    Harnessing CRISPR-Cas for Targeted Epigenetic Manipulations: a physiological study of Gene Regulation

    Genome engineering, epigenome editing, as one of the prominent research techniques, has also become instrumental in discovering the gene expression regulation mechanisms and exploring their physiological correlates in health and pathology. This review summarizes the state-of-the-art CRISPR-Cas strategies that focus on epigenetics modification and projects its utility in molecular physiology of gene regulation. In this course we will delve into how CRISPR-Cas systems interact with epigenetic signals through DNA methylation, histone modifications and accessibility to chromatin. Then we look at the physiological effects of epigenetic modulations in cellular differentiation and development, cellular signal and homeostasis, disease pathogenesis, and therapeutic for application. Issues and perspectives on how epigenome manipulations can revolutionize therapeutics are also emphasized exploring the potential of precision medicine and individualized therapies. Overall, the review stresses the pivotal role of epigenetic modulation in the development of our understanding of gene expressions/mediation and cellular physiology. It also provides some inputs on the potential directions that the field of genetic engineering might take in the future

    EVALUATE THE EFFECT OF ORLISTAT, LIRAGLUTIDE, AND THE HERBAL SUPPLEMENT ON THE MOTHERS OF EXPERIMENTALLY FATTENED WHITE RATS

    The current study aimed to evaluate the effect of orlistat, liraglutide, and the herbal supplement on the mothers of experimentally fattened white rats. The female rats were divided into two main groups, the first fed a standard ration, in which two groups were the first pregnant and the second non-pregnant, while the second main group was fed a special diet containing 35%. Fat for the purpose of fattening as a first stage and for a period of 6 weeks, then the second stage began, which is the pregnancy and treatment stage and included eight secondary groups. The first group was not treated with any drug, the second group was orlistat at a therapeutic dose, the third group was liraglutide at a therapeutic dose, the fourth group was an herbal supplement, and the fifth group was a supplement with a therapeutic dose. Of Orlistat, the sixth group is supplemented with a therapeutic dose of liraglutide, the seventh group is a high dose of orlistat, and the eighth group is a high dose of liraglutide. The current study showed that the obesity leads to changes in lipid profiles compared to control groups, as the effect of fattening is represented by an increase in the concentration of total cholesterol, triglycerides, low-density lipoproteins, very low-density lipoproteins, and a decrease in high-density lipoproteins, histological examinations. Pathological changes appeared in each of the liver tissues, represented by a large proliferation of fat droplets, steatosis, Müller bodies, collapse of the central vein wall, and lymphocytic infiltration. The two best groups were when the herbal supplement was used with both drugs, and the herbal supplement had a synergistic role. With the two drugs, it reduces the accumulation of fat in the cells

    EVALUATE THE EFFECT OF SALVIA OFFICINALIS AND CARDAMOM ON ANIMAL PRODUCTIVITY OF BROILER CHICKENS

    It is in considerable demand to produce poultry meat and eggs of superior quality at affordable costs without using growth promoters or antibiotics, so the current study aimed to evaluate the effect of Salvia officinalis and Cardamom on animal productivity of broiler chickens. This experiment was carried out at the poultry field of the Agricultural Experiment Station, College of Agriculture, Tikrit University. Ninety unsexed, one-day-old Ross 308 broiler chicks were employed. The results showed that the hemoglobin concentration, RBC counts and PCV of S. officinalis boilers and cardamom boilers indicated a significant (P≤0.05) elevated compared to the standard diet boilers. While, WBC count of S. officinalis boilers and cardamom boilers indicated non-significant (P≤0.05) elevated compared to the standard diet boilers. For antioxidant enzymes, the glutathione and catalase levels of S. officinalis boilers and cardamom boilers indicated a significant (P≤0.05) elevated compared to the standard diet boilers. Finally, total protein and albumin activities of S. officinalis boilers and cardamom boilers indicated a significant (P≤0.05) reduced compared to the standard diet boilers. It is concluded from the current study that both S. offcinalis and cardamom showed a stimulating effect on blood parameters and raising protein levels. S. offcinalis and cardamom can be considered an enhancer of the performance of broiler chickens

    IDENTIFICATION AND EXTRACTION PROCESS OF CARDIAC GLYCOSIDES FROM FOXGLOVE PLANTS

    Foxglove (Digitalis spp.) is a plant known for its cardiac glycoside content, which has been used in medicine for centuries. Cardiac glycosides, such as digoxin and digitoxin, are the main components that have a therapeutic effect on the cardiovascular system. This research examines the cardiac glycoside content of various Digitalis species, with a focus on extraction methods, isolation and characteristics of these compounds. Methods used include high-performance liquid chromatography (HPLC), mass spectrometry (MS), and nuclear magnetic resonance (NMR). The analysis results showed that Digitalis purpurea and Digitalis lanata were the main sources of cardiac glycosides. Digoxin and digitoxin were isolated and identified as the main active components. Pharmacological studies show that this compound interacts with the sodium-potassium ATPase pump, which increases cardiac contractility and is used in the treatment of heart failure and arrhythmias. Cardiac glycosides from the foxglove plant play an important role in cardiovascular therapy. Further research is needed to develop more efficient extraction methods and understand the deeper mechanisms of action, as well as potential side effects associated with long-term use

    STUDY AND ANALYSIS OF FETAL HEART RATE MONITORING SYSTEM

    Analysis of FHR variability, together with uterine muscle contractions and fetal movement activity, subject to cardiotocographic monitoring, currently play a fundamental role in fetal assessment. The strength of cardiotocography lies in that normal ranges of the results of the FHR signal analysis almost always (>95%) confirm fetal wellbeing. Unfortunately, questionable or abnormal signal features may indicate both fetal distress and its absence. The classic fetal heart rate variability analysis consists in determining the FHR baseline and then in identifying bradycardia/tachycardia, acceleration/deceleration patterns as well as the type and amplitude of oscillations. A more advanced FHR analysis is aimed at evaluating instantaneous FHR variability, especially with beat-to-beat approach, which is very important in fetal condition assessment. The most common technique of recording the FHR signal is the Doppler ultrasound method, which makes it difficult to correctly estimate the instantaneous heart rate variability because the measurements are averaged. On the other hand, instantaneous heart rate variability plays a crucial role in the diagnosis of fetal condition [1]. The increased interest in monitoring the fetal cardiac bioelectrical activity is due to the fact that the analysis of the fetal electrocardiogram ensures high efficiency and accuracy of the heart rate measurement even on a beat-to-beat level. The first fetal electrocardiogram was recorded as early as in the 1960-ies by means of a special spiral electrode enabling direct signal registration from a fetal head. Due to the way the electrode is placed, its use is limited only to the advanced labour stage [1].

    PSYCHOLOGICAL TREATMENT APPROACHES FOR OBSESSIVE-COMPULSIVE DISORDERS DURING THE COVID-19 PANDEMIC

    This study investigates the psychocorrection of obsessive-compulsive disorders (OCD) exacerbated by the COVID-19 pandemic. The research involved a cohort of 40 patients diagnosed with OCD, whose symptoms intensified during the pandemic. Participants underwent a series of psychocorrective interventions, including cognitive-behavioral therapy (CBT), exposure and response prevention (ERP), and mindfulness-based techniques. The efficacy of these interventions was assessed through pre- and post-treatment evaluations using standardized OCD symptom scales and patient self-reports. Results indicated significant improvements in OCD symptoms and overall mental well-being. Patients reported reduced anxiety and compulsive behaviors, and increased ability to manage obsessive thoughts. The findings suggest that targeted psychocorrective strategies can be effective in alleviating pandemic-induced exacerbations of OCD, highlighting the importance of adapting therapeutic approaches to current global health crise
