Sandalwood (Santalum album), which belongs to Santalaceae family, is a commercially important tree in Indonesia due to its essential oil contents used for important essence of perfume in the perfumery industry. However, its population has significantly depleted since the planting materials of this tree using conventional methods are difficult to be provided. This study was conducted to mass propagate sandalwood using in vitro methods through friable embryogenic callus (FEC). The somatic embryos were formed using leaves in length of 1-3 and 4-7 mm cultured in MS medium containing 0.5 mg/l and 1 mg/l indole acetic acid (IAA), MS medium supplemented with 1 mg/l IAA and 0.2 mg/l kinetin and half concentration of MS medium supplemented with 1 mg/l Gibberellic acid (GA3). Primary somatic embryos (PSE) and secondary somatic embryos (SSE) then formed friable embryogenic callus when it repetitively transferred into MS medium supplemented with 1.7 mg/l BAP, 1 mg/l proline or 1.5 mg/l BAP and 1.2 mg/l kinetin every 3 weeks. The FEC shows its optimum maturation and regeneration in the MS medium supplemented with 1.5 mg/l BAP and 1.2 mg/l kinetin for 4-8 weeks. The acclimatization of sandalwood plantlets were perfectly conducted in the medium containing soil, sand and compos in ratio of 1:1:1 with the companion plant namely Murraya paniculata, (L) Jack which gave the best percentage of survival rate and the lowest percentage of fallen leaves. These findings may improve the massive propagation of sandalwood through FEC as well as a useful material for further genetic improvement of sandalwood by using FEC as material for genetic transformation

    No full text
Recombinant human Granulocyte Colony Stimulating Factor (G-CSF) has been produced in a soluble form in Escherichia coli BL21 (DE3) as a fusion protein. The open reading frame of G-CSF was synthetically constructed in previous work and was codon optimized for best expression in E. coli. In this research, the gene was fused to thioredoxin (Trx) at the N-terminal in pET32 vector. The purpose of this research was to optimize the overproduction and purification processes to obtain high yield recombinant protein in soluble form, and to characterize the Trx-G-CSF fusion protein. Overproduction was performed using IPTG induction method for 3 and 6 hours. The protein was purified by Ni-NTA affinity chromatography and separated using gradient concentration of imidazole. The purified protein was then characterized by SDS-PAGE and Western Blot analysis. Further, enterokinase was used to separate G-CSF from the fusion protein. The purified form of G-CSF was subsequently characterized using Western Blot and mass spectrometry using MALDI-TOF. The results showed that the fusion protein was successfully produced in soluble part as much as 48.25% were obtained after 3 hours of induction. The yield of fusion protein was 67.37% from total protein (229.65 mg protein/L culture). The Western Blot analysis showed the G-CSF band at around 18.6 kDa. Mass spectrometry with MALDI-TOF/ TOF revealed that 25.86% of amino acid residue was recognized as part of human G-CSF sequence

This study uses a qualitative approach with a case study method through interviews, field observations, and documentation with the research subject of 5 new students of SMK Taman Wisata from the four skill competencies. Based on the findings, the rational choice of new students in choosing a tourism vocational school in the Covid-19 era is divided into three factors. Sociologically, new students choose tourism vocational schools because they are influenced by success stories from alumni and those closest to them. Economically, choosing the tourism vocational high school is considered less costly than other high school options. Psychologically, there is a desire to realize their dreams and want to work immediately after graduation. The goals of each of the five new students choosing a tourism vocational school are different, including working in accordance with their field and pursuing their goals. However, this goal is not a single goal; there are other goals as an alternative strategy if the main goal is not achieved

A previous study has shown that households with both the head and the spouse joining the labor force tend to exit from poverty. In Indonesia, women that actively participated in labor market were relatively small, only around 50% in 2017. Meanwhile, most of the women in Indonesia were married in 2017. A husband was one of the factors affecting their wife’s decision to participate in labor market. This study investigates married women’s employment conditions and the effects of husband’s occupation and education on their labor market participation. The research sample consists of women aged 15-year-old and above, married, and living with their husband. We found that the higher the husband’s education, the lower married women’s probability of participating in the labor market. Husbands with an informal occupation increased married women’s probability to be in the labor force

Creative economy is a potential sector that can be a source of renewable economic growth as a solution for the government to prevent the slowing down economy. Export from the creative economy indicates a good result. Export is also one of the components to count Gross Domestic Product (GDP). That means the rise in export from the creative industry will increase the economic growth from the creative economy, which may lead to the increasing of economic growth as a whole. The objective of this paper is to know the impact of the creative industry to GDP growth from the creative industry. In order to get creative economy export in 2019, we forecast creative economy export using the ARIMA model. The result showed that ARIMA (0,1,1)(2,0,1)12 is the best model to get creative economy export 2019, which is 20.87 million USD or there is an increase in creative economy exports of 0.70 billion USD from 2018. The additional value of creative economy exports is equivalent to 15.54 trillion rupiahs. Using Input-Output analysis table of creative economy, it is obtained that the additional creative economy exports of 15.54 trillion rupiahs will provide an additional creative economy GDP of 7.81 trillion rupiahs. Hence, the creative economic growth in 2019 will be 0.71 percent. Unfortunately, this creative economic growth is smaller 0.38 percent from 2018
