    Study of classical molecular dynamics of viral proteins

    The new coronavirus strain, SARS-CoV-2, highlighted the lack of tools to address new challenges in structure analysis. The understanding of the sequence-structure-function paradigm proved to be insufficient when developing new drugs based on molecules with similar structures and sequences. Therefore, extensive studies of molecular interactions and molecular functions are needed to understand the interaction processes of viral proteins when invading an infected cell. Proteins, when involved in physiological processes, maintain movements to allow them to interact with their environment, thus establishing a relationship between structure and function. Normal mode analysis has emerged as a powerful tool to elucidate the dynamics of structures and how it affects their basic functions. However, several studies have exposed different conformations of SARS-CoV-2 structures, which can obscure the study of significant structures. The application of principal component analysis allows to classify and reduce the dimensions of analyzed structures to obtain those that maintain a better fit. In this sense, this thesis project proposes an exploratory study of principal component analysis to characterize and classify the structures with better coupling to physical models to later analyze the influence of the structure on its internal dynamics.La nueva cepa de coronavirus, SARS-CoV-2, puso de manifiesto la falta de herramientas para afrontar los nuevos retos del análisis de estructuras. La comprensión del paradigma secuencia-estructura-función resultó insuficiente a la hora de desarrollar nuevos fármacos basados en moléculas con estructuras y secuencias similares. Por lo tanto, se necesitan estudios exhaustivos de las interacciones y funciones moleculares para comprender los procesos de interacción de las proteínas virales cuando invaden una célula infectada. Las proteínas, cuando participan en procesos fisiológicos, mantienen movimientos que les permiten interactuar con su entorno, estableciendo así una relación entre estructura y función. El análisis del modo normal ha surgido como una poderosa herramienta para dilucidar la dinámica de las estructuras y cómo afecta a sus funciones básicas. Sin embargo, varios estudios han puesto de manifiesto diferentes conformaciones de las estructuras del SARS-CoV-2, lo que puede dificultar el estudio de estructuras significativas. La aplicación del análisis de componentes principales permite clasificar y reducir las dimensiones de las estructuras analizadas para obtener aquellas que mantienen un mejor ajuste.     Effects of imidacloprid and acetamiprid during the in vitro neuronal differentiation of SH-SY5Y neuroblastoma cells

    Neonicotinoids, acetamiprid (ACE) and imidacloprid (IMI), are widely used insecticides in agriculture, and their application has exponentially increased in the recent years. In insects, these compounds act on the central nervous system causing death after a few hours of exposure. Their mode of action is based on the affinity and agonist activity for nicotinic acetylcholine receptors (nAChR). In humans, neonicotinoids were initially thought to have low toxicity due to their poor affinity for the nAChRs of vertebrate organisms. However, recent evidence has revealed neurotoxicity in these organisms. One of the key stages for studying their toxicity is neurodevelopment due to the greater vulnerability to exogenous compounds. The SH-SY5Y cell line was used in this work as an in vitro model for the study of neurodevelopmental toxicity due to its basal expression of functional nAChRs, as well as its differentiation into neuronal-type cells induced by retinoic acid or staurosporine. The SH-SY5Y cell line was exposed to neonicotinoids at three different stages: non-Differentiated cells, On-the-Go-Differentiating cells, and Differentiated cells. En insectos, estos compuestos actúan sobre el sistema nervioso central provocando la muerte después de pocas horas de exposición. Su modo de acción se basa en la afinidad y actividad agonista por los receptores nicotínicos de acetilcolina (nAChRs, por sus siglas en inglés). En humanos, los neonicotinoides inicialmente fueron denominados como insecticidas de baja toxicidad debido a su poca afinidad por los nAChRs de organismos vertebrados. Sin embargo, evidencia reciente ha revelado neurotoxicidad en estos organismos. Una de las etapas claves para estudiar su toxicidad es el neurodesarrollo debido a la mayor vulnerabilidad a compuestos exógenos. La línea celular SH-SY5Y se utilizó en este trabajo como modelo in vitro para el estudio de la toxicidad durante el neurodesarrollo debido a su expresión basal de nAChRs funcionales, así como por su diferenciación en células de tipo neuronal inducida por ácido retinoico o estaurosporina.     Increasing of photochemical activity of bioentities by the combination of Algae and Grass-chloroplasts when Encapsulated into Hierarchical silica- Monoliths

    The photosynthetic activity of plants and microalgae can be widely exploited to produce biomaterials with varied properties such as the absorption of CO2 and the removal of metals such as Cu and Ni in wastewater. In this study, microalgae species Chlorella Vulgaris and chloroplasts extracted from Pennisetum Clandestinum turfgrass residues were immobilized into silica monoliths with hierarchical porosity by the high internal phase emulsion (HIPE) method. Additionally, in this research, a new monolith model in which microalgae and chloroplasts are encapsulated in the same silica matrix is presented. In order to verify its effectiveness, two other monolith models were synthesized under the same conditions using chloroplasts and microalgae separately. Optical and fluorescence microscopy as well as UV-Vis, diffuse reflectance and fluorescence spectroscopy were used to monitor the preservation of photosynthetic activity in the monoliths. The capacity of the monoliths to produce O2, was verified by cyclic voltammetry. Moreover, the bio remotion activity of monoliths for copper and nickel was evaluated performing a kinetic test. The results demonstrate that the three types of monoliths synthesized are capable of maintaining their photosynthetic activity and producing O2 even after 90 days of encapsulation. The proposed new monolith model presented higher activity compared to the others after 60 days. Kinetic tests showed that separately encapsulated microalgae are more effective in removing Cu and Ni.La actividad fotosintética de las plantas y microalgas puede ser ampliamente aprovechada para producir biomateriales con propiedades variadas tales como como la absorción de CO2 y la remoción de metales tales como Cu y Ni en aguas residuales. En este estudio, la especie de microalga Chlorella Vulgaris y cloroplastos extraídos residuos de césped Pennisetum Clandestinum fueron inmovilizados en monolitos de sílice con porosidad jerárquica mediante el método de emulsión de fase interna alta (HIPE). Adicionalmente, en esta investigación presentamos un nuevo modelo de monolito en el cual se encapsulan microalgas y cloroplastos en una misma matriz de sílice. Para poder comprobar su efectividad, se sintetizaron bajo las mismas condiciones otros dos modelos de monolitos utilizando cloroplastos y microalgas por separado. Para monitorear la preservación de la actividad fotosintética en los monolitos se utilizaron microscopia óptica y de fluorescencia, así como espectroscopias de UV-Vis, reflectancia difusa y fluorescencia. Para comprobar la capacidad de producir O2 de los monolitos se utilizó voltamperometría cíclica y también se realizaron pruebas cinéticas para comprobar la bio extracción de cobre y níquel. Los resultados demostraron que los tres tipos de monolitos sintetizados son capaces de mantener su actividad fotosintética y producir O2 hasta después de 90 días de encapsulación. El modelo de monolito propuesto presento mayor actividad en comparación a los otros después de 60 días.     Drug delivery system: designing hydrogels as machinery for anti-inflammatory therapeutics from plant diversity of Ecuador

    Nowadays, several medicinal plants have been a natural source of pharmacological compounds. The present study extracted plant extracts (L001-M, L002-E, L003-M, and L004- E) by maceration in methanol and ethanol as solvents. The bioactive compounds were characterized by phytochemical screening to identify their secondary metabolites content. The functional groups corresponding to alkaloids, saponins, terpenes, flavonoids, phenols, and tannins were identified by FT-IR. In addition, thin layer chromatography techniques and UV/Vis spectra were used to analyze the components in the extracts. This research also evaluated the biological activity of the extracts in antimicrobial, antioxidant, and anti-inflammatory assays to analyze their potential pharmaceutical application. The minimum bactericidal concentration of samples L001-M and L002-E on Gram-negative bacteria Escherichia Coli was 2.5 mg/mL. The in vitro antioxidant activity was evaluated by the Ferric Reducing Power and Total Antioxidant Activity assay showing that the methanolic extracts (L001-M and C003-M) were better antioxidant compounds compared to the ethanolic extracts (L002-E and C004-E). In addition, inhibition of hypotonicity-induced hemolysis was positive for L001-M, L002-E, and C004-E. Cotton cellulose (NRC) and microcrystalline cellulose (MCC) hydrogels formulated with the four extracts were studied kinetically, structurally, and morphologically. The incorporation of the extracts into the hydrogels was verified by FT-IR assay and their analysis by stereoscope. Kinetic studies revealed that the MCC type showed better swelling values (45% to 140%) than the NRC hydrogels (49% to 95%). The release profiles of the extracts were also studied in PBS at pH 7.4, where the NRC type (50%) showed better results than the MCC hydrogel (30%). The effective biological activity of the hydrogels on the DH5-α cell line (E. coli) was recorded. The overall results suggest the effective encapsulation of the natural extracts in hydrogels as controlled release systems suggesting their possible biomedical application.Recientemente, diversas plantas medicinales han sido una fuente natural de compuestos farmacológicos. En el presente estudio, extractos vegetales (L001-M, L002-E, L003-M and L004-E) se obtuvieron mediante extracción por maceración en metanol y etanol como solvente. Los compuestos bioactivos se caracterizaron mediante screening fitoquímico para identificar sus metabolitos secundarios. Se identifico a través de FT-IR los grupos funcionales correspondientes a alcaloides, saponinas, terpenos, flavonoides, fenoles y taninos. Además, se utilizó técnicas de cromatografía de capa fina y el espectro UV/Vis espectro para referir la diversidad de componentes en los extractos. Esta investigación también evaluó la actividad biológica de los extractos en ensayos antimicrobianos, antioxidantes y anti-inflamatorios con el fin analizar su potencial aplicación farmacéutica. La concentración mínima bactericida de las muestras L001-M y L002-E en bacteria Gram negativa Escherichia Coli fue a 2.5 mg/mL. La actividad antioxidante in vitro se evaluó mediante el ensayo del Poder Reductor Férrico y la Actividad Antioxidante Total demostrando que los extractos metanolicos (L001-M y C003-M) fueron mejores compuestos antioxidantes comparado con los extractos etanolicos (L002-E y C004-E). Además, la inhibición de la hemolisis inducida por hipotonicidad resultó positiva para L001-M, L002-E y C004-E. Los hidrogeles de celulosa de algodón (NRC) y celulosa microcristalina (MCC) formulados con los cuatro extractos se estudiaron cinética, estructural y morfológicamente. La incorporación de los extractos en los hidrogeles se verificó mediante el ensayo FT-IR, y su análisis mediante estereoscopía. Los estudios cinéticos revelaron que el tipo MCC mostró mejores valores de hinchamiento (45% a 140%) que los hidrogeles NRC (49% a 95%). También se estudiaron los perfiles de liberación de los extractos en PBS con pH 7.4 donde el tipo NRC (50%) mostró mejores resultados que el hidrogel MCC (30%). La actividad biológica eficaz de los hidrogeles en la línea celular DH5-α (E. Coli) fue registrada.     Cellulose-based hydrogels towards an antibacterial wound dressing

    Wound healing is one of the most recently studied topics in biomedicine, mainly due to the risk of skin infections and scar formation. Different wound dressings are available on the market, but most of them are dry, which does not help avoid scars formation, and others are not antibacterial. Therefore, there is a need for new and better biomaterials that can help prevent infections and repair damaged skin tissue. Cellulose is the most abundant natural polymer, and the production of cellulose-based hydrogels in the biomedical field is increasing due to its convenient properties and biocompatibility. In addition, further research reported that cellulose properties depend on the extraction source, such as leaves or fruit shells. Some show significant antibacterial and healing properties essential for tissue repair. Then, in this research, we developed cellulose-based hydrogels for the treatment of wounds. For this purpose, different sources of cellulose from Ecuadorian plants were selected. Characterization techniques such as X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FT-IR) was carried out to know their structure and composition. For the antibacterial tests, in-vitro and ex-vivo experiments were performed to evaluate the efficacy of the synthesized hydrogels against two Gram-negative bacteria strains. Hydrogels from cellulose samples F53 and F12 showed the best bacteria inhibition activity, and F53 presents the bigger liquid absorption capacity. Finally, prototypes of wound dressings were developed with the hydrogels that exhibited the best antibacterial properties.La cicatrización de heridas es uno de los temas más recientemente estudiados en biomedicina, especialmente debido al riesgo de infecciones cutáneas y formación de cicatrices. Hay diferentes apósitos para heridas disponibles en el mercado, pero la mayoría son secos, lo cual no ayuda a evitar la formación de cicatrices, y otros no son antibacterianos. Por lo tanto, existe la necesidad de desarrollar biomateriales nuevos y mejores que puedan ayudar a evitar infecciones, así como mejorar la reparación del tejido cutáneo dañado. Por otro lado, la celulosa es el polímero natural más abundante y la producción de hidrogeles a base de celulosa en el campo biomédico está aumentando debido a sus ventajosas propiedades y biocompatibilidad. Además, se ha reportado que las propiedades de la celulosa dependen de la fuente de su extracción, como pueden ser, hojas, troncos o cáscaras de frutos. Algunos de ellos muestran importantes propiedades antibacterianas y cicatrizantes, que son muy importantes para la reparación de tejidos. Por ello, el en esta investigación se desarrollaron hidrogeles a base de celulosa para el tratamiento de heridas. Para ello, se procesaron diferentes fuentes de celulosa para formar hidrogeles. Para conocer su estructura y composición se realizaron diferentes técnicas de caracterización como la difracción de rayos X (XRD) y la espectroscopia infrarroja por transformada de Fourier (FT-IR) así como la biodegradación. Para las pruebas antibacterianas se realizaron experimentos in vitro y ex vivo para evaluar la eficacia de los hidrogeles sintetizados frente a dos cepas de bacterias Gram negativas. Finalmente, se desarrollaron prototipos de apósitos para heridas con los hidrogeles que presentaron las mejores propiedades antibacteriales.Ingeniero/a Biomédico/

    Preliminary study of Rubrene-Pluronic Soft Assemblies using Small-Angle Neutron Scattering

    Nanomaterials have become the norm nowadays, and industries are showing interest in these materials. Particularly, the search for precise mechanisms for their application in obtaining bio-images for the identification and localization of tumors has increased over the last decades, as well as in the evaluation of the efficacy of cancer treatments. Currently, the use of organic nanoparticles (ONPs) for bioimaging is being explored by researchers, taking advantage of their convenient spectral properties. To provide greater stability to these ONPs, and allow their transport within the human body without losing their properties, the incorporation of polymeric materials, such as Pluronic F127, has been suggested with which their biocompatibility is highly enhanced. Rubrene, an inorganic semiconductor material derived from a tetracene, has demonstrated interesting properties that could be exploited for bioimaging. This work presents a fundamental structural and spectroscopic study of Rubrene-Pluronic soft assemblies to evaluate their potential for applications in bioimaging. Understanding in detail how these hybrid materials form and what their structure is, with particular focus on temperature dependency, will open new paths towards tailoring and customization of such materials. For this work, Rubrene-Pluronic soft assemblies were synthesized using the well-known reprecipitation method. The structure of the materials was investigated using Dynamic Light Scattering, and Small-Angle Neutron Scattering. The results showed that micellation occurs well below the Critical Micellation Temperature of Pluronic F127 due to the presence of Rubrene and the formed structures are larger than regular micelles from Pluronic F127 alone.Los nanomateriales se han convertido en la norma hoy en día, y las industrias están mostrando interés en estos materiales. En particular, la búsqueda de mecanismos precisos para su aplicación en la obtención de bioimágenes para la identificación y localización de tumores se ha incrementado en las últimas décadas, así como en la evaluación de la eficacia de tratamientos contra el cáncer. Actualmente, los investigadores están explorando el uso de nanopartículas orgánicas (NPOs) para la obtención de imágenes biológicas, aprovechando sus convenientes propiedades espectrales. Para dotar de mayor estabilidad a estas NPOs, y permitir su transporte dentro del cuerpo humano sin perder sus propiedades, se ha sugerido la incorporación de materiales poliméricos, como Pluronic F127, con el que se potencia mucho su biocompatibilidad. Este trabajo presenta un estudio estructural y espectroscópico de ensamblaje de Rubrene-Pluronic para evaluar su potencial para aplicaciones en bioimagen. Comprender en detalle cómo se forman estos materiales y cuál es su estructura, con un enfoque en la dependencia de la temperatura, abrirá nuevos caminos hacia la adaptación y personalización de dichos materiales. Para este trabajo se sintetizaron ensamblajes de Rubrene-Pluronic utilizando el método de reprecipitación. La estructura de los materiales se investigó mediante la dispersión de luz dinámica y la dispersión de neutrones de ángulo pequeño. Los resultados mostraron que la micelación ocurre muy por debajo de la temperatura crítica de micelación de Pluronic F127 debido a la presencia de Rubrene y que las estructuras formadas son más grandes que las micelas regulares de Pluronic F127.Químico/

    Theoretical study of interaction of COVID-19 palliative chlorine dioxide with hemoglobin analogs

    The use of chlorine dioxide (ClO2) has taken an important role as a palliative to combat viral diseases such as COVID-19. Even though organizations and regulatory entities have not endorsed its use and have warned of its possible harmful effects on health, its consumption continues to be popular among the population. ClO2 in aqueous solution gradually produces by-products such as chlorites and can act as a powerful oxidizing agent of different biomolecules that play important biological roles in humans, such is the case of hemoproteins whose active site, the heme group, contains an iron atom in which the control of its chemical environment and its electronic structure are very important for their biological functions. For this reason, this work aims to study the interactions of ClO2 and its main derivative, the chlorite ion ClO2− with the heme-b group, as well as to compare the changes in structural and electronic properties that occur because of these interactions. To carry out this study, computational tools have been used to predict the behavior of the complexes of interest. Through the DFT density functional theory and the B3LYP functional, the geometry of the complexes was optimized, as well as the structural and electronic properties. On the other hand, an experimental study of the interaction of such chlorinated derivatives with the bifunctional enzyme catalase peroxidase from Neurospora crassa, CAT2, was carried out. This enzyme was used as a structural model because it contains a heme-b prosthetic group in its active site. The computational results obtained show that the ClO2 and ClO2− ligands coordinate to iron through the oxygen atom and with less energy than the oxygenated form. Regarding its geometry, it is evident that in the presence of the ligands H2O, O2, ClO2 and ClO2− there is a distortion of the octahedral geometry that is in accordance with that reported in the literature; furthermore, the Mulliken population analysis shows a significant change in iron charge. Finally, from the experimental analysis of the interaction of the CAT-2 enzyme with a commercial solution of chlorine dioxide, significant spectral changes were documented, demonstrating the degradation of the heme group caused by such interaction. Both types of results support the hypothesis of this work, in the sense that the use of chlorine dioxide as a palliative agent for the treatment of COVID-19 can cause important alterations in the functioning of biomolecules relevant to the human body such as hemoglobin.El uso de dióxido de cloro (ClO2) ha tomado protagonismo como un paliativo para combatir enfermedades virales como el COVID-19. A pesar de que las organizaciones y entes regulatorios no han avalado su uso y han advertido de sus posibles efectos dañinos en la salud, su consumo sigue siendo popular entre la población. El ClO2, en solución acuosa produce subproductos como cloritos y puede actuar como un potente agente oxidante hacia diferentes biomoléculas que juegan roles biológicos importantes en el ser humano, tal es el caso de las hemoproteínas cuyo sitio activo, el grupo hemo, alberga un átomo de hierro en el que el control de su ambiente químico y su estructura electrónica son muy importantes para que sus funciones biológicas no sean alteradas. Por ello, este trabajo tiene como objetivo estudiar las interacciones del ClO2 y su el ion clorito ClO2− con el grupo hemo-b, así como comparar los cambios en las propiedades estructurales y electrónicas que se producen por efecto de dichas interacciones. Para llevar a cabo este estudio se ha utilizado herramientas computacionales que permiten predecir, el comportamiento de los complejos de interés. A través de la teoría funcional de densidad DFT y el funcional B3LYP se realizó la optimización de geometría de los complejos, así como también se estudiaron las propiedades estructurales y electrónicas. Por otro lado, se realizó un estudio experimental de la interacción de dichos derivados clorados con la enzima bifuncional catalasa peroxidasa de Neurospora crassa, CAT-2. Dicha enzima se utilizó como modelo estructural debido a que contiene un grupo prostético hemo-b en su sitio activo. Los resultados computacionales obtenidos demuestran que los ligantes ClO2 y ClO2− se coordinan al hierro mediante el átomo de oxígeno y con una energía menor a la forma oxigenada. Además, los ligantes H2O, O2, ClO2 y ClO2- producen una distorsión de la geometría octaédrica que está en concordancia con lo reportado en la literatura. El análisis poblacional de Mulliken muestra un cambio significativo en la carga del hierro. Finalmente, del análisis experimental de interacción de la enzima CAT-2 con una disolución comercial de dióxido de cloro, se documentaron cambios espectrales significativos que revelan la degradación del grupo hemo. Ambos tipos de resultados apoyan la hipótesis de este trabajo, en el sentido de que el uso de dióxido de cloro como agente paliativo para el tratamiento de COVID-19 puede causar alteraciones importantes en el funcionamiento de biomoléculas relevantes para el cuerpo humano como la hemoglobina.Químico/

    Magnetic Separation and Enrichment of Fe–Ti Oxides from Iron Titaniferous Beach Sands: Process Design Applied to Coastal Ecuador

    Iron titaniferous sands, also called black sands, are a source of various magnetic minerals, such as iron and titanium (Fe–Ti) oxides, with countless scientific and industrial applications. Ecuador is deemed a geo-diverse country that contains deposits of black sands in the Andean and coastal regions; therefore, the industrialization of these magnetic sands might be of high interest. This study presents a preliminary industrial design for the magnetic separation process of Fe–Ti oxides from iron titaniferous Ecuadorian beach sands. Four stages are considered for the process, involving collecting, drying, screening, and magnetic separation. This proposal returns the large particles (>150 µm) and the non-agnetic fraction to the original place, generating a minimum environmental impact with the support of natural marine and coastal processes. The process design criteria are based on engineering guidelines, sampling, and characterization of eleven black sand samples. Using conventional techniques, the water content, granulometric distribution, particle size, and semiquantitative Fe–Ti oxide concentration were determined for the different sand samples. It is estimated that Fe–Ti oxide production may reach 5.835 metric tons per day (5.835 mtpd) with a magnetic content of 97.50%, starting from 100 mtpd of black sands. Based on an economic analysis (Class V), a net profit of USD 835,875.63 is expected during the first year of production. Thus, the magnetic separation and enrichment of Fe–Ti oxides from iron titaniferous coastal sands exploitation should allow the commercial valorization of these resources in an eco-friendly way, i.e., with economic benefits and minimization of environmental impact in the source area

    Theoretical Approach on the Comparison between the Oxidative Dehydrogenation of Amines and MOF Mediated Amine Oxidation

    Oxidative dehydrogenation play an important role in the production of raw materials for chemical, petrochemical, polymers, pharmacological and agrochemical industry. Moreover, studies in different investigations have been performed in relation to the production of formaldehyde, nitrogen heterocycles, imines, and biochemical systems such as alcohol dehydrogenase, galactose oxidase, and amine oxidase. The oxidative dehydrogenation of amines to produce imines have attracted a great interest within the scientific community because imines can react as electrophiles reductions, condensations, additions and cycloadditions reactions. One of the most important characteristic of oxidative dehydrogenation is its high selectivity to form imines from amines and for that reason several solvents, environmental conditions, metal organic frameworks and metal complexes in conjunction with different transition metals have been used as catalysts for the purposes of increasing the reaction yields. Copper (III) in Cu-BTC and iron (III) in tetracyano(1,2-diamine)ferrate(III) have been used as metallic center in oxidative dehydrogenation of amines because these metals are abundant, cheaper and easily access. In this work, a comparison study between the reaction mechanisms to the imine formation through oxidative dehydrogenation promoted by the coordination of the amine ligand to iron (III) and the catalyzed oxidation of amines by a Cu(II)-MOF was carried out by using geometry optimizations, frequencies and Mülliken charges based on DFT computational calculations. The presence of Fe(III) and Cu(III) as transition metals in tetracyano (1,2-diamine) ferrate(III) and Cu-BTC respectively are the responsible to promote the oxidation of the coordinate amine which influence the imine formation. The results obtained from the DFT simulations for each step of the reaction mechanism of tetracyano (1,2-diamine) ferrate(III) and Cu-BTC reveal that high energies are required to form the imine for each step of the reaction mechanism but Cu- BTC mechanism for the formation of imines was more thermodynamically favored.La deshidrogenación oxidativa juega un papel importante en la producción de materias primas para la industria química, petroquímica, polímeros, farmacológica y agroquímica. Además, se han realizado estudios en diferentes investigaciones relacionadas con la producción de formaldehído, heterociclos de nitrógeno, iminas y en sistemas bioquímicos como la alcohol deshidrogenasa, la galactosa oxidasa y la amina oxidasa. La deshidrogenación oxidativa de aminas para producir iminas ha generad un gran interés en la comunidad científica ya que las iminas pueden reaccionar como reducciones, condensaciones, adiciones y reacciones de cicloadición electrófilas. Una de las características más importantes de la deshidrogenación oxidativa es su alta selectividad para formar iminas a partir de aminas y por ello se han utilizado diferentes catalizadores, disolventes, condiciones ambientales, armazones metal-orgánicos (MOF) y complejos metálicos en combinación con diferentes metales de transición con el fin de aumentar los rendimientos de la reacción. El cobre (III) en Cu-BTC y el hierro (III) en tetraciano(1,2-diamina)ferrato(III) se han utilizado como centro metálico en la deshidrogenación oxidativa de aminas porque estos metales son abundantes, más baratos y de fácil acceso. En este trabajo se ha realizado un estudio comparativo entre los mecanismos de reacción para la formación de iminas a través la deshidrogenación oxidativa promovida por la coordinación del ligando amina al hierro (III) y la oxidación de aminas catalizada por un MOF de cobre (II) mediante la optimización de las geometrías, frecuencias y las cargas de Mülliken basadas en cálculos computacionales DFT. La presencia de Fe(III) y Cu(II) como metales de transición en el tetraciano (1,2-diamina) ferrato(III) y Cu-BTC respectivamente son los responsables de promover la oxidación de la amina coordinada que influye en la formación de iminas. Los resultados obtenidos de las simulaciones DFT para cada paso del mecanismo de reacción del tetraciano (1,2-diamina) ferrato(III) y el Cu-BTC revelan que se requieren altas energías para formar la imina para cada paso del mecanismo de reacción, pero el mecanismo del Cu-BTC para la formación de iminas fue más favorecido termodinámicamente.Químico/

    An alternative to the global climate model (GCM) is the regional climate model (RCM). The experiments obtained from regional climate change models have recently become popular and are widely used in the Statistical literature to analyze and understand the evolution of natural phenomena such as climate change in a limited area, using discretized versions of physical processes that they are modeled by systems of differential equations. This methodology is appropriate for the analysis of time series of climatic variables; therefore, the present work is motivated by the need to compare sources of variability in the projections made by the computational models used to model the climate. We propose to use a functional data modeling through an ANOVA from a Bayesian statistical point of view, assigning prior distributions of Gaussian processes in each batch of the functional effects. Some alternatives to specify the model will be discussed, and computationally efficient strategies will be considered for the simulation of the samples and estimation of the posterior distribution using a Markov Chain Monte Carlo algorithm (MCMC) for the first case and a Laplace approximation (INLA) for the second case. The methodology will be illustrated through two cases: in the first case, the effect of the geographical region on the temperature profiles in the meteorological stations of the Republic of Ecuador is considered; In the second case, the sources of variability in the output of regional climate models will be examined considering a designed experiment.Una alternativa al modelo climático global (GCM) es el modelo climático regional (RCM). Los experimentos obtenidos a partir de modelos de cambio climático regionales se han popularizado recientemente y son ampliamente utilizados en la literatura Estadística para analizar y comprender la evolución de fenómenos naturales tales como es el cambio climático en un área limitada, utilizando versiones discretizadas de procesos físicos que son modelados por sistemas de ecuaciones diferenciales. Esta metodología es apropiada para el análisis de series temporales de variables climáticas; por lo tanto, el presente trabajo está motivado por la necesidad de comparar fuentes de variabilidad en las proyecciones realizadas por los modelos computacionales utilizados para modelar el clima. Proponemos utilizar un modelado de datos funcionales a través de un ANOVA desde un punto de vista de la estadística Bayesiana, asignando distribuciones a priori de procesos Gaussianos en cada batch de los efectos funcionales. Se discutirán algunas alternativas para especificar el modelo, y se considerarán estrategias computacionalmente eficientes para la simulación de las muestras y estimación de la distribución posterior utilizando un algoritmo Markov Chain Monte Carlo (MCMC) para el primer caso y una aproximación de Laplace (INLA) para el segundo caso. La metodología se ilustrará mediante dos casos: en el primer caso se considera el efecto de la región geográfica en los perfiles de temperatura en las estaciones meteorológicas de la República del Ecuador; en el segundo caso se examinarán las fuentes de variabilidad en el resultado de los modelos climáticos regionales considerando un experimento diseñado.Matemático/
