief DISCERN when publishing online information
College students food choices and social environment surrounding food consumption at Oklahoma State University: A photovoice project
This research sought to explore the intricate dynamics influencing snack choices among college students, emphasizing healthiness, taste, convenience, and price. By synthesizing existing literature on food insecurity, Photovoice methodology, and empirical studies, this study aimed to contribute to a comprehensive understanding of factors shaping food decisions. Through the innovative Photovoice research method, it strove to capture authentic insights into the real-world complexities of snack choices and the social environment surrounding food consumption among college students at Oklahoma State University and what students suggested to better suit their consumption needs
Ultramicrotomy and infrared imaging applied to the forensic examination of automotive paint
In the forensic examination of automotive paint, each layer of paint is analyzed individually by infrared (IR) spectroscopy. Laboratories in North America typically hand section each layer and present each separated layer to an IR spectrometer for analysis, which is time consuming. In addition, sampling too close to the boundary between adjacent layers can pose a problem as it produces an IR spectrum that is a mixture of two layers. Not having a “pure” IR spectrum of each layer will prevent a meaningful comparison between each paint layer or in the situation of searching an automotive database will prevent the forensic paint examiner from developing an accurate hit list of potential suspects. These two problems have been addressed by collecting concatenated IR data from all paint layers in a single analysis by interrogating the cross sectioned layers of the paint sample using Fourier transform infrared (FTIR) imaging microscopy. Using an ultramicrotome instead of a conventional microtome allows for so-called “small” paint chips which are often found on the clothing of a pedestrian in a hit-and-run to be cross sectioned and analyzed than what is practical by conventional FTIR. Decatenation of the concatenated IR data is achieved by multivariate curve resolution using alternating least squares (ALS) or modified alternating least squares (MALS) to obtain an IR spectrum of each automotive paint layer. A new baseline correction algorithm that integrates three well known baseline correction methods developed a snapshot of the background and minimized error in the MALS recovered spectra. In addition, limiting the FTIR spectrum of each layer to the region corresponding to the binder is shown to be sufficient to identify the assembly plant of the automotive vehicle and perform accurate library searches to identify the vehicle manufacturer, line, and model of the vehicle from which the original equipment manufacturer paint chip originated
Exploring Belizean secondary English language arts teachers’ experiences during a place-based writing professional development workshop in the Toledo district
The concept of place is more than where people reside. Places are important in shaping the identity and experiences of people. Exploring the idea of relationships with place creates a context that ignites transformative thoughts and actions. “Place’ is formed out of the particular set of social relations which interact at a particular location” (Massey, 2005, p. 168). The purpose of this qualitative case study design was to investigate selected secondary ELA teachers’ experiences, pedagogical shift, and envision in implementing place-based writing pedagogy during and after a place-based writing professional development in Toledo District, Belize, Central America. The study’s design offered the researcher a detailed description of the data collected from the four cases. The artifacts used to collect the data were pre-survey, an open ended post-survey, observations, poem, short story, three written reflections, and a focus group interview. The data was analyzed through thematic analysis. The study was framed in the critical pedagogy of place theoretical framework and added significantly to the responsive writing practices of cultural education research in Belize. The findings revealed the participants’ successfully experienced place-based writing workshop. The data revealed to the extent to which the participants attributed their writing pedagogy shift to the place-based writing workshop. The participants’ uptake of place-based writing practices included critical place-based lens, peer-review, and writer’s notebook. Finally, the data identified how participants envision implementing place-based writing in their lessons. The participants acknowledged how the place-based writing workshop provided them with knowledge and skills needed to make writing more relevant for their students. Additionally, the participants were immersed in a variety of learning strategies that supported their writing knowledge. These findings confirmed the importance of a place-based writing workshop and the need to provide a professional development that is content specific for ELA teachers in Belize
