    Cellular dynamics and molecular mechanisms underlying the 3D organization and connectivity of the statoacoustic ganglion

    The statoacoustic ganglion (SAG) is a complex 3D structure composed by neurons in charge of transmitting the information from inner ear hair cells to the CNS. During development, SAG copes with otic tissue demands to maintain functionality. However, SAG development in coordination with otic development has not been addressed in detail. We use high resolution confocal imaging of otic neuroblasts (NB), photoconversion, photoablation, transgenic lines, CRISPR/Cas9 and Cas13 to address SAG development at the molecular and cellular levels. We find a population of pioneer SAG neurons specified outside the otic placode, which play an attracting role over delaminating NB affecting its coalescence. NB from the otic epithelium delaminate in an EMT-like manner, and non-collectively and actively migrate depending on RhoGTPases to establish the anterior SAG lobe. Followingly, NB crawl onto pioneer axons to form the posterior SAG lobe. Lack of both pioneer SAG neurons and pioneer axons alters SAG shape. Finally, we uncover the cell adhesion molecule Cntn2 and the chemokine Cxcl14 as two new molecules required for correct pioneer axon targeting to HC and posterior lobe formation. We confirm that HC and neurotrophin signaling is not required for directed axon targeting but stabilization and neuronal survival. In conclusion, SAG development is a complex process in which several mechanisms interplay, prime and scaffold further SAG developmental steps.El gangli estatoacústic (SAG) és una estructura 3D altament complexa formada per neurones que transmeten la informació rebuda de les cèl·lules ciliades (HC) del l’oïda interna al sistema nerviós central (CNS). Durant el desenvolupament, el SAG s’organitza en coordinació amb l’oïda interna per tal de mantenir la seva funcionalitat. No obstant això, es té un lleu coneixement com aquest s’organitza en 3D. Per tal d’analitzar a nivell molecular i cel·lular la formació del SAG, hem emprat microscopia confocal d’alta resolució amb marcatges de neuroblasts (NB) individuals, fotoconversió, fotoablació, CRISPR/Cas9 i Cas13. Aquí descrivim el paper de les neurones pioneres del SAG, que atrauen els NB que delaminen de l’epiteli òtic. Els NB delaminen fora de l’epiteli seguint un procés EMT. Un cop fora, els NB migren activament dependent de RhoGTPases i no col·lectiva per tal de formar un lòbul anterior del SAG. Seguidament, els NB migren sobre axons pioners per tal de formar un lòbul posterior del SAG. La manca de neurones pioneres i d’axons pioners altera la formació del SAG. Finalment, hem descobert que la molècula d’adhesió Cntn2 i la quemoquina Cxcl14 son relevants per a la correcta extensió i orientació dels axons pioners a les HC de l’oïda i la formació del lòbul posterior. Confirmem que les HC i la senyalització per neurotrofines no és requerida per a la correcta formació dels axons pioners, però sí per a estabilitzar-los i promoure la supervivència neuronal. En conclusió, l’estudi proporciona nova informació sobre els comportaments que permeten als NB establir un gangli estructurat i les claus moleculars implicades en la formació de connexions neuronals amb l’oïda interna.Programa de doctorat en Biomedicin

    Rethinking Thatcher and Europe: an alternative reading

    The goal of this doctoral dissertation is to establish the raison d’être of the policy options that Margaret Thatcher adopted on European integration in the years of her premiership. She was the Prime Minister of the United Kingdom from 1979 to 1990, years of major developments in European integration. The thesis wishes to clarify the expectations Thatcher had over the European Community and the decisions she took as British Prime Minister about the integration process through archival research and by placing Thatcher’s European options within the broader context of the 1980s. I aim to show that she balanced ideology and pragmatism when reacting to the positions of the other European leaders, often very different from hers, and to the proposal and decisions of the main institutions of the Community. This research is original to the point it deconstructs the mainstream narrative of Thatcher as Eurosceptical and reconstructs a more comprehensive and nuanced outlook of her as British Prime Minister, pragmatic in her adapting to circumstances but not incoherent in her overall attitude.El objetivo de esta tesis doctoral es establecer la razón de ser de las opciones políticas que Margaret Thatcher adoptó sobre la integración europea en los años de su presidencia. Fue primera ministra del Reino Unido de 1979 a 1990, años de importantes avances en la integración europea. La tesis desea aclarar las expectativas que Thatcher tenía sobre la Comunidad Europea y las decisiones que tomó como primera ministra británica sobre el proceso de integración a través de la investigación de archivos y colocando sus opciones europeas en el contexto más amplio de la década de 1980. Mi objetivo es demostrar que ha equilibrado la ideología y el pragmatismo al reaccionar ante las posiciones de los demás dirigentes europeos, a menudo muy diferentes de la suya, y ante la propuesta y las decisiones de las principales instituciones de la Comunidad.     The gastrulating zebrafish under heat waves and cold spells: developmental robustness and vulnerabilities under changing temperatures

    Temperature affects all processes, including development, and yet, it remains unknown how and/or if underlying mechanical and biochemical events are coordinated to slow down development robustly at lower temperatures. Here, using a variety of methods from molecular biology to 5D live imaging, I uncover the vulnerabilities of the zebrafish epithelial during gastrulation at sub-optimal temperatures (20° C). The enveloping layer (EVL) in the cold displays large multi-nucleated cells stemming from cytokinesis failures. Differences in tissue tension during late epiboly also led to rupturing of the EVL in certain embryos. Furthermore, mesendoderm patterning reveals convergence-extension defects under varying temperatures, suggesting that different aspects of gastrulation are affected in parallel. Proteome profiling across five different temperatures also reveals processes affected under temperature fluctuations. This PhD project is one of the first studies focusing on the effect of temperature on gastrulation and exposes mechanisms behind developmental failures under sub-optimal temperatures.La temperatura afecta a tots els processos i, no obstant això, es desconeix com i/o si els esdeveniments mecànics i bioquímics es coordinen per alentir el desenvolupament a temperatures més baixes. Aquí, utilitzant des de la biologia molecular fins a la microscòpia en 5D, descobrim les vulnerabilitats de la gastrulació en l'epiteli del peix zebra a temperatures subòptimes (20° C). La capa envoltant (EVL) en fred mostra cèl·lules multinucleades derivades de fallades de citocinesi. Diferències en la tensió tissular durant l'epibòlia tardana també provoquen la ruptura de l'EVL en certs embrions. A més, el patró de mesoderma revela defectes d'extensió-convergència sota temperatures variables, suggerint que diferents aspectes de la gastrulació es veuen afectats. L'anàlisi proteòmic en cinc temperatures diferents també revela processos afectats per les fluctuacions de temperatura. Aquest projecte de doctorat és dels primers en centrar-se en l'efecte de la temperatura sobre la gastrulació i exposa els mecanismes rere conseqüents fallades de desenvolupament.Programa de Doctorat en Biomedicin

    Genetics, energetics, and allostery in proteins with randomized cores and surfaces

    A lack of systematic experimental data limits our understanding of protein evolution. In this study, we experimentally characterized proteins with randomized sequences. Vast numbers of amino acid combinations constitute stable protein cores and surfaces. However, alternative cores frequently disrupt protein function by indirect allosteric effects. Both protein stability and binding can be predicted using simple additive energy models with a small contribution from pairwise energetic couplings. Indeed, energy models trained on one protein can predict functional cores and surfaces across more than a billion years of evolution, with only rare energetic couplings that we experimentally identify limiting the transplantation of cores between highly diverged proteins. Curs 2024-2025Tutor: Pablo Cruz Mantilla de los RíosLa cláusula de identidad nacional, consagrada en el artículo 4.2 del Tratado de la Unión Europea (TUE), ha adquirido una relevancia creciente en el debate jurídico europeo contemporáneo. Inicialmente concebida como una fórmula de respeto a la diversidad constitucional de los Estados miembros, su invocación reciente en contextos de conflicto con el Derecho de la Unión plantea interrogantes sobre su alcance y su compatibilidad con los valores fundamentales recogidos en el artículo 2 TUE. Este trabajo tiene como objetivo analizar críticamente el uso y los límites de dicha cláusula, especialmente cuando es empleada por ciertos gobiernos nacionales para justificar reformas que amenazan el Estado de Derecho o para impugnar la primacía del Derecho europeo. En este contexto, se examina la evolución normativa de la cláusula, su interpretación por parte del Tribunal de Justicia de la Unión Europea (TJUE) y su utilización por Tribunales Constitucionales nacionales. Asimismo, se valora si la cláusula actúa como un mecanismo legítimo de equilibrio constitucional dentro de la Unión Europea (UE) o si, por el contrario, está siendo instrumentalizada como una herramienta de confrontación que socava la cohesión normativa y axiológica del ordenamiento europeo

    Enhancing human NK cell antitumor function by knocking out SMAD4 to counteract TGFβ and activin A suppression

    Transforming growth factor beta (TGFβ) and activin A suppress natural killer (NK) cell function and proliferation, limiting the efficacy of adoptive NK cell therapies. Inspired by the partial resistance to TGFβ of NK cells with SMAD4 haploinsufficiency, we used CRISPR-Cas9 for knockout of SMAD4 in human NK cells. Here we show that SMAD4KO NK cells were resistant to TGFβ and activin A inhibition, retaining their cytotoxicity, cytokine secretion and interleukin-2/interleukin-15-driven proliferation. They showed enhanced tumor penetration and tumor growth control, both as monotherapy and in combination with tumor-targeted therapeutic antibodies. Notably, SMAD4KO NK cells outperformed control NK cells treated with a TGFβ inhibitor, underscoring the benefit of maintaining SMAD4-independent TGFβ signaling. SMAD4KO conferred TGFβ resistance across diverse NK cell platforms, including CD19-CAR NK cells, stem cell-derived NK cells and ADAPT-NK cells.     Integrative methods for linking genotype to phenotype: an application to intervertebral disc degeneration

    Intervertebral disc degeneration (IDD) is a complex condition influenced by genetics, environment, and lifestyle. Despite advancements in sequencing technologies, challenges remain in understanding the biology of complex diseases. The increasing use of artificial intelligence has led to innovative approaches in studying these conditions, benefiting translational medicine, drug discovery, and personalized treatment. This thesis introduces novel computational frameworks for studying the genetics of complex disorders, including a tool based on knowledge graph embeddings and Genopyc, a Python library for variant analysis. We exploited these tools in the study of biological underpinnings of IDD. Additionally, we investigated the autoimmune basis of Modic change, a comorbidity of IDD, using TwinsUK, the largest twin cohort worldwide. Our findings demonstrate the successful utility of these computational tools in advancing the understanding of IDD biology, emphasizing the importance of integrating diverse data sources to achieve a comprehensive understanding of this intricate condition.La degeneració del disc intervertebral (IDD) és una condició complexa influïda per la genètica, l'entorn i l'estil de vida. Malgrat els avanços en tecnologies de seqüenciació, persisteixen els reptes en comprendre la biologia de malalties complexes. L'augment de l'ús de la intel·ligència artificial (IA) ha portat a enfocaments innovadors en l'estudi d'aquestes condicions, beneficiant la medicina translacional, la descoberta de medicaments i el tractament personalitzat. Aquesta tesi presenta nous marcs computacionals per estudiar la genètica de trastorns complexes, incloent-hi una eina basada en "knowledge-graph embeddings” (KGE) i Genopyc, una llibreria de Python per a l'anàlisi de variants. Hem aprofitat aquestes eines en l'estudi de les bases biològiques de l'IDD i hem investigat la base autoimmunitària del canvi de Modic, una comorbiditat de l'IDD, utilitzant TwinsUK, la cohort de bessons més gran del món. Els nostres resultats demostren la implementació reeixida d'aquestes eines computacionals en l'avenç de la comprensió de la biologia de l'IDD, posant èmfasi en la importància d'integrar fonts de dades diverses per assolir una comprensió completa d'aquesta condició complexa.Programa de Doctorat en Biomedicin

    Advanced microscopy to resolve the higher-order organization of the exocyst

    The exocyst is an heterooctameric protein complex that tethers secretory vesicles to the plasma membrane during exocytosis. Each exocytic event is mediated by 10-15 exocysts. However, how multiple exocysts organize in time and space to accomplish their function remains unknown. We resolved the dynamic higher-order structure of the exocyst by correlating live-cell imaging and super-resolution microscopy. Fluorescent temporal markers demonstrated that multiple exocysts follow dynamic higher-order structures that switch from small and compact clusters to wider ring-like organizations with an asymmetric distribution of the exocysts around a central cavity. Conditional mutants linked conformational changes with defined molecular functions and cryo-electron tomography provided accurate descriptors of the vesicle geometry. We propose a model were the multiple exocysts bind the vesicle as compact clusters for four seconds. In a Sec9-dependent process, the exocysts open to form stable arcs and ring-like architectures that are disassembled by Sec18 eight seconds after tethering initiation.L’exocyst és un complex proteic que uneix vesícules de secreció a la membrana plasmàtica durant l’exocitosi. Entre 10 i 15 exocysts participen en cada esdeveniment d’exocitosi, però en desconeixem la seva l’organització espacial i temporal. La combinació de microscòpia en cèl·lules vives i de super-resolució ens ha permès resoldre la supra-estructura de l’exocyst. Mitjançant l’ús de marcadors temporals hem demostrat que aquesta supra-estructura és dinàmica: comença formant clústers compactes que es transformen en estructures anulars al voltant de les quals els exocysts es distribueixen asimètricament. Gràcies als mutants condicionals hem assignat canvis conformacionals a funcions moleculars concretes, i la crio-tomografia electrònica ens ha permès descriure geomètricament les vesícules. Proposem un model on els exocysts s’uneixen a la vesícula formant clústers durant 4 segons. Sec9 indueix l’obertura d’aquests exocysts, que passen a formar arcs i anells estables. 8 segons després de l’inici del procés, l’acció de Sec18 desmunta aquests anells.Programa de doctorat en Biomedicin

    Nonanatomic posteromedial bundle augmentation of the posterior cruciate ligament after hyperextension trauma

    The surgical management of posterior cruciate ligament (PCL) injuries can be challenging. As most PCL injuries occur in a flexed knee position, the anterolateral bundle is thought to be more commonly injured than the posteromedial bundle (PMB); however, in hyperextension, the PMB plays a more significant role. The smaller size of the PMB compared with the anterolateral bundle and its lower strength may explain why isolated hyperextension PMB injuries can be easily overlooked. In this Technical Note, a surgical technique to perform a nonanatomic PMB augmentation of the PCL using a gracilis tendon autograft or allograft is reported. These technical features aim to overcome current limitations in existing techniques to address the symptoms after partial PCL hyperextension injuries

    Evaluating the environmental and systemic viability of SCRAP for textile waste management in Catalonia

Curs 2024-2025Tutora: Rosa ColoméAquest estudi avalua la viabilitat ambiental i sistèmica del sistema de gestió de residus tèxtils SCRAP (Sistema Col·lectiu de Responsabilitat Ampliada del Productor) coordinat per RE-VISTE a Catalunya. D'acord amb la Directiva 2008/98/CE de la UE i la Llei 7/2022 d'Espanya, SCRAP té com a objectiu promoure la responsabilitat ampliada del productor per a la circularitat tèxtil. La recerca adopta una lent multidimensional per avaluar l'impacte ambiental dels sistemes i el paper crucial dels stakeholders. Es va dur a terme una Anàlisi del Cicle de Vida per simular diversos escenaris d'implementació. Els resultats mostren que augmentar la recollida selectiva de residus tèxtils (STW) redueix substancialment els impactes climatics, fins a un 75% de reduccions de CO₂- eq en escenaris d'alta recollida. Un model d'infraestructura descentralitzada basanse en les vegueria supera constantment les alternatives provincials a causa de la reducció de les emissions del transport. No obstant això, els residus tèxtils recollits no selectivament (UTW), que encara dominen, continuen sent la principal càrrega ambiental, i abordar-los requereix canvis de comportament del consumidors. Els grups de debat van revelar que els ciutadans no tenen clar quines peces de roba es poden reciclar, sovint exclouen la roba danyada i desconfien del destí dels tèxtils recollits. Per altre banda, la participació de les empreses ha d'anar més enllà del compliment normatiu per invertir en ecodisseny i materials reciclables. La participació de MANGO il·lustra un compromís prometedor però encara parcial. L'estudi conclou que SCRAP té un fort potencial d'implementació, però el seu èxit depèn de la millora de la infraestructura de recollida, les campanyes de sensibilització i l'adaptació del disseny per part de les empreses. Les limitacions metodològiques, com ara l'exclusió dels impactes de la reutilització/reciclatge i l'absència de comparacions de referència, destaquen àrees per a futures investigacions.This study evaluates the environmental and systemic viability of the SCRAP (Sistema Colectivo de Responsabilidad Ampliada del Productor) textile waste management system coordinated by RE-VISTE in Catalonia. In line with EU Directive 2008/98/EC and Spain’s Law 7/2022, SCRAP aims to promote extended producer responsibility (EPR) for textile circularity. The research adopts a multi-dimensional lens to assess the systems environmental impact and crucial role of stakeholders. A Life Cycle Assessment (LCA) was performed to simulate various implementation scenarios. Results show that increasing the selective collection of textile waste (STW) substantially reduces climate-related impacts, up to 75% CO₂-eq reductions in highcollection scenarios. A decentralized “vegueria” infrastructure model consistently outperforms provincial alternatives due to reduced transport emissions. However, unselectively collected textile waste (UTW), which still dominates, remains the key environmental burden, and addressing it requires behavioural shifts. Focus groups revealed that citizens lack clarity on what garments qualify for recycling, often exclude damaged clothing, and mistrust the fate of collected textiles. Simultaneously, company participation must go beyond compliance to invest in eco-design and recyclable materials. MANGO’s engagement illustrates promising but still partial commitment. The study concludes that SCRAP holds strong implementation potential but its success depends on improved collection infrastructure, awareness campaigns, and design adaptation on the companies side. Methodological limitations, such as exclusion of reuse/recycling impacts and absence of baseline comparisons, highlight areas for future research.Este estudio evalúa la viabilidad ambiental y sistémica del sistema de gestión de residuos textiles SCRAP (Sistema Colectivo de Responsabilidad Ampliada del Productor), coordinado por RE-VISTE en Cataluña. De acuerdo con la Directiva 2008/98/CE de la UE y la Ley 7/2022 de España, SCRAP busca promover la responsabilidad ampliada del productor para la circularidad textil. La investigación adopta una perspectiva multidimensional para evaluar el impacto ambiental de los sistemas y el papel crucial de los stakeholders. Se realizó un Análisis del Ciclo de Vida para simular varios escenarios de implementación. Los resultados muestran que el aumento de la recogida selectiva de residuos textiles (STW) reduce sustancialmente los impactos climáticos, con reducciones de hasta un 75 % de CO₂-eq en escenarios de alta recogida. Sin embargo, los residuos textiles recogidos de forma no selectiva (UTW), que aún predominan, siguen siendo la principal carga ambiental, y abordarlos requiere cambios de comportamiento. Los grupos de debate revelaron que la ciudadanía desconoce qué prendas son aptas para el reciclaje, a menudo excluyen la ropa dañada y desconfían del destino de los textiles recogidos. Simultáneamente, la participación de las empresas debe ir más allá del cumplimiento normativo e invertir en ecodiseño y materiales reciclables. La participación de MANGO ilustra un compromiso prometedor, aunque aún parcial.
