Disease-specific neuropathological alterations of the locus coeruleus in Alzheimer's disease, Down syndrome, and Parkinson's disease
Introduction: The locus coeruleus (LC), the brain's primary source of noradrenaline (NA), undergoes early neurodegeneration in Parkinson's disease (PD), Alzheimer's diseases (AD), and Down syndrome (DS); however, differences have not been examined in parallel. Methods: Post mortem brains (n = 67) from individuals with AD, DS-AD, and PD without and with dementia (PD-D) and controls were analyzed for amyloid beta (Aβ), phosphorylated tau (pTau), α-synuclein, endo-lysosomal alterations, biogenic amines, and selective biomarkers. Results: LC degeneration correlated with age, peaking in AD and PD-D, while NA and dopaminergic metabolites were significantly reduced only in PD-D. DS-AD, the youngest group, showed the highest Aβ and pTau levels but the least noradrenergic neuron loss. We demonstrated for the first time that endosomal alterations were present in AD, lysosomal changes were present in PD-D/DS-AD, and DYRK1A, a key protein from chromosome 21, was elevated only in DS-AD. Discussion: Loss of noradrenergic neurons may occur independently of amyloid and tau pathologies. Highlights: We provide the first analysis of neuropathological and biochemical features including biogenic amines of the LC in AD, DS, and PD. Loss of noradrenergic neurons was most severe in AD and PD. Only in DS, levels of DYRK1A - a kinase encoded on chromosome 21 and implicated in neurodegenerative processes - were elevated and negatively correlated to biogenic amine levels. Residential exposure to noise, green space, and children's language acquisition
Background: Noise pollution has been linked to impaired development in a variety of language-related skills in laboratory settings. While studies have focused on school environments, residential noise exposure's impact remains underexplored. Methods: We used multilevel regression models to examine the association between noise exposure measured using deterministic noise modelling and language development in kindergarten-aged children in Vancouver, Canada, between April 1, 2000 and December 31, 2005, measured through a questionnaire completed by kindergarten teachers (the Early Development Instrument). The models were adjusted for median income level and English as a Secondary Language (ESL) status, as well as random effects on teachers, and we explored the potential interaction effects of greenness, measured using satellite imagery. Results: The study included 33,153 children for which there were data on noise exposure and indicators of language development. The mean noise level was 63.5 dB(A), and the mean percentage of greenness within a 250-meter radius buffer zone was 31.8 %. We found that an increase in residential exposure to noise independently increased the odds of not meeting developmental expectations in basic literacy (OR: 1.18, 95 % CI: 1.12-1.25), advanced literacy (OR: 1.11, 95 % CI: 1.07-1.16), and communication and general knowledge (OR: 1.10, 95 % CI: 1.06-1.14). Greenness was found to have interaction effects with basic and advanced literacy skills in noisy environment. Conclusion: This study found residential exposure to noise was associated with poorer language development outcomes, with interaction effects of greenness observed in literacy skills. Future studies should also examine the long-term effects of residential exposure to noise on language development
Las máscaras proteicas de la estética de la crueldad: una propuesta de definición y de los principios fundamentales de la estética de la crueldad en la narrativa de ficción contemporánea
En la presente investigación se propone el análisis de la estética de la crueldad en la narrativa de ficción como un sistema de juegos del lenguaje a partir de un estudio de un corpus literario de 146 obras. Se formulan un conjunto de principios ─como el principio de soberanía agentiva─ y una tipología de elementos de primer grado ─el agente y el sujeto cruel, la víctima y el lector─ y de segundo grado ─el neronismo─ que permiten analizar la naturaleza y la composición de escenas literarias en las que se manifiesta lo cruel. Asimismo, se relaciona la estética de la crueldad con otras expresiones de estética negativa, como la estética de lo grotesco y de lo demoníaco, y se investigan múltiples aspectos de la gnoseología de la estética de la crueldad. Finalmente, estos instrumentos analíticos se aplican en el estudio de aspectos concretos de la novela de Roberto Bolaño 2666.In the present research the analysis of the aesthetics of cruelty in fictional narrative is proposed as a system of language-game based on a study of a literary corpus of 146 works. A set of principles are formulated, such as the principle of agentive sovereignty, and a typology of first-degree elements (the agent, the cruel subject, the victim and the reader) and of second-degree elements (like neronism) that allow to analyze the nature and the composition of literary scenes in which cruelty is manifested. Likewise, the aesthetics of cruelty are related to other expressions of negative aesthetics, such as the aesthetics of the grotesque and the demonic, and multiple aspects of the gnoseology of the aesthetics of cruelty are investigated. Finally, these analytical instruments are applied in the study of specific aspects of Roberto Bolaño’s novel 2666.In der vorliegenden Promotionsschrift wird die Analyse der Ästhetik der Grausamkeit in fiktionalen Erzählungen als System von Sprachspielen, ausgehend von einer Untersuchung eines literarischen Korpus von 146 Werken, vorgeschlagen. Es werden eine Reihe von Prinzipien formuliert ─beispielsweise das Prinzip der handelnden Souveränität─ und eine Typologie von Elementen ersten Grades ─dem Agens, dem grausamen Subjekt, dem Opfer und dem Leser─ und von Elementen zweiten Grades ─z. B. dem Neronismus─, die eine Analyse der Eigenschaften und Elementen literarischer Szenen ermöglichen, in denen sich das Grausame offenbart. Ebenso wird die Ästhetik der Grausamkeit mit anderen Ausdrucksformen negativer Ästhetik in Zusammenhang gesetzt, u. A. mit der Ästhetik des Grotesken und des Dämonischen, und es werden mehrere Aspekte der Erkenntnistheorie der Ästhetik der Grausamkeit untersucht. What role will refugee labour mobility play in the future of safe and legal pathways?: a case study of the Spanish experience
The 2018 Global Compact on Refugees acknowledged the restricted nature of resettlement and the need “to expand its base” by diversifying labour migration opportunities for refugees. Spain’s recent labour mobility project under its National Resettlement Plan offers a case study in this approach, making a shift in legal pathways for refugees. However, the relevance of refugee labour pathways and their normative implications has mainly been ignored. To address the research gap, this article aims to articulate the need for complementary pathways, contribute to theories of refugee labour mobility, and assess whether refugee labour mobility can and should be used as a tool to access territory, with a focus on the Spanish context. To this end, the analysis involves analysing multi-sited policy documents and conducting semi-structured interviews with stakeholders in the field of safe and legal pathways. The analysis demonstrated that, while refugee labour pathways have some potential to expand admission for displaced talent, the line between addition and substitution risks diluting long-term refugee protection. Conditioning admission on labour market utility excludes many in need of international protection and introducing the logic of cost-effectiveness undermines the solidarity inherent in the right to seek asylum, risking the commodification of the international protection system
Chronic circadian disruption in adolescent mice impairs hippocampal memory disrupting gene expression oscillations
Chronodisruption, the misalignment of internal biological systems with external environmental changes, negatively impacts health and cognition. A prevalent form, social jet lag, affects a large portion of adolescents and is associated with reduced academic performance, and impaired emotional regulation. Despite its prevalence, the mechanism by which adolescent chronodisruption results in cognitive deficits remains unexplored. Here, we induced chronic chronodisruption in adolescent mice through light/dark cycle manipulation. We performed a battery of established behavioral tests alongside an analysis of daily rhythms of expression of genes involved in relevant physiological functions. We report that adolescent circadian disruption induces impairments in short-term, social, and spatial memory without prompting anxiety-like behavior. Gene expression analyses revealed altered oscillation patterns of circadian clock genes in the hypothalamus and hippocampus. Furthermore, we identified alterations in daily patterns of genes involved in glucocorticoid and endocannabinoid transmissions as well as synaptic plasticity. This work emphasizes the importance of circadian homeostasis for cognitive function and hippocampal homeostasis. Evaluation of topoisomers of snake venom-derived peptides as potential antimicrobial and antitumor agents
This thesis reports an investigation of the effect of topoisomeric modification on the activity of two peptides: crotalicidin (Ctn), a cathelicidin from the venom of a South American rattlesnake, and Ctn[15-34], a Ctn segment with interesting antimicrobial and antitumoral properties, as well as excellent serum stability.
Topoisomeric refers to composition-identical (hence isomeric) yet three-dimensionally distinct (hence topo-) variants of a biomolecule where the structural maneuvers bring about potentially useful physiological effects. For peptides, the topoisomer term is often reserved to describe all-D-amino acid versions with sequences identical [i.e., enantio (e) form] or fully reversed [i.e., retroenantio (re)] relative to the native primary structure.
To comprehensively assess the properties and activities of Ctn and Ctn[15-34] e and re topoisomers, I have compared them with both the parent peptide and its sequence-inverted variant [retro (r) form, also treated as a topoisomer]. While all topoisomers were active against gram-negative bacteria and tumor cells, those featuring D-amino acids were more cytotoxic than their L- counterparts towards normal cells. Even so, the higher stability of the e and re versions in human fluids prompted further exploration.
Thus Ctn[15-34], Ctn re and Ctn[15-34] re were selected for an in vivo efficacy evaluation of their anti-infective potential in a murine model of Acinetobacter baumannii systemic infection. Following an in vivo toxicology study in CD1 mice, Ctn[15-34] re was deemed too toxic at the examined doses and excluded. For the remaining two topoisomers, Ctn[15-34] and Ctn re, a single intraperitoneal dose did not ensure mice survival. For Ctn[15-34] in particular, reasonably good results warranted further investigation despite some animals' deaths. Unfortunately, a three-day regimen of Ctn[15-34] intraperitoneal injections was lethal for all animals after the first day.
Based on these results, this thesis is a sobering reminder that topoisomeric modification of bioactive peptides, which has proven quite succesful in several other cases, cannot be taken for granted to provide viable antimicrobial leads and must be therefore pursued with caution until confirmed by in vivo evidence. Aside from the unexpectedly adverse outcomes, the thesis illustrates a cogent approach for evaluating antimicrobial peptide topoisomeric leads from bench to in vivo infection model stages.Aquesta tesi presenta un estudi sobre l'efecte de la modificació topoisomèrica en l'activitat de dos pèptids antimicrobians: la crotalicidina (Ctn), una catelicidina del verí d'una serp de cascavell sud-americana, i Ctn[15-34], un segment de Ctn amb propietats antimicrobianes i antitumorals interessants, així com una excel·lent estabilitat en sèrum.
El terme topoisomeria fa referència a variants d'una biomolècula que són d’idèntica composició (isomeria) però tridimensionalment diferents (topo-), on les modificacions estructurals poden produir efectes fisiològics potencialment útils. En el cas dels pèptids, el terme topoisòmer s’utilitza sovint per descriure versions amb tots els aminoàcids en configuració D, amb seqüències idèntiques (forma enantio, e) o totalment invertides (forma retroenantio, re) respecte de l'estructura primària nativa.
Per avaluar de manera exhaustiva les propietats i activitats dels topoisòmers Ctn i Ctn[15-34] en forma e i re, es van comparar amb el pèptid progenitor i amb la seva variant d'inversió de seqüència (forma retro, r), que també es considera un topoisòmer. Tot i que tots els topoisòmers mostraven activitat contra bacteris gramnegatius i cèl·lules tumorals, aquells amb aminoàcids en configuració D eren més citotòxics envers les cèl·lules normals que les versions amb aminoàcids en configuració L. Malgrat això, l'elevada estabilitat de les versions e i re en fluids humans va motivar una exploració més detallada.
Per tant, es van seleccionar els topoisòmers Ctn[15-34], Ctn re i Ctn[15-34] re per a una avaluació in vivo del seu potencial antiinfecciós en un model murí d'infecció sistèmica per Acinetobacter baumannii. Després d’un estudi de toxicologia in vivo en ratolins CD1, es va determinar que Ctn[15-34] re era massa tòxic en les dosis examinades i es va excloure. Per als altres dos topoisòmers restants, Ctn[15-34] i Ctn re, una única dosi intraperitoneal no va garantir la supervivència dels ratolins. En particular, per a Ctn[15-34], els resultats van ser raonablement bons però es van observar morts en alguns animals. Malauradament, una administració de tres vegades al dia d’injeccions intraperitoneals amb Ctn[15-34] no va resultar eficient per a tots els animals.
Basat en aquests resultats, aquesta tesi serveix com a recordatori clar que la modificació topoisomèrica de pèptids bioactius, que ha demostrat ser força reeixida en altres casos, no es pot assumir com a estratègia general per a obtenir compostos antimicrobians sense confirmació prèvia in vivo. Telomere length and emotional and behavioral problems in children from the prospective birth cohort Infancia y Medio Ambiente (INMA) study
Background/Objectives: This study aimed to examine the association between leukocyte telomere length (TL) measured at ages 4 and 8 and emotional and behavioral problems at age 8. We also explored whether changes in leukocyte TL between ages 4 and 8 were associated with outcomes. Methods: Data were obtained from a population-based birth cohort and included 647 children with TL at age 4 and emotional and behavioral assessments at age 8, 673 with TL and outcomes at age 8, and 315 with TL measured at both ages. TL was determined using quantitative PCR on blood samples and converted into z-scores for analysis. Emotional and behavioral problems—including internalizing, externalizing, and total difficulties—were assessed using the Strengths and Difficulties Questionnaire. Regression models were conducted using zero-inflated and negative binomial, adjusting for sociodemographic and lifestyle covariates. Results: No statistically significant associations were observed between leukocyte TL at ages 4 or 8, or TL changes over this period, and emotional and behavioral outcomes at age 8. Rapid and accurate demultiplexing of direct RNA nanopore sequencing data with SeqTagger
Nanopore direct RNA sequencing (DRS) enables direct measurement of RNA molecules, including their native RNA modifications, without prior conversion to cDNA. However, commercial methods for molecular barcoding of multiple DRS samples are lacking, and community-driven efforts, such as DeePlexiCon, are not compatible with newer RNA chemistry flowcells and the latest generation of graphics processing units (GPUs). To overcome these limitations, we introduce SeqTagger, a rapid and robust method that can demultiplex DRS data sets with 99% precision and 95% recall. We demonstrate the applicability of SeqTagger in both RNA002/R9.4 and RNA004/RNA chemistries and show its robust performance both for long and short RNA libraries, including custom libraries that do not contain standard poly(A) tails, such as Nano-tRNAseq libraries. Finally, we demonstrate that increasing the multiplexing up to 96 barcodes yields highly accurate demultiplexing models. SeqTagger can be executed in a standalone manner or through the MasterOfPores NextFlow workflow. Leveraging melodic context for improved Svara representation
For the South Indian musical tradition known as Carnatic music, embeddings of svara (note) pitch time series have proven useful for tasks such as svara classification and performance analysis. In this paper, we extend an existing embedding method by incorporating findings from musicological research on the relationship between the performance of a svara and its immediate melodic context, in order to improve the learning of these embedding models. We present a context-aware GRUbased model, adapting the existing DeepGRU architecture to encode both svara and its surrounding melodic context, before combining them via a co-attention mechanism prior to classification. For a ground truth dataset of 2,077 expert svara annotations across two performances in r¯aga Bhairavi, we observe that the inclusion of melodic context leads to a 6.6% absolute increase in F1 score for svara label classification (from 78.3% to 84.9%), and an 7.8% absolute increase (from 59.9% to 67.7%) for classification of svara-form: sub-svara clusters that capture gamaka (ornamentation) variations in the performed svara.This research was carried out as part of the "IA y Música: Cátedra en Inteligencia Artificial y Música" (TSI-100929-2023-1), funded by the Secretaría de Estado de Digitalización e Inteligencia Artificial, and the European Union-Next Generation EU, under the program Cátedras ENIA 2022 para la creación de cátedras universidad-empresa en IA
Dietary patterns and exposure to non-persistent endocrine-disrupting chemicals during pregnancy
Background: Exposure to non-persistent endocrine disrupting chemicals (EDCs) is widespread, causing various adverse health effects, especially during pregnancy. Diet is a major source of EDCs and the identification of dietary patterns that give rise to higher exposure levels is crucial. We aimed to explore the relationship between dietary patterns during pregnancy and exposure to non-persistent EDCs. Methods: We included 640 pregnant women from the Barcelona Life Study Cohort (BiSC), Spain. Using food frequency questionnaires (2nd trimester), we estimated ultraprocessed food (UPF) consumption and Mediterranean diet adherence, and derived data-driven dietary patterns. Metabolites of phthalates, 1,2-cyclohexane dicarboxylic acid di(isononyl) ester (DINCH), chlorpyrifos and pyrethroid, polycyclic aromatic hydrocarbons (PAHs, including phenanthrenes), and organophosphates, and parent compounds of phenols and parabens were measured in weekly urine pools (3rd trimester). Associations between dietary patterns and EDCs were evaluated through multivariable linear regressions. Results: Higher compliance with a "Vegetarian" pattern or the Mediterranean diet was associated with lower levels of some of the phthalate metabolites (e.g., di-iso-nonyl phthalates [Geometric mean ratio (GMR) per unit increases in the "Vegetarian" score = 0.97, 95 %CI 0.95, 1.00]). Conversely, "Western" and "Animal protein" patterns, and UPF intake were associated with higher PAHs (e.g., "Western" and 2-Hydroxynaphthalene [GMR(95 %CI) 1.07(1.01, 1.13)]), pesticides (e.g., "Animal protein" and 3-Phenoxybenzoic acid [1.15(1.01, 1.32]), and phthalates metabolites levels (e.g., UPF and Monoethyl phthalate [1.05(1.01, 1.09)]). No association remained significant after correcting for multiple testing. Conclusions: Following a plant-based diet may help reduce the exposure to certain phthalate and organophosphate metabolites, whereas higher consumption of foods from animal origin and high in fat/sugar/salt may increase exposure to PAHs, pesticides, and phthalates metabolites in pregnancy.This project has also received funding from the European Union’s Horizon 2020 research and innovation programme under grant agreement No 874583 (ATHLETE project).
