    Listening for the Interior in Hip-Hop and R&B Music

    This article analyzes how four Black musical artists make “quiet,” or the inner life of African Americans, legible. Specifically, we consider ways that the quiet found within the lyrics of recent acclaimed albums from two hip-hop artists and two neo-soul artists—Kendrick Lamar’s DAMN (2017) and Rapsody’s Laila’s Wisdom (2017), Solange’s A Seat at the Table (2016) and Maxwell’s blackSUMMERS’night(2016), respectively—offer subtle, quotidian challenges to oppression, dehumanization, and objectification. We find that quiet occurs as artists describe the use of metaphysical space, or how place is used to make and take space for the self and to find peace, the protection of the interior self, and the gifts of quiet to the struggle for resistance. These lyrics speak to the interior safe space that Blacks seek as refuge from oppression by the dominant culture and demands from within their community. We contend that Blacks exercise power through their dominion over their interior selves, which in turn expresses their humanity. It is their control of the content of inner life, whatever those contents may be, that is an expression of sovereignty

    This book was developed as a clinical primer to provide the reader with concrete examples of how to apply multiple family systems theories and techniques to stepfamily concerns using a single case study approach. Given the ongoing escalation in the numbers of stepfamilies, and the probability that clinicians, regardless of the setting of counseling practice, will experience growing numbers of stepfamily concerns in their clinical caseloads, the availability of multiple approaches to support stepfamily success will be a welcome resource. This is a practitioner book. It assumes the reader has basic knowledge of theory-based treatment planning, systems thinking, and family theory.     Indirect effects in a planktonic disease system

    Indirect effects, both density- and trait-mediated, have been known to act in tandem with direct effects in the interactions of numerous species. They have been shown to affect populations embedded in competitive and mutualistic networks alike. In this work, we introduce a four-dimensional system of ordinary differential equations and investigate the interplay between direct density-effects and density- and trait-mediated indirect effects that take place in a yeast parasite–zooplankton host–incompetent competitor system embedded in a food web which also includes resources and predators. Among our main findings is the demonstration that indirect effects cause qualitative and quantitative changes almost indistinguishable from direct effects and the corroboration through our analysis of the fact that the effects of direct and indirect mechanisms cannot be disentangled. Our results underpin the conclusions of past studies calling for comprehensive models that incorporate both direct and indirect effects to better describe field data

    A mutation affecting polycystin-1 mediated heterotrimeric G-protein signaling causes PKD

    Autosomal dominant polycystic kidney disease (ADPKD) is characterized by the growth of renal cysts that ultimately destroy kidney function. Mutations in the PKD1 and PKD2 genes cause ADPKD. Their protein products, polycystin-1 (PC1) and polycystin-2 (PC2) have been proposed to forma calcium-permeable receptor-channel complex; however the mechanisms by which they function are almost completely unknown. Most mutations in PKD1 are truncating loss-of-function mutations or affect protein biogenesis, trafficking or stability and reveal very little about the intrinsic biochemical properties or cellular functions of PC1. An ADPKD patient mutation (L4132D or DL), resulting in a single amino acid deletion in a putative G-protein binding region of the PC1 C-terminal cytosolic tail, was found to significantly decrease PC1-stimulated, G-protein-dependent signaling in transient transfection assays. Pkd1DL/DL mice were embryo-lethal suggesting that DL is a functionally null mutation. Kidney-specific Pkd1DL/cond mice were born but developed severe, postnatal cystic disease. PC1DL protein expression levels and maturation were comparable to those of wild type PC1, and PC1DL protein showed cell surface localization. Expression of PC1DL and PC2 complexes in transfected CHO cells failed to support PC2 channel activity, suggesting that the role of PC1 is to activate G-protein signaling to regulate the PC1/PC2 calcium channel

    Regulated Capture of Vκ Gene Topologically Associating Domains by Transcription Factories

    Expression of vast repertoires of antigen receptors by lymphocytes, with each cell expressing a single receptor, requires stochastic activation of individual variable (V) genes for transcription and recombination. How this occurs remains unknown. Using single-cell RNA sequencing (scRNA-seq) and allelic variation, we show that individual pre-B cells monoallelically transcribe divergent arrays of Vκ genes, thereby opening stochastic repertoires for subsequent Vκ-Jκ recombination. Transcription occurs upon translocation of Vκ genes to RNA polymerase II arrayed on the nuclear matrix in transcription factories. Transcription is anchored by CTCF-bound sites or E2A-loaded Vκ promotors and continues over large genomic distances delimited only by topological associating domains (TADs). Prior to their monoallelic activation, Vκ loci are transcriptionally repressed by cyclin D3, which prevents capture of Vκ gene containing TADs by transcription factories. Cyclin D3 also represses protocadherin, olfactory, and other monoallelically expressed genes, suggesting a widely deployed mechanism for coupling monoallelic gene activation with cell cycle exit
