    PSYCHOLOGICAL CHARACTERISTICS OF MOTIVATION AND PROFESSIONAL INTEREST IN MEDICAL STUDENTS AT TASHKENT STATE MEDICAL UNIVERSITY

    Background: The formation of sustainable motivation for professional activity is a critical task in medical education, directly influencing the quality of future healthcare. In the context of the ongoing reforms in the healthcare system of the Republic of Uzbekistan, understanding the motivational structure of students at Tashkent State Medical University (TSMU) is of particular importance. Aim: To identify and analyze the psychological characteristics and structure of motivation for professional activity among future doctors studying at Tashkent State Medical University. Materials and Methods: A cross-sectional study was conducted involving 320 students from the 2nd to 5th years of the Faculty of General Medicine at TSMU. The diagnostic complex included: "The Motivation of Professional Activity" method (K. Zamfir in modification by A. Rean), "The Scale of Academic Motivation" (T.O. Gordeeva et al.), and a specially designed questionnaire to assess factors influencing the choice of profession. Statistical analysis was performed using SPSS 26.0. Results: The study revealed a polymotivational structure. The leading motives were cognitive (interest in medical science - 78%) and altruistic-humanistic (desire to help people - 82%). However, a significant proportion of students demonstrated pronounced pragmatic motives, such as the prestige of the profession (65%) and financial stability (58%). A correlation was found between the year of study and the dynamics of motivation: senior students showed an increase in "internal" motivation associated with clinical practice and a decrease in "external" positive motivation based solely on academic success. Key demotivating factors were identified as academic overload (45%), uncertainty about future employment conditions (40%), and emotional burnout in initial clinical contact. Conclusion: The motivation of TSMU students is characterized by a complex interplay of internal (cognitive, altruistic) and external (pragmatic) motives. The findings necessitate the development of a comprehensive psychological and pedagogical program at the university level aimed at strengthening internal motivation through early clinical immersion, mentorship programs, and psychological support to prevent burnout, thereby contributing to the training of highly qualified and dedicated medical specialists for the healthcare system of Uzbekistan.   KEYWORDS: medical education, motivation, professional development, psychology, Tashkent State Medical University, internal and external motives, burnout

    THE INTERRELATIONSHIP BETWEEN CHANGES IN IMMUNE AND BIOCHEMICAL INDICATORS IN AUTOIMMUNE LIVER DISEASE

    Significant hypergammaglobulinemia and the presence of many autoantibodies in the blood are symptoms of autoimmune hepatitis, a chronic liver illness with an unclear etiology that is characterized by periportal or more widespread inflammation. Current theories about the pathophysiology of AH include the interplay of genetic predisposition, immunological tolerance abnormalities, and environmental variables. The liver develops progressive non-inflammatory and fibrotic alterations as a result of this interaction, which triggers T-cell immune responses against hepatocyte antigens. With an annual incidence of 0.1–1.9 cases per 100,000 and a frequency of 3–17 instances per 100,000 in both Europe and the US, AH is regarded as a rather uncommon disease. Therefore, it is estimated that there are between 10,000 and 20,000 of these patients in the Russian Federation.     This article provides a detailed study of the participation of young people from the Republic of Uzbekistan in modern political processes. Special attention is given to analyzing the key trends and factors that determine the political activity of young citizens, as well as the forms of their involvement in the country's social and political life. The paper examines the level of political awareness and culture among young people, and the influence of state youth policy, educational institutions, and civil society organizations in shaping an active civic stance. Furthermore, the article highlights the existing problems, barriers, and prospects for further expanding youth participation in political decision-making. The author emphasizes the importance of a comprehensive approach to developing the political consciousness of the younger generation, which considers not only formal institutions but also informal channels of socialization such as family, media, social networks, and youth associations. It is noted that the formation of a stable political identity among youth is closely linked to their level of trust in the authorities, the degree of openness of the political system, and the availability of real mechanisms for participation. In the context of the modernization of Uzbek society, the need to improve educational programs aimed at developing legal literacy, civic responsibility, and critical thinking among young people becomes more urgent.     A new stability indicating UPLC-MS/MS methods for the simultaneous estimation of Abacavir, Dolutegravir and Lamivudine in pharmaceutical dosage forms

    Abacavir, Dolutegravir and Lamivudine are anti-viral drugs. A new stability indicating LC-ESI-MS method has been proposed and validated for the quantification of Abacavir, Dolutegravir and Lamivudine in pharmaceutical formulations as per ICH guidelines. WATERS, Quattro Premier XE Mass Spectrophotometer (MS/MS) with Software: Mass Lynx, Ver 4.1 and WATERS, Acquity UPLC with PDA detector was employed for the present study. A mixture of 0.2% Formic acid: Acetonitrile was used as mobile phase on gradient mode using Phenomenex Gemini C18, 50*4.6mm*5um with 0.5mL/min and run time of 3.5 min. A mixture of Acetonitrile: Water (80:20) was used as diluent. Linearity was observed over the concentration range 10.0-150ng/ml for Abacavir, Dolutegravir and Lamivudine by the proposed method and the methods are found to be simple, precise and accurate.     Structure-Based Virtual Screening and Molecular Dynamics Validation of Tryptoline as a Parasite-Selective PfPNP Inhibitor for Antimalarial Development

    Artemisinin resistance in Plasmodium falciparum has prompted the search for new antimalarial targets. Purine nucleoside phosphorylase (PNP) from P. falciparum offers real promise as a target because the parasite needs purine salvage to survive and lacks adenosine kinase. This study examined whether PfPNP could be selectively targeted through structural comparison and computational drug discovery protocol. BLAST searches with an e-value threshold of 0.001 found no sequence similarity between PfPNP and human PNP. When we compared their 3D structures, we found only 9% sequence identity with an RMSD of 3.08 Å and TM-score of 0.7, indicating they diverge substantially. A key difference emerged in their structure: PfPNP forms a six-unit complex while human PNP forms a three-unit complex, reflecting how differently they've evolved. Screening 326 compounds identified tryptoline as the best lead, showing superior binding efficiency (GLE: -0.638, GLE-SA: -1.500, GLE-IN: -2.326) compared to other candidates. Molecular docking revealed that tryptoline binds through multiple contact points, including aromatic interactions with Tyr160 and electrostatic interactions with Asp206. Molecular dynamics simulation over 100 ns confirmed that tryptoline stays firmly bound, with critical interactions persisting throughout the trajectory. These results support PfPNP as a selective and viable drug target. Cell-based studies and binding assays are needed to validate tryptoline's potential as a new antimalarial

    Method development and validation of a new RP-UFLC method for the simultaneous estimation of Sofosbuvir and Velpatasvir in tablet dosage form using an internal standard

    Sofosbuvir is an anti-viral drug used for the treatment of chronic hepatitis C virus infection and Velpatasvir is also an anti-viral drug used for the inhibition of hepatitis C virus and both the drugs acts directly. The combination of Velpatasvir and Sofosbuvir works by stopping the hepatitis C genotype 1, 2, 3, 4, 5, or 6 infection. A new RP-UFLC method has been proposed for the simultaneous estimation of Sofosbuvir and Velpatasvir in tablet dosage forms in presence of an internal standard. Shimadzu Model CBM-20A/20 Alite UFLC system with PDA detector and Zorbox Eclipse Plus C18 column was used for the chromatographic study. Mobile phase mixture consisting of 0.1 % Tri fluoro acetic acid: Acetonitrile (45: 55, v/v) with a flow rate 0.8 ml/min was chosen for the simultaneous determination of Sofosbuvir and Velpatasvir and the method was validated as per ICH guidelines.   KEYWORDS Sofosbuvir, Velpatasvir, RP-UFLC, validation, ICH guidelines

    The following case report is alopecia areata in 23 years old male who was treated successfully with individualised homoeopathic medicine within 6 months of treatment. Syphilinum was prescribed as a constitutional remedy on the basis of totality of symptoms and after the repertorisation. Homoeopathic medicines have wonderful results in such type of cases, where the modern medicines have very less effective treatment.
